Characteristics of the memory loss of a patient with Wernicke-Korsakoff's syndrome without alcoholism.

The memory loss in alcoholic Wernicke-Korsakoff syndrome has been well characterized. However, it has been suggested that some of the neuropsychological defects seen in these patients are due to frontal lobe dysfunction resulting from chronic alcohol abuse. The present report details the nature and extent of the amnesia in a Wernicke-Korsakoff patient who did not have a history of alcoholism. In spite of her normal performance on measures of frontal lobe function, this patient showed many characteristics similar to those seen in alcoholic Wernicke-Korsakoff syndrome including a graded loss of

The spatial learning task of Lhermitte and Signoret: Marker of bilateral hippocampal dysfunction in temporal lobe epilepsy.

OBJECTIVE: Bilateral hippocampal compromise in temporal lobe epilepsy (TLE) substantially increases the risk of significant adverse memory outcomes following unilateral anterior temporal lobectomy and may contraindicate surgery. The Spatial Learning Task (SLT) of Lhermitte and Signoret (1972) is a simple object-location arbitrary associative learning task proposed as a marker of bitemporal dysfunction. Here, we test whether the SLT might serve as a useful marker in TLE by examining, in individuals with TLE and hippocampal sclerosis (HS), whether performance on the SLT depends on the laterality

Divergent effects of win-paired cues on learning from timeout penalties in female and male rats.

In both males and females, linking rewards with salient audiovisual cues in simulated gambling games increases risky choice in humans and rats. However, the prevalence and severity of gambling problems differs in men and women. In previous work, reinforcement learning (RL) models were applied to data from male rats performing the rat gambling task (rGT) to investigate the computational processes promoting risky choice. In the rGT, the optimal strategy is to favor options paired with smaller per-trial gains but shorter and less frequent time-out penalties. Rewards are either delivered with (cue

Developmental Trajectories of Letter and Speech Sound Integration During Reading Acquisition.

Reading acquisition in alphabetic languages starts with learning the associations between speech sounds and letters. This learning process is related to crucial developmental changes of brain regions that serve visual, auditory, multisensory integration, and higher cognitive processes. Here, we studied the development of audiovisual processing and integration of letter-speech sound pairs with an audiovisual target detection functional MRI paradigm. Using a longitudinal approach, we tested children with varying reading outcomes before the start of reading acquisition (T1, 6.5 yo), in first grad

Learning-Dependent and -Independent Enhancement of Mitral/Tufted Cell Glomerular Odor Responses Following Olfactory Fear Conditioning in Awake Mice.

Associative fear learning produces fear toward the conditioned stimulus (CS) and often generalization, the expansion of fear from the CS to similar, unlearned stimuli. However, how fear learning affects early sensory processing of learned and unlearned stimuli in relation to behavioral fear responses to these stimuli remains unclear. We subjected male and female mice expressing the fluorescent calcium indicator GCaMP3 in olfactory bulb mitral and tufted cells to a classical olfactory fear conditioning paradigm. We then used awake, in vivo calcium imaging to quantify learning-induced changes in

Automated Determination of Left Ventricular Function Using Electrocardiogram Data in Patients on Maintenance Hemodialysis.

BACKGROUND AND OBJECTIVES: Left ventricular ejection fraction is disrupted in patients on maintenance hemodialysis and can be estimated using deep learning models on electrocardiograms. Smaller sample sizes within this population may be mitigated using transfer learning. DESIGN, SETTING, PARTICIPANTS, & MEASUREMENTS: We identified patients on hemodialysis with transthoracic echocardiograms within 7 days of electrocardiogram using diagnostic/procedure codes. We developed four models: (1) trained from scratch in patients on hemodialysis, (2) pretrained on a publicly available set of natural ima

The basolateral amygdala complex and perirhinal cortex represent focal and peripheral states of information processing in rats.

We previously identified the roles of the basolateral amygdala complex (BLA) and perirhinal cortex (PRh) in sensory preconditioning in male and female rats (Wong et al., 2025). Here, we used variations of a sensory preconditioning protocol to test a general theory that the BLA and PRh represent focal and peripheral states of attention, respectively. We specifically tested predictions derived from the theory regarding when learning about a stimulus that signals danger will be disrupted by BLA or PRh infusions of the N-methyl-D-aspartate receptor (NMDAR) antagonist, DAP5. Consistent with the the

Multimodal Skin Lesion Diagnosis via Dynamic Consensus and Self-Correcting Representation Learning.

Multimodal skin-lesion classification requires integrating complementary clinical and dermoscopic images while addressing inter-branch disagreement and the semantic gap. We propose MDCL-SCRL, a modality-specific dual-stream framework using paired images and patient metadata. Multimodal Dynamic Consensus Learning (MDCL) derives detached sample-level weights from inter-branch prediction disagreement to modulate the consensus classification loss, whereas Self-Correcting Representation Learning (SCRL) uses concept scores as training-only auxiliary targets. Inference requires only the paired images

Imaging biomarkers of post-COVID dyspnea: insights from machine learning CT patterns and parametric response mapping.

BACKGROUND: Dyspnea is one of the most common symptoms in the post-acute phase of COVID-19 pneumonia. Conventional pulmonary function tests and computed tomography (CT) scores often fail to show correlation with symptom severity, highlighting the need for more sensitive imaging biomarkers. Machine-learning–based quantitative CT analysis and parametric response mapping (PRM) can capture subtle structural and functional abnormalities that may be associated with persistent dyspnea. METHODS: We analyzed inspiratory and paired inspiratory–expiratory CT scans of early (3–6 months) post-COVID-19 pneu

Repeated heroin in rats produces locomotor sensitization and enhances appetitive Pavlovian and instrumental learning involving food reward.

We tested the hypothesis that sensitization to heroin enhances appetitive motivational processes involving food reward. In Experiment 1, sixteen rats were exposed to positive pairings of a light stimulus and food for 4 consecutive daily sessions. Then, the rats received either saline or heroin (2 mg/kg) injections before placement in activity monitors for 9 consecutive daily sessions. Rats were then tested in operant conditioning chambers where one lever produced the light stimulus previously paired with food and another lever produced a tone stimulus not paired with anything. Heroin produced

Computational Modelling Reveals Slower Safety Learning and Threat Extinction are Associated With Higher Anxiety Severity in Remote Fear Conditioning.

Anxiety disorders are chronic, pervasive, and debilitating; characterised by a persistent or exaggerated response to distal or abstract threats. Impaired threat discrimination (distinguishing safe from threatening stimuli) and impaired threat extinction (learning a once threatening stimulus is now safe), are known risk factors in the development and persistence of anxiety disorders. These effects can be experimentally elicited through fear conditioning. First, repeated trials of paired aversive and neutral stimuli are delivered during a fear acquisition phase, followed by repeated trials with

AI-assisted vs. textbook-based vs. blended learning for acute abdomen diagnosis: a retrospective cohort study of emergency interns.

BACKGROUND: Large language models (LLMs) are reshaping medical education. OBJECTIVE: To examine whether learning approach-AI-assisted, textbook-based, or blended-was associated with diagnostic accuracy for acute abdominal conditions among emergency interns. METHODS: We reviewed 720 clinical decisions by 72 emergency interns at a tertiary center over 12 months. Interns were classified into three groups: textbook-based (n = 27), AI-assisted (n = 21), and blended (n = 24). Propensity score matching with pair-stratified GEE addressed selection bias and clustering. RESULTS: Diagnostic accuracy w

Mapping functional traces of opioid memories in the rat brain.

Addiction to psychoactive substances is a maladaptive learned behaviour. Contexts surrounding drug use integrate this aberrant mnemonic process and hold strong relapse-triggering ability. Here, we asked where context and salience might be concurrently represented in the brain during retrieval of drug-context paired associations. For this, we developed a morphine-conditioned place preference protocol that allows contextual stimuli presentation inside a magnetic resonance imaging scanner and investigated differences in activity and connectivity at context recall. We found context-specific respon

Neurocognitive mechanisms of mental imagery-based disgust learning.

Disgust imagery represents a potential pathological mechanism for disgust-related disorders. However, it remains controversial as to whether disgust can be conditioned with disgust-evoking mental imagery serving as the unconditioned stimulus (US). Therefore, we examined this using a conditioned learning paradigm in combination with event-related potential (ERP) analysis in 35 healthy college students. The results indicated that the initial neutral face (conditioned stimulus, CS+) became more disgust-evoking, unpleasant, and arousing after pairing with disgust-evoking imagery (disgust CS+), com

Impaired cognitive plasticity and goal-directed control in adolescent obsessive-compulsive disorder.

BACKGROUND: Youths with obsessive-compulsive disorder (OCD) experience severe distress and impaired functioning at school and at home. Critical cognitive domains for daily functioning and academic success are learning, memory, cognitive flexibility and goal-directed behavioural control. Performance in these important domains among teenagers with OCD was therefore investigated in this study. METHODS: A total of 36 youths with OCD and 36 healthy comparison subjects completed two memory tasks: Pattern Recognition Memory (PRM) and Paired Associates Learning (PAL); as well as the Intra-Extra Dimen

Predictable and precise template-free CRISPR editing of pathogenic variants.

Following Cas9 cleavage, DNA repair without a donor template is generally considered stochastic, heterogeneous and impractical beyond gene disruption. Here, we show that template-free Cas9 editing is predictable and capable of precise repair to a predicted genotype, enabling correction of disease-associated mutations in humans. We constructed a library of 2,000 Cas9 guide RNAs paired with DNA target sites and trained inDelphi, a machine learning model that predicts genotypes and frequencies of 1- to 60-base-pair deletions and 1-base-pair insertions with high accuracy (r = 0.87) in five human a

The impact of mental health symptoms on relational memory performance.

Broad memory challenges arise for people with clinically diagnosed mental health disorders, in part due to changes in hippocampal structure and/or function. However, individuals with mental health symptoms often show memory impairments, even when symptoms do not reach the level of a formal clinical diagnosis. Building on this, the present study examined how mental health symptoms influence performance on two relational memory tasks known to be critically dependent on hippocampal function. Participants from across the adult lifespan completed a transverse patterning task for which they learned
