Automated determination of asulam by enhanced chemiluminescence using luminol/peroxidase system.

A flow injection system with chemiluminescence detection for the determination of asulam, enhancer of the system luminol-H(2)O(2)-horseradish peroxidase, is proposed. The method shows a moderate selectivity against other pesticides usually present in formulations of herbicides and in water. The procedure was applied to the determination of asulam in tap water samples and a recovery study was carried out in order to validate the method. The obtained results show acceptable recovery values (between 88.3 and 93.9%). The detection limit for asulam was 0.12 ng/mL. The precision of the method expres

Expression pattern of cytosolic glutathione peroxidase (cGPx) mRNA during mouse embryogenesis.

The selenoprotein cytosolic glutathione peroxidase (cGPx) is ubiquitously distributed in a variety of organs, and its primary function is to protect oxidative damage. To investigate the spatial and temporal expression pattern of cGPx mRNA in embryogenesis, as this has not been studied before, reverse transcription-polymerase chain reaction (RT-PCR) was carried out in a thermal cycler using mouse-specific cGPx primers, and in situ hybridization was performed in whole embryos or embryonic tissues using digoxigenin-labeled mouse cGPx riboprobes. Expression of cGPx mRNA was detected in all the emb

Glutathione peroxidase activity in blood cells from aspirin-induced asthma patients.

Aspirin and other inhibitors of cyclo-oxygenase precipitate acute bronchospasm in about 10% of adult asthmatics. It has been assumed that cyclo-oxygenase is involved in the pathogenesis of bronchospasm, but the precise mechanism remains poorly understood. The oxygenation products of arachidonic acid include hydroperoxyeicosatetraenoic acid and cyclic endoperoxides. Glutathione peroxidase (GPX) reduces semi-stable hydroperoxides to less reactive alcohols and removes H(2)O(2) involved in inflammation. GPX could, therefore, modulate oxidation of arachidonic acid in the cyclo-oxygenase and lipoxyg

Cytochrome c peroxidase from a methylotrophic yeast: physiological role and isolation.

Mutant strains of the methylotrophic yeast Hansenula polymorpha defective in catalase (cat) and in glucose repression of alcohol oxidase synthesis (gcr1) have been isolated following multiple UV mutagenesis steps. One representative gcr1 cat mutant C-105 grows during batch cultivation in a glucose/methanol medium. However, growth is preceded by a prolonged lag period. C-105 and other gcr1 cat mutants do not grow on methanol medium without an alternative carbon source. A large collection of second-site suppressor catalase-defective (scd) revertants were isolated with restored ability for methyl

Thyroid peroxidase autoantibody fingerprints. II. A longitudinal study in postpartum thyroiditis.

It is not known whether epitopes recognized by autoantibodies in an individual remain constant or change over time, especially during perturbations of the humoral immune response. To address this question, we studied the epitopic profile ("fingerprint") of autoantibodies to thyroid peroxidase (TPO) in the sera of 19 women during the postpartum period. Fingerprints were determined in competition studies using 4 recombinant F(ab). At delivery and at 3 time intervals over the subsequent 9-12 months, the pool of F(ab) inhibited autoantibody binding to TPO by 80-100%, consistent with the definition

Immunophenotyping of peroxidase-negative acute leukaemias using immunoalkaline phosphatase (APAAP) method.

Immunophenotyping of acute leukaemias has become an important diagnostic tool in haematology laboratories as it is now well recognised that the presence of certain surface markers has prognostic significance. In 1988, we experimented with the alkaline phosphatase anti-alkaline phosphatase (APAAP) method for immunophenotyping of leukaemic cells in our laboratory. 48 cases of peroxidase-negative acute leukaemias were studied. Our study showed that 2 peroxidase-negative cases carried myeloid surface markers, 44% were negative for the markers studied and 5% were unclassified due to technical probl

The peroxidase-promoted metabolic activation of acetaminophen produces electronically excited species.

When the analgesic and antipyretic drug acetaminophen is exposed to the horseradish peroxidase/hydrogen peroxide system, light emission is observed. Both the rate of reaction and the emission intensity are increased in the presence of the cationic detergent CTAB. The emission spectrum indicates the presence of at least three emitters. The peak at 445 nm is close to the position of the only band observed in the fluorescence spectrum of the spent reaction mixture and is ascribed to excited N-acetyl-p-benzoquinoneimine. The other two emission bands (505 and 580 nm) must be due to transients. Tota

von Willebrand factor multimer analysis using a sensitive peroxidase staining method.

A peroxidase staining method for von Willebrand factor (vWF) multimer analysis was modified for comparison with an autoradiographic method. This method consists of SDS-agarose electrophoresis followed by blotting to a nitrocellulose membrane and a sensitive peroxidase staining method. The resolution of vWF multimers on the nitrocellulose membrane is comparable to that on the conventional autoradiography. Results can be obtained in 3 days. This nonradioisotopic method will be useful for the determination of the type of von Willebrand's disease in clinical laboratories.

Methyl ethyl ketone peroxide damage to cytochrome P-450 peroxidase activities.

Some new aspects of damage to cytochrome P-450 peroxidase activities by a toxic peroxide were investigated. Methyl ethyl ketone peroxide (MEKP) damage to cytochrome P-450 and inhibition of cytochrome P-450-mediated peroxidase activities were studied. In vivo, hepatic microsomal NADH- and NADPH-peroxidase activities were induced in rats by phenobarbital and 3-methylcholanthrene. Phenobarbital treatment induced more NADH- and NADPH-peroxidase activity than did 3-methylcholanthrene treatment. In vitro, microsomal cytochrome P-450 from rat liver was more sensitive to damage by MEKP than was this p

Peroxidase-catalysed iodotyrosine formation in dispersed cells of mouse extrathyroidal tissues.

A method has been developed for the isolation of cells, high in iodine uptake and peroxidase activity, from the stomach and submaxillary gland of mice. The isolated cells could produce protein-bound monoiodotyrosine, di-iodotyrosine and an unknown iodocompound. The reactions were catalysed by peroxidase and were sensitive to antithyroid drugs and haemoprotein inhibitors but were insensitive to TSH. In-vitro iodination of stomach or submaxillary soluble proteins with the respective peroxidase yielded similar iodocompounds while thyroxine was produced when thyroglobulin was used instead.

Anion binding to resting and half-reduced Pseudomonas cytochrome c peroxidase.

The anion-binding characteristics of resting and half-reduced Pseudomonas cytochrome c peroxidase (ferrocytochrome c-551: hydrogen peroxide oxidoreductase, EC 1.11.1.5) have been examined by EPR and optical spectroscopy with cyanide, azide and fluoride as ligands. The resting enzyme was found to be essentially inaccessible for ligation, which indicates that it has a closed conformation. In contrast, the half-reduced enzyme has a conformation in which the low-potential heme is easily accessible for ligands, a behavior parallel to that towards the substrate hydrogen peroxide (Rönnberg, M., Arais

Development of glutathione peroxidase activity during dietary and genetic copper deficiency.

Copper deficiency was produced in developing rodents to study a possible interaction between copper and the selenoenzyme, glutathione peroxidase (GSH-Px). Dietary copper deficiency was investigated in Sprague-Dawley rats and in three mouse strains (C57BL, C3H/HeJ, C58); genetic copper deficiency was studied in two of the mouse strains, C57BL and C3H/HeJ, using brindled mice. In certain cases it appeared that copper deficiency was associated with depressed liver GSH-Px activity; values from copper-deficient livers were 40-70% of control values. However, the decrease in liver GSH-Px in both rats

Polymorphonuclear leukocyte bactericidal activity and oxidative metabolism during glutathione peroxidase deficiency.

Glutathione peroxidase (GPx) deficiency has been proposed as a cause of some instances of chronic granulomatous disease (CGD). GPx activity varies greatly among species, and specific deficiency of this selenium-dependent enzyme can be produced by dietary selenium deficiency in rats. Bactericidal activity of polymorphonuclear (PMN) leukocytes from normal rats, humans, and guinea pigs (GPx high, intermediate, and nearly absent, respectively), selenium-deficient rats (GPx absent), and a patient with CGD were compared. There was no correlation between natural levels of GPx and bactericidal activit

Influence of the distal his in imparting imidazolate character to the proximal his in heme peroxidase: (1)h NMR spectroscopic study of cyanide-inhibited his42-->ala horseradish peroxidase.

The functional higher oxidation states of heme peroxidases have been proposed to be stabilized by the significant imidazolate character of the proximal His. This is induced by a "push-pull" combination effect produced by the proximal Asp that abstracts ("pulls") the axial His ring N(delta)H, along with the distal protonated His that contributes ("pushes") a strong hydrogen bond to the distal ligand. The molecular and electronic structure of the distal His mutant of cyanide-inhibited horseradish peroxidase, H42A-HRPCN, has been investigated by NMR. This complex is a valid model for the active s

Biomimetic Dichalcogenide-Hybrid Nanozymes With Glutathione Peroxidase-Like Activity for Liver Protection.

Glutathione peroxidase (GPX)-mimetic nanozymes are a promising strategy to restore redox homeostasis, but many existing systems rely on metals and remain limited by cytotoxicity, poor selectivity, and susceptibility to catalytic deactivation. Inspired by the active-site chemistry of native GPX, we designed dichalcogenide-hybrid mesoporous silica nanoparticles (MSNs) as inorganic-organic nanozymes with tunable sulfur and selenium environments. Structure-activity analysis showed that GPX-like performance is governed by chalcogen composition, chalcogen density, and spatial proximity of S/Se withi

Electrochemical Dopamine Biosensor Based on Plant-Derived Peroxidase Immobilized on Titanate Nanowires.

Dysregulation of dopamine (DA) levels is strongly associated with a range of neurodegenerative and psychiatric disorders, including Parkinson's disease. Limitations inherent to conventional analytical techniques have prompted the development of alternative methods, particularly electrochemical biosensors, which provide rapid response and real-time analysis. Among enzyme-based biosensors, peroxidases are notable for their catalytic efficiency and versatility. In this study, gherkin (Cucumis anguria L.) extract was immobilized onto titanate nanowires (TNWs) to fabricate four electrochemical bios

MnTi bimetallic MOF as peroxidase mimic for colorimetric detection of acid phosphatase.

Sensitive and visual detection of acid phosphatase (ACP) is essential for clinical diagnostics. However, the analytical performance of current colorimetric assays is often restricted by the limited catalytic efficiency of conventional single-component nanozymes. To address this issue, we designed a highly active bimetallic metal-organic framework (MnTi-MOF) to serve as a robust peroxidase mimic. By introducing Mn2+ into the Ti-MOF structure, the natural electronegativity difference between the heterometallic nodes generates an internal electric field. This built-in field drives the efficient s

Radical diffusion, not lifetime, determines the range of peroxidase-based proximity labelling.

Proximity labelling offers a powerful strategy for mapping the molecular composition in the vicinity of a protein of interest. Here, we employed APEX2- and HRP-mediated biotinylation in the Drosophila follicular epithelium to analyse the apical, lateral and basal membrane proteomes, using Cadherin99c (Cad99c), Fasciclin 3 (Fas3) and Nidogen (Ndg) as baits. We unexpectedly found that standard peroxidase-based labelling conditions produced a strong basal biotinylation signal, even when the tagged cargo localized apically or laterally. This arises from the long-range diffusion of phenoxy radicals
