Spatial working memory among middle-aged and older patients with schizophrenia and volunteers using fMRI.

OBJECTIVE: Goldman-Rakic and Selemon (Schizophr. Bull. 23 (1997)) hypothesized that many of the symptoms of schizophrenia can be explained by deficits of working memory (WM) that are, in turn, caused by dysfunction of dorsolateral prefrontal cortex (DLPFC). We examined whether older patients with schizophrenia would show an aberrant neural response in the DLPFC or other brain sites when performing a spatial working memory (WM) task adapted from McCarthy et al. (Proc. Natl. Acad. Sci. U. S. A. 91 (1994)). METHOD: Middle-aged and older patients with schizophrenia or schizoaffective disorder and

Effects of vasopressin on histamine H(1) receptor antagonist-induced spatial memory deficits in rats.

The effects of [Arg(8)] vasopressin on histamine H(1) receptor antagonist-induced memory deficits were investigated using the eight-arm radial maze performance test in rats. Pyrilamine and diphenhydramine as well as scopolamine induced memory deficits characterized by increases in the number of total errors, reference memory errors and working memory errors. [Arg(8)] vasopressin improved not only scopolamine--but also pyrilamine--and diphenhydramine-induced memory deficits, although a high dose of [Arg(8)] vasopressin was needed to antagonize pyrilamine-induced memory deficits. The effects of

Dopamine and spatial working memory in rats and monkeys: pharmacological reversal of stress-induced impairment.

The anxiogenic benzodiazepine inverse agonist FG7142 increases dopamine turnover in rodent prefrontal cortex but not in other dopamine terminal field areas. FG7142-induced increases in prefrontal cortical dopamine receptor stimulation impair prefrontal-dependent, but not nonprefrontal-dependent, cognitive tasks in rats and monkeys. The degree of impairment correlates with levels of prefrontal cortical dopamine turnover in rats and can be blocked in rats and monkeys with dopamine receptor antagonists, suggesting that increased dopamine turnover is directly related to the cognitive deficits. The

Chemogenetic activation and inhibition of CaMKⅡα-positive neurons in the dorsal hippocampus modulate spatial working memory and hippocampus-dependent memory in the hemiparkinsonian rat.

Parkinson's disease (PD) is frequently associated with cognitive impairments, especially deficits in working memory and hippocampus-dependent memory. However, the contribution of dorsal hippocampus (dHipp) Ca2+/calmodulin-dependent protein kinase Ⅱ α (CaMKⅡα)-positive neurons to these deficits remains unclear. In this study, we investigated the impacts of chemogenetic modulation of dHipp CaMKⅡα-positive neurons on cognitive function, hippocampal theta rhythm, and monoamine levels in both sham-operated and unilateral 6-hydroxydopamine (6-OHDA)-lesioned rats. 6-OHDA lesions of the medial forebra

Small lesions of the dorsal or ventral hippocampus subregions are associated with distinct impairments in working memory and reference memory retrieval, and combining them attenuates the acquisition rate of spatial reference memory.

The importance of the hippocampus in spatial learning is well established, but the precise relative contributions by the dorsal (septal) and ventral (temporal) subregions remain unresolved. One debate revolves around the extent to which the ventral hippocampus contributes to spatial navigation and learning. Here, separate small subtotal lesions of dorsal hippocampus or ventral hippocampus alone (destroying 18.9 and 28.5% of total hippocampal volume, respectively) spared reference memory acquisition in the water maze. By contrast, combining the two subtotal lesions significantly reduced the rat

Parvalbumin-positive neurons in the medial septum participate in the formation of hippocampal-dependent spatial memory.

GABAergic medial septal neurons play a key role in regulating hippocampus-dependent spatial memory, but the underlying mechanisms are not yet fully understood. In male mice, we establish an object-place recognition impairment model using an acute sleep deprivation protocol. Here we show that parvalbumin-positive neurons in the medial septum regulate object-location memory specificity during encoding by modulating hippocampal place cell population activity. Using in vivo electrophysiology combined with optogenetics, we characterize the role of medial septal parvalbumin-positive neurons in objec

Psilocybin restores spatial memory but not fronto-hippocampal synchrony in a rat model of Alzheimer's disease.

BACKGROUND: Fronto-hippocampal desynchronization is linked to cognitive decline in Alzheimer's-like pathology, but whether psilocybin can restore this dysfunction is unknown. METHODS: Male rats received intracerebroventricular streptozotocin (ICV-STZ) to induce neurotoxicity, followed by a single dose of psilocybin (2.5 mg/kg, the maximum tolerated dose). Spatial memory, recognition memory, spatial learning, anxiety-like behavior, and depressive-like behavior were assessed (Y-maze, novel object recognition, Barnes maze, elevated plus maze, forced swim test), alongside resting-state local fiel

Extended trajectory of spatial memory errors in typical and atypical development: The role of binding and precision.

Spatial reconstruction, a method for evaluating how individuals remember the placement of objects, has traditionally been evaluated through the aggregate estimation of placement errors. However, this approach may obscure the nature of task errors. Specifically, recent data has suggested the importance of examining the precision of responses, as well as absolute performance on item-context bindings. In contrast to traditional analysis approaches based on the distance between the target and the reconstructed item, in this study we further explored three types of errors (swap error, global error,

Different Forms of AMPA Receptor Mediated LTP and Their Correlation to the Spatial Working Memory Formation.

Spatial working memory (SWM) and the classical, tetanus-induced long-term potentiation (LTP) at hippocampal CA3/CA1 synapses are dependent on L-α-amino-3-hydroxy-5-methylisoxazole-4-propionate receptors (AMPARs) containing GluA1 subunits as demonstrated by knockout mice lacking GluA1. In GluA1 knockout mice LTP and SWM deficits could be partially recovered by transgenic re-installation of full-length GluA1 in principle forebrain neurons. Here we partially restored hippocampal LTP in GluA1-deficient mice by forebrain-specific depletion of the GluA2 gene, by the activation of a hypomorphic GluA2

Disruption of component processes of spatial working memory by electroconvulsive shock but not magnetic seizure therapy.

Self-ordered spatial working memory measures provide important information regarding underlying cognitive strategies, such as stereotypy. This strategy is based on repetitive sequential selection of a spatial pattern once a correct sequence has been identified. We previously reported that electroconvulsive shock (ECS) but not magnetic seizure therapy (MST) impaired performance on a spatial working memory task in a preclinical model. Here we tested the hypothesis that ECS disrupted stereotyped patterns in the selection of spatial stimuli. In a within-subject study design, we assessed the effect

Amygdaloid kindling is anxiogenic but fails to alter object recognition or spatial working memory in rats.

Kindling in rats produces enduring behavioral changes that parallel the psychobehavioral disturbances frequently accompanying temporal lobe epilepsy. Some evidence suggests that the site of kindling is an important determinant of the type of behavioral changes observed following kindling, although this variable has not been systematically investigated. In the present experiments, the effects of amygdaloid kindling were assessed on a battery of behavioral tests we used previously to assess the effects of kindling in dorsal hippocampus or perirhinal cortex. Three generalized seizures were kindle

Spatial working memory in Asperger's syndrome and in patients with focal frontal and temporal lobe lesions.

Spatial working memory (SWM) was investigated in 15 patients with Asperger's syndrome (AS) comparing their performance to 18 age- and IQ-matched control subjects. An additional comparison was made with 20 unilateral frontal excision patients [9 right (RFL); 11 left (LFL)] and with 38 unilateral temporal lobectomy patients [18 right (RTL); 18 left (LTL)], the frontal and temporal lobe patients having separate matched control groups. SWM was tested using the Executive Golf Task, a test that also measures spatial strategy formation. The AS group showed a substantial deficit on SWM, but no impairm

Ginsenoside Rg3 modulates spatial memory and fear memory extinction by the HPA axis and BDNF-TrkB pathway in a rat post-traumatic stress disorder.

Post-traumatic stress disorder (PTSD) is a serious mental disorder that can develop after exposure to extreme stress. Korean red ginseng, whose major active component is ginsenoside Rg3 (Rg3), is a widely used traditional antioxidant that has anti-inflammatory, anti-apoptotic and anxiolytics effects. This study investigated whether the administration of Rg3 ameliorated the memory deficit induced by a single prolonged stress (SPS) in rats. Male rats were dosed with Rg3 (25 or 50 mg/kg) once daily for 14 days after exposure to SPS. Rg3 administration improved fear memory and spatial memory might

The effects of insulin-regulated aminopeptidase inhibition with HA08 on recognition memory acquisition and spatial working memory under reversed circadian conditions in male rats.

In 1995, a new M1 aminopeptidase member was discovered in fat and muscle cells, the insulin regulated aminopeptidase (IRAP; oxytocinase; placental leucine aminopeptidase). IRAP can be found in glucose transporter type 4 (GLUT4) vesicles and is believed to regulate cellular glucose uptake by mediating trafficking of GLUT4 to the plasma membrane. It is expressed in various tissues, including areas of the brain associated with cognition, and it cleaves multiple endogenous substrates like oxytocin, vasopressin, somatostatin and cholecystokinin-8. In the beginning of the century, IRAP was identifie

Functional loss of PKMζ in the dorsal hippocampus potentiates the time-dependent increase in false contextual fear memory and impairs spatial recognition memory in mice.

INTRODUCTION: False contextual fear memory has been attributed to a time-dependent loss of precision in contextual memory representations. In this study, we investigated the role of protein kinase M zeta (PKMζ), a key molecule in the maintenance of hippocampus-dependent long-term memory, in false contextual fear memory within the dorsal (dHPC) and ventral hippocampus (vHPC). METHODS: Two weeks prior to behavioral testing, male C57BL/6J mice (7-10 weeks old) received bilateral injections of adeno-associated virus (AAV PHP.eB) into the dHPC or vHPC to induce PKMζ knockdown (PKMζ KD), overexpres

Effects of transcranial magnetic stimulation on spatial memory: a systematic review across animal and human studies.

INTRODUCTION: Spatial memory relies on distributed hippocampal-cortical networks that are highly sensitive to modulation of excitability, connectivity, and synaptic plasticity. Transcranial magnetic stimulation (TMS) has emerged as a promising tool to experimentally probe and therapeutically modulate these networks, although its effects on spatial memory remain heterogeneous. METHODS: We conducted a PRISMA-guided systematic review synthesizing evidence from rodent and human studies examining the effects of TMS on spatial memory. A total of 35 studies (23 animal, 12 human) identified through s

Visual-spatial working memory in ADHD: new evidence for a storage rather than a processing deficit.

The ability to voluntarily control eye movements is part of the "executive function" domain, which includes working memory and is impaired in neuro-developmental disorders such as ADHD. The present study aimed to investigate two types of "executive" saccades - anti-saccades (AS) and memory-guided saccades (MGS) - as well as their relationships in samples of children and adolescents with ADHD and neurotypical controls. To that end, anti-saccades and memory-guided saccades were recorded in n = 65 participants with ADHD (age 9.2-17.1 years) and n = 74 neurotypical controls (aged 9.1-17.3 years) a

Hippocampal astrocyte St6galnac5 silencing improves spatial memory and preserves synaptic integrity in an AD mouse model.

Cognitive resilience in Alzheimer's disease (AD) requires the maintenance of synaptic integrity despite progressive pathological insults. Reactive astrocytes can switch between neuroprotective and neurotoxic states, and their maladaptive transition significantly accelerates neurodegeneration, yet the molecular drivers of this shift remain elusive. Here, using published single-nucleus transcriptomic data, we identified the sialyltransferase St6galnac5 as a candidate regulator associated with reactive, pro-inflammatory astrocyte states. We further show that astrocyte-specific, AAV-mediated knock

Effects of reward magnitude on rats' long-term spatial memory retention in the radial-arm maze.

Our study examined whether reward magnitude affects long-term spatial memory retention in rats performing a radial-arm maze task. Previous studies, using a forced-choice learning phase followed by a free-choice test, have reported retention of up to 72 hours, but they typically used a large reward (10 pellets per arm; 45 mg each) rather than the standard reward (one pellet per arm), leaving open a reward-based explanation. In Experiment 1, we conducted a between-subjects comparison of two reward conditions: a standard-reward group (one pellet per arm) and a large-reward group (10 pellets per a

Selective Impairment of Spatial Recognition Memory and Reduced Frontal Corticothalamic Spine Density Following Adolescent Alcohol Consumption.

BACKGROUND: Heavy alcohol use is common during adolescence and is associated with an increased risk of alcohol use disorder and prevalence of residual cognitive deficits, especially in behaviors associated with the latently developing prefrontal cortex (PFC). A major need for advancing our understanding of this relationship is accessible preclinical behavioral batteries that can be used to disassociate the effects of adolescent alcohol on select PFC circuits. METHODS: The present study uses a retrograde viral approach to identify PFC projection neurons to the mediodorsal thalamus (MdT), follo
