Cell-Free Supernatant of Porphyromonas gingivalis Promotes the Malignancy of Oral Squamous Cell Carcinoma by Suppressing Ferroptosis.

BACKGROUND: Oral squamous cell carcinoma (OSCC) is a series global health concern. Porphyromonas gingivalis (P.g), a major pathogenic bacterium, promotes tumorigenesis and progression. Ferroptosis is also closely related to the progression of tumors. Therefore, this study aimed to investigate whether P.g cell-free supernatant (Pg-SN) alters ferroptosis and malignant phenotypes in OSCC cells. METHODS: CCK-8 assays were used to compare the effects of cell-free Pg-SN and Streptococcus salivarius cell-free supernatant (Ss-SN) on OSCC cell viability. Wound healing and Transwell invasion assays wer

Multiple alcohol dehydrogenases but no functional acetaldehyde dehydrogenase causing excessive acetaldehyde production from ethanol by oral streptococci.

Ethanol consumption and poor oral hygiene are risk factors for oral and oesophageal cancers. Although oral streptococci have been found to produce excessive acetaldehyde from ethanol, little is known about the mechanism by which this carcinogen is produced. By screening 52 strains of diverse oral streptococcal species, we identified Streptococcus gordonii V2016 that produced the most acetaldehyde from ethanol. We then constructed gene deletion mutants in this strain and analysed them for alcohol and acetaldehyde dehydrogenases by zymograms. The results showed that S. gordonii V2016 expressed t

Prevalent bacterial species and novel phylotypes in advanced noma lesions.

The purpose of this study was to determine the bacterial diversity in advanced noma lesions using culture-independent molecular methods. 16S ribosomal DNA bacterial genes from DNA isolated from advanced noma lesions of four Nigerian children were PCR amplified with universally conserved primers and spirochetal selective primers and cloned into Escherichia coli. Partial 16S rRNA sequences of approximately 500 bases from 212 cloned inserts were used initially to determine species identity or closest relatives by comparison with sequences of known species or phylotypes. Nearly complete sequences

On salivary film formation and bacterial retention to solids. A methodological and experimental in vitro study.

A parallel-plate flow cell system was developed to assess individual and combined parameters relevant to oral microbial retention. In this system, internal reflection germanium prisms represented oral solid surfaces including tooth surfaces. The system used, allowed adsorbed protein films to be analyzed in situ by multiple non-destructive surface analytical techniques, without provoking surface associated biofilms. Using a range of biochemical and biophysical techniques, germanium prisms of medium critical surface tension were noted to accurately model film formation at tooth-saliva interfaces

Oligosaccharides with different glycosidic bonds regulate gut microbiota: differential modulation of bacterial and fungal communities and their metabolism in vitro.

Oligosaccharides significantly impact host health by modulating gut microbial composition and metabolic activities. However, the influence of oligosaccharides with different glycosidic bonds on intestinal microecology remains unclear, hindering their functional food applicability. This study examined the impact of six disaccharides, including trehalose (α-1,1), maltose (α-1,4), isomaltose (α-1,6), cellobiose (β-1,4), β-lactose (β-1,4), and gentiobiose (β-1,6), at concentrations of 10 mM. The results showed that α-disaccharides (maltose and isomaltose) were metabolized faster than β-disaccharid

Trichuris muris egg-hatching assay for anthelminthic drug discovery and characterization.

Trichuriasis is a neglected tropical disease widely distributed among tropical and sub-tropical areas and associated with poverty and lack of access to safe drinking water, sanitation and hygiene. Existing drugs have limited efficacy and face a constant risk of developing resistance, necessitating the search for alternative treatments. However, drug discovery efforts are sparse and little research has been performed on anthelminthic effects on embryonated eggs, the infectious life stage of Trichuris spp. We examined bacterial species dependent egg hatching of the murine model parasite Trichuri
