BcSTP6, a sugar transporter of STP family, modulates flowering time and plant growth by regulating sugar signaling in flowering Chinese cabbage.

Flowering Chinese cabbage (Brassica campestris L. ssp. chinensis var. utilis Tsen et Lee.), an cultivated variant of Brassica rapa var. chinensis in the Brassicaceae family, is distinguished from Chinese cabbage (Brassica rapa var. chinensis) by its basal leaves, which have non-stippled petioles and slender, grooved leaf stalks. Sugar transport proteins (STPs), a key subfamily within the plant monosaccharide transporter (MST) superfamily, serves as primary hexose transporters that facilitate the transmembrane movement of glucose, fructose, and other hexoses. These proteins play a crucial role

Expression of genes coding for sugar transporters and sugar metabolizing enzymes during dynamic fructan metabolism in chicory (Cichorium intybus L.).

Carbohydrate partitioning and intracellular sugar transport are central to plant energy management, stress adaptation, and storage metabolism. In fructan-accumulating species such as Cichorium intybus, however, the underlying transport processes remain poorly understood. Because fructan metabolism occurs in the vacuole, coordinated regulation of tonoplast transporters and fructan-metabolizing enzymes is required to control sugar fluxes across the vacuolar membrane. Here, we analyzed the expression of genes encoding sugar transporters, including TSTs, ERDL6, SUC4, SWEETs, and SUT1, during veget

"Pour some sugar on me"-Environmental Candida albicans isolates and the evolution of increased pathogenicity and antifungal resistance through sugar adaptation.

Candida albicans is an opportunistic fungal pathogen that colonizes mucosal surfaces of most humans. Only in rare cases, C. albicans isolates are found in the environment. This study investigated whether environmental isolates differ in their virulence potential from clinical strains and how adaptation to a human diet influences key virulence attributes. We examined three C. albicans isolates from oak trees in the United Kingdom, and observed that one exhibited high host cell damage, increased hypha formation, invasion capacity, and candidalysin production, along with an intrinsic resistance t

Process-integrated engineered resting cells for biocatalytic production of rare natural sugars from sole methanol molecules.

The biocatalytic conversion of green methanol, a promising feedstock, into high value-added products is seen as an attractive way to establish sustainable biomanufacturing. Although natural and engineered microorganisms can utilize methanol, efficiently converting C1 carbon into valuable chemicals in growing cells remains challenging because non-native pathways often suffer from low carbon utilization and limited catalytic stability. Here, we report the integration of an engineered enzyme cascade into resting cells and its coupling with a screened alcohol oxidase enzyme to develop an efficient

Non-Equilibrium Growth Processes to Glyceraldehyde and Glycerol as Building Blocks of Interstellar Sugars and Phospholipids.

Complex organic molecules (COMs) such as aldoses and polyols are essential prebiotic compounds and serve as fundamental molecular building blocks for key biomolecules, including sugars and lipids. Despite their detection in star-forming regions and carbonaceous meteorites, the mechanisms of formation of these COMs in extraterrestrial environments remain largely controversial. Here, we demonstrate the bottom-up synthesis of a diverse set of COMs in the formaldehyde-water (H2CO-H2O) ice analogs subjected to simulated galactic cosmic rays (GCRs), including aldoses and polyols (glycolaldehyde, eth

Room temperature synthesis of 3D-nanocrystalline graphitic carbon from biomass-derived sugars, alcohols, and polyphenolic compounds.

Nanocrystalline carbon materials exhibit promising potential for sustainable and high-performance applications in electronics, energy storage, and environmental technologies. While sugars are abundant and renewable, converting them to graphitic carbon usually requires high temperature treatment. Here, we present a groundbreaking approach for synthesizing nanocrystalline carbon from readily available sugars such as glucose, fructose, and sucrose at ambient pressure and temperature. This novel method involves electrochemical reduction on a negatively charged Ag surface coupled with intermolecula

Co-substrate model development and validation on pure sugars and corncob hemicellulosic hydrolysate for xylitol production.

A co-substrate model of Candida tropicalis TISTR 5306 cultivated in 10 - 100 g/L xylose and 1 - 10 g/L glucose at the ratio of 10:1 was developed based in part on modified Monod equation. The kinetic parameters include substrate limitation as well as substrate and product inhibitions with inclusion of threshold values. A general good fitting with average RSStotal, R2, and MStotal values of 162, 0.979, and 10.8, respectively, was achieved between ten simulated profiles and experimental kinetics data. The implementation of developed model on xylitol production from non-detoxified corncob hemicel

Amino sugars influence Aspergillus fumigatus cell wall polysaccharide biosynthesis, and biofilm formation through interfering galactosaminogalactan deacetylation.

Aspergillus fumigatus is a ubiquitous fungal pathogen responsible for a significant number of deaths annually due to invasive aspergillosis infection. While the utilization of diverse carbon sources, including amino sugars, has been explored in other fungi, its impact on A. fumigatus remains uncharted territory. In this study, we investigated A. fumigatus responses to glucose (Glc), glucosamine (GlcN) and N-acetylglucosamine (GlcNAc) as carbon sources. GlcN inhibited growth, reduced sporulation and delayed germination, while GlcNAc had no such effects. Both amino sugars induced alterations in

Unifying Scheme for the Biosynthesis of Acyl-Branched Sugars: Extended Substrate Scope of Thiamine-Dependent Enzymes.

Thiamine diphosphate (ThDP) dependent enzymes are useful catalysts for asymmetric C-C bond formation through benzoin-type condensation reactions that result in α-hydroxy ketones. A wide range of aldehydes and ketones can be used as acceptor substrates; however, the donor substrate range is mostly limited to achiral α-keto acids and simple aldehydes. By using a unifying retro-biosynthetic approach towards acyl-branched sugars, we identified a subclass of (myco)bacterial ThDP-dependent enzymes with a greatly extended donor substrate range, namely functionalized chiral α-keto acids with a chain l

Stool Reducing Sugars and Stool pH Are Poor Screening Tests for Intestinal Disaccharidase Deficiencies in Children.

BACKGROUND: Tests for stool reducing sugars and stool pH are ordered for children with osmotic diarrhea to screen for carbohydrate malabsorption. METHODS: We compared the results of the two screening tests, stool reducing sugars and stool pH, with a more definitive result from an intestinal tissue disaccharidase activity assay ordered for pediatric patients (<18 years old). Overall, 159 patients had results for tissue disaccharidase and stool reducing sugars, but only 115 had additional results of stool pH. Forty-six of the 159 patients had mild, moderate, or severe disaccharidase deficiencie

Comparison of Anti-Obesity Effect between Two Types of Syrup Containing Rare Sugars in Wistar Rats.

D-Allulose-containing rare sugar sweeteners have been categorized into two types, rare sugar syrup (RSS), consisting of 4 rare monosaccharides, and modified glucose syrup (MGS), rich in D-allulose, which was previously referred to D-psicose. The anti-obesity effect of RSS and D-allulose has been already clarified, but that of rare monosaccharides other than D-allulose in RSS has not yet been well understood. Here, we investigated and compared the anti-obesity effect of RSS and MGS in rats. Male Wistar rats were divided into 4 dietary groups: a high-sucrose control diet group (S), a high-fructo

Quantitative HPLC-MS analysis of nucleotide sugars in plant cells following off-line SPE sample preparation.

An analytical workflow was developed for the absolute quantification of uridine diphosphate (UDP)-sugars in plant material in order to compare their metabolism both in wild-type Arabidopsis thaliana and mutated plants (ugd2,3) possessing genetic alterations within the UDP-glucose dehydrogenase genes involved in UDP-sugar metabolism. UDP-sugars were extracted from fresh plant material by chloroform-methanol-water extraction and further purified by solid-phase extraction with a porous graphitic carbon adsorbent with extraction efficiencies between 80 ± 5 % and 90 ± 5 %. Mixed antimony(V) complexes with different sugars to modulate the oral bioavailability of pentavalent antimonial drugs.

Previous studies have shown that the association of the drug meglumine antimoniate (MA) with β-cyclodextrin can improve its bioavailability by the oral route. In this work, ribose and maltose were investigated for their ability to form mixed or association complexes with MA, release MA and modulate the serum levels of Sb after oral administration in mice. Analysis of the MA/ribose composition by high performance liquid chromatography coupled to mass spectrometry (LCMS-IT-TOF) revealed the presence of mixed meglumine-Sb-ribose and Sb-ribose complexes. Analysis of the MA/maltose composition sugg

Evaluation of thermodynamic parameters of some amphiphilic drugs in presence of sugars at the cloud point.

In the present investigation, we report the thermodynamics of clouding in four amphiphilic drugs viz., two tricyclic antidepressants: amitriptyline hydrochloride (AMT) and imipramine hydrochloride (IMP) and two phenothiazines: chlorpromazine hydrochloride (CPZ) and promethazine hydrochloride (PMT) in the absence and presence of sugars. For an amphiphile, cloud point (CP) can be considered as the limit of its solubility as the phase separates at temperatures above the CP. The clouding components release their solvated water and separate out from the solution. For all the cases, the standard Gib

Separation of furans and carboxylic acids from sugars in dilute acid rice straw hydrolyzates by nanofiltration.

This work studied the concentration of hydrolyzates obtained from dilute acid hydrolysis of rice straw using nanofiltration (NF). In order to minimize the Donnan exclusion effect of the membrane, the hydrolyzate solution was controlled at low pH value. Negative retentions of both furans and carboxylic acids were observed. The maximum separation factor of acetic acid over xylose was 49, while the maximum separation factor of acetic acid over arabinose was 52, when the system was operated at pH 2.9 and an applied pressure of 24.5-34.3 bar. The separation factors of inhibitors over glucose became

Sugars containing alpha,beta-unsaturated carbonyl systems: synthesis and their usefulness as scaffolds in carbohydrate chemistry.

The alpha,beta-unsaturated carbonyl function occurs in a wide variety of bioactive natural products. It is usually associated with the bioactivities of these compounds and acts as Michael acceptors for the addition of protein nucleophilic groups. The design and synthesis of sugars containing this functionality has provided a wide range of compounds, which can serve as building blocks of high synthetic versatility. This review deals with the chemistry of sugar-based molecules bearing singly linked or fused unsaturated lactones and ketones along with that of pyranoid enones and enonolactones. Ex

High-throughput HPAEC-PAD for the analysis of neutral sugars and uronic acids in biomass hydrolysates.

Lignocellulosic and marine biomasses are valuable renewable sources of cellulose and alginate polysaccharides. Accurate quantification of the neutral sugars and uronic acids content in complex biomass hydrolysates is essential for effective characterization and use. High-performance anion-exchange chromatography with pulsed amperometric detection (HPAEC-PAD) provides sensitive, derivatization-free analysis with good isomer separation. However, traditional methods often involve lengthy gradients and demanding column maintenance, which limit throughput. In this study, we introduce and validate a
