Rapid and precise diagnosis of T. marneffei pulmonary infection in a HIV-negative patient with autosomal-dominant STAT3 mutation: a case report.

BACKGROUND: Talaromyces marneffei, also named Penicillium marneffei, is an opportunistic pathogen that can cause systemic or limited infection in human beings. This infection is especially common in human immunodeficiency virus (HIV)-infected hosts; however, it has also been recently reported in HIV-negative hosts. Here, we report a very rarely seen case of T. marneffei pulmonary infection in a non-HIV-infected patient with signal transducer and activator of transcription 3 (STAT3) mutation. CASE PRESENTATION: A 34-year-old woman was admitted to our hospital for uncontrollable nonproductive c

Synthetic Lethality of Fungal Catabolite Repressors Reveals Essential Coupling of Carbon Repression With Mitochondrial and Ribosomal Gene Expression.

Carbon catabolite repression (CCR) enables fungi to preferentially utilize favorable carbon sources through global transcriptional control. In filamentous fungi, CCR is classically attributed to the C2H2 zinc-finger repressor Mig1/CreA; however, residual repression following Mig1 disruption suggests additional regulatory layers in Talaromyces pinophilus (previously called Penicillium funiculosum NCIM1228). Here, we identify Mig3, a previously uncharacterized, Eurotiales-specific C2H2 transcription factor that functions redundantly with Mig1 in CCR. Mig3 contains conserved N-terminal zinc-finge

Clinically Important Emerging and Uncommon Fungal Pathogens in Asia: Epidemiology, Antifungal Resistance and Challenges in Diagnosis and Management.

Fungal infections in Asia are influenced by warm, humid climates, high population density and expanding healthcare systems, which together facilitate the emergence, transmission and detection of both common and rare pathogens. While Candida and Aspergillus species remain the leading causes of invasive fungal disease, increasing attention has focused on uncommon yeasts and rare moulds, many of which show intrinsic or acquired antifungal resistance. Advances in molecular diagnostics and susceptibility testing have improved identification of emerging pathogens, including uncommon Candida species

Recurrent Disseminated Talaromycosis Mimicking Liver Disease in a STAT3-Mutated HIES Patient: A Case Report.

BACKGROUND: Talaromycosis is increasingly recognized in immunocompromised individuals beyond those with HIV, including patients with primary immunodeficiencies such as Hyper-IgE syndrome (HIES). However, diagnosing disseminated infection remains challenging due to nonspecific clinical manifestations and limitations of conventional diagnostic methods. CASE PRESENTATION: We report a rare case of recurrent disseminated Talaromyces marneffei (T. marneffei) infection in a 25-year-old male with STAT3-mutated HIES. Initially presenting with abnormal liver function tests, the patient had a history of

Wild-type MIC distributions and epidemiological cut-off values for ibrexafungerp, manogepix, and rezafungin against WHO fungal priority pathogens.

OBJECTIVE: This study gathered public data regarding the in vitro activity of three novel antifungal agents, ibrexafungerp, manogepix, and rezafungin, against the fungi listed in the WHO fungal priority pathogens list. METHODS: Epidemiological cut-off values (ECVs/ECOFFs) were established for species with sufficient data (≥100 MIC values) and when distributions met further essential criteria. For pathogens in which data did not fulfil one or more essential criteria, tentative ECVs (T-ECVs/T-ECOFFs) were proposed. The values were determined by a comprehensive analysis that included visual insp

Regulatory functions of AcuK and AcuM transcription factors in fungal metabolic adaptation, stress response, and virulence.

Fungal species thrive in diverse ecological niches and must dynamically adjust their metabolism, growth, and development in response to environmental fluctuations. Transcriptional regulation plays a pivotal role in these adaptive processes, with Zn2Cys6 transcription factors constituting the largest family of fungal-specific regulators that orchestrate metabolism, development, and pathogenicity. Among these, AcuK and AcuM represent a distinct subfamily with diverse key regulatory functions. In this review, we provide a comprehensive analysis of AcuK and AcuM functions across various fungal spe

Gut microbiota and mycobiota change with feeding duration in mice on a high-fat and high-fructose diet.

BACKGROUND: Metabolic dysfunction-associated fatty liver disease (MAFLD), formerly known as nonalcoholic fatty liver disease (NAFLD), is becoming the most common chronic liver disease. The gut microbiome is regarded to play a crucial role in MAFLD, but the specific changes of gut microbiome, especially fungi, in different stages of MAFLD are not well understood. This study aimed to observe the longitudinal changes of colon bacteria and fungi of mice at different feeding duration of a high-fat and high-fructose diet (HFHFD), and explore the association between the changes and the progression of

The indigenous microbial diversity involved in the spontaneous fermentation of red dragon fruit (Hylocereus polyrhizus) identified by means of molecular tools.

Red dragon fruit (RDF) is well-known for its high nutritional content, especially the red pigment betacyanins that possess high antioxidant activity. Natural fermentation is an ancient yet outstanding technique that relies on the autochthonous microbiota from fruits and vegetables surfaces to preserve and improve the nutritional values and quality of the food product. The present study was to evaluate and identify the indigenous microbial community (bacteria and fungi) that are involved in the natural fermentation of RDF. Results revealed a total of twenty bacterial pure cultures and nine fung

Microbial degradation as a powerful weapon in the removal of sulfonylurea herbicides.

Sulfonylurea herbicides have been widely used worldwide and play a significant role in modern agricultural production. However, these herbicides have adverse biological effects that can damage the ecosystems and harm human health. As such, rapid and effective techniques that remove sulfonylurea residues from the environment are urgently required. Attempts have been made to remove sulfonylurea residues from environment using various techniques such as incineration, adsorption, photolysis, ozonation, and microbial degradation. Among them, biodegradation is regarded as a practical and environment

Central Nervous System Infection with Other Endemic Mycoses: Rare Manifestation of Blastomycosis, Paracoccidioidomycosis, Talaromycosis, and Sporotrichosis.

The central nervous system (CNS) is not a major organ involved with infections caused by the endemic mycoses, with the possible exception of meningitis caused by Coccidioides species. When CNS infection does occur, the manifestations vary among the different endemic mycoses; mass-like lesions or diffuse meningeal involvement can occur, and isolated chronic meningitis, as well as widely disseminated acute infection that includes the CNS, are described. This review includes CNS infection caused by Blastomyces dermatitidis, Paracoccidioides brasiliensis, Talaromyces marneffei, and the Sporothrix

Identification of Paecilomyces variotii in clinical samples and settings.

Paecilomyces variotii is a commonly occurring species in air and food, but it is also associated with many types of human infections and is among the emerging causative agents of opportunistic mycoses in immunocompromised hosts. Paecilomyces can cause hyalohyphomycosis, and two species, Paecilomyces lilacinus and P. variotii, are the most frequently encountered organisms. In the present study, a set of 34 clinical isolates morphologically identified as P. variotii or P. lilacinus were formally identified by sequencing intergenic transcribed spacer regions 1 and 2 (including 5.8S rDNA) and a pa

Effects of different intercropping patterns on tea rhizosphere environment and leaf quality.

Intercropping can improve tea soils and leaf quality, but partner-specific responses remain unclear. We compared four systems-Paris polyphylla-tea (CT), leguminous green manure-tea (LF), Pleurotus ostreatus-tea (CJ), and Polygonatum kingianum-tea (HJ)-with matched monoculture controls. Tea and soil traits were integrated with high-throughput sequencing, LEfSe, PICRUSt2 functional prediction, and correlation analyses. LF significantly increased soil available and total phosphorus, enriched the potentially beneficial fungi Mortierella and Trichoderma in the tea rhizosphere, and enhanced the pred
