Dietary oxidized frying oil activates hepatic stellate cells and accelerates the severity of carbon tetrachloride- and thioacetamide-induced liver fibrosis in mice.

Deep-frying is a common cooking practice worldwide, and after repeated heating's, the oil undergoes various chemical reactions, including hydrolysis, polymerization, lipid oxidation, and the Maillard reaction. Studies have pointed out that oxidized dietary frying oil may cause teratogenesis in mice and increase cancer and cardiovascular risks. The liver is the main organ involved in dietary nutrient catabolism, detoxification, bile production, and lipid metabolism. Nevertheless, the effects of oxidized frying oil exposure on the activation of hepatic stellate cells (HSCs) and liver fibrosis ar

Simultaneous intervention against oxidative stress and inflammation by targeting Nrf2/ARE and NLRP3 inflammasome pathway mitigates thioacetamide-induced liver fibrosis in rat.

Liver fibrosis is a typical pathological state/stage involved in most chronic liver diseases and its persistence results in cirrhosis. Inflammasomes are cytoplasmic sensors that induce inflammation in response to stress. Glibenclamide (GLB) is an USFDA-approved drug for type 2 diabetes and is reported to possess anti-inflammatory activity by inhibiting inflammatory cytokines. Dimethyl fumarate (DMF) is an USFDA-approved drug for multiple sclerosis and has been reported to activate the Nrf2/ARE pathway to maintain the cellular antioxidant balance. A total of 36 rats were randomized into six gro

Saxagliptin Attenuates Thioacetamide-Induced Liver Fibrosis in Rats in Association with Changes in SIRT1/Nrf2/HO-1 Signaling, Oxidative Stress, and Apoptosis.

PURPOSE: Currently, no universally effective treatment exists to stop or reverse the development of liver fibrosis. Saxagliptin (SAX), a dipeptidyl peptidase-4 (DPP-4) inhibitor, has demonstrated extra-glycemic benefits, including anti-inflammatory and anti-apoptotic properties, but its role in hepatic fibrogenesis has not been characterized. This study examined the protective potential and underlying mechanisms of saxagliptin against thioacetamide (TAA)-induced hepatic fibrogenesis in rats. METHODS: Twenty-four male Wistar rats were randomly allocated to four groups: control, TAA-induced fib

The Effects of Hepatogomax Enteral Formula on Systemic Inflammation, Caecum Short-Chain Fatty Acid Levels, and Liver Histopathology in Thioacetamide-Induced Rats.

Liver damage characterized by fibrosis and necrosis can worsen the condition of liver disease. Liver disease is associated with impaired immune response and may affect short-chain fatty acid (SCFA) gut metabolites. Hepatogomax enteral formula was developed, which contains brain-chain amino acids (BCAAs) and middle-chain triglycerides (MCTs), which could repair liver tissue damage, improve the inflammatory status, and modulate SCFA in liver damage. The study aimed to determine the effect of hepatogomax on liver tissue repair, inflammation (TNF-α and IL-6), and SCFA levels in thioacetamide (TAA)

Effect of Empagliflozin on Thioacetamide-Induced Liver Injury in Rats: Role of AMPK/SIRT-1/HIF-1α Pathway in Halting Liver Fibrosis.

Hepatic fibrosis causes severe morbidity and death. No viable treatment can repair fibrosis and protect the liver until now. We intended to discover the empagliflozin's (EMPA) hepatoprotective efficacy in thioacetamide (TAA)-induced hepatotoxicity by targeting AMPK/SIRT-1 activity and reducing HIF-1α. Rats were treated orally with EMPA (3 or 6 mg/kg) with TAA (100 mg/kg, IP) thrice weekly for 6 weeks. EMPA in both doses retracted the serum GGT, ALT, AST, ammonia, triglycerides, total cholesterol, and increased serum albumin. At the same time, EMPA (3 or 6 mg/kg) replenished the hepatic content

Altered renal disposition of adefovir in metabolic dysfunction-associated steatohepatitis using the fast-food diet and thioacetamide-induced model to extrapolate renal secretion.

Metabolic dysfunction-associated fatty liver disease and metabolic dysfunction-associated steatohepatitis (MASH) are the most prevalent liver diseases globally. As MASH progresses, changes in expression of xenobiotic transporters significantly alter drug disposition and subsequent increased risk of toxicity. Patients with MASH and co-occurring chronic hepatitis B virus are becoming increasingly prevalent, presenting a unique challenge in clinical practice. Adefovir (ADEF) is an antiviral drug used to treat hepatitis B virus infection and is a substrate of organic anion transporters (OATs). Usi
