INTRODUCTION: Many patients acutely self-poisoned with organophosphorus insecticides have co-ingested ethanol. Currently, profenofos 50% emulsifiable concentrate (EC50) is commonly ingested for self-harm in Sri Lanka. Clinical experience suggests that ethanol co-ingestion makes management more difficult. Therefore, we aimed to determine the relationships between plasma ethanol concentration, plasma profenofos concentration and its toxicokinetics, and clinical outcome in acute profenofos self-poisoning.
METHODS: Demographic and clinical data, including a history of ethanol ingestion and blood

Sinoporphyrin sodium (DVDMS) is a novel hematoporphyrin-like photosensitizer developed for photodynamic therapy (PDT), an effective therapeutic modality for tumor treatment; however, the safety of photosensitizer-based PDT is always of great concern. The purpose of the current study was to investigate the potential repeated-dose toxicity and describe the toxicokinetic process of DVDMS-based PDT in Beagle dogs. The dogs were randomly allocated to six groups, and then were administrated a DVDMS preparation intravenously at dose levels of 0, 1, 3, 9, 1 and 9 mg per kg body weight, respectively; t

BACKGROUND: Methylmercury (MeHg), known for well over a century as a neurotoxin in adults, has more recently been studied for potential detrimental effects during early brain development. While several studies have estimated mercury exposure, they usually rely on either a single biomarker or questionnaire data, each of which has limitations. The goal of this paper was to develop a toxicokinetic model that incorporates both biomarker and questionnaire data to estimate the cumulative exposure to MeHg through seafood consumption using data collected from the Childhood Autism Risks from Genetics a

The management of patients suspected, but not confirmed, as being poisoned is challenging. The Royal College of Emergency Medicine and National Poisons Information Service have produced this guidance to provide a generalised clinical approach to any poisoned patient in the emergency department. This guideline provides a clinical approach to support the initial assessment of a patient, identification of potential toxic agents and emergency management. A toxidromic approach is used, with emphasis on consideration of the toxicokinetics of potential poisons and how the patient's clinical condition

Tetrodotoxin (TTX) is one of the most potent naturally occurring neurotoxins and continues to attract considerable scientific, clinical, forensic, and regulatory attention. Historically associated with pufferfish poisoning in East and Southeast Asia, TTX has recently emerged as a global food safety concern due to its detection in an increasing number of marine and terrestrial organisms and its expanding geographical distribution, including European coastal waters. Despite decades of research, important questions regarding its biosynthesis, ecological origin, toxicokinetics, and environmental d

Background/Objectives: Nutritional intake plays an important role in modulating lead absorption and toxicity. In addition to micronutrient status, emerging evidence suggests that body fat distribution may influence heavy metal toxicokinetics, yet this aspect remains poorly explored in occupational settings. This study aimed to investigate the associations of dietary intake of zinc, calcium, vitamin D, and protein, as well as anthropometric indicators, with blood lead levels (BLLs) among lead-exposed male workers. Methods: A cross-sectional study was conducted involving 144 male workers from fi

Opiates produce analgesia via G-protein signaling, and adverse effects, such as respiratory depression and decreased bowel motility, by β-arrestin pathway. Oliceridine, a G protein-biased MOR agonist, only presents modest safety advantages as compared to other opiates in clinical trials, possibly due to its limited bias. Our previous study shown that LPM3480392, a full MOR biased agonist, is selective for the Gi pathway over the β-arrestin-2. In the present article, we evaluated the subacute toxicity of LPM3480392 in rats. The rats were administered with control article or LPM3480392 0.6, 1.2

Thioacetamide (TA) undergoes saturation toxicokinetics in ad libitum (AL) fed rats. Diet restriction (DR) protects rats from lethal dose of TA despite increased bioactivation-mediated liver injury via CYP2E1 induction. While a low dose (50 mg TA/kg) produces 6-fold higher initial injury, a 12-fold higher dose produces delayed and mere 2.5-fold higher injury. The primary objective was to determine if this less-than-expected increase in injury is due to saturation toxicokinetics. Rats on AL and DR for 21 days received either 50 or 600 mg TA/kg i.p. T(1/2) and AUCs for TA and TA-S-oxide were cons

Authors present a simple small bowel surgical wash-out technique that can be used in the treatment of severe chloroquine poisoning. This toxicokinetic treatment can improve the prognosis of this poisoning in under-developed countries, without substitution with modern toxico-dynamic treatment. Toxico-dynamic treatment needs intensive medical care, which is unavailable in these countries. Surgical wash-out of the small bowel could also be a complement to medical treatment in very massive chloroquine poisoning.

Systemic insecticides, comprising neonicotinoids, systemic organophosphates, phenylpyrazoles (fipronil), and anthranilic diamides, now dominate global insecticide use. Their high water solubility and systemic translocation also drive migration into paddy fields, wetlands, and agricultural waterways and persistence in soils and sediments, the habitats amphibians depend on. Permeable skin, gill-dominated respiration, and obligate aquatic larval phases make amphibians especially vulnerable, yet assessments linking field exposure to internal toxicity remain limited. Focusing on these four classes,

Perfluorobutanoic acid (PFBA) is of increasing concern due to its widespread occurrence, high environmental mobility, and potential for human exposure. This study investigated the toxicokinetics of PFBA in female Sprague Dawley rats and assessed implications for human exposure. Under repeated oral exposure (0.2 mg/kg/day for 16 days), concentrations of PFBA in blood increased rapidly and stabilized within the range of 230-300 ng/mL. Owing to its large tissue mass, muscle served as the major reservoir for the overall body burden of PFBA. Among the various organs, kidney, stomach, liver and gut

Absorption in the gastrointestinal tract is a key factor determining the bioavailability of chemicals after oral exposure but is frequently assumed to have a conservative value of 100% for environmental chemicals, particularly in the context of high-throughput toxicokinetics for in vitro-to-in vivo extrapolation (IVIVE). For pharmaceutical compounds, the physiologically based advanced compartmental absorption and transit (ACAT) model has been used extensively to predict gut absorption but has not generally been applied to environmental chemicals. Here we develop a probabilistic environmental c

Human umbilical cord mesenchymal stem cells (hUC-MSCs) are proposed for the treatment of acute lung injury and atopic dermatitis. To advance hUC-MSC entry into clinical trials, the effects of hUC-MSCs on the general toxicity, immune perturbation and toxicokinetic study of hUC-MSCs in cynomolgus monkeys were assessed. hUC-MSCs were administered to cynomolgus monkeys by intravenous infusion of 3.0 × 106 or 3.0 × 107cells/kg or by subcutaneous injection of 3.0 × 107cells/kg twice a week for 3 weeks followed by withdrawal and observation for 6 weeks. Toxicity was assessed by clinical observation,

BACKGROUND: Topiramate is an anticonvulsant from sulfamate-substituted monosaccharides that is increasingly used to treat migraines. Serious topiramate intoxications have been described. Unfortunately, indications for and the effect of interventions, including hemodialysis, in severe intoxications seem expert-based and lack empirical evidence. We aim to review the literature on topiramate intoxication cases and to describe the first topiramate intoxication with toxicokinetic data following treatment with hemodialysis.
METHODS: A literature review was conducted using the PubMed database. Inclu

Nontarget aquatic organisms residing in wetlands are commonly exposed to current-use pesticides through spray drift and runoff. However, it is frequently challenging to measure exposure because of rapid dissipation of pesticides from water and reduced bioavailability. The authors' hypothesis is that freshwater snails can serve as bioindicators of pesticide exposure based on their capacity to passively accumulate tissue residues. Helisoma trivolvis snails were evaluated as biomonitors of pesticide exposure using a fungicide formulation that contains pyraclostrobin and metconazole and is frequen

Per- and polyfluoroalkyl substances (PFAS) are persistent chemicals with substantial bioaccumulation potential, but their distribution between blood and urine in humans remains poorly characterized. In this review, we assessed the urine-to-blood concentration ratio (UtBCR) as a potential indicator of PFAS bioaccumulation by integrating evidence from human biomonitoring studies and protein-binding data. We summarized PFAS concentrations in human serum and urine across general and highly exposed populations and identified clear compound-specific differences in blood-urine partitioning. We furthe

The rapidly expanding use of novel brominated flame retardants (NBFRs), combined with limited knowledge of their effects, raises concerns regarding their potential hazards. To address this gap, screening-level toxicokinetic and toxicity profiling was first conducted for 27 environmentally prevalent NBFRs, followed by zebrafish verification using decabromodiphenyl ethane (DBDPE) as a representative compound. The study further integrated transcriptomic analysis and ferrostatin-1 (Fer-1) intervention to investigate whether ferroptosis is involved in DBDPE-induced neurodegeneration-related cogniti

The purpose of this study was to use chemical similarity evaluations, transcriptional profiling, in vitro toxicokinetic data, and physiologically based pharmacokinetic (PBPK) models to support read-across for a series of branched carboxylic acids using valproic acid (VPA), a known developmental toxicant, as a comparator. The chemicals included 2-propylpentanoic acid (VPA), 2-ethylbutanoic acid, 2-ethylhexanoic acid (EHA), 2-methylnonanoic acid, 2-hexyldecanoic acid, 2-propylnonanoic acid (PNA), dipentyl acetic acid or 2-pentylheptanoic acid, octanoic acid (a straight chain alkyl acid), and 2-e

Ethanol and medical psychotropics (MPs) are legal psychoactive substances widely consumed in Western countries that are routinely detected in standard toxicological analyses at compulsory autopsies, and toxicokinetic interactions between these drugs have been described. However, studies assessing the consequences of this co-consumption are scarce in Europe. We performed a retrospective study on toxicological results from compulsory autopsies in Spain. Thirty-five legal psychotropics, including ethanol, were measured in blood samples from 380 decedents to assess the determinants of such co-cons

Data concerning the monomethylmercury (MeHg) bioaccumulation in marine biota from Southeast Atlantic Ocean are scarce. This study purchased large specimens of demersal fishes from an upwelling region: Warsaw grouper (Epinephelus nigritus), Dusky grouper (Epinephelus marginatus) and Namorado sandperch (Pseudopercis numida). The authors addressed the bioaccumulation and toxicokinetic of mercury in fish organs, and the toxicological risk for human consumption of this metal in the muscle tissues accessed. Additionally, the present study discussed the possible implications of shifts in key variable
