The ontogeny of the shell in side-necked turtles, with emphasis on the homologies of costal and neural bones.

Although we are starting to understand the molecular basis of shell development based on the study of cryptodires, basic comparative ontogenetic data for the other major clade of living turtle, the pleurodires, are largely missing. Herein, the developmental and phylogenetic relation between the bony shell and endoskeleton of Pleurodira are examined by studying histological serial sections of nine specimens of three different species, including an ontogenetic series of Emydura subglobosa. Emphasis is given to the portion of the carapace in which ribs and vertebral spinous processes become part

An M-like outward current regulates the excitability of spinal motoneurones in the adult turtle.

The excitatory action of muscarine on spinal motoneurones was investigated with intracellular recordings in a slice preparation from adult turtles. In these cells muscarine is known to facilitate a persistent inward current mediated by L-type Ca(2+) channels. When this effect was blocked by nifedipine, muscarine still increased the excitability. In voltage clamp, a slowly activating outward current, generated during depolarizing voltage commands and deactivating as a tail current on return to the holding voltage, was reduced by muscarine. This outward current was activated when the voltage was

Presynaptic inhibition of primary olfactory afferents mediated by different mechanisms in lobster and turtle.

Presynaptic regulation of transmission at the first olfactory synapse was investigated by selectively imaging axon terminals of receptor neurons in the lobster olfactory lobe and turtle olfactory bulb. In both species, action potential propagation into axon terminals after olfactory nerve stimulation was measured using voltage-sensitive dyes. In addition, in the turtle, calcium influx into terminals was measured by selectively labeling receptor neurons with dextran-conjugated calcium indicator dyes. In the lobster, application of the inhibitory transmitters GABA or histamine suppressed action

[Development of the subclavian artery in the loggerhead turtle (Caretta caretta) studied by the injection method].

In the turtle, the left aorta and the pulmonary trunk originate from the right ventricle, while the right aorta takes its origin from the left ventricle as a functional systematic arch. The subclavian artery arises from the brachiocephalic artery on each side, and passes ventral to the vagus nerve and the jugular vein. These features are basically the same as in birds, and the subclavian artery of the adult turtle corresponds to a secondary artery from the viewpoint of comparative anatomy. Many investigators, including one of the present authors (Suzuki, 1987), have studied the development of

Pineal 5-methoxytryptophol rhythms in the box turtle: effect of photoperiod and environmental temperature.

The effect of different photoperiods and temperatures on pineal 5-methoxytryptophol (ML) content was investigated in male box turtles, Terrapene carolina triunguis. A rhythm in pineal ML was evident in the long photoperiod (18 h light (L)-6 h dark (D] with high daytime levels of 178 +/- 48 pg/gland (means +/- S.E.M.) which dropped to 38 +/- 6 pg/gland during lights off. In the short photoperiod (8L:16D) no clearcut ML rhythm was observed. Diurnal (10.00-12.00 h) ML concentrations rose linearly (P less than 0.05) with increasing ambient temperatures (5, 15, 20 and 27 degrees C). Day/night diffe

Temperature dependence of in vitro pituitary, testis, and thyroid secretion in a turtle, Pseudemys scripta.

In vitro culture was used to examine the direct actions of temperature at the level of pituitary hormone [luteinizing hormone (LH), thyrotropin (TSH), growth hormone (GH), prolactin (PRL)] responses to neuropeptides and two related peripheral endocrine responses [thyroid hormone (T4) and testicular androgen secretion] to pituitary hormones (TSH and gonadotropins) in a turtle, Pseudemys scripta. All these responses were fully suppressed at very low temperatures (5-6 degrees) and maximal near the species' preferred body temperature (28 degrees), but sensitivities differed markedly in intermediat

Glycemia and immunohistochemical changes in the endocrine pancreas of the turtle Chrysemys dorbigni treated with streptozotocin.

The diabetogenic action of streptozotocin (SZ) was investigated in the turtle Chrysemys dorbigni after a 1- or 14-day fast. SZ (130 or 250 mg/kg) was injected intravenously, and blood glucose and plasma insulin were measured. Pancreatic endocrine cells were stained immunohistochemically by the immunoperoxidase avidin-biotin-peroxidase complex method. Only 14% of the SZ-treated turtles showed hyperglycemia. Prolonged fasting did not increase the percentage of hyperglycemic animals. In control turtles, insulin (beta)-, glucagon (alpha)- and somatostatin (delta)-immunoreactive cells were detected

Suppression of evoked potentials with continued ion transport during anoxia in turtle brain.

A key to turtle brain survival during anoxia is continued ion transport and avoidance of anoxic depolarization. Previous findings that ATP concentration remained constant during prolonged anoxia and calculations that ATP production decreased indicate that compensatory processes, other than consumption of energy stores or increased anaerobic glycolysis, must also contribute to ion homeostasis and brain survival. To determine whether preservation of ion transport is associated with changes in electrophysiology during loss of oxidative metabolism, the brains of pentobarbital sodium-anesthetized t

Carbonic anhydrase function and the epithelial organization of H+ secretion in turtle urinary bladder.

The function of carbonic anhydrase in H(+) secretion by the turtle bladder was studied in vitro. Dose response curves were obtained for the carbonic anhydrase inhibitors, acetazolamide and ethoxzolamide, with and without addition of CO(2) to the system. In addition, carbonic anhydrase was assayed in homogenates of mucosal cells. The activity in the homogenates was 155+/-16 U/g dry wt, of which only 11 U represented contamination from erythrocytes; after addition of 5 x 10(-6)m acetazolamide, no enzyme activity was detectable. In the intact preparation free of exogenous CO(2), the dose response

Forty years of monitoring increasing sea turtle relative abundance in the Gulf of Mexico.

Longitudinal data sets for population abundance are essential for studies of imperiled organisms with long life spans or migratory movements, such as marine turtles. Population status trends are crucial for conservation managers to assess recovery effectiveness. A direct assessment of population growth is the enumeration of nesting numbers and quantifying nesting attempts (successful nests/unsuccessful attempts) and emergence success (number of hatchlings leaving the nest) because of the substantial annual variations due to nest placement, predation, and storm activity. We documented over 133,

Morphological characterization of the digestive tube of hawksbill sea turtle (Eretmochelys imbricata) hatchlings.

Morphological studies concerning the digestive system can further information on animal diets, thus aiding in the understanding of feeding behavior. Given the scarcity of information on sea turtle digestive system morphology, the aim of the present study was to describe the digestive tube (DT) morphology of Eretmochelys imbricata hatchlings to further understand the diet of these individuals in the wild. DT samples from 10 stillborn turtles (undefined sex) were analyzed at the macro and microscopic levels. The esophagus, stomach, small intestine (SI), and large intestine (LI) are described. Hi

Leucism: the prevalent congenital malformation in the olive ridley sea turtle of northwestern Mexico.

Despite being the most abundant sea turtle in the world, the olive ridley turtle Lepidochelys olivacea is classified as Vulnerable by the IUCN. There is evidence of congenital malformations in hatchlings, and the associated causes are multifactorial, with both genetic and environmental sources. Santuario Playa Ceuta (SPC) is a sanctuary for the olive ridley, located at the northernmost region of its nesting range in the Mexican Pacific. The objective of this study was to identify and quantify the prevalence and severity of congenital malformations in olive ridley embryos/hatchlings in SPC duri

Fatal Bronchopneumonia and Tracheitis in a Green Turtle (Chelonia mydas) Caused by Serratia proteamaculans.

A free-ranging subadult, male green turtle (Chelonia mydas) presented with radiographic evidence of pneumonia and died acutely. On necropsy, the trachea and bronchi were plugged by diphtheritic membranes, comprised of fibrin, necrotic debris, and colonies of bacilli, identified as Serratia proteamaculans. S. proteamaculans, typically considered an opportunistic plant pathogen, has rarely been described as causing disease in animals. This is the first report of S. proteamaculans causing severe necrotizing tracheitis and bronchopneumonia in a reptile.

COMPUTED TOMOGRAPHY OF CUTANEOUS MYIASIS IN AN ORNATE BOX TURTLE (TERRAPENE ORNATA ORNATA).

A juvenile intact female ornate box turtle (Terrapene ornata ornata) was referred for evaluation of right-sided appendicular swelling that was nonresponsive to antimicrobial therapy. Computed tomography (CT) revealed severe soft tissue swelling of the limbs and neck, with multiple associated tortuous gas-filled sinus tracts extending from the cutaneous surface, which contained oblong soft tissue attenuating structures. These CT findings were most consistent with migrating dipterous larvae. Surgical exploration was performed and six sarcophagid larvae (Superfamily Oestroidea) were extracted. To

Learning-Dependent Transcriptional Regulation of BDNF by its Truncated Protein Isoform in Turtle.

The vertebrate brain-derived neurotrophic factor (BDNF) gene produces a number of alternatively spliced transcripts only some of which generate the BDNF protein required for synaptic plasticity and learning. Many of the transcripts are uncharacterized and are of unknown biological significance. Previously, we described alternative splicing within the protein-coding sequence of the BDNF gene in the pond turtle (tBDNF) that generates a functionally distinct truncated protein isoform (trcBDNF) that is regulated during a neural correlate of eyeblink classical conditioning in ex vivo brainstem prep

Computationally modelling the mechanical behaviour of turtle shell sutures-A natural interlocking structure.

Sutures, the soft collagenous tissue joining interdigitating bony protrusions on the edges of bone plates, play a significant mechanical role in allowing a turtle shell to respond optimally to a range of loading regimes. In this contribution, qualitative and quantitative aspects of the mechanical behaviour of turtle shell suture regions are investigated by means of mathematical modelling. Notable features of the model include: (i) a geometrically realistic three dimensional model for the suture geometry; (ii) taking the hyperelastic, anisotropic and incompressible nature of the suture material

SURGICAL ANATOMY OF CELIOTOMY APPROACHES TO THE STOMACH IN THE COMMON SNAPPING TURTLE (CHELYDRA SERPENTINA).

Entanglement in or ingestion of fishing gear is a common cause of morbidity and mortality in chelonians. Commercial and recreational fishing activities exert bycatch pressures sufficient to cause population declines in the common snapping turtle (Chelydra serpentina) and the alligator snapping turtle (Macrochelys spp.). Gastrotomy for the removal of fish hook foreign bodies from the stomach of freshwater turtles typically requires a plastron osteotomy but in sea turtles has been successfully accomplished via soft tissue approaches. This study compared the anatomy and feasibility of soft tissue

Gut bacteria of Cuora amboinensis (turtle) produce broad-spectrum antibacterial molecules.

Antimicrobial resistance is a major threat to human health, hence there is an urgent need to discover antibacterial molecule(s). Previously, we hypothesized that microbial gut flora of animals are a potential source of antibacterial molecules. Among various animals, Cuora amboinensis (turtle) represents an important reptile species living in diverse ecological environments and feed on organic waste and terrestrial organisms and have been used in folk medicine. The purpose of this study was to mine turtle's gut bacteria for potential antibacterial molecule(s). Several bacteria were isolated fro

Histochemical and ultrastructural analyses of the lubrication systems in the olfactory organs of soft-shelled turtle.

In general, the nasal cavity of turtles is divided into two chambers: the upper chamber, lined with the olfactory epithelium containing ciliated olfactory receptor cells, and the lower chamber, lined with the vomeronasal epithelium containing microvillous receptor cells. In the nasal cavity of soft-shelled turtles, however, differences between the upper and lower chamber epithelia are unclear due to the presence of ciliated receptor cells in both epithelia. In the olfactory organ of vertebrates, the surface of sensory epithelium is covered with secretory products of associated glands and suppo
