Tissue selective modulation of redox and phosphate potentials by beta,beta'-methyl-substituted hexadecanedioic acid.

beta,beta'-Methyl-substituted hexadecanedioic acid (MEDICA 16) shares some of the calorigenic-hypolipidemic characteristics of thyroid hormones. In light of this similarity, MEDICA 16 was further evaluated here as a modulator of rat liver redox and phosphate potentials as well as an effector of cardiac high energy intermediates level in comparison to thyroid hormone. 1) Similarly to thyroid hormone, MEDICA 16 treatment resulted in 3.5-fold decrease in cytosolic redox potential. With both treatment modes, the induced decrease in cytosolic redox potential resulted in a concomitant increase in th

Oral effects of sugars and sweeteners.

Increasingly, sweeteners are being used in the diets of caries-prone individuals to reduce sugar intake. Such substitution deprives the bacteria in the dental plaque of the sugars many of them use to produce the acids that cause demineralization of tooth tissue and development of the caries lesion. Sweeteners are particularly effective replacements for dietary sugars because they also stimulate the flow of saliva which can, through several mechanisms, prevent demineralization and even bring about remineralization of already demineralized enamel, dentine or cementum. More saliva means that more

The mechanism of reducing the oil content of fried instant noodles by adding sugar alcohol: microstructure, surface properties and molecular structure.

Sugar alcohols are characterized by anti-caries properties, low glycemic response, low calorie value and non-participation in the Maillard reaction. Our preliminary experimental demonstrated that erythritol (ET), xylitol (XT), and lactitol (LT) significantly reduce the oil content of fried instant noodles, though the mechanism was unclear. This study therefore aimed to elucidate how these sugar alcohols reduce oil content by examining the alterations in oil distribution, pore characteristics, and molecular structures of starch and protein from macro-, micro-, and molecular levels. Low-field nu

Regulation of Streptococcus mutans biofilm virulence by dietary sugars: Mechanistic basis of sucralose-mediated suppression.

Streptococcus mutans is a major cariogenic pathogen whose proliferation, acidogenesis, and pathogenicity are strongly impacted by dietary sugars, resulting in dental biofilm-associated illnesses. This study evaluated the effects of sucrose, glucose, and alternative sweeteners (allulose, erythritol, stevia, sucralose, and xylitol) on S. mutans biofilm formation, adhesion, autoaggregation, acidogenicity, metabolic activity, and virulence-related gene expression across different dental surfaces. Sucrose significantly enhanced acid production and biofilm formation, with 3% sucrose resulting in the

Unveiling the impact of allulose on oral microbiota and biofilm formation via a cariogenic potential assessment platform.

INTRODUCTION: The increased consumption of refined carbohydrates, particularly sucrose, has contributed to metabolic disorders and oral diseases such as dental caries by promoting dysbiotic biofilm formation and reducing microbial diversity. Allulose, a rare sugar with physicochemical properties similar to sucrose, has been suggested to offer metabolic health benefits; however, its impact on oral biofilm ecology remains unclear. METHODS: We evaluated the cariogenic potential of allulose using a multi-tiered in vitro platform consisting of single-species planktonic and biofilm models, a dual-s

Impact of concerning excipients on animal safety: insights for veterinary pharmacotherapy and regulatory considerations.

OBJECTIVES: Veterinarians and pharmacists are familiar with the efficacy and safety aspects attributed to active pharmaceutical ingredients included in medicines, but they are rarely concerned with the safety of excipients present in medicines. Although generally recognized as safe, excipients are not chemically inert and may produce adverse events in certain animal populations. This review aims to present excipients of concern to these populations and highlight their relevance for rational veterinary pharmacotherapy. EVIDENCE ACQUISITION: A comprehensive review of the literature about the ex

Action of agents on glucosyltransferases from Streptococcus mutans in solution and adsorbed to experimental pellicle.

Glucosyltransferase (Gtf) activity mediates sucrose-dependent adherence of mutans streptococci to the tooth surface, is essential for the cariogenicity of these micro-organisms, and contributes significantly to the exopolysaccharide component of the dental-plaque matrix. Clearly, agents that inhibit Gtfs could have therapeutic benefit. Here the effects of agents that inhibit Gtfs in solution and adsorbed to a surface were explored. Various classes of chemical reagents were tested for their ability to inhibit the enzymes responsible for insoluble-glucan synthesis (GtfB), insoluble/soluble gluca

[Hypocaloric peripheral parenteral nutrition in postoperative patients (the Europan Project) (II)].

Hypocaloric peripheral parenteral nutrition (HPPN) appears to be indicated in patients in a situation of moderate malnutrition who are to undergo a short period of fasting following surgery. Our aim was to determine the utility of the contribution of parenteral solutions of amino acids (AA) with limited caloric supply in the post-surgical patient, using different nutritional evaluation parameters. We examined 75 post-surgical patients who met at least two of the three criteria established as malnutrition: 1) albumin < 3 g/dl; 2) pre-albumin < 21 mg/dl; 3) bodyweight of less than 95% of the ide

Manganese Deficiency Is Required for High Itaconic Acid Production From D-Xylose in Aspergillus terreus.

Itaconic acid is used as a bio-based, renewable building block in the polymer industry. It is produced by submerged fermentations of the filamentous fungus Aspergillus terreus from molasses or starch, but research over the efficient utilization of non-food, lignocellulosic plant biomass is soaring. The objective of this study was to test whether the application of two key cultivation parameters for obtaining itaconic acid from D-glucose in high yields - Mn2+ ion deficiency and high concentration of the carbon source - would also occur on D-xylose, the principal monomer of lignocellulose. To th
