Prediction of bone strength by μCT and MDCT-based finite-element-models: how much spatial resolution is needed?

OBJECTIVES: Finite-element-models (FEM) are a promising technology to predict bone strength and fracture risk. Usually, the highest spatial resolution technically available is used, but this requires excessive computation time and memory in numerical simulations of large volumes. Thus, FEM were compared at decreasing resolutions with respect to local strain distribution and prediction of failure load to (1) validate MDCT-based FEM and to (2) optimize spatial resolution to save computation time. MATERIALS AND METHODS: 20 cylindrical trabecular bone specimens (diameter 12 mm, length 15-20mm) we

Synergistic Modification of Static Chemical Agent with Fly Ash and Hemihydrate Gypsum for Mechanically Compatible Gas Drainage Borehole Sealing.

Borehole sealing quality plays a critical role in coal seam gas extraction efficiency, while the mechanical mismatch between conventional cement-based sealing materials and surrounding coal masses often leads to interface damage and sealing failure under mining-induced stress conditions. To address this issue, a modified static chemical agent (MSCA) was developed by incorporating fly ash (FA) and hemihydrate gypsum (HG) into a static chemical agent (SCA) system. The hydration behavior, expansion characteristics, mechanical properties, and engineering applicability of MSCA were systematically i

Mechanical, translucency, tribological resistance, and biocompatibility of lithium disilicate glass-ceramics produced by ZrO2 doping and digital light processing for dental prosthesis.

BACKGROUND: Lithium disilicate glass-ceramics (LDGCs) have become one of the most widely used dental prosthesis materials in clinical practice due to their excellent esthetics, biocompatibility, and enamel-like wear resistance. However, their inherent brittleness, limited mechanical strength, and progressive wear under long-term service have restricted their application in high-stress posterior regions. Herein, this study aims to develop zirconia (ZrO2)-reinforced LDGCs and systematically investigate the effects of ZrO2 content on microstructure evolution and mechanical, translucency, tribolog

Pharmacologic Mobilization and Chemokine-Directed Recruitment of Mesenchymal Stromal Cells to the Surgically Repaired Rotator Cuff.

BACKGROUND: Mesenchymal stromal cell (MSC) techniques represent a promising method to enhance the surgical repair of rotator cuff tears. To eliminate the resource-intensive process of cell isolation and culture expansion, a method to recruit endogenous MSCs was investigated in an established rat model of rotator cuff repair. HYPOTHESIS: MSCs can be pharmacologically mobilized from the peripheral blood and recruited to the operative rotator cuff to enhance tendon-bone healing. STUDY DESIGN: Controlled laboratory study. METHODS: The rat model of supraspinatus tendon detachment and acute surgi

Associations of circulating T-cell subsets with carotid artery stiffness: the multiethnic study of atherosclerosis.

Arterial stiffness measured by total pulse wave velocity (T-PWV) is associated with an increased risk of multiple age-related diseases. T-PWV can be described by structural (S-PWV) and load-dependent (LD-PWV) arterial stiffening. T-cells have been implicated in arterial remodeling, arterial stiffness, and hypertension in humans and animals; however, it is unknown whether T-cells are risk factors for T-PWV or its components. Therefore, we evaluated the cross-sectional associations of peripheral T-cell subpopulations with T-PWV, S-PWV, and LD-PWV. Peripheral blood T-cells were characterized usin

Strain-mode-specific mechanical testing and the interpretation of bone adaptation in the deer calcaneus.

The artiodactyl (deer and sheep) calcaneus is a model that helps in understanding how many bones achieve anatomical optimization and functional adaptation. We consider how the dorsal and plantar cortices of these bones are optimized in quasi-isolation (the conventional view) versus in the context of load sharing along the calcaneal shaft by "tension members" (the plantar ligament and superficial digital flexor tendon). This load-sharing concept replaces the conventional view, as we have argued in a recent publication that employs an advanced analytical model of habitual loading and fracture ri

Six-minute, in vivo MRI quantification of proximal femur trabecular bone 3D microstructure.

BACKGROUND: Assessment of proximal femur trabecular bone microstructure in vivo by magnetic resonance imaging has recently been validated for acquiring information independent of bone mineral density in osteoporotic patients. However, the requisite signal-to-noise ratio (SNR) and resolution for interrogation of the trabecular microstructure at this anatomical location prolongs the scan duration and renders the imaging protocol clinically infeasible. Parallel imaging and compressed sensing (PICS) techniques can reduce the scan duration of the imaging protocol without substantially compromising

Intra-articular injection of epigallocatechin (EGCG) crosslinks and alters biomechanical properties of articular cartilage, a study via nanoindentation.

Osteoarthritis and rheumatoid arthritis are debilitating conditions, affecting millions of people. Both osteoarthritis and rheumatoid arthritis degrade the articular cartilage (AC) at the ends of long bones, resulting in weakened tissue prone to further damage. This degradation impairs the cartilage's mechanical properties leading to areas of thinned cartilage and exposed bone which compromises the integrity of the joint. No preventative measures exist for joint destruction. Discovering a way to slow the degradation of AC or prevent it would slow the painful progression of the disease, allowin

Bone matrix development in steroid-induced osteoporosis is associated with a consistently reduced fibrillar stiffness linked to altered bone mineral quality.

UNLABELLED: Glucocorticoid-induced osteoporosis (GIOP) is a major secondary form of osteoporosis, with the fracture risk significantly elevated - at similar levels of bone mineral density - in patients taking glucocorticoids compared with non-users. The adverse bone structural changes at multiple hierarchical levels in GIOP, and their mechanistic consequences leading to reduced load-bearing capacity, are not clearly understood. Here we combine experimental X-ray nanoscale mechanical imaging with analytical modelling of the bone matrix mechanics to determine mechanisms causing bone material qua

Characterisation of a novel light activated adhesive scaffold: Potential for device attachment.

The most common methods for attaching a device to the internal tissues of the human body are via sutures, clips or staples. These attachment techniques require penetration and manipulation of the tissue. Tears and leaks can often be a complication post-attachment, and scarring usually occurs around the attachment sites. To resolve these issues, it is proposed to develop a soft tissue scaffold impregnated with Rose Bengal/Chitosan solution (RBC-scaffold, 0.01% w/v Rose Bengal, 1.7% w/v Medium Molecular Weight Chitosan). This scaffold will initially attach to the tissue via a light activation me

Food material properties and mandibular load resistance abilities in large-bodied hominoids.

Numerous comparative studies have sought to demonstrate a functional link between feeding behavior, diet, and mandibular form in primates. In lieu of data on the material properties of foods ingested and masticated, many investigators have relied on qualitative dietary classifications such as "folivore" or "frugivore." Here we provide the first analysis of the relationship between jaw form, dietary profiles, and food material properties in large-bodied hominoids. We employed ratios of area moments of inertia and condylar area to estimate moments imposed on the mandible in order to evaluate and

Three-dimensional modeling of the effects of parathyroid hormone on bone distribution in lumbar vertebrae of ovariectomized cynomolgus macaques.

Biomechanical and quantitative computed tomography (QCT) analyses showed beneficial effects of parathyroid hormone (PTH (1-34)) on lumbar vertebrae from ovariectomized monkeys, even after withdrawal of treatment for 6 months. Adult cynomolgus monkeys were randomized, ovariectomized (except for sham ovariectomy controls), and treated subcutaneously with vehicle (OVX) or 5 micrograms/kg per day PTH (1-34) (PTH5) for 18 months. An additional group was treated subcutaneously with 5 micrograms/kg per day PTH (1-34) (PTH5W) for 12 months and then switched to vehicle for the remaining 6 months. Lumba

Gold Nanorod-Doped Acrylic Bone Cement: Mechanical Stability and Feasibility of Photothermal Therapy.

PURPOSE: Photothermal therapy (PTT) involves the conversion of electromagnetic radiation into heat to achieve targeted destruction of cells or tissue. Gold nanorods (GNRs) are commonly used nanoparticles in PTT due to their efficient photothermal conversion properties. The purposes of this biomechanical study were 1) to evaluate the in vitro feasibility of PTT using GNR-doped acrylic bone cement and 2) to assess the mechanical properties of the GNR-doped bone cement before and after PTT exposure. METHODS: Forty-eight polymethyl methacrylate (PMMA) bone cement tensile specimens were prepared,

Structural and mechanical insights into the extracellular matrix of the aging human lacrimal gland for tissue engineering applications.

Aqueous-deficient dry eye disease (ADDE) frequently results from structural and functional impairment of the lacrimal gland. Regenerative approaches require biomaterials that replicate the extracellular matrix (ECM) composition and biomechanical properties of native lacrimal gland tissue. However, quantitative data on the ECM composition and large-strain biomechanics of the human lacrimal gland remain limited. In this study, histological and immunofluorescence-based ECM analyses were combined with large-strain mechanical testing and quantitative enzyme-linked immunosorbent assay (ELISA)-based
