The incidence of squamous cell carcinoma of the skin is rising worldwide for decades. Chronic exposure to sunlight is the most important environmental risk factor for this type of skin cancer. This is predominantly due to the DNA damaging effect of ultraviolet-B (UVB) in sunlight. UVB induces also sunburn cells, i.e. apoptotic keratinocytes, which is a crucial protective mechanism against the carcinogenic effects of UVB irradiation. This process is regulated by a wide range of molecular determinants involved in the balance between pro- and anti-apoptotic pathways. Growing evidence suggests tha

Ultraviolet (UV) radiation can induce acute and chronic photobiologic reactions in the absence of exogenous chromophores. Nuclear DNA is a major chromophore to initiate UV-induced physiologic reactions. The XPA-gene deficient mouse, an animal model of xeroderma pigmentosum, develops increased photobiologic reactions including acute inflammation, immunosuppression and skin cancers, because of the defect in the excision repair of UV-induced DNA lesions.

The inflamed synovium of rheumatoid arthritis exhibits many features typical for neoplastic tissue implying that the photodynamic therapy might be an efficient modality for chronic poliarthritis. The accumulation of endogenously produced porphyrins after administration of exogenous 5-aminolevulinic acid (ALA) in a rabbit model of rheumatoid arthritis was evaluated by fluorescence spectroscopy. Independent of the way, intravenously or intra-articularly, in which ALA was administered to the experimental animals, the highest fluorescence intensity of endogenously produced porphyrins was detected

Photooxidative processes play a role in the pathobiochemistry of various disorders of light-exposed tissue. After irradiation of skin with UV light, erythema (sunburn) is an initial effect suitable for monitoring the direct biological response. Carotenoids are efficient in photoprotection, scavenging singlet oxygen and peroxyl radicals. Intervention studies with supplements or a carotenoid-rich diet documented efficiency in systemic photoprotection, measuring a decreased sensitivity against UV-induced erythema. For successful intervention, treatment with carotenoids is needed for a period of a

Phototoxic lesions generated in tumor tissue by photodynamic therapy (PDT) are recognized by the host as a threat to the integrity and homeostasis at the affected site. Among the canonical pathways invoked by the host for dealing with this type of challenge is the activation of the complement system, integrating proteins that serve as molecular sensors of danger signals produced by PDT and those initiating signalling cascades coupled into the network of inflammatory and immune responses. Since the activated complement system is a salient participant of the antitumor response produced by PDT, i

The photodynamic therapy involving saturation of triplet state of a photosensitizer by short high-energy light pulses was studied theoretically. The possibility of creating a uniform therapeutic dose distribution throughout a large tumor using both surface and interstitial irradiation is shown. Possible thickness of the treated tissue layer and the required duration of the PDT session are estimated.

Human exposure to sunlight promotes the formation of pre-vitamin D in the skin. Low or marginal levels of vitamin D has been linked to a wide range of human health outcomes, including the development of various types of cancer. However, few data exist on the actual exposure to human due to vitamin D producing ultraviolet radiation. Most studies of human disease and vitamin D have linked latitude and location of residence to expected exposure form the available ambient UV radiation. Human UV exposure for the development of vitamin D depends on a variety of factors such as time spent outdoors, p

The immunological effects of visible and infrared light from laser and non-laser sources have remained insufficiently studied, which has restricted the use of light in the treatment of diseases associated with immune system disorders. The present randomised, placebo-controlled double-blind trial was designed to study changes in the humoral immunity of a large group of volunteers after exposure of a small body area to polychromatic visible and infrared polarized (VIP) and non-polarized (VInP) light (400-3400 nm, 95% polarization, 40 mW cm(-2), 12 J cm(-2) and 400-3400 nm, no polarization, 38 mW

Photoactive yellow protein, a small, water-soluble blue-light absorbing photoreceptor protein from Ectothiorhodospira(Halorhodospira)[space]halophila has a structure with two hydrophobic cores, of which the main one houses its light-sensitive chromophore (p-coumaric acid), separated by a central [small beta]-sheet. This photoreceptor protein contains a single tryptophan residue (W119) that is situated at the interface between the central beta-sheet and its N-terminal cap. The fluorescence properties of W119 in the dark state pG (lambda(max)= 328 nm; Phi(fl)= 0.01; nearly pH-independent) are ty

Chemical antibacterial agents are increasingly being used in prophylactic and therapeutic regimes for dental plaque-related diseases, which are among the most common human infections. As these agents are difficult to maintain at a therapeutic concentration in the oral cavity and can be rendered ineffective by resistance development in the target organisms, there is a need to develop alternative antimicrobial approaches. Bacteria and other microbes can be sensitised to light through prior treatment with a chemical photosensitising agent. Lethal photosensitisation of a wide range of bacteria res

Beta-carotene is a strong singlet oxygen quencher and antioxidant. Epidemiologic studies have implied that an above average intake of the carotenoid might reduce cancer risks. Earlier studies found that the carotenoid, when added to commercial closed-formula rodent diets, provided significant photoprotection against UV-carcinogenesis in mice. Clinical intervention trials found that beta-carotene supplementation evoked no change in incidence of nonmelanoma skin cancer. However, when smokers were supplemented with the carotenoid a significant increase in lung cancer resulted. Recently, employing

The photophysical properties of 2,3-diazabicyclo[2.2.2]oct-2-ene (DBO) were determined in 15 solvents, two supramolecular hosts (cucurbit[7]uril and beta-cyclodextrin) as well as in the gas phase. The oscillator strength and radiative decay rate of DBO as a function of refractive index i.e. polarizability have been analyzed. The oscillator strength increases by a factor of 10 upon going from the gas phase to the most polarizable carbon disulfide, while the corresponding radiative decay rates increase by a factor of 40. There is a good empirical correlation between the oscillator strength of th

The fluorescence spectra of acidified aqueous solutions of tetracycline (tcH3+) exhibit three components with slightly different degrees of anisotropy: the 'blue' component (lambdad approximately equal to 475 nm) decays on the timescale of a few picoseconds; the second, most intense component (lambdad approximately equal to 530 nm) shows decay times of about 25 (H3O+) and 70 ps (D3O+); the third component (lambdad approximately equal to 650 nm) is longer lived (tau approximately equal to 200 ps). All three fluorescence components appear quasi-instantaneously, thus providing evidence that the r

The effects of ozone depiction on global biogeochemical cycles, via increased UV-B radiation at the Earth's surface, have continued to be documented over the past 4 years. In this report we also document various effects of UV-B that interact with global climate change because the detailed interactions between ozone depletion and climate change are central to the prediction and evaluation of future Earth environmental conditions. There is increasing evidence that elevated UV-B has significant effects on the terrestrial biosphere with important implications for the cycling of carbon, nitrogen an

Although being an efficient photochromic compound which absorbs in the blue in its stable form and in the orange in its photoactivated form, the mercury dithizonate complex is shown to be a poor optical limiter for nanosecond laser pulses at the wavelengths where both isomers absorb. Optical limiting effect, which is a consequence of reverse saturable absorption due to the photoactivated form, is demonstrated to be weak because of the back photobleaching of this form, which is important all the more as the laser intensity is high. The photochemical fingerprint for the N-acetyl methyl ester of the aliphatic amino acid tert-leucine 1 was investigated. This reaction path was compared with the electron transfer active amino acids methionine (N-acetyl methyl ester derivative 2a as well as the methyl ester derivative 2b) and the cysteine derivatives 3a and 3b (penicillamine derivative). Photofragmenation of the ester group dominated the photolysis of 1, whereas loss of methylmercaptane was observed for 2a and 3a. Vinylglycine derivatives 11 and 14 were formed to a minor extent. The gem-dimethylated compound 3b gave a solvent-

The potential health effects of elevated levels of ambient UV-B radiation are diverse, and it is difficult to quantify the risks, especially as they are likely to be considerably modified by human behaviour. Nevertheless epidemiological and experimental studies have confirmed that UV radiation is a definite risk factor for certain types of cataract, with peak efficacy in the UV-B waveband. The causal link between squamous cell carcinoma and cumulative solar UV exposure has been well established. New findings regarding the genetic basis of skin cancer, including studies on genetically modified

Sunscreens are known to-protect against sunlight-induced erythema and sunburn, but their efficiency at protecting against skin cancer is still a matter of debate. Specifically, the capacity of physical sunscreens to prevent or reduce tissue and DNA damage has not been thoroughly investigated. Our objective was to assess the ability of a broad-spectrum sunscreen containing TiO2 to protect human skin against tissue and DNA damage following ultraviolet radiation. For this purpose, engineered human skin (EHS) was generated and either treated or not treated with an SPF 28 physical sunscreen and exp

We have developed a versatile tool for the delivery of inhibitors of carbonic anhydrase II, which allows modification of a hydrophobic drug with either a water-solubilizing, photolabile cage or a hydrophobic, photolabile cage. The former mask is useful for direct delivery of hydrophobic molecules in an aqueous prodrug form. The latter may find application if delivery from a surface is desirable. In our system, where the target enzyme is found in the eye, both approaches may be useful for the delivery of hydrophobic drugs having subnanomolar dissociation constants from the enzyme.

Zein is a type of prolamine protein that is derived from corn, and it has been recognized by the US FDA as one of the safest biological materials available. Zein possesses valuable characteristics that have made it a popular choice for the preparation of drug carriers, which can be administered through various routes to improve the therapeutic effect of antitumor drugs. Additionally, zein contains free hydroxyl and amino groups that offer numerous modification sites, enabling it to be hybridized with other materials to create functionalized drug delivery systems. However, despite its potential
