Assessment of seroprevalence of Mpox virus infection among sex workers in Chiang Mai, Thailand.

OBJECTIVES: The 2022 global Mpox outbreak revealed major surveillance gaps, particularly among marginalized populations such as sex workers, whose role in undetected MPXV transmission remains poorly understood. This study aims to assess the seroprevalence of MPXV IgG antibodies and identify associated risk factors among sex workers in Chiang Mai, Thailand. METHODS: In this cross-sectional study, 262 sex workers recruited from March to December 2022 were tested for prior MPXV exposure using an in-house indirect ELISA targeting the recombinant MPXV E8L antigen. Logistic regression models were u

Characterising household transmission dynamics of clade Ib mpox in Burundi: a prospective cohort study.

BACKGROUND: Knowledge of intrahousehold transmission dynamics of clade Ib mpox, especially in recently epidemic African contexts, is scarce. Our study aimed to quantify household transmission patterns of clade Ib mpox in Burundi, with a focus on children. METHODS: We conducted a prospective cohort study in two health districts, Bujumbura and Kayanza, in Burundi from Jan 23 to March 20, 2025, enrolling 88 laboratory-confirmed primary mpox cases and 432 of their household contacts. We estimated household secondary attack rates (SARs), serial intervals, and basic reproduction number (R0), includ

Associations Between Both HIV and Metabolic Comorbidity and Self-Reported Mpox Among Men Who Have Sex With Men: Multicenter Cross-Sectional Study.

BACKGROUND: Men who have sex with men (MSM) face a disproportionately high risk of mpox infection, and China has recently experienced a rapid increase in the reported cases. This population also has a high prevalence of HIV, which has been identified as a critical factor in understanding the vulnerability to mpox. In addition, metabolic diseases frequently co-occur with HIV and share immunometabolic pathways, raising concerns that they may interact to confer additional risk of mpox infection. OBJECTIVE: This study examines the potential interaction between HIV and metabolic comorbidity in rel

Survey of pain and stigma experiences in people diagnosed with mpox in Baltimore, Maryland during 2022 global outbreak.

A high prevalence of mpox in men who have sex with men and in people with HIV, plus visually striking and contagious lesions, have raised concerns for mpox stigma. 24 PCR-confirmed mpox patients were surveyed over the course of three months, utilizing an mpox stigma scale adapted from the HIV Stigma Scale plus assessment of pain, analgesic efficacy, and healthcare experiences. Participants were cis-male (100%), with male sexual partners (96%), mostly African-American (88%), and living with HIV (79%). Patients answered 4-16 of 24 (mean 10) stigma questions affirmatively, particularly related to

Social Determinants of Message Exposure and Health Anxiety Among Young Sexual Minority Men in the United States During the 2022 Mpox Outbreak.

This study tested the structural influence model of communication in the context of the 2022 global outbreak of the Mpox virus among young sexual minority men. The primary objective was to understand how distinct social determinants, including education, race/ethnicity, and interpersonal discrimination, influenced exposure to Mpox messages in daily life and affected health anxiety concerning the Mpox virus in the United States. We also explored the significance of LGBTQ+ community connectedness as a crucial form of social capital during the outbreak. We collected a three-wave longitudinal data

Risk of Clade II Mpox Associated with Intimate and Nonintimate Close Contact Among Men Who Have Sex with Men and Transgender Adults - United States, August 2022-July 2023.

A global outbreak of clade II mpox associated with sexual contact, disproportionately affecting gay, bisexual, and other men who have sex with men (MSM), has been ongoing since May 2022. Information on types of contact most associated with transmission is limited. This report used data from a multijurisdictional vaccine effectiveness case-control study of sexually active persons aged 18-49 years who identified as MSM or transgender, collected during August 2022-July 2023. Odds of mpox associated with selected types of intimate and nonintimate close contact with a person with mpox were estimate

Characterization of macaque pulmonary fluid proteome during monkeypox infection: dynamics of host response.

Understanding viral pathogenesis is challenging because of confounding factors, including nonabrasive access to infected tissues and high abundance of inflammatory mediators that may mask mechanistic details. In diseases such as influenza and smallpox where the primary cause of mortality results from complications in the lung, the characterization of lung fluid offers a unique opportunity to study host-pathogen interactions with minimal effect on infected animals. This investigation characterizes the global proteome response in the pulmonary fluid, bronchoalveolar lavage fluid, of macaques dur

Transmission of atypical varicella-zoster virus infections involving palm and sole manifestations in an area with monkeypox endemicity.

During a suspected monkeypox outbreak in the Republic of Congo, we documented transmission of varicella-zoster virus (VZV) infection with palm and sole manifestations among 5 family members. Genotyping results confirmed the VZV strain European E2, a genotype not previously reported in Africa. VZV with palm and sole involvement should be considered when differentiating a monkeypox diagnosis.

Oral Tecovirimat for the Treatment of Smallpox.

BACKGROUND: Smallpox was declared eradicated in 1980, but variola virus (VARV), which causes smallpox, still exists. There is no known effective treatment for smallpox; therefore, tecovirimat is being developed as an oral smallpox therapy. Because clinical trials in a context of natural disease are not possible, an alternative developmental path to evaluate efficacy and safety was needed. METHODS: We investigated the efficacy of tecovirimat in nonhuman primate (monkeypox) and rabbit (rabbitpox) models in accordance with the Food and Drug Administration (FDA) Animal Efficacy Rule, which was in

A mouse model of lethal infection for evaluating prophylactics and therapeutics against Monkeypox virus.

Monkeypox virus (MPXV) is an orthopoxvirus closely related to variola, the etiological agent of smallpox. In humans, MPXV causes a disease similar to smallpox and is considered to be an emerging infectious disease. Moreover, the use of MPXV for bioterroristic/biowarfare activities is of significant concern. Available small animal models of human monkeypox have been restricted to mammals with poorly defined biologies that also have limited reagent availability. We have established a murine MPXV model utilizing the STAT1-deficient C57BL/6 mouse. Here we report that a relatively low-dose intranas

Detection of monkeypox virus with real-time PCR assays.

BACKGROUND: Human monkeypox, a zoonotic disease, was first reported outside of Africa during the 2003 US outbreak. OBJECTIVES: We present two real-time PCR assays critical for laboratory diagnosis of monkeypox during the 2003 US outbreak. STUDY DESIGN: A TaqMan-based assay (E9L-NVAR) targets the orthopoxvirus DNA polymerase gene and detects Eurasian orthopoxviruses other than Variola. A hybridization assay, utilizing a MGB Eclipsetrade mark (Epoch Biosciences) probe, targets an envelope protein gene (B6R) and specifically detects monkeypox virus (MPXV). Assays were validated using coded orth

Mpox knowledge, attitudes toward Mpox vaccine and hesitancy for Mpox vaccination among teachers in Kenya.

BACKGROUND: Sixteen African countries, including Kenya, reported Mpox cases over a two-month period, prompting the Africa Centers for Disease Control and Prevention to declare the outbreak a Public Health Emergency of Continental Security on 13th August 2024. The World Health Organization issued guidance for Mpox prevention in schools, where teachers are expected to support infection prevention. This study assessed teachers' knowledge of Mpox and attitudes toward Mpox vaccination, given these factors that may influence teachers' confidence in promoting preventive behaviours and vaccination awa

Health insurance status and severe mpox disease outcomes among sexual minority men in NYC: a retrospective cohort study.

BACKGROUND: The 2022-2023 global mpox outbreak predominantly affected sexual minority men, with notable racial/ethnic disparities in the USA. While the current literature has established the clinical predictors of severe mpox disease, little is known about the role of insurance status on clinical outcomes. We sought to characterize patients diagnosed with mpox in New York City (NYC) and examine associations between insurance status and mpox severity score (mpox-SS). METHODS: We included 143 patients aged 18 years and older between May 1, 2022, and December 31, 2023, with confirmed mpox identi

Exploring Viral Genome Profile in Mpox Patients during the 2022 Outbreak, in a North-Eastern Centre of Italy.

In 2022, an unprecedented outbreak of mpox raged in several nations. Sequences from the 2022 outbreak reveal a higher nucleotide substitution if compared with the estimated rate for orthopoxviruses. Recently, intra-lesion SNVs (single nucleotide variants) have been described, and these have been suggested as possible sources of genetic variation. Until now, it has not been clear if the presence of several SNVs could represents the result of local mutagenesis or a possible co-infection. We investigated the significance of SNVs through whole-genome sequencing analysis of four unrelated mpox case
