Host survival during sepsis depends not only on pathogen burden but also on inflammatory thresholds calibrated by the gut microbiota. Here, we show that different survival outcomes were observed in genetically equivalent female C57BL/6 mouse populations depending on their specific gut microbiota configuration. A Muribaculaceae-enriched gut microbiota, characterized by the dominance of Sangeribacter muris KT1-3, predisposed mice to fatal sepsis caused by Acinetobacter baumannii via TLR4-dependent hyperinflammation. This lethal phenotype, reproduced by colonization with S. muris strain KT1-3, wa

Antimicrobial resistance (AMR) in Acinetobacter baumannii represents a critical global health challenge, particularly in intensive care settings where the pathogen causes severe, refractory infections. As a leading member of the ESKAPE group, A. baumannii has accumulated extensive resistance to multiple antibiotic classes, including carbapenems, resulting in the widespread emergence of multidrug-resistant (MDR), extensively drug-resistant (XDR), and pan-drug-resistant (PDR) strains. This review provides a chronological overview of the evolution of antimicrobial therapies used against A. bauman

Acinetobacter baumannii-calcoaceticus complex (ABC complex) is recognized as one of the most critical multidrug drugs resistant (MDR) pathogens worldwide and remains a major cause of hospital-acquired infections, particularly in intensive care settings. Members of this complex are associated with ventilator-associated pneumonia, bloodstream infections, wound infections, urinary tract infections, and meningitis, often affecting critically ill and immunocompromised patients. Their clinical importance is primarily driven by their remarkable ability to acquire, accumulate, and maintain resistance

Healing of traumatic wounds is complicated by infecting pathogens, with Pseudomonas aeruginosa and members of the Acinetobacter calcoaceticus-baumannii complex among the most common infectious agents. However, a full understanding of genotypic and phenotypic differences between circulating wound isolates is lacking. To address this, traumatic wounds were sampled at Emory University Hospital, and 24 isolates were characterized; we focused on P. aeruginosa and Acinetobacter due to their prevalence and tendency for antibiotic resistance. Even though these species are renowned for antibiotic resis

UNLABELLED: Acinetobacter baumannii is a clinically prevalent multidrug-resistant bacterium, while Elizabethkingia, an emerging pathogen, has shown increasing infection and mortality rates in recent years. Although co-infections involving both pathogens have been reported, their interactions remain poorly understood. In this study, clinical data from 15 co-infected patients revealed that such infections were more frequent in elderly individuals with underlying diseases requiring intensive care and prolonged hospitalization. The two isolates exhibited similar antibiotic resistance profiles. In

Carbapenem-resistant Acinetobacter baumannii (CRAb) causes hard-to-control healthcare outbreaks, but the mobile resistance determinants that sustain regional spread are often underrecognized. We characterized 70 CRAb isolates from 11 hospitals in southern Poland (2019-2022) using whole-genome sequencing and core genome multilocus sequence typing (cgMLST), with reconstruction of carbapenemase context and plasmid structure. All isolates belonged to international clone 2 (IC2; ST2, Pasteur scheme) or to ST425 (n = 47), ST195 (n = 19), ST208 (n = 2), and ST348 (n = 1) according to the Oxford schem

Acinetobacter baumannii is a priority bacterial pathogen and leading cause of nosocomial infections, particularly in intensive care units (ICUs). The average incidence of carbapenem-resistant A. baumannii infections in ICUs is 41.7 cases/1,000 patients, highlighting the urgent need for more effective alternative therapies to replace carbapenems. Thus, this study aimed to investigate for the first time the antibacterial activity of curcumin in combination with the novel phage vB_AbaSI_1 to combat multidrug-resistant (MDR) A. baumannii in vitro. Phage vB_AbaSI_1 (capsid diameter 91 nm, contracti

Acinetobacter baumannii is an opportunistic pathogen and a major cause of nosocomial infections worldwide. This study aimed to isolate and characterize phages with lytic activity against multidrug-resistant A. baumannii strains to enable antibacterial alternatives. Eight phages (AKO8a, PS118, B612, MCR, IDQ7, 89P13, CRL20, and CIM23) were isolated and subjected to genomic, phylogenetic, and functional analyses. Antibacterial activity was assessed in vitro against A. baumannii strain AbAK04 by measuring optical density over 17 h at multiplicities of infection (MOIs) of 0.1, 1, and 10, using a r

This study analyzed the antimicrobial resistance profiles and risk factors for carbapenem-resistant Acinetobacter baumannii (CRAB) in a tertiary hospital and developed a predictive model for infection control. Among 64 Acinetobacter baumannii isolates collected in 2024 from the First People's Hospital of Chuzhou, CRAB accounted for 40.63% (26/64), with sputum being the most common specimen source (85.94%) and the highest isolation rate observed in respiratory wards. CRAB exhibited significantly higher resistance to most antibiotics compared to carbapenem-sensitive strains (CSAB), except for po

The increasing antimicrobial resistance (AMR) of Acinetobacter baumannii presents a challenge to clinical management and underscores its role as a critical pathogen in refractory urinary tract infections (UTIs). Nitroxoline has gained renewed interest as a potential therapeutic option. Raman deuterium stable isotope probing (Raman-DIP), which allows for the analysis of bacterial metabolic activity, has shown promise as a tool for assessing antimicrobial efficacy. The aim of this study was to evaluate the bactericidal activity of nitroxoline against A. baumannii and the potential of Raman-DIP t

INTRODUCTION: With the rising incidence of infectious diseases, the overuse of antibiotics has become a serious problem in clinical practice. In recent years, metagenomic next-generation sequencing (mNGS) has emerged as a promising alternative, offering advantages such as rapid turnaround, broad-spectrum detection, and comprehensive coverage. However, its clinical utility for antimicrobial resistance testing remains to be fully established.
METHODS: In this study, we evaluated the effectiveness of mNGS in detecting antimicrobial resistance in Acinetobacter baumannii using 53 clinical samples.

BACKGROUND: Pleural effusion (PE) is a frequent clinical condition with diverse etiologies including heart failure, infections, and malignancies. While the color of pleural fluid is rarely considered diagnostic, unusual discolorations may offer important clinical clues. We report what appears to be only the third documented case of purple PE (PPE) in the medical literature. This case was associated with Acinetobacter baumannii infection, a rarely reported cause of PPE, providing additional insight into its potential pathophysiology.

Rhamnolipids, a major category of glycolipid biosurfactant, have recently gained enormous attention in medical field because of their relevance as effective antibacterial agents against a wide variety of pathogenic bacteria. Our previous studies have shown that rhamnolipids from an environmental isolate of Pseudomonas aeruginosa UKMP14T possess antibacterial, anti-adhesive and anti-biofilm activity against multidrug-resistant ESKAPE (Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa and Enterobacter sp.) pathogens. However, the

Acinetobacter baumannii is an opportunistic pathogen with ability to cause serious infection such as bacteremia, ventilator associated pneumonia, and wound infections. As strains of A. baumannii are resistant to almost all clinically used antibiotics and with the emergence of carbapenems resistant phenotypes warrants the search for novel antibiotics. Considering this, herein, a series of computer aided drug designing approach was utilized to search novel chemical scaffolds that bind stronger to MurE ligase enzyme of A. baumannii, which is involved peptidoglycan synthesis. The work identified L

Acinetobacter baumannii is one of the most successful pathogens that can cause difficult-to-treat nosocomial infections. Outbreaks and infections caused by multi-drug resistant A. baumannii are prevalent worldwide, with only a few antibiotics are currently available for treatments. Plasmids represent an ideal vehicle for acquiring and transferring resistance genes in A. baumannii. Five extensively drug-resistant A. baumannii clinical isolates from three major Jordanian hospitals were fully sequenced. Whole-Genome Sequences (WGS) were used to study the antimicrobial resistance and virulence gen

The emergence of multidrug-resistant Gram-negative bacteria underscores the need to define genetic vulnerabilities that can be therapeutically exploited. The Gram-negative pathogen, Acinetobacter baumannii, is considered an urgent threat due to its propensity to evade antibiotic treatments. Essential cellular processes are the target of existing antibiotics and a likely source of new vulnerabilities. Although A. baumannii essential genes have been identified by transposon sequencing, they have not been prioritized by sensitivity to knockdown or antibiotics. Here, we take a systems biology appr
