Distinguishing the molecular diversity, nutrient content, and energetic potential of exometabolomes produced by macroalgae and reef-building corals.

Metabolites exuded by primary producers comprise a significant fraction of marine dissolved organic matter, a poorly characterized, heterogenous mixture that dictates microbial metabolism and biogeochemical cycling. We present a foundational untargeted molecular analysis of exudates released by coral reef primary producers using liquid chromatography-tandem mass spectrometry to examine compounds produced by two coral species and three types of algae (macroalgae, turfing microalgae, and crustose coralline algae [CCA]) from Mo'orea, French Polynesia. Of 10,568 distinct ion features recovered fro

Comparison of the Sulfonamide Inhibition Profiles of the α-Carbonic Anhydrase Isoforms (SpiCA1, SpiCA2 and SpiCA3) Encoded by the Genome of the Scleractinian Coral Stylophora pistillata.

The ubiquitous metalloenzymes carbonic anhydrases (CAs, EC 4.2.1.1) are responsible for the reversible hydration of CO₂ to bicarbonate (HCO₃-) and protons (H⁺). Bicarbonate may subsequently generate carbonate used in many functional activities by marine organisms. CAs play a crucial role in several physiological processes, e.g., respiration, inorganic carbon transport, intra and extra-cellular pH regulation, and bio-mineralization. Multiple transcript variants and protein isoforms exist in the organisms. Recently, 16 α-CA isoforms have been identified in the coral Stylophora pistillata. Here,

Long-term impacts of rising sea temperature and sea level on shallow water coral communities over a ~40 year period.

Effects of combined rising sea temperature and increasing sea level on coral reefs, both factors associated with global warming, have rarely been addressed. In this ~40 y study of shallow reefs in the eastern Indian Ocean, we show that a rising relative sea level, currently estimated at ~11 mm y-1, has not only promoted coral cover but also has potential to limit damaging effects of thermally-induced bleaching. In 2010 the region experienced the most severe bleaching on record with corals subject to sea temperatures of >31 °C for 7 weeks. While the reef flats studied have a common aspect and a

Lobophorin K, a New Natural Product with Cytotoxic Activity Produced by Streptomyces sp. M-207 Associated with the Deep-Sea Coral Lophelia pertusa.

The present article describes the isolation of a new natural product of the lobophorin family, designated as lobophorin K (1), from cultures of the marine actinobacteria Streptomyces sp. M-207, previously isolated from the cold-water coral Lophelia pertusa collected at 1800 m depth during an expedition to the submarine Avilés Canyon. Its structure was determined using a combination of spectroscopic techniques, mainly ESI-TOF MS and 1D and 2D NMR. This new natural product displayed cytotoxic activity against two human tumor cell lines, such as pancreatic carcinoma (MiaPaca-2) and breast adenoca

Shagenes A and B, new tricyclic sesquiterpenes produced by an undescribed Antarctic octocoral.

The isolation and characterization of two new tricyclic sesquiterpenoids, shagenes A (1) and B (2) are presented. These compounds were isolated from an undescribed soft coral collected from the Scotia Arc in the Southern Ocean. One- and two-dimensional NMR spectroscopy and mass spectrometry provided the data necessary to characterize the compounds and their relative stereochemical configurations. Exploration of the bioactivity of shagenes A and B found 1 active against the visceral leishmaniasis causing parasite, Leishmania donovani, with no cytotoxicity against the mammalian host.

Disturbance driven colony fragmentation as a driver of a coral disease outbreak.

In September of 2010, Brewer's Bay reef, located in St. Thomas (U.S. Virgin Islands), was simultaneously affected by abnormally high temperatures and the passage of a hurricane that resulted in the mass bleaching and fragmentation of its coral community. An outbreak of a rapid tissue loss disease among coral colonies was associated with these two disturbances. Gross lesion signs and lesion progression rates indicated that the disease was most similar to the Caribbean coral disease white plague type 1. Experiments indicated that the disease was transmissible through direct contact between colon

Global gradients of coral exposure to environmental stresses and implications for local management.

BACKGROUND: The decline of coral reefs globally underscores the need for a spatial assessment of their exposure to multiple environmental stressors to estimate vulnerability and evaluate potential counter-measures. METHODOLOGY/PRINCIPAL FINDINGS: This study combined global spatial gradients of coral exposure to radiation stress factors (temperature, UV light and doldrums), stress-reinforcing factors (sedimentation and eutrophication), and stress-reducing factors (temperature variability and tidal amplitude) to produce a global map of coral exposure and identify areas where exposure depends on

Heterotrophy in tropical scleractinian corals.

The dual character of corals, that they are both auto- and heterotrophs, was recognized early in the twentieth Century. It is generally accepted that the symbiotic association between corals and their endosymbiotic algae (called zooxanthellae) is fundamental to the development of coral reefs in oligotrophic tropical oceans because zooxanthellae transfer the major part of their photosynthates to the coral host (autotrophic nutrition). However, numerous studies have confirmed that many species of corals are also active heterotrophs, ingesting organisms ranging from bacteria to mesozooplankton. H

Forecasting models to quantify three anthropogenic stresses on coral reefs from marine recreation: anchor damage, diver contact and copper emission from antifouling paint.

This research focuses on damage to coral reefs from three anthropogenic stresses: the dropping of anchors and their chains, human contact, and emission of copper from antifouling paints. Forecasting models are described that quantify degradation in terms of percentage of coral cover damaged/year or increasing levels of water toxicity/year. The models utilize a Monte Carlo simulation that applies a range of values or a probability distribution to each of the numerous uncertain variables. This model has the flexibility to adapt, and become more accurate, when users input assumptions specific to

Mosaic Accumulation of Somatic Genetic Variation and Estimates of Age in the Long-Lived Reef-Building Coral Acropora palmata.

Somatic genetic variation (SoGV) accumulates during an organism's lifetime and is traditionally viewed as detrimental rather than adaptive due to links with cancer and senescence. However, in modular organisms like corals, deleterious mutations can be purged at the cellular or polyp level, while adaptive mutations may rise in frequency as polyps create genetically distinct modules. Quantifying the somatic genetic landscape in corals is necessary to understand the role these mutations may have in coral and clonal animal development and evolution. Here, we catalogue somatic genetic variation in

An integrated deep learning framework for automated coral health assessment using a custom-annotated database.

Coral reefs face accelerating threats from climate-induced bleaching events, underscoring the need for scalable and automated health monitoring tools. This paper presents a comprehensive deep learning framework integrating object detection and image classification with a rigorously evaluated preprocessing pipeline for automated coral health assessment. Building on the custom annotated coral bleaching dataset introduced in Weraikat et al. [1], comprising 2,640 underwater coral images, four preprocessing strategies, RGB normalization, HSV conversion, histogram equalization, and color jittering,

Ocean acidification modulates material flux linked with coral calcification and photosynthesis.

Coral reefs are essential for the foundation of marine ecosystems. However, ocean acidification (OA), driven by rising atmospheric carbon dioxide (CO2) threatens coral growth and biological homeostasis. This study examines two Hawaiian coral species-Montipora capitata and Pocillopora acuta to elevated pCO2 simulating OA. Utilizing pH and O2 microsensors under controlled light and dark conditions, this work characterized interspecific concentration boundary layer (CBL) traits and quantified material fluxes under ambient and elevated pCO2. The results of this study revealed that under increased

Impacts of hurricanes and disease on Diadema antillarum in shallow water reef and mangrove locations in St John, USVI.

The 1983-1984 mortality event of the long-spined sea urchin Diadema antillarum reduced their population by up to 99% and was accompanied by a phase shift from coral dominated to algal dominated reefs in the Caribbean. Modest rebounds of D. antillarum populations in the Caribbean have been noted, and here we document the impacts of two major hurricanes (2017, Irma and Maria) and the 2022 disease outbreak on populations of D. antillarum found by targeted surveys in the urchin zone at nine fringing reef and three mangrove sites on St. John, USVI. D. antillarum populations at the reef sites had de

Decision analysis to support wastewater management in coral reef priority area.

A cocktail of land-based sources of pollution threatens coral reef ecosystems, and addressing these has become a key management and policy challenge in the State of Hawai'i, other US territories, and globally. In West Maui, Hawai'i, nearly one quarter of all living corals were lost between 1995 and 2008. Onsite disposal systems (OSDS) for sewage leak contaminants into drinking water sources and nearshore waters. In recognition of this risk, the Hawai'i State Department of Health (DOH) is prioritizing areas for cesspool upgrades. Independently, we applied a decision analysis process to identify

Zoanthid mucus as new source of useful biologically active proteins.

Palythoa caribaeorum is a very common colonial zoanthid in the coastal reefs of Brazil. It is known for its massive production of mucus, which is traditionally used in folk medicine by fishermen in northeastern Brazil. This study identified biologically active compounds in P. caribaerum mucus. Crude mucus was collected during low tides by the manual scraping of colonies; samples were maintained in an ice bath, homogenized, and centrifuged at 16,000 g for 1 h at 4 °C; the supernatant (mucus) was kept at -80 °C until use. The enzymatic (proteolytic and phospholipase A2), inhibitory (metallo, cys

Metagenomic analysis reveals a green sulfur bacterium as a potential coral symbiont.

Coral reefs are ecologically significant habitats. Coral-algal symbiosis confers ecological success on coral reefs and coral-microbial symbiosis is also vital to coral reefs. However, current understanding of coral-microbial symbiosis on a genomic scale is largely unknown. Here we report a potential microbial symbiont in corals revealed by metagenomics-based genomic study. Microbial cells in coral were enriched for metagenomic analysis and a high-quality draft genome of "Candidatus Prosthecochloris korallensis" was recovered by metagenome assembly and genome binning. Phylogenetic analysis show

Antibody characterization and immunoassays for palytoxin using an SPR biosensor.

Palytoxin (PLTX), a polyether marine toxin originally isolated from the zoanthid Palythoa toxica, is one of the most toxic non-protein substances known. Fatal poisonings have been linked to ingestion of PLTX-contaminated seafood, and effects in humans have been associated with dermal and inhalational exposure to PLTX containing organisms and waters. Additionally, PLTX co-occurrence with other well-characterized seafood toxins (e.g., ciguatoxins, saxitoxins, tetrodotoxin) has hindered direct associations of PLTX to seafood-borne illnesses. There are currently no validated methods for the quanti

Visualization of the ultrastructural interface of cells with the outer and inner-surface of coral skeletons.

Crystalline, porous biomaterials, such as marine invertebrate skeletons, have been widely used for functional reconstruction of human tissues like bone and dental implants. Since in such an abrasive microenvironment adequate cell-material interactions are crucial for a successful treatment, it is of great importance to improve the means to examine these interactions. We developed a method that reveals the ultrastructure of the interface between coral skeletons and cultured neural cells to a higher quality than do traditional methods as it does not include damaging procedures like decalcificati

Dynamics and impact of the coral disease white plague: insights from a simulation model.

Coral disease is playing a significant role in structuring today's coral reef communities. While monitoring programs document declines associated with coral disease, there is a lack of tools that can test hypotheses of disease incidence and control. Here, we describe a modeling tool developed to test hypotheses about the spread and impact of white plague disease in diverse coral populations distributed across heterogeneous reef landscapes. The model Simulation of Infected Corals (SICO) was based on the dynamics of white plague over the course of 6 yr of monitoring on the fore-reefs of Little C
