The reproducibility of scrapie-associated fibril and PrPSc detection methods after long-term cold storage of natural ovine scrapie-affected brain tissue.

A pool of grey matter (medulla/brain stem, cerebellum and frontal cerebral cortex) was prepared from the brains of 16 sheep with scrapie, diagnosed clinically and by the demonstration of spongiform encephalopathy. Aliquots from the pool of tissue were finely chopped or homogenized and stored at +4 degrees C or -70 degrees C, after undergoing one of several specific pre-treatments (storage with or without protease inhibitors or, alternatively, with or without the cryoprotectant, dimethyl sulphoxide). At intervals over a period of 2 years, the stored extracts were examined by electron microscopy

A carboxy-terminal processing protease gene is located immediately upstream of the invasion-associated locus from Bartonella bacilliformis.

A gene with homology to those encoding an unusual class of C-terminal processing proteases that flanks the invasion-associated locus ialAB of Bartonella bacilliformis has been identified. The 1302 bp gene, termed ctpA, is located immediately upstream of the ialA gene and encodes a predicted nascent product of 434 amino acids, producing a mature protein of 411 amino acid residues. The Bartonella CtpA appears to undergo autolysis in vitro, producing multiple products of 43-46 kDa, and a second group of products of 36-37 kDa. Production of CtpA in vivo gives a single product of 41.8 kDa. In addit

Effect of early ovariectomy and steroid hormone replacement of embryo transport, development and implantation in mice.

Bilateral ovariectomy on Day 1 of pregnancy increased abnormal embryo numbers on Day 4 and delayed passage of embryos to the uterus. Progestins given on Day 1 reversed these effects; given on Day 3 they reduced numbers of abnormal embryos, but did not restore normal transport. Oestrogen given alone after ovariectomy increased embryo loss, but restored preimplantation embryo development to normal when given on Day 3 after progestins on Day 1. The results suggested that both oestrogen and progesterone were necessary for normal preimplantation embryo development in vivo. However, although Day-1 p

The pathology of Rift Valley fever. II. Lesions occurring in field cases in adult cattle, calves and aborted foetuses.

Since the original description of Rift Valley fever in sheep, cattle and man in the Rift Valley in Kenya in 1931, very little has been published on the disease in cattle. This report deals with the macroscopic and microscopic pathology of field cases of Rift Valley fever in 22 adult cattle, 8 calves and 8 aborted foetuses. The microscopic liver lesions in 13 adult cattle were characterized by marked centri- and midzonal eosinophilic necrosis, involving almost 2/3 of the lobules, and accompanied by sparsely distributed primary foci of necrosis. In 3 animals, however, the hepatic lesions were mo

Purification and characterization of Clostridium difficile toxin.

Recent evidence indicates that toxigenic Clostridium difficile strains are a major cause of antimicrobial-associated ileocecitis in laboratory animals and pseudomembranous colitis in humans. C. difficile ATCC 9689 was cultivated in a synthetic medium to which 3% ultrafiltrated proteose peptone was added. Purification of the toxin from broth filtrate was accomplished through ultrafiltration (100,000 nominal-molecular-weight-limit membrane), precipitation with 75% (NH4)2SO4, and chromatographic separation using Bio-Gel A 5m followed by ion-exchange chromatography on a diethylaminoethyl-Sephadex

A human-cell recombinant prethrombin-2 platform producing thrombin with enhanced fibrinogen processing for in vitro applications.

Thrombin is a central mediator of haemostasis, immune responses, and inflammation. It arises from prothrombin upon prothrombinase cleavage at R320-I321, which triggers the allosteric maturation typical of chymotrypsin-like serine peptidases. Further processing yields the physiologically dominant α-thrombin (αT), whose competent active site centres on catalytic S525. Despite widespread biomedical and biotechnological use, plasma-derived αT is heterogeneous and risks pathogen or prion transmission, motivating recombinant alternatives. These have largely relied on suboptimal bacterial or non-huma

A Robust, Self-Digestion-Resistant LysN with Superior Activity and Cleavage Fidelity for Advanced Proteomic Workflows.

LysN is a valuable protease in proteomics because it cleaves peptide bonds N-terminal to lysine, generating peptides with physicochemical properties complementary to those produced by LysC and trypsin. However, the broader adoption of LysN in proteomic workflows has been limited by the lack of commercially available enzymes that combine high activity, low missed-cleavage rates, and sufficient stability under practical sample-processing conditions. Here, we report the recombinant production and proteomic characterization of a self-digestion-resistant and highly active LysN from Shewanella loihi

Valorization of Miscanthus × giganteus hydrolysates into heme-enriched yeast biomass and extract using xylose-fermenting Saccharomyces cerevisiae.

Heme-enriched microbial products represent value-added bioproducts that can be produced through sustainable fermentation processes. Current microbial heme production platforms rely mainly on refined sugars and downstream purification, limiting cost-effectiveness. Here, we developed a lignocellulosic hydrolysate-based bioprocess for producing heme-enriched yeast extract using Saccharomyces cerevisiae. The xylose-fermenting S. cerevisiae SR8 accumulated approximately twofold higher specific intracellular heme on xylose than on glucose, reaching 0.25 ± 0.02 mg/g dry cell weight (DCW) compared wit

Reduced habenular volumes and neuron numbers in male heroin addicts: a post-mortem study.

The Habenula is increasingly being investigated in addiction. Reduced volumes of other relevant brain regions in addiction, such as nucleus accumbens, globus pallidus and hypothalamus have been reported. Reduced volumes of the habenula as well as reduced neuronal cell count in the habenula have also been reported in mood disorders and an overlap between mood disorders and addiction is clinically widely recognized. Thus, our aim was to investigate possible volume and neuronal cell count differences in heroin addicts compared to healthy controls. Volumes of the medial (MHB) and lateral habenula

Persistent bacteremia due to methicillin-resistant Staphylococcus aureus infection is associated with agr dysfunction and low-level in vitro resistance to thrombin-induced platelet microbicidal protein.

BACKGROUND: The causes of persistent bacteremia (PB) due to methicillin-resistant Staphylococcus aureus (MRSA) are poorly understood. This investigation examined potential associations between PB with key clinical features and several in vitro bacterial genotypic and phenotypic characteristics, in isolates from 1 institution. METHODS: Pulsed-field gel electrophoresis (PFGE) relatedness, thrombin-induced platelet microbicidal protein (tPMP)-susceptibility phenotype, accessory gene regulator (agr) genotype and functionality (via delta-lysin production), and autolysis phenotypes were assessed in

[Heterologous biosynthesis and production optimization of p-coumaric acid in Streptomyces coelicolor].

p-coumaric acid is an important bioactive natural product and a crucial precursor for the synthesis of downstream phenylpropanoids. Compared with plant extraction and chemical synthesis methods, microbial production of p-coumaric acid offers unique advantages, including environmental friendliness, short production cycles, and high conversion efficiency. Streptomyces species possess a complete shikimate pathway and an extensive aromatic amino acid metabolic network, being promising chassis for the production of p-coumaric acid. This study aims to achieve efficient heterologous biosynthesis of p

Engineering an Escherichia coli Autolytic System for Overexpression of a Collagen-Mimetic Peptide with Enhanced Hemostatic Activity.

We previously identified that the collagen mimetic peptide (CMP)-E3 is sufficient to induce in vitro blood clotting. In this study, we further demonstrated its ability to induce in vivo blood coagulation through a rat tail bleeding assay. However, intracellular expression of CMP-E3 negatively impacted Escherichia coli growth, causing cell density to fluctuate within an optical density at 600 nm (OD600) range of 1.8-2.2, which limited high-level expression. To address this challenge, we developed a programmed autolysis system by regulating the expression of lysis proteins. We identified Colicin

Impaired Autophagic Flux in Glucose-Deprived Cells: An Outcome of Lysosomal Acidification Failure Exacerbated by Mitophagy Dysfunction.

Autophagy dysfunction is associated with human diseases and conditions including neurodegenerative diseases, metabolic issues, and chronic infections. Additionally, the decline in autophagic activity contributes to tissue and organ dysfunction and aging-related diseases. Several factors, such as down-regulation of autophagy components and activators, oxidative damage, microinflammation, and impaired autophagy flux, are linked to autophagy decline. An autophagy flux impairment (AFI) has been implicated in neurological disorders and in certain other pathological conditions. Here, to enhance our

Mutation of mltG increases peptidoglycan fragment release, cell size, and antibiotic susceptibility in Neisseria gonorrhoeae.

Infection with the Gram-negative species Neisseria gonorrhoeae leads to inflammation that is responsible for the disease symptoms of gonococcal urethritis, cervicitis, and pelvic inflammatory disease. During growth these bacteria release significant amounts of peptidoglycan (PG) fragments which elicit inflammatory responses in the human host. To better understand the mechanisms involved in PG synthesis and breakdown in N. gonorrhoeae, we characterized the effects of mutation of mltG. MltG has been identified in other bacterial species as a terminase that stops PG strand growth by cleaving the

Histologic Study of the Esophagogastric Junction of Organ Donors Reveals Novel Glandular Structures in Normal Esophageal and Gastric Mucosae.

INTRODUCTION: Whether cardiac mucosa at the esophagogastric junction is normal or metaplastic is controversial. Studies attempting to resolve this issue have been limited by the use of superficial pinch biopsies, abnormal esophagi resected typically because of cancer, or autopsy specimens in which tissue autolysis in the stomach obscures histologic findings. METHODS: We performed histologic and immunohistochemical studies of the freshly fixed esophagus and stomach resected from 7 heart-beating, deceased organ donors with no history of esophageal or gastric disease and with minimal or no histo

Vitrification technique for female germinative tissue cryopreservation and banking.

OBJECTIVE: To report on a device designed for the vitrification of germinative tissue, and a systematic vitrification/warming protocol. METHODS: We obtained six fragments of cortical germinative tissue from a human ovary. We randomly chose two fragments and sent them to histological analysis. We vitrified four test samples and stored them for one week in liquid nitrogen (LN), and warmed one week later. We sent the vitrified/warmed fragments to the pathology laboratory, where they analyzed them morphologically under an optical microscope (10-40X). They analyzed the nuclear and cytoplasmic char

The mysterious "mental illness" of a philosopher: the case of Blaise Pascal.

Blaise Pascal (1623-1662) was a French philosopher, who wrote the Pensées, a collection of "thoughts" about the apparent insignificance of human existence. In the last three centuries, it was claimed that his disease was mental. Hysteria, melancholia, and post-traumatic neurosis were taken into consideration, but none of the proposed diagnoses seems to be satisfactory. The aim of our work is to identify Pascal's mysterious illness. We correlated the symptoms of the indirect anamnesis (Pascal's letters to friends, letters and biographies made by his sisters and granddaughter) and autopsy data.

Reduced volumes of the external and internal globus pallidus in male heroin addicts: a postmortem study.

Deep brain stimulation (DBS) of the globus pallidus internus was recently proposed as a potential new treatment target for opioid addiction. DBS requires computer-assisted-3D planning to implant the stimulation electrode precisely. As volumes of brain regions may differ in addiction compared to healthy controls, our aim was to investigate possible volume differences in addicts compared to healthy controls. Volumes of the globus pallidus externus (PE) and internus (PI) in heroin addicts (n = 14) and healthy controls (n = 12) were assessed using morphometry of serial whole-brain sections. Total

Non-Saccharomyces in Wine: Effect Upon Oenococcus oeni and Malolactic Fermentation.

This work is a short review of the interactions between oenological yeasts and lactic acid bacteria (LAB), especially Oenococcus oeni, the main species carrying out the malolactic fermentation (MLF). The emphasis has been placed on non-Saccharomyces effects due to their recent increased interest in winemaking. Those interactions are variable, ranging from inhibitory, to neutral and stimulatory and are mediated by some known compounds, which will be discussed. One phenomena responsible of inhibitory interactions is the media exhaustion by yeasts, and particularly a decrease in L-malic acid by s
