Protocells Featuring Membrane-Bound and Dynamic Membraneless Organelles.

Living cells, especially eukaryotic ones, use multicompartmentalization to regulate intra- and extracellular activities, featuring membrane-bound and membraneless organelles. These structures govern numerous biological and chemical processes spatially and temporally. Synthetic cell models, primarily utilizing lipidic and polymeric vesicles, have been developed to carry out cascade reactions within their compartments. However, these reconstructions often segregate membrane-bound and membraneless organelles, neglecting their collaborative role in cellular regulation. To address this, we propose

New players in actin polymerization--WH2-domain-containing actin nucleators.

Actin nucleators promote the polymerization of the different types of actin arrays formed in a variety of cellular processes, such as cell migration, cellular morphogenesis and membrane trafficking processes. Several novel nucleators have been discovered recently. They all contain Wiskott-Aldrich syndrome protein (WASP) homology 2 (WH2 or W) domains for actin nucleation but seem to employ different molecular mechanisms and serve distinct cellular functions. Here, we summarize what is currently known about the different molecular mechanisms that Spire, Cordon-Bleu and Leiomodin seem to use and,

Design and Standardization of a Mandibular Injury Model for the Study of Craniofacial Tissue Regeneration in the Ambystoma mexicanum Model.

The craniofacial complex is an anatomical region that, in humans, can be affected by developmental malformations, trauma, and pathologies such as cancer, leading to the loss of affected tissues. This creates a need to seek strategies for regenerating these lost tissues. This has promoted the study of cellular and molecular mechanisms that orchestrate the regenerative response of injured craniofacial structures in regenerating species, with Ambystoma mexicanum (A. mexicanum) being a species of great interest. In accordance with the above, a reproducible surgical lesion model was designed and st

Cellular Interaction of Bone Marrow Mesenchymal Stem Cells with Polymer and Hydrogel 3D Microscaffold Templates.

Biomimicking biological niches of healthy tissues or tumors can be achieved by means of artificial microenvironments, where structural and mechanical properties are crucial parameters to promote tissue formation and recreate natural conditions. In this work, three-dimensional (3D) scaffolds based on woodpile structures were fabricated by two-photon polymerization (2PP) of different photosensitive polymers (IP-S and SZ2080) and hydrogels (PEGDA 700) using two different 2PP setups, a commercial one and a customized one. The structures' properties were tuned to study the effect of scaffold dimens

Cholesterol manipulation by West Nile virus perturbs the cellular immune response.

Complex membrane structures induced by West Nile virus (WNV), an enveloped RNA virus, are required for efficient viral replication. How these membranes are induced and how they facilitate the viral life cycle are unknown. We show that WNV modulates host cell cholesterol homeostasis by upregulating cholesterol biosynthesis and redistributing cholesterol to viral replication membranes. Manipulating cholesterol levels and altering concentrations of cellular geranylgeranylated proteins had a deleterious effect on virus replication. Depletion of the key cholesterol-synthesizing enzyme 3-hydroxy-met

Ciliopathies - from rare inherited cystic kidney diseases to basic cellular function.

BACKGROUND: Primary cilia are membrane-bound microtubule-based protuberances of the cell membrane projecting to the extracellular environment. While little attention was paid to this subcellular structure over a long time, recent research has highlighted multiple cellular functions of primary cilia and has brought cilia to the focus of medical and cell biological research. FINDINGS: Cilia are nowadays considered to be crucial cellular structures controlling diverse intracellular signaling cascades. Dysfunction of cilia leads to a pleiotropic group of diseases ranging from cystic kidney diseas

Intracellular crystal formation as a mechanism of cytotoxicity in murine pulmonary Cryptococcus neoformans infection.

Rod-like crystalline structures formed during eosinophilic Cryptococcus neoformans pneumonia in C57BL/6 mice. Crystals were found associated with yeast cells and free in host cell cytoplasm. The crystals apparently formed because of the interaction of a host protein with the cryptococcal polysaccharide. Crystal formation likely contributes to pathogenesis by causing cellular damage.

Glycosaminoglycans in the blood of hereditary multiple exostoses patients: Half reduction of heparan sulfate to chondroitin sulfate ratio and the possible diagnostic application.

Hereditary multiple exostoses (HME) is an autosomal dominant skeletal disorder with wide variation in clinical phenotype and is caused by heterogeneous germline mutations in two of the Ext genes, EXT-1 and EXT-2, which encode ubiquitously expressed glycosyltransferases involved in the polymerization of heparan sulfate (HS) chains. To examine whether the Ext mutation could affect HS structures and amounts in HME patients being heterozygous for the Ext genes, we collected blood from patients and healthy individuals, separated it into plasma and cellular fractions and then isolated glycosaminogly

BRCA1 : BARD1 induces the formation of conjugated ubiquitin structures, dependent on K6 of ubiquitin, in cells during DNA replication and repair.

The N-terminus of the BRCA1 protein bears a RING finger domain that functions as an E3 ubiquitin ligase in vitro where it is able to catalyse the synthesis of monoubiquitin and polyubiquitin targeted proteins. This activity is greatly increased when BRCA1 is in a complex with its N-terminal binding partner BARD1. In this report we use an immunohistochemical approach to demonstrate the association of cellular BRCA1 with the end product of the ubiquitin conjugation and ligation pathway, conjugated ubiquitin. Association is apparent at DNA replication structures in S-phase and following treatment

Modelling kidney cystogenesis using human kidney tubuloid cultures.

BACKGROUND: Renal cyst formation, as observed in autosomal dominant polycystic kidney disease (ADPKD), is a life-threatening condition with no effective cure yet. The molecular mechanisms underlying primary cilia dysfunction, which causes cyst formation and disease development, are not well understood. Human kidney tubuloids offer a promising model system to investigate the disease mechanisms of PKD in physiologically relevant 3D structures. However, their inherent cystic morphology poses a challenge in effectively modelling kidney cystogenesis. Therefore, our study aims to refine the culture

Expansion Microscopy: High-Resolution Fluorescent Imaging with a Conventional Microscope.

Expansion microscopy (ExM) is an innovative super-resolution fluorescence microscopy technique that physically enlarges biological specimens, enabling imaging beyond the diffraction limit using conventional systems such as confocal microscopes. By embedding samples in a swellable hydrogel matrix, ExM allows isotropic expansion of the sample, enhancing spatial resolution without requiring specialized nanoscopy equipment. Advanced variants, such as Ultrastructure Expansion Microscopy (U-ExM), preserve fine cellular details, including organelles and macromolecular assemblies. The observation of s

In vivo uptake of antisense oligonucleotide drugs predicted by ab initio quantum mechanical calculations.

Liver and kidney uptake and antisense activity is studied for a series of Locked Nucleic Acid (LNA) oligonucleotides with fully stereo-defined, internucleoside linkages. These stereo-specific phosphorothioates are made with a newly developed synthesis method and are being analyzed both theoretically and experimentally. Their structures are obtained theoretically by using many-body Schrödinger equations applied to a group of 11 stereo-defined LNA antisense oligonucleotides selected for biological experiments. The fully converged electronic structures were obtained from ab initio quantum calcula

Structural diversity in the membrane-bound hIAPP dimer correlated with distinct membrane disruption mechanisms.

Amyloid deposits of the human islet amyloid polypeptide (hIAPP) have been identified in 90% of patients with type II diabetes. Cellular membranes accelerate the hIAPP fibrillation, and the integrity of membranes is also disrupted at the same time, leading to the apoptosis of β cells in pancreas. The molecular mechanism of hIAPP-induced membrane disruption, especially during the initial membrane disruption stage, has not been well understood yet. Herein, we carried out extensive all-atom molecular dynamics simulations investigating the hIAPP dimerization process in the anionic POPG membrane, to

Gene Expression-Related Changes in Morphologies of Organelles and Cellular Component Organization in Mucopolysaccharidoses.

Mucopolysaccharidoses (MPS) are inherited metabolic diseases characterized by accumulation of incompletely degraded glycosaminoglycans (GAGs) in lysosomes. Although primary causes of these diseases are mutations in genes coding for enzymes involved in lysosomal GAG degradation, it was demonstrated that storage of these complex carbohydrates provokes a cascade of secondary and tertiary changes affecting cellular functions. Potentially, this might lead to appearance of cellular disorders which could not be corrected even if the primary cause of the disease is removed. In this work, we studied ch

Phase Transitions Drive the Formation of Vesicular Stomatitis Virus Replication Compartments.

RNA viruses that replicate in the cell cytoplasm typically concentrate their replication machinery within specialized compartments. This concentration favors enzymatic reactions and shields viral RNA from detection by cytosolic pattern recognition receptors. Nonsegmented negative-strand (NNS) RNA viruses, which include some of the most significant human, animal, and plant pathogens extant, form inclusions that are sites of RNA synthesis and are not circumscribed by a membrane. These inclusions share similarities with cellular protein/RNA structures such as P granules and nucleoli, which are ph

Localization of superoxide dismutases in Alzheimer's disease and Down's syndrome neocortex and hippocampus.

Abnormalities in the cellular regulation and expression of antioxidant enzymes may have a role in mechanisms of central nervous system aging and neurodegeneration. We therefore examined, using isozyme-specific antibodies and immunohistochemistry, the localization of copper, zinc-superoxide dismutase and manganese-superoxide dismutase in the frontal and temporal neocortices and hippocampi of aged controls and individuals with Alzheimer's disease or Down's syndrome. Two different antibodies to copper, zinc-superoxide dismutase and one antibody to manganese-superoxide dismutase were evaluated by

Using stochastic cellular automata to model and define sufficient conditions for the survival of Enterococcus faecalis biofilms with the pCF10 plasmid under erythromycin treatment.

A biofilm is a community of microorganisms adhered to a surface, bound together by extracellular polymeric substances (EPS). They are ubiquitous in nature and develop on a range of surfaces including living tissue. Biofilms themselves typically do not negatively affect their host, but under certain conditions they can retain pathogenic features and cause a wide range of illnesses including persistent or chronic infections. In this study, we look at the bacterium Enterococcus faecalis. E. faecalis is a gram-positive, commensal bacterium commonly found in the human gastrointestinal tract. Genera

Comparing convolutional neural networks and preprocessing techniques for HEp-2 cell classification in immunofluorescence images.

Autoimmune diseases are the third highest cause of mortality in the world, and the identification of an anti-nuclear antibody via an immunofluorescence test for HEp-2 cells is a standard procedure to support diagnosis. In this work, we assess the performance of six preprocessing strategies and five state-of-the-art convolutional neural network architectures for the classification of HEp-2 cells. We also evaluate enhancement methods such as hyperparameter optimization, data augmentation, and fine-tuning training strategies. All experiments were validated using a five-fold cross-validation proce

Tertiary Lymphoid Structures in Human Melanoma: Molecular Mechanisms and Therapeutic Opportunities.

Tertiary lymphoid structures (TLSs) are ectopic lymphoid aggregates often found in chronic inflammatory conditions, including cancer. These structures, which share many cellular and functional features with secondary lymphoid organs, can profoundly influence the tumor microenvironment by promoting local anti-tumor immune activation. TLSs have been observed in various cancers, including melanoma, and are associated with improved responses to immunotherapy and clinical outcomes. However, our understanding of the molecular mechanisms underlying TLS formation and function remains incomplete. This
