The gut microbial metabolite phenylacetylglutamine exacerbates severe acute pancreatitis by promoting ferroptosis and TLR4/NF-κB signaling pathways.

Severe acute pancreatitis (SAP) is a life-threatening abdominal disease. In recent years, multiple studies have suggested that gut microbiota metabolites play a key role in regulating SAP disease outcomes. The gut microbial metabolite phenylacetylglutamine (PAGln) exerts important effects in inflammation and oxidative stress-induced injury; however, its role in SAP remains unexplored. In this study, caerulein and LPS were used to induce experimental SAP models in mice and AR42J cells, followed by histopathological, biochemical, and molecular biological analyses. SAP induced hemorrhagic necrosi

Rutin protects the pancreas from inflammatory injury and oncogene-driven tumorigenesis by inhibiting acinar to ductal metaplasia.

Rutin is a valuable traditional Chinese medicine known for its anti-inflammatory and anticancer effects. It has been shown to be effective in treating various inflammation-associated diseases. Here, we investigated the influence of rutin on acute pancreatitis and tumorigenesis. Using C57BL/6J mice and Kras mutant transgenic mice, we induced pancreatitis and acinar regeneration models. Pancreatic malondialdehyde (MDA), superoxide dismutase (SOD) activity and reduced glutathione (GSH) contents were measured for oxidative stress. Histological staining and a pancreatic acinar 3D culture model were

HMG Box-containing Protein 1 (HBP1) Protects Against Pancreatic Injury in Acute Pancreatitis but Promotes Neoplastic Progression.

BACKGROUND & AIMS: Pancreatitis is an inflammatory disease of the exocrine pancreas and a known risk factor for pancreatic ductal adenocarcinoma (PDAC). Previously, we identified HMG-box transcription factor 1 (HBP1) as a potential master transcription factor (TF) in the early progression of PDAC, with its expression associated with poor patient survival, underscoring its significance in pancreatic disease. However, the functional role of HBP1 in the onset and progression of acute pancreatitis (AP) remains unknown. METHODS: We examined HBP1 expression in human pancreatitis samples and a cerul

Mouse Models for Pancreatic Ductal Adenocarcinoma are Affected by the cre-driver Used to Promote KRASG12D Activation.

BACKGROUND & AIMS: The fundamental biology of pancreatic ductal adenocarcinoma has been greatly impacted by the characterization of genetically engineered mouse models that allow temporal and spatial activation of oncogenic KRAS (KRASG12D). One of the most commonly used models involves targeted insertion of a cre-recombinase into the Ptf1a gene. However, this approach disrupts the Ptf1a gene, resulting in haploinsufficiency that likely affects sensitivity to oncogenic KRAS (KRASG12D). This study aims to determine if Ptf1a haploinsufficiency affected the acinar cell response to KRASG12D before

Transfection of 12/15-lipoxygenase effectively alleviates inflammatory responses during experimental acute pancreatitis.

BACKGROUND: Acute pancreatitis (AP), the initially triggered inflammatory process in the pancreas, can be life-threatening. It has been reported that 15-lipoxygenase may promote the removal of damaged intracellular components, maintain intracellular homeostasis, and promote apoptosis by upregulating the activity of caspases. Despite an increased understanding of the lipoxygenase pathway in inflammation and immune diseases, the role of the Alox15 gene product in modulating the inflammatory changes during AP is not well defined. AIM: To investigate the effect of Alox15 expression in cerulein-in

Inhibition of Rb phosphorylation leads to H2S-mediated inhibition of NF-kB in acute pancreatitis and associated lung injury in mice.

BACKGROUND: Acute pancreatitis (AP), an inflammatory condition of pancreas, destructs the exocrine cells by releasing various pro-inflammatory cytokines that activates the stellate cells. However, the underlying molecular mechanism remains unclear. The present study investigated the role of retinoblastoma (Rb), hydrogen sulphide and nuclear factor-κB (NF-κB) in the regulation of exocrine cell proliferation under inflammatory condition. METHODS: The randomly grouped male swiss mice were administered with 6 consecutive hourly i.p injections of caerulein to induce AP. Palbociclib (PD) (25 mg/kg

The proteinase/proteinase-activated receptor-2/transient receptor potential vanilloid-1 cascade impacts pancreatic pain in mice.

AIMS: Proteinase-activated receptor-2 (PAR2) and transient receptor potential vanilloid-1 (TRPV1) are co-localized in the primary afferents, and the trans-activation of TRPV1 by PAR2 activation is involved in processing of somatic pain. Given evidence for contribution of PAR2 to pancreatic pain, the present study aimed at clarifying the involvement of TRPV1 in processing of pancreatic pain by the proteinase/PAR2 pathway in mice. MAIN METHODS: Acute pancreatitis was created by repeated administration of cerulein in conscious mice, and the referred allodynia/hyperalgesia was assessed using von

Disulfiram Mitigates Acinar Cell Damage in the Caerulein-Induced Acute Pancreatitis Model through Inhibition of the NLRP3/Caspase-1/GSDMD Pathway.

Acute pancreatitis (AP) is marked by severe inflammatory injury to pancreatic acinar cells, with pyroptosis playing a critical role in its progression. Disulfiram (DSF), traditionally prescribed for alcohol dependence, has recently attracted attention for its anti-inflammatory potential. In this study, caerulein-stimulated AR42J cells were used to model AP in vitro and to evaluate whether DSF could alleviate cellular injury and inflammatory responses. Treatment with DSF improved cell viability, suppressed lactate dehydrogenase release, and significantly reduced pro-inflammatory cytokines IL-1β

Inhibition of a Disintegrin and Metalloproteinase With Thrombospondin Type 1 Motif, Member 13 Activity by Peptidylarginine Deiminase IV Exacerbates Pancreatic Necrosis in Acute Pancreatitis.

BACKGROUND & AIMS: Dysregulation of the von Willebrand factor and its protease, a disintegrin and metalloproteinase with thrombospondin type 1 motif, member 13, has been implicated in the pathogenesis of pancreatic necrosis during acute pancreatitis, but the underlying mechanisms remain incompletely understood. NETosis and their enzyme component, peptidylarginine deiminase type IV, have been proposed to inhibit a disintegrin and metalloproteinase with thrombospondin type 1 motif, member 13. We aimed to determine whether peptidylarginine deiminase type IV-mediated inhibition of a disintegrin an

Irisin-mediated KEAP1 degradation alleviates oxidative stress and ameliorates pancreatitis.

Oxidative stress (OS) injury is pivotal in acute pancreatitis (AP) pathogenesis, contributing to inflammatory cascades. Irisin, a ubiquitous cytokine, exhibits antioxidant properties. However, the role of irisin in AP remains inconclusive. Our study aims to elucidate irisin expression in AP patients and investigate its mechanism of action to propose a novel treatment strategy for AP. Serum irisin levels in 65 AP patients were quantified using an enzyme-linked immunosorbent assay and correlated with disease severity scores. Core genes implicated in AP-related oxidative stress were identified an

Hydrostatic pressure induces mitochondrial oxidative stress and mtDNA-mediated cGAS-STING activation in acute pancreatitis.

Increased mechanical pressure is a well-recognized feature of acute pancreatitis (AP), but its pathological mechanisms remain elusive. Although previous studies have emphasized shear stress-induced pancreatic injury, the effects of static hydrostatic pressure have been underappreciated. Mitochondria act as mechanosensitive organelles, and mechanical stimuli can induce mitochondrial oxidative stress. Release of mitochondrial DNA (mtDNA) could trigger activation of the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway in immune cells. However, whether increased hydros

p8 Deficiency causes siderosis in spleens and lymphocyte apoptosis in acute pancreatitis.

OBJECTIVES: The gene p8 was initially described in pancreatic tissue during acute experimental pancreatitis, a disease that is characterized by a systemic immune response. Although early reports suggested that p8 affects leukocyte migration during acute pancreatitis (AP), no studies revealing its immune-modulatory effects have been performed. METHODS: We investigated the composition of the cellular immune system in naive p8 knockout (p8(−/−)) mice and compared with matched wild-type mice during pancreatitis. RESULTS: In young mice, there were no relevant differences in the composition of per

Adult pancreatic acinar cells dedifferentiate to an embryonic progenitor phenotype with concomitant activation of a senescence programme that is present in chronic pancreatitis.

OBJECTIVE: Acinar cells display plasticity in vitro and in vivo and can activate a variety of differentiation programmes that may contribute to pancreatic diseases. The aims were to determine: (1) the differentiation potential of acinar cells under conditions which favour stem cell survival, and (2) its relationship to the phenotypes acquired by pancreatic epithelial cells in chronic pancreatitis. DESIGN: Murine acinar cells were cultured in suspension and their molecular phenotype was characterised by qRT-PCR, chromatin immunoprecipitation, immunocytochemistry and global transcriptome analys

Inflammation-Induced Claudin-2 Up-Regulation Limits Pancreatitis Development by Enhancing Pancreatic Ductal Transport.

BACKGROUND & AIMS: Pancreatitis is influenced by environmental and genetic factors. Genome-wide association studies identified polymorphisms at the X-linked CLDN2 locus as risk factors for chronic pancreatitis. CLDN2 encodes claudin-2 (CLDN2), a paracellular cation-selective channel at tight junctions. However, its role in pancreatitis susceptibility remains unclear. We aimed to determine the role of CLDN2 in pancreatitis onset and progression. METHODS: Human pancreatic tissue, pancreatitis mouse models, pancreatic ductal epithelial organoids, and isolated mouse pancreatic ducts were used. CL

Repeated photobiological regulation therapy alleviates inflammation in mice with experimental acute pancreatitis through ROS/NF-κB pathway.

The prevalence of acute pancreatitis (AP) is increasing, but current treatments remain inadequate. Photobiomodulation (PBM) therapy has demonstrated anti-inflammatory effects under various conditions; however, its role in AP has not been explored. Evaluating this could introduce a noninvasive treatment option for AP. This study aimed to assess PBM’s potential of PBM in reducing cerulein-induced AP inflammation and elucidate the underlying mechanisms. Animal Experiment: Mice were categorized into control, model, and experimental groups. AP was induced with cerulein injections in depilated mice,

Repurposing BCL2 inhibitors: Venetoclax protects against acinar cell necrosis in acute pancreatitis by promoting apoptosis.

Acute pancreatitis (AP), a severe inflammatory disorder of the pancreas, lacks effective pharmacological treatment. The disease is primarily driven by necrosis of pancreatic acinar cells (PACs), which intensifies inflammation and organ injury. This study explores the potential of BCL2 inhibitors, specifically Navitoclax and Venetoclax, to shift cell death pathways from necrosis to apoptosis and thereby mitigate disease severity. Ex vivo models using cerulein or ethanol/palmitoleic acid (EtOH/POA) showed that both inhibitors significantly reduced necrosis, increased apoptosis, and improved PAC
