Immunogenicity and Efficacy of a Trivalent HSV-2 gC2, gD2, gE2 Nucleoside-Modified mRNA-LNP Vaccine Against HSV-1 Eye Infection and Neuroinvasion in Mice.

BACKGROUND/OBJECTIVES: Eye infection with herpes simplex virus type 1 (HSV-1) can result in keratitis, a leading cause of corneal blindness. We evaluated whether an experimental vaccine containing HSV-2 immunogens to prevent genital herpes also protects against HSV-1 eye infection and neuroinvasion. METHODS: Mice were immunized twice, one month apart, with PBS or a nucleoside-modified lipid nanoparticle vaccine containing mRNA encoding for gC2, gD2, and gE2. One month later, 106 plaque forming units (PFU) (10 lethal dose 50, LD50) of the HSV-1 McKrae strain were added to the intact cornea of

Necrotizing Scleritis, Conjunctivitis, and Other Pathologic Findings in the Left Eye and Brain of an Ebola Virus-Infected Rhesus Macaque (Macaca mulatta) With Apparent Recovery and a Delayed Time of Death.

A 3.5-year-old adult female rhesus macaque (Macaca mulatta) manifested swelling of the left upper eyelid and conjunctiva and a decline in clinical condition 18 days following intramuscular challenge with Ebola virus (EBOV; Kikwit-1995), after apparent clinical recovery. Histologic lesions with strong EBOV antigen staining were noted in the left eye (scleritis, conjunctivitis, and peri-optic neuritis), brain (choriomeningoencephalitis), stomach, proximal duodenum, and pancreas. Spleen, liver, and adrenal glands, common targets for acute infection, appeared histologically normal with no evidence

An ocular infection model using suckling hamsters inoculated with equine herpesvirus 9 (EHV-9): kinetics of the virus and time-course pathogenesis of EHV-9-induced encephalitis via the eyes.

By using a new member of the neurotropic equine herpesviruses, EHV-9, which induced encephalitis in various species via various routes, an ocular infection model was developed in suckling hamsters. The suckling hamsters were inoculated with EHV-9 via the conjunctival route and were sacrificed after 6, 12, 24, 36, 48, 72, 96, 120, and 144 hours (h) post inoculation (PI). Three horizontal sections of the brains, including the eyes and cranial cavity, were examined histologically to assess the viral kinetics and time-course neuropathological alterations using a panoramic view. At 6 to 24 h PI, th

CCL25 chemokine promotes antiviral tissue resident CD4+ and CD8+ effector memory TRM cells associated with a reduction of ocular herpes infection and disease: a potential gut-eye axis in herpes immunity.

CCL25, a mucosal chemokine mainly produced in the gut, is known for its role in intestinal immunity through CCR9. In this study, we tested whether CCL25 also contributes to ocular immunity within the gut-eye axis during herpes simplex virus type 1 (HSV-1) infection. Using CCL25-deficient (CCL25(-/-) deficient) mice and wild-type C57BL/6J controls, which were infected ocularly with HSV-1 (McKrae strain), we found that CCL25 deficiency led to more severe corneal disease, increased viral replication, and reduced survival. CCL25(-/-) deficient mice also had fewer antiviral effector memory CD44+CD4

Characterization of the Life Cycle of a Fish Eye Fluke, Austrodiplostomum ostrowskiae (Digenea: Diplostomidae), with Notes on Two Other Diplostomids Infecting Biomphalaria havanensis (Mollusca: Planorbidae) from Catfish Aquaculture Ponds in Mississippi, USA.

Ocular diplostomiasis is caused by trematode species in the family Diplostomidae, specifically those in the genera Austrodiplostomum, Diplostomum, and Tylodelphys. Diplostomid trematodes are globally distributed parasites of fish. Heavy infections of diplostomids that parasitize the eyes of fish can result in acute mortality while chronic infections are often characterized by impaired vision or blindness. In the southeastern United States, commercial catfish production is threatened by piscivorous birds and the many trematode species that parasitize them. The life cycles typically involve a pi

Ocular infections in international travelers.

INTRODUCTION: Ophthalmological conditions in international travelers may be associated with low mortality but high morbidity. Eye involvement in travelers is less frequently reported than febrile, gastrointestinal and respiratory infections, but data probably represent a degree of under-notification. METHODS: an extensive narrative review of the main viral, bacterial, fungal and parasitic infections affecting the eye in travelers was performed. MAIN FINDINGS: Common respiratory tract viral infections may cause ocular complications in travelers, human influenza viruses have been associated wi

[Transmission of conventional agents during outpatient care].

Outpatient care bring together in a single location a large number of patients potentially bearing conventional infectious agents (virus, bacteria, fungi, parasites). This report details the germs potentially involved. We emphasize adenovirus epidemic keratoconjunctivitis, which is particularly prevalent in ophthalmology outpatient care. We analyze the modes of contamination and the means to prevent iatrogenic infections.

Common ocular infections. A prescriber's guide.

While most ocular infections are benign, others can be associated with devastating visual consequences. Most patients present with either ocular discharge, visual symptoms or a red or painful eye. The primary care physician is usually the first to evaluate these patients. We have separated ocular infections into 3 groups. Infections affecting the cornea and conjunctiva often present with eye pain and a red eye; noninfectious aetiologies can have a similar presentation. Infections inside the eye (endophthalmitis) often have devastating consequences. They usually occur following penetrating ocul

Endogenous Panophthalmitis in a case of Multiple Myeloma and Diabetes Mellitus.

Multiple myeloma cripples the human body in many ways, one of them being decreased immunity. Infections occurring spontaneously can increase the morbidity. We report a case of an elderly lady with multiple myeloma on treatment and uncontrolled diabetes, who developed loss of vision, swelling and redness of left eye of 4 days duration. There was no history of injury or entry of a foreign body. She also had left arm cellulitis. Ocular examination revealed visual acuity of 6/36 in right eye and no perception of light in left eye. Anterior segment of the right eye was insignificant while the left

Fluoroquinolones for the eye.

Two fluoroquinolone antibiotics, ciprofloxacin (Ciloxan-Alcon) and ofloxacin (Exocin-Allergan), are available in the UK as topical preparations for the eye. Ciprofloxacin eyedrops are licensed for the treatment of "corneal ulcers and superficial infections of the eye", and ofloxacin eyedrops for "external ocular infections (such as conjunctivitis and keratoconjunctivitis)". In the UK chloramphenicol is commonly recommended as the drug of first choice for the treatment of superficial bacterial eye infections. Do fluoroquinolone preparations offer any advantage over this or other established dru

Intraocular delivery of anti-infective drugs-bacterial, viral, fungal and parasitic.

This article reviews the current literature about the use of intraocular drugs for treatment of bacterial, fungal, viral and protozoal intraocular infection. This route of administration has the advantage of delivering antimicrobials directly into the eye achieving a high therapeutic concentration above the MIC(90) of most pathogens and with minimal systemic side effects. It is usually well tolerated but carries a small risk of intraocular complications. Intraocular therapy is now the main therapeutic approach for treatment of bacterial endophthalmitis and has an important adjunctive role in t

Ocular manifestations of systemic infection.

Increased research and awareness of various systemic infections places a greater emphasis on the ophthalmologist's knowledge of ocular manifestations of these diseases. New advances in the diagnosis and treatment, as well as studies of the pathogenesis and histological features of different infectious processes are continually being reported. Recent publications focusing on ophthalmic findings of infectious diseases are reviewed. This article discusses new reports on herpes simplex, herpes zoster, Lyme disease, malaria, onchocerciasis, cysticercosis, and toxocariasis.

Human immunodeficiency virus and opportunistic ocular infections.

Opportunistic ocular infections are a common and important complication of the acquired immunodeficiency syndrome. Cytomegalovirus retinitis is by far the most frequent infection, but other viral, bacterial, and parasitic infections of the posterior segment of the eye may be seen. Infections of the anterior segment are less common but may cause visual loss if not treated. Prompt recognition of these diseases is essential because ocular involvement may be an early sign of systemic dissemination.

The pathogenesis of herpetic ocular disease in the guinea pig.

A detailed study of the pathogenesis of herpetic eye disease in the guinea pig was undertaken to further develop this animal model. Several well-known HSV-1 strains were tested for their ability to produce disease and cause acute and latent infections of the trigeminal ganglion: McKrae, KOS, McIntyre, RE, and Shealey. Two HSV-2 strains failed to cause eye infections. The Shealey strain [HSV-1 (Sh)] produced the most severe eye infections, characterized by epithelial and stromal disease, corneal vascularization and ulcerative blepharitis. Consequently, HSV-1 (Sh) was selected as the prototype s

Acute ocular infection by type 2 herpes simplex virus in adults.

Acute ocular infections in two adults were caused by type 2 herpes simplex virus (HSV) ("genital herpesvirus"). One patient had an acute blepharoconjunctivitis, the other an acute keratoconjunctivitis. Genital infections had preceded the eye infections, and type 2 HSV was isolated from the eyes of both patients and from the genital lesions of one patient. This strongly suggests transmission of type 2 HSV from the genital site to the eye.

An Eye on Staphylococcus aureus Toxins: Roles in Ocular Damage and Inflammation.

Staphylococcus aureus (S. aureus) is a common pathogen of the eye, capable of infecting external tissues such as the tear duct, conjunctiva, and the cornea, as well the inner and more delicate anterior and posterior chambers. S. aureus produces numerous toxins and enzymes capable of causing profound damage to tissues and organs, as well as modulating the immune response to these infections. Unfortunately, in the context of ocular infections, this can mean blindness for the patient. The role of α-toxin in corneal infection (keratitis) and infection of the interior of the eye (endophthalmitis) h
