Antidepressant-like effects of alarin produced by activation of TrkB receptor signaling pathways in chronic stress mice.

Alarin is a newly identified member of the galanin family of neuropeptides. Until now, research on alarin is limited compared with other members of the galanin family. Unearthing the new biological effects of alarin and its unidentified receptor(s) interests us. We previously showed that alarin has an effect on depression-like behaviors, although the underlying mechanisms are not fully clarified. The present study verified the antidepressant-like effects of alarin using the unpredictable chronic mild stresses (UCMS) paradigm, and explored the mechanism that underlies antidepressant-like effect

Neuropeptides in human salivary (submandibular and parotid) glands.

The existence, distribution and density of various neuropeptides in human submandibular and parotid glands were investigated using immunocytochemistry and radioimmunoassay. Numerous nerve fibers containing vasoactive intestinal polypeptide (VIP) and peptide histidine methionine (PHM), or neuropeptide Y (NPY) and C-flanking peptide of NPY (CPON) immunoreactivities (ir) were found in close association to acini, ducts and blood vessels. Only few calcitonin gene-related peptide (CGRP)- and substance P (SP)-ir nerve fibers could be demonstrated, mainly localized around blood vessels and ducts. Gala

Chronic treatment with carboplatin induces mechanical hypersensitivity and neurochemical changes in the mouse dorsal root ganglia and spinal cord.

PURPOSE: Carboplatin is a widely used antineoplastic drug, but it is associated with severe adverse effects, including peripheral neuropathy. However, the neurobiological mechanisms underlying this side effect are not fully known. Thus, we determined neurochemical changes in the lumbar dorsal root ganglia (DRG) and spinal cord following carboplatin treatment. METHODS: Male BALB/c (23 weeks-old) mice received carboplatin (i.p.; 60 mg/kg/week for 4 weeks) or vehicle. Hindpaw mechanical sensitivity was evaluated at baseline and after carboplatin administration. On day 30 post-first carboplatin a

Critical role of somatostatin receptor 2 in the vulnerability of the central noradrenergic system: new aspects on Alzheimer's disease.

Alzheimer's disease and other age-related neurodegenerative disorders are associated with deterioration of the noradrenergic locus coeruleus (LC), a probable trigger for mood and memory dysfunction. LC noradrenergic neurons exhibit particularly high levels of somatostatin binding sites. This is noteworthy since cortical and hypothalamic somatostatin content is reduced in neurodegenerative pathologies. Yet a possible role of a somatostatin signal deficit in the maintenance of noradrenergic projections remains unknown. Here, we deployed tissue microarrays, immunohistochemistry, quantitative morp

A molecular study of hippocampus in dogs with convulsion during canine distemper virus encephalitis.

In this study, we have investigated the expression of the nuclear transcription factor (c-Fos, NFkB), growth factors (nerve growth factor--NGF, brain-derived neurotrophic factor--BDNF), peptides (enkephalin, galanin) and glutamate transporter (AA 504-523 rat EAAC1) in 6 dogs sacrificed immediately after seizure attack during encephalomyelitis due to canine distemper virus (CDV) (as assessed by clinical examination, RT-PCR and viral RNA detection either in blood or brain tissue and CDV immunohistochemistry in brain slices). In all these CDV affected dogs, the observed neurological signs include

Leptin and the regulation of food intake, energy homeostasis and immunity with special focus on periparturient ruminants.

The biology of leptin has been studied most extensively in rodents and in humans. Leptin is involved in the regulation of food intake, energy homeostasis and immunity. Leptin is primarily produced in white adipose tissue and acts via a family of membrane bound receptors, including an isoform with a long intracellular domain (OB-Rb), and many isoforms with short intracellular domains (Ob-Rs). OB-Rb is predominantly expressed in the hypothalamic regions involved in the regulation of food intake and energy homeostasis. The other isoforms are distributed ubiquitously and are found in most peripher

Neuropeptide systems in temporal lobe epilepsy.

Neuropeptides commonly function as co-transmitters alongside classical neurotransmitters within the same neuron. In temporal lobe epilepsy (TLE), the expression of numerous neuropeptides in the brain is profoundly altered. These changes may reflect activity-dependent regulation of the respective neuronal populations-often associated with enhanced neuronal firing and co-release of neuropeptides with classical transmitters-but may also exert direct functional effects through activation of peptide-specific receptors. This review summarizes alterations in neuropeptide systems in experimental model

Neuropeptide levels in murine liver and biliary pathways.

Neuropeptide concentrations in the liver, gallbladder and bile duct of 30 male mice were determined by radioimmunoassay of tissue extracts. Vasoactive intestinal peptide (VIP), substance P, somatostatin, neurotensin, neuropeptide Y (NPY), galanin, gastrin-releasing peptide (GRP) and enkephalin were found in extracts of liver, gallbladder and bile duct. The level of enkephalin was low in all tissues investigated. The most predominant neuropeptide in liver and bile duct was VIP and in the gallbladder, NPY. The function of these neuropeptides in the liver and biliary pathways is unclear. One can

Effects of CYP-induced cystitis on PACAP/VIP and receptor expression in micturition pathways and bladder function in mice with overexpression of NGF in urothelium.

We have previously demonstrated nerve growth factor (NGF) regulation of pituitary adenylate cyclase-activating polypeptide (PACAP)/receptors in bladder reflex pathways using a transgenic mouse model of chronic NGF overexpression in the bladder using the urothelial-specific uroplakin II promoter. We have now explored the contribution of target-derived NGF in combination with cyclophosphamide (CYP)-induced cystitis to determine whether additional changes in neuropeptides/receptors are observed in micturition reflex pathways due to the presence of additional inflammatory mediators in the urinary

Immunohistochemical evaluation of a malignant phecochromocytoma in a wolfdog.

A malignant pheochromocytoma with multiple metastases was diagnosed in a 7-year-old male wolfdog that resulted from a cross between an eastern timber wolf (Canis lupus lycaon) and an Alaskan malamute. A yellowish white neoplastic mass approximately 10 cm diameter was found in the right adrenal gland. The neoplasm penetrated through the wall of the caudal vena cava. A diagnosis of pheochromocytoma was established by histopathologic and immunohistochemical procedures. Immunohistochemically, the neoplastic cells expressed chromogranin A, substance P, synaptophysin, Leu-7, protein gene product 9.5

Effects of Galp and alarin peptides on HPA axis gene expression and adrenal function: In vivo experiments.

BACKGROUND: Many experimental data indicate interactions between peptides involved in the control of food intake, energy homeostasis and adrenocortical hormone release. Glucocorticoids stimulate or inhibit the secretion of orexigenic and anorexigenic peptides, which in turn are involved in the regulation of adrenal growth, structure and function. Galanin-like peptide (Galp) and alarin (Ala) are involved in the regulation of food intake. Galp and Ala mRNAs have already been shown to be present in the arcuate nucleus (ARC) of the hypothalamus in both rats and mice. OBJECTIVES: To investigate th

Neuroendocrine Determinants of Polycystic Ovary Syndrome.

Polycystic ovary syndrome (PCOS) is the most common endocrine disorder in women and a major cause of anovulatory infertility. A diagnosis of PCOS is established based the presence of two out of three clinical symptoms, which are criteria accepted by the ESHRE/ASRM (European Society of Human Reproduction and Embryology/American Society for Reproductive Medicine). Gonadotropin-releasing hormone (GnRH) is responsible for the release of luteinizing hormone, and follicle stimulating hormone from the pituitary and contributes a leading role in controlling reproductive function in humans. The goal of

Neuropeptide messenger plasticity in the CNS neurons following axotomy.

Neuronal peptides exert neurohormonal and neurotransmitter (neuromodulator) functions in the central nervous system (CNS). Besides these functions, a group of neuropeptides may have a capacity to create cell proliferation, growth, and survival. Axotomy induces transient (1-21 d) upregulation of synthesis and gene expression of neuropeptides, such as galanin, corticotropin releasing factor, dynorphin, calcitonin gene-related peptide, vasoactive intestinal polypeptide, cholecystokinin, angiotensin II, and neuropeptide Y. These neuropeptides are colocalized with "classic" neurotransmitters (acety

A new pharmacological strategy against treatment-resistant depression.

Major depressive disorder affects more than 50 million people in the world. However, 50% of patients don't respond to two or more drugs or psychotherapeutic treatments, named treatment-resistant depression (TRD). In this work, we propose a new augmentation treatment against TRD based on combining Fluoxetine (FLX) and the N-terminal fragment Galanin, GAL(1-15). In Wistar Kyoto (WKY) rats, akin to endogenous depression genetically, we evaluate GAL(1-15)'s impact on FLX-induced behaviours on tests measuring despair and anhedonia. We explored GALR2 involvement using the antagonist M871 and an in v

Neuropeptides in the human penis: an immunohistochemical study.

In the present study, the distribution of neuropeptides in the human penis is demonstrated by immunohistochemistry (IHC). IHC screening detected a complex network of nerve fibers containing vasoactive intestinal polypeptide (VIP), peptide histidine-methionine (PHM), prepro-VIP (111-122), neuropeptide Y (NPY), C-flanking peptide of NPY (C-PON), calcitonin gene-related peptide, substance P, and galanin immunoreactivities. Special attention was also given to the recently isolated, VIP-related lizard peptide helospectin, which could also be detected in neuronal elements in the penis. Colocalizatio

[Premature ejaculation: the main physiological mechanisms and new approaches to the treatment].

Premature ejaculation (PE) is characterized by an early ejaculation, which occurs within 1 minute after or before vaginal penetration. PE is the most common sexual problem and leads to a number of negative psychological consequences. To understand the etiopathogenesis of PE, it is necessary to understand the physiological mechanisms of the ejaculatory reflex. A group of galanine-ergic lumbar spinothalamic cells (LSt cells) with their numerous projections to various structures of the central nervous system plays a crucial role. The main approaches to the treatment of PE are based on the inhibit

Inhibition of insulin secretion: a fail-safe system.

Insulin secretion from the beta-cells of the endocrine pancreas is subject to a pot-pourri of stimulatory, modulatory and inhibitory influences. beta-Cell secretion is reduced or blocked by a variety of inhibitors (including galanin, somatostatin and noradrenaline) which reach the cells either via the islet vascular system or are released locally from sympathetic and peptidergic nerves terminating in the pancreas. It is now becoming clear that among these many inhibitors, several have multiple mechanisms by which they inhibit release at the cellular level. Indeed, with multiple inhibitors (som
