The white-footed mouse (Peromyscus leucopus) has expanded its northern limit into southern Québec over the last few decades. P. leucopus is a great disperser and colonizer and is of particular interest because it is considered a primary reservoir for the spirochete bacterium that causes Lyme disease. There is no current information on the gene flow between mouse populations on the mountains and forest fragments found scattered throughout the Montérégie region in southern Québec, and whether various landscape barriers have an effect on their dispersal. We conducted a population genetics analysi

Medical image computing and computer-assisted intervention : MICCAI ... International Conference on Medical Image Computing and Computer-Assisted Intervention2012eng
With the advent of advanced imaging techniques, genotyping, and methods to assess clinical and biological progression, there is a growing need for a unified framework that could exploit information available from multiple sources to aid diagnosis and the identification of early signs of Alzheimer's disease (AD). We propose a modeling strategy using supervised feature extraction to optimally combine high-dimensional imaging modalities with several other low-dimensional disease risk factors. The motivation is to discover new imaging biomarkers and use them in conjunction with other known biomark

Between the 16th and 18th centuries, Aedes aegypti (Diptera: Culicidae), a mosquito native to Africa, invaded the Americas, where it was successively responsible for the emergence of yellow fever (YF) and dengue (DEN). The species was eradicated from numerous American countries in the mid-20th century, but re-invaded them in the 1970s and 1980s. Little is known about the precise identities of Ae. aegypti populations which successively thrived in South America, or their relation with the epidemiological changes in patterns of YF and DEN. We examined these questions in Bolivia, where Ae. aegypti

Human African trypanosomiasis (HAT) or sleeping sickness is a major public health problem in sub-Saharan Africa and is due to the kinetoplastid parasite Trypanosoma brucei gambiense in West and Central Africa. The exact role of multiple infections, the basis of clinical diversity observed in patients and the determinism that leads trypanosomes into different body fluids of the host remain opened questions to date. In this paper we investigate, in three Guinean foci, whether strains found in blood, lymph or cerebrospinal fluid (CSF) or in patients at different phase of HAT (phase 1, early phase

Despite the importance of population structure for the epidemiology of pathogenic bacteria, the spatial and ecological heterogeneity of these populations is often poorly characterized. Here, we investigated the genetic diversity and population structure of the Lyme borreliosis (LB) spirochaete Borrelia garinii in its marine cycle involving colonial seabirds and different host races of the seabird tick Ixodes uriae. Multilocus sequence analyses (MLSA) on eight chromosomal and two plasmid loci (ospA and ospC) indicate that B. garinii circulating in the marine system is highly diverse. Microevolu

Although Vibrio cholerae is an important human pathogen, little is known about its populations in regions where the organism is endemic but where cholera disease is rare. A total of 31 independent isolates confirmed as V. cholerae were collected from water, sediment, and oysters in 2008 and 2009 from the Great Bay Estuary (GBE) in New Hampshire, a location where the organism has never been detected. Environmental analyses suggested that abundance correlates most strongly with rainfall events, as determined from data averaged over several days prior to collection. Phenotyping, genotyping, and m

Plant transformation protocols generally involve the use of selectable marker genes for the screening of transgenic material. The bacterial gene nptII, coding for a neomycin phosphotransferase, and the hpt gene, coding for a hygromycin phosphotransferase, are frequently used. These enzymes detoxify aminoglycoside antibiotics by phosphorylation, thereby permitting cell growth in the presence of antibiotics. Nevertheless, the screening for transgenic regenerated shoots is often partial and difficult due to regeneration of escapes and chimeras. These difficulties can be caused, in part, by an inc

Familial Mediterranean Fever (FMF) is an autosomal, recessively inherited disease, characterized by recurrent and short attacks of fever with serosal inflammation that are caused by mutations in MEFV gene that encodes pyrin protein. To date, more than 70 disease-associated mutations have been identified, almost all of them representing missense nucleotide changes. FMF is very common among patients with Mediterranean ancestry, although the exact prevalence is not yet known, Greeks are considered to be at 'intermediate risk'. In the present study, we studied FMF patients in natives of Crete, a p

Genetic diversity and population structure of seven populations of Schistosoma mansoni sampled in Kenya were assessed using five microsatellite markers. The mean number of alleles per locus, expected heterozygosity in Hardy-Weinberg equilibrium and pairwise F(ST) values ranged from 5.2 to 10.7, 0.5-0.8 and 3.6-27.3%, respectively. These data reveal that S. mansoni populations in Kenyan have relatively high levels of genetic diversity and is significantly differentiated. Our data combined with information on biogeography support the hypothesis that the strong genetic structure in Kenyan schisto

In Vietnam, dengue hemorrhagic fever has been detected since the 1950s. In Southeast Asia, urban centers expanded rapidly in an uncontrolled and unplanned way. The Aedes aegypti populations and dengue viruses thrived in these new ecological and demographic settings. The result of these changes was a greatly extended geographic distribution, increased densities of Ae. aegypti and the maintenance of the four dengue serotypes leading to a dramatic increase in dengue transmission. To assess the role of the vector in the changing pattern of the disease in Southeast Asia, we studied the ecology of A

Data on comparative molecular genetic analysis of pFra plasmids from plague bacillus belonging to either two biovars (antiqua and orientalis) are presented. It was established that during evolution these replicons were rearranged, which resulted in the differences between pFra plasmids of plague bacillus of antiqua biovar (Yersinia pestis 231 and Y. pestis 358/12) and that of orientalis biovar (Y. pestis A1122 and Y. pestis EV). These included inversions of two plasmid DNA regions and size differences of 3 kb.

The isoenzymes of the 3 beta-hydroxysteroid dehydrogenase/5-ene-4-ene-isomerase (3 beta-HSD) gene family catalyse the transformation of all 5-ene-3 beta-hydroxysteroids into the corresponding 4-ene-3-keto-steroids and are responsible for the interconversion of 3 beta-hydroxy- and 3-keto-5 alpha-androstane steroids. The two human 3 beta-HSD genes and the three related pseudogenes are located on the chromosome 1p13.1 region, close to the centromeric marker D1Z5. The 3 beta-HSD isoenzymes prefer NAD+ to NADP+ as cofactor with the exception of the rat liver type III and mouse kidney type IV, which

Hb M Hyde Park disease was detected in a girl who for several years was thought to have cyanotic heart disease. The problems of recognizing the condition are outlined and clues to diagnosis are discussed. Evidence for heme loss from the aberrant beta chains of Hb M Hyde Park and production of an unstable molecule is presented. The normal haematological findings in the patient's parents, as well as their blood groups and isozymes, suggest that the occurrence of her Hb M Hyde Park was the result of a fresh mutation.

This study characterizes the genomic diversity and population structure of Mexican Gir cattle. We sampled 226 animals from three tropical herds and isolated DNA from hair samples was genotyped using the GGP Indicus 50K array. After quality control, 212 animals and 28,394 autosomal SNPs remained for a within-breed analysis. We merged the Mexican Gir dataset with an available online database involving 134 worldwide breeds. The combined dataset was used for across-breed comparisons, with 9091 shared SNPs. Within-breed diversity was high (Heterozygosity Observed = 0.40; Heterozygosity Expected = 0

Callery pear (Pyrus calleryana Decne.; PC) possesses many desirable characteristics valued in managed landscapes. This has driven the release of numerous cultivars, including both hybrids and selections derived from native populations. The extensive planting of PC cultivars in managed areas has contributed to the widespread occurrence of invasive individuals across a broad range of habitats in the eastern United States (US). Self-incompatibility, tolerance to various environmental conditions, pathogen and pest resistance, intraspecific hybridization among the cultivars, possible interspecific

Understanding the factors shaping parasite population structure is key to unravelling host-parasite coevolution and transmission dynamics. Respiratory mites of the genus Orthohalarachne (Acari: Halarachnidae) are obligate parasites infecting the respiratory tract of otariids, yet their population genetics remain largely unexplored. Here, we examined the genetic structure of O. attenuata and O. diminuata infecting two sympatric hosts, the South American fur seal (Arctocephalus australis; SAFS) and the South American sea lion (Otaria byronia; SASL), across their distribution ranges. Genetic anal

The Hetian Gray Donkeys (China_HTQD) and Xinjiang Donkeys (China_XJD) are representative local donkey breeds in Xinjiang, China, that show significant differences in production phenotypes, such as body size and coat color. However, their genetic background and the molecular mechanisms influenced by artificial selection have not been systematically elucidated. This study integrated whole-genome data from 60 newly sequenced China_HTQD and China_XJD individuals, along with 155 global reference datasets from the NCBI database, to construct a modern donkey whole-genome variation set covering domest

Post-Acute Sequelae of COVID-19 (PASC) is increasingly associated with long-term cardiovascular complications, yet the underlying molecular mechanisms, remain poorly understood. Host genetic susceptibility is likely to modulate post-viral cardiac vulnerability, but its interaction with persistent molecular remodelling after SARS-CoV-2 infection has not been systematically investigated. We employed an integrative multi-omics framework combining a pilot genome-wide association study (GWAS) of Indian PASC patients with post-acute cardiac transcriptomic profiling from a SARS-CoV-2-infected Golden
