High glucose condition aggravates inflammatory response induced by Porphyromonas gingivalis in THP-1 macrophages via autophagy inhibition.

BACKGROUND: Porphyromonase gingivalis (P. gingivalis) is a type of bacteria that causes periodontitis, which is strongly correlated with systemic diseases such as diabetes. However, the effect of hyperglycemia on periodontitis are unclear. The present study examined the effects of high glucose levels on the response to P. gingivalis infection. RESULTS: The expression of P. gingivalis-induced interleukin-1β (IL-1β) and inflammasomes increased as the glucose concentration increased. High glucose conditions suppressed P. gingivalis-induced autophagy in human acute monocytic leukemia cell line (T

Prospective randomized double-blind comparative study of topical acetyl zingerone with tetrahexyldecyl ascorbate versus tetrahexyldecyl ascorbate alone on facial photoaging.

BACKGROUND: Tetrahexydecyl ascorbate (THDA) is a lipophilic precursor to ascorbic acid that may be stabilized by acetyl zingerone (AZ). Studies have shown that the topical application of THDA may have photoprotective effects. Similarly, AZ has been shown to mitigate oxidative and inflammatory stress, thereby improving the appearance of photoaging. AIMS: To examine the effects of THDA and AZ (THDA-AZ) on skin photoaging compared to THDA alone. PATIENTS/METHODS: In this double-blind, randomized controlled trial, healthy individuals aged 30 to 65 were included and 44 participants were randomize

Identification and molecular characterization of a Bacillus subtilis IS13 strain involved in the biodegradation of 4,5,6-trichloroguaiacol.

4,5,6-Trichloroguaiacol (4,5,6-TCG) is a recalcitrant organochlorine compound produced during pulp bleaching and a potential environmental hazard in paper mill effluents. We report here the identification by biochemical tests and molecular biological analysis, using 16S ribotyping, of a 4,5,6-TCG-degrading bacterium, identified as a strain of Bacillus subtilis that is most closely related according to the phylogenetic analysis to B. subtilis strain Lactipan (alignment score 99%). Biodegradation of 4,5,6-TCG by this organism in a mineral salts medium was shown to occur only when the inoculum wa

Rescue of the catalytic activity of an H42A mutant of horseradish peroxidase by exogenous imidazoles.

His-42 plays a critical role in the H2O2-dependent catalytic turnover of horseradish peroxidase (HRP). This is clearly illustrated by the finding that an H42A mutation decreases the rate of Compound I formation by a factor of approximately10(6). As shown here, the addition of 2-substituted imidazoles partially rescues both the rate of formation of Compound I and the peroxidase activity of the H42A mutant. 2-Substituted imidazoles are the most effective because they do not coordinate to the iron. In contrast to native HRP, which exhibits a parabolic pH profile, and the H42A mutant, for which th

Coumarin application modulates antioxidant defense to enhance resistance against Fusarium in mung bean.

Plant-derived bioactive compounds contribute a major role in plant defense against fungal pathogens. Among these, coumarin proved to be a major controller in root-pathogen interactions, exhibiting diverse protective mechanisms. This study investigates the effects of coumarin (50 mg.L- 1), applied through seed priming and foliar spray, on the biochemical responses of Mung bean and its impact on root colonization by Fusarium solani. Mung bean seeds pre-treated with coumarin were sown under both infected and non-infected conditions. Once the seedling established up to two-leaf stage, weekly folia

Mixtures modeling identifies chemical inducers versus repressors of toxicity associated with wildfire smoke.

Exposure to wildfire smoke continues to be a growing threat to public health, yet the chemical components in wildfire smoke that primarily drive toxicity and associated disease are largely unknown. This study utilized a suite of computational approaches to identify groups of chemicals induced by variable biomass burn conditions that were associated with biological responses in the mouse lung, including pulmonary immune response and injury markers. Smoke condensate samples were collected and characterized, resulting in chemical distribution information for 86 constituents across ten different e

Protecting cows in small holder farms in East Africa from tsetse flies by mimicking the odor profile of a non-host bovid.

BACKGROUND: For the first time, differential attraction of pathogen vectors to vertebrate animals is investigated for novel repellents which when applied to preferred host animals turn them into non-hosts thereby providing a new paradigm for innovative vector control. For effectively controlling tsetse flies (Glossina spp.), vectors of African trypanosomosis, causing nagana, repellents more powerful than plant derived, from a non-host animal the waterbuck, Kobus ellipsiprymnus defassa, have recently been identified. Here we investigate these repellents in the field to protect cattle from nagan

Cadmium and lead interactive effects on oxidative stress and antioxidative responses in rice seedlings.

Interactive effects of two heavy metal pollutants Cd and Pb in the growth medium were examined on their uptake, production of reactive oxygen species (ROS), induction of oxidative stress and antioxidative defence responses in Indica rice (Oryza sativa L.) seedlings. When rice seedlings in sand culture were exposed to 150 μM Cd (NO3)2 or 600 μM Pb (CH3COO)2 individually or in combination for 8-16 days, a significant reduction in root/shoot length, fresh weight, relative water content, photosynthetic pigments and increased production of ROS (O2˙- and H2O2) was observed. Both Cd and Pb were readi

Comparative Evaluation of Traditional and Controlled Drying Methods of Chestnuts (Castanea sativa Mill.): Impact on the Chemical Composition, Aromatic, and Sensory Profile of Flour.

Chestnut flour, obtained through drying and milling of Castanea sativa fruits, has evolved from a subsistence food into a sought-after niche product, appreciated for its naturally gluten-free profile, high starch content, and richness in micronutrients. Over the past decade, its demand has steadily increased due to consumer perception of the health benefits associated with chestnut consumption. As the market for chestnut flour expanded from small-scale to large-scale production, alternative methods to the traditional process were developed. Its distinctive aroma and flavor are strongly influen

Biochemical Responses to the Long-Term Impact of Copper Sulfate (CuSO4) in Tobacco Plants.

Metabolic changes under stress are often studied in short-term experiments, revealing rapid responses in gene expression, enzyme activity, and the amount of antioxidants. In a long-term experiment, it is possible to identify adaptive changes in both primary and secondary metabolism. In this study, we characterized the physiological state of tobacco plants and assessed the amount and spectrum of phenolic compounds and the lignification of axial organs under excess copper stress in a long-term experiment (40 days). Plants were treated with 100 and 300 μM CuSO4, as well as a control (Knop solutio

Hydrogen bond network in the distal site of peroxidases: spectroscopic properties of Asn70 --> Asp horseradish peroxidase mutant.

The distal His in peroxidases forms a hydrogen bond with the adjacent Asn, which is highly conserved among many plant and fungal peroxidases. Our previous work [Nagano, S., Tanaka, M., Ishimori, K., Watanabe, Y., & Morishima, I. (1996) Biochemistry 35, 14251-14258] has revealed that the replacement of Asn70 in horseradish peroxidase C (HRP) by Val (N70V) and Asp (N70D) discourages the oxidation activity for guaiacol, and the elementary reaction rate constants for the mutants was decreased by 10-15-fold. In order to delineate the structure-function relationship of the His-Asn couple in peroxida

Bioconversion of phenolic acids from hydrolyzed sugarcane bagasse into vinyl derivatives by recombinant phenolic acid decarboxylase.

Several commercially important vinyl phenolic compounds can be produced by enzymatic decarboxylation of phenolic acids such as ferulic and p-coumaric acids which can be extracted from agro-industrial waste. Phenolic acid decarboxylase is an enzyme that acts in the decarboxylation of these acids to 4-vinylguaiacol and 4-vinylphenol, respectively. In this study, the gene encoding phenolic acid decarboxylase from Klebsiella pneumoniae TD 4.7 was isolated and identified as a 504 bp fragment, encoding a polypeptide of 167 amino acid residues. A 98 % predicted amino acid sequence identity between fe

Phenotypic Plasticity of Photochemical Traits and Antioxidant Responsiveness Confer Photosynthetic Resilience in Peanut (Arachis hypogaea L.) Under Phosphorus Deficiency: The Pivotal Role of Cyclic Electron Flow.

Phosphorus (P) deficiency is a major factor governing peanut (Arachis hypogaea L.) productivity, and the physiological mechanisms by which different genotypes (with contrasting photosynthetic capacities) coordinate carbon assimilation and photoprotection remain elusive. This study elucidated the strategic divergence among different peanut genotypes in their foliar photosystems to perform physiological homeostasis under low-phosphorus (LP) conditions. Based on a peanut mini-core collection, six representative accessions with contrasting photosynthetic capacities were selected and categorized in

Nano-iron modulates salt stress responses in Lycium barbarum via enhanced nutrient uptake, antioxidant defense, and phenolic metabolism.

While goji berry (Lycium barbarum) exhibits moderate to high salt tolerance, prolonged or high salinity can still impair its growth, water relations, and metabolism, especially in arid and saline regions. Despite its agronomic and pharmaceutical value, the combined effects of salinity and foliar-applied nano-iron remain largely unexplored. This study investigated the interactive impacts of salinity (0, 150, and 300 mM NaCl) and foliar nano-iron (0, 15, and 30 µM Fe-NPs) on physiological performance, nutrient dynamics, antioxidant responses, and secondary metabolite accumulation in hydroponical

A study on effects of cell phone tower-emitted non-ionizing radiations in an Allium cepa test system.

Considering enormous growth in population, technical advancement, and added reliance on electronic devices leading to adverse health effects, in situ simulations were made to evaluate effects of non-ionizing radiations emitted from three cell phone towers (T1, T2, and T3) of frequency bands (800, 1800, 2300 MHz), (900, 1800, 2300 MHz), and (1800 MHz), respectively. Five sites (S1-S5) were selected near cell phone towers exhibiting different power densities. The site with zero power density was considered as control. Effects of radiations were studied on morphology; protein content; antioxidant
