Itraconazole in the Treatment of Aberrantly Active Hedgehog and/or PI3K Recurrent Ovarian Cancer.

BACKGROUND: Treatment options for recurrent ovarian cancer (OC) are limited, leading to poor prognosis. Targeting tumor-promoting signaling transduction pathways (STPs), such as Hedgehog (HH) and Phosphoinositide-3-kinase (PI3K) STPs, might be an option for treatment. This study evaluates the efficacy of itraconazole as a targeted treatment in HH and/or PI3K active recurrent OC. METHODS: We assessed HH and PI3K STP activity in recurrent OC patients. If activity was aberrantly high in either STP, patients received itraconazole treatment, which has been shown to inhibit both HH and PI3K pathway

Gastrointestinal adverse events associated with Hedgehog pathway inhibitors: a disproportionality analysis.

OBJECTIVE: The Hedgehog pathway inhibitors (HPIs) are effectively used to treat basal cell carcinoma (BCC). Despite the benefits, HPIs are reported to cause various gastrointestinal adverse events (GIAEs). The study was conducted to detect possible safety signals of HPIs concerning GIAE through disproportionality analysis. METHODS: A retrospective case/non-case disproportionality analysis was performed in the USFDA Adverse Event Reporting System database using OpenVigil 2.1, a web-based pharmacovigilance tool. All the GIAEs reported due to the HPIs, like vismodegib, sonidegib, and glasdegib,

RXR Gamma Enables Oligodendrocyte Differentiation by Suppressing Sonic Hedgehog Signaling.

Overcoming remyelination failure is one of the main targets in therapeutic strategies for multiple sclerosis. This process requires the differentiation of oligodendrocyte precursor cells (OPCs) to mature myelinating oligodendrocytes (OLs), a process known to be controlled by thyroid hormone, nuclear receptors, and sonic hedgehog (SHH). Retinoid X receptor gamma (RXRg) is one of the nuclear receptors acting as a positive regulator of remyelination, but little is known about its mechanisms of function. Using transcriptomic and pharmacological analysis of primary neural stem cell-derived OPCs, we

Discovery of a new selective ERAP1 inhibitor for Hedgehog-dependent cancer treatment.

Inappropriate activation of the Hedgehog (HH) signaling pathway drives the pathogenesis of several cancers, including medulloblastoma (MB), the most common malignant brain tumor in children. HH group MB (HH-MB) is highly heterogeneous and resistant to current treatments. Understanding the molecular events underlying the control of the HH pathway is critical for the development of more effective and tailored interventions. Endoplasmic reticulum aminopeptidase 1 (ERAP1), a key regulator of the immune response, has emerged as a promising therapeutic target for HH-MB, but the lack of clinically vi

Targeting the PKC-Gli Axis: Bryostatin 1 as a Hedgehog Pathway Inhibitor.

Aberrant Hedgehog (Hh) signaling contributes to multiple cancers, yet current Smoothened (Smo)-targeted therapies are limited by resistance and relapse. This study investigates bryostatin 1, a clinically relevant natural product from Bugula neritina, for its ability to inhibit Hh signaling downstream of Smo. Bryostatin 1 robustly suppresses Gli transcriptional activity and significantly reduces the expression of key Hh pathway components in a time- and dose-dependent manner. Additionally, it selectively impairs viability in Hh-driven cancer cells while sparing healthy fibroblasts. These compel

Ectopic pregnancy in a European hedgehog (Erinaceus europaeus) - the first documented case.

BACKGROUND: Ectopic pregnancy (EP) is defined as the implantation of a fertilized ovum outside the endometrial lining of the uterine cavity. It occurs rarely in animals and has been most frequently documented in companion and farm animals. The classifications of pregnancies were adapted from human Medicine, as only basic classifications exist in veterinary Medicine. The criteria for classifying EP in both humans and animals have been unified. The classification is based on the site of occurrence, distinguishing between tubal and abdominal pregnancies, each of which can be primary or secondary.

Primary cilia mediate skeletogenic BMP and Hedgehog signaling in heterotopic ossification.

Heterotopic ossification (HO), defined as the formation of extraskeletal bone in muscle and soft tissues, is a diverse pathological process caused by either genetic mutations or inciting trauma. Fibrodysplasia ossificans progressiva (FOP) is a genetic form of HO caused by mutations in the bone morphogenetic protein (BMP) type I receptor gene activin A receptor type 1 (ACVR1). These mutations make ACVR1 hypersensitive to BMP and responsive to activin A. Hedgehog (Hh) signaling also contributes to HO development. However, the exact pathophysiology of how skeletogenic cells contribute to endochon

Myxofibrosarcoma of the penis in an African pygmy hedgehog (Atelerix albiventris) - A clinical case.

A 3-year-old, 420 g, intact male African pygmy hedgehog (Atelerix albiventris) was presented with a sudden appearance of a mass protruding from its preputium. A detailed physical examination revealed the presence of a polyp-like mass, connected to the mucous membrane of the penis and a second, multilobular mass with a larger base. Both masses were surgically removed. While the histopathological examination of the polyp-like mass revealed only a chronic active inflammatory reaction, the histopathological examination of the multilobular mass revealed a tumorous tissue composed of spindle-shaped

Impact of Sonic Hedgehog-dependent sphenoid bone defect on craniofacial growth.

OBJECTIVES: The main objective of this study was to evaluate how an apparently minor anomaly of the sphenoid bone, observed in a haploinsufficient mouse model for Sonic Hedgehog (Shh), affects the growth of the adult craniofacial region. This study aims to provide valuable information to orthodontists when making decisions regarding individuals carrying SHH mutation. MATERIALS AND METHODS: The skulls of embryonic, juvenile and adult mice of two genotypes (Shh heterozygous and wild type) were examined and measured using landmark-based linear dimensions. Additionally, we analysed the clinical c

Hedgehog signaling regulates Wolffian duct development through the primary cilium†.

Primary cilia play pivotal roles in embryonic patterning and organogenesis through transduction of the Hedgehog signaling pathway (Hh). Although mutations in Hh morphogens impair the development of the gonads and trigger male infertility, the contribution of Hh and primary cilia in the development of male reproductive ductules, including the epididymis, remains unknown. From a Pax2Cre; IFT88fl/fl knock-out mouse model, we found that primary cilia deletion is associated with imbalanced Hh signaling and morphometric changes in the Wolffian duct (WD), the embryonic precursor of the epididymis. Downregulation of Sonic hedgehog signaling induces G2-arrest in genital warts.

BACKGROUND: Human papillomavirus (HPV) infected keratinocyte dysfunction results in the formation of genital warts, and the specific role of Sonic hedgehog (SHh) signaling in genital warts remains elusive. Thus, this study aimed to identify the correlation between wart formation and SHh signaling. MATERIALS AND METHODS: In this study, nine male patients with genital warts were recruited, and the expression of SHh and its downstream signal molecules Patched-1 and GLI family zinc finger 1 (Ptch1 and Gli1) was detected. Moreover, G2-phase cells in the collected genital warts samples were assesse

Sonic hedgehog is Essential for Proximal-Distal Outgrowth of the Limb Bud in Salamanders.

The developing forelimb has been a foundational model to understand how specified progenitor cells integrate genetic information to produce the tetrapod limb bauplan. Although the reigning hypothesis is that all tetrapods develop limbs in a similar manner, recent work suggests that urodeles have evolved a derived mode of limb dvelopment. Here, we demonstrate through pharmacological and genetic inactivation of Sonic hedgehog (Shh) signaling in axolotls that Shh directs expansion and survival of limb progenitor cells in addition to patterning the limb across the proximodistal and antero-posterio

Hedgehog Signaling as a Therapeutic Target for Airway Remodeling and Inflammation in Allergic Asthma.

Genome-wide association studies (GWAS) have shown that variants of patched homolog 1 (PTCH1) are associated with lung function abnormalities in the general population. It has also been shown that sonic hedgehog (SHH), an important ligand for PTCH1, is upregulated in the airway epithelium of patients with asthma and is suggested to be involved in airway remodeling. The contribution of hedgehog signaling to airway remodeling and inflammation in asthma is poorly described. To determine the biological role of hedgehog signaling-associated genes in asthma, gene silencing, over-expression, and pharm

GAB1 overexpression identifies hedgehog-activated anterior skull base meningiomas.

AIMS: Mutations activating the hedgehog (Hh) signalling pathway have been described in anterior skull base meningiomas, raising hope for the use of targeted therapies. However, identification of Hh-activated tumours is hampered by the lack of a reliable immunohistochemical marker. We report the evaluation of GAB1, an immunohistochemical marker used to detect Hh pathway activation in medulloblastoma, as a potential marker of Hh-activated meningiomas. METHODS: GAB1 staining was compared to SMO mutation detection with Sanger and NGS techniques as well as Hh pathway activation study through mRNA

Ciliary Hedgehog signaling regulates cell survival to build the facial midline.

Craniofacial defects are among the most common phenotypes caused by ciliopathies, yet the developmental and molecular etiology of these defects is poorly understood. We investigated multiple mouse models of human ciliopathies (including Tctn2, Cc2d2a, and Tmem231 mutants) and discovered that each displays hypotelorism, a narrowing of the midface. As early in development as the end of gastrulation, Tctn2 mutants displayed reduced activation of the Hedgehog (HH) pathway in the prechordal plate, the head organizer. This prechordal plate defect preceded a reduction of HH pathway activation and Shh

The Hedgehog Signaling Pathway is Expressed in the Adult Mouse Hypothalamus and Modulated by Fasting.

The hedgehog signaling pathway is best known for its role in developmental patterning of the neural tube and limb bud. More recently, hedgehog signaling has been recognized for its roles in growth of adult tissues and maintenance of progenitor cell niches. However, the role of hedgehog signaling in fully differentiated cells like neurons in the adult brain is less clear. In mammals, coordination of hedgehog pathway activity relies on primary cilia and patients with ciliopathies such as Bardet-Biedl and Alström syndrome exhibit clinical features clearly attributable to errant hedgehog such as p

Hedgehog Signaling, a Critical Pathway Governing the Development and Progression of Hepatocellular Carcinoma.

Hedgehog (Hh) signaling is a classic morphogen in controlling embryonic development and tissue repairing. Aberrant activation of Hh signaling has been well documented in liver cancer, including hepatoblastoma, hepatocellular carcinoma (HCC) and cholangiocarcinoma. The present review aims to update the current understanding on how abnormal Hh signaling molecules modulate initiation, progression, drug resistance and metastasis of HCC. The latest relevant literature was reviewed with our recent findings to provide an overview regarding the molecular interplay and clinical relevance of the Hh sign

The Role of Sonic Hedgehog in Human Holoprosencephaly and Short-Rib Polydactyly Syndromes.

The Hedgehog (HH) signalling pathway is one of the major pathways controlling cell differentiation and proliferation during human development. This pathway is complex, with HH function influenced by inhibitors, promotors, interactions with other signalling pathways, and non-genetic and cellular factors. Many aspects of this pathway are not yet clarified. The main features of Sonic Hedgehog (SHH) signalling are discussed in relation to its function in human development. The possible role of SHH will be considered using examples of holoprosencephaly and short-rib polydactyly (SRP) syndromes. In

Hedgehog-Transmitted Trichophyton erinaceid Causing Painful Bullous Tinea Manuum.

Trichophyton is a species of fungus that commonly infects hedgehogs. As the popularity of keeping hedgehogs as pets increases, there have been reports of Trichophyton species infection in humans. Diagnosis and appropriate treatment of Trichophyton infection can be delayed owing to physicians' low index of suspicion and unfamiliarity with the physical manifestations. We describe a case of a 27-year-old healthy man who developed tinea manuum after handling a pet hedgehog. Presenting symptoms included a worsening bullous eruption and severe pain. Both fungal and histopathological study confirmed
