Evaluation of Amoxicillin and Amoxicillin-Clavulanate (Augmentin) for Antimicrobial Postexposure Prophylaxis Following Bacillus anthracis Inhalational Exposure in Cynomolgus Macaques.

Amoxicillin is a broad-spectrum antibiotic used to treat a variety of gram-positive and gram-negative infections, such as infections of the ear, nose, and throat, genitourinary tract, skin, and lower respiratory tract; gonorrhea; and Helicobacter pylori. The prophylactic benefit of both amoxicillin and Augmentin (amoxicillin-clavulanate for use against β-lactamase-expressing bacteria) was evaluated for inhalation anthrax in cynomolgus macaques in 2 studies. A pilot study on amoxicillin-clavulanate that used a portion of the study animals demonstrated empirically that dosing twice a day was eff

Vascular effects of physical activity are not modified by short-term inhaled diesel exhaust: Results of a controlled human exposure study.

BACKGROUND: The combined effects of physical activity and air pollution exposure on vascular function are insufficiently understood, particularly after the inhalation of a β2-agonist, a vasodilating agent. OBJECTIVE: To assess the micro- and macrovascular response to physical activity after β2-agonist use while breathing diesel exhaust (DE) in individuals with exercise-induced bronchoconstriction. METHODS: On four exposure visits, eighteen adults inhaled either 400 μg of the β2-agonist salbutamol or placebo before resting for 60 min, followed by a 30-min cycling bout. During rest and cycling

Work Tasks as Determinants of Respirable and Inhalable Indium Exposure among Workers at an Indium-Tin Oxide Production and Reclamation Facility.

Increased global demand for touch screens, photovoltaics, and optoelectronics has resulted in an increase in the production of indium-tin oxide (ITO). Occupational exposure to indium compounds is associated with the development of indium lung disease. Although many previous epidemiologic investigations highlight an excess of lung abnormalities in workplaces where ITO is produced, few assessments of occupational exposure to respirable and inhalable indium are reported to date. The objective of this study was to identify the determinants of respirable and inhalable indium at an ITO production fa

Evaluation of the Nose-to-Brain Transport of Different Physicochemical Forms of Uranium after Exposure via Inhalation of a UO4 Aerosol in the Rat.

BACKGROUND: Health-risk issues are raised concerning inhalation of particulate pollutants that are thought to have potential hazardous effects on the central nervous system. The brain is presented as a direct target of particulate matter (PM) exposure because of the nose-to-brain pathway involvement. The main cause of contamination in nuclear occupational activities is related to exposure to aerosols containing radionuclides, particularly uranium dust. It has been previously demonstrated that instilled solubilized uranium in the rat nasal cavity is conveyed to the brain via the olfactory nerve

On the contribution of the phagocytosis and the solubilization to the iron oxide nanoparticles retention in and elimination from lungs under long-term inhalation exposure.

The aim of our study was to test a hypothesis according to which the pulmonary clearance vs. retention of metal oxide nanoparticles (NPs) is controlled not only by physiological mechanisms but also by their solubilization which in some cases may even prevail. Airborne Fe2O3 NPs with the mean diameter of 14±4nm produced by sparking from 99.99% pure iron rods were fed into a nose-only exposure tower. Rats were exposed to these NPs for 4h a day, 5days a week during 3, 6 or 10 months at the mean concentration of 1.14±0.01mg/m(3). NPs collected from the air exhausted from the exposure tower proved

Integrating in vitro aerosol characterization and cross-species PBPK modeling to predict human lung exposure of inhaled therapeutic proteins.

Inhalation therapy provides a rapid onset and avoids first-pass metabolism, making it a key non-invasive delivery strategy for therapeutic proteins. However, predicting pulmonary exposure is challenging due to aerosol properties and lung anatomy. This study is the first to construct an integrated in vitro aerosol characterization and cross-species extrapolation pulmonary physiologically-based pharmacokinetic (PBPK) model for inhaled therapeutic proteins. Using recombinant human interferon-α1b (rhIFN-α1b) as a model drug, aerosol aerodynamic properties and delivery performance were evaluated at

A method for simultaneously determining both the inhalable fraction and vapor concentration to assess worker exposure to tri-n-butyl phosphate.

OBJECTIVES: This study aimed to develop a personal exposure measurement method that concurrently determines both the inhalable fraction and vapor concentration of tri-n-butyl phosphate (TBP). METHODS: A personal sampler, the IFV Pro, equipped with a glass-fiber filter (GFF) and a Chromosorb 106 adsorption tube was used. Postsampling, TBP was extracted or desorbed separately from the GFF and Chromosorb 106 using dichloromethane containing tri-n-amyl phosphate as an internal standard. The solutions obtained were analyzed via gas chromatography with a flame ionization detector. The evaluation pa

User-oriented independent analysis of the toxic load model's ability to predict the effects of time-varying toxic inhalation exposures.

Toxic industrial chemicals and chemical warfare agents present an acute inhalation hazard to exposed populations. The hazardous materials consequence assessment modeling community requires toxicity models to estimate these hazards. One popular phenomenological toxicity model is the toxic load model. Although this model is only well-defined for constant-concentration exposures, several generalizations have been proposed for the case of time-varying exposures. None of them, however, were validated by experimental evidence at the time they were proposed. Accordingly, the Defense Threat Reduction

Integrated Transcriptomics, Metabolomics, and Lipidomics Profiling in Rat Lung, Blood, and Serum for Assessment of Laser Printer-Emitted Nanoparticle Inhalation Exposure-Induced Disease Risks.

Laser printer-emitted nanoparticles (PEPs) generated from toners during printing represent one of the most common types of life cycle released particulate matter from nano-enabled products. Toxicological assessment of PEPs is therefore important for occupational and consumer health protection. Our group recently reported exposure to PEPs induces adverse cardiovascular responses including hypertension and arrythmia via monitoring left ventricular pressure and electrocardiogram in rats. This study employed genome-wide mRNA and miRNA profiling in rat lung and blood integrated with metabolomics an

Variation in echocardiographic and cardiac hemodynamic effects of PM and ozone inhalation exposure in strains related to Nppa and Npr1 gene knock-out mice.

Elevated levels of ambient co-pollutants are associated with adverse cardiovascular outcomes shown by epidemiology studies. The role of particulate matter (PM) and ozone (O3) as co-pollutants in this association is unclear. We hypothesize that cardiac function following PM and O3 exposure is variably affected by genetic determinants (Nppa and Npr1 genes) and age. Heart function was measured before and after 2 days each of the following exposure sequence; (1) 2-h filtered air (FA) and 3-h carbon black (CB; 0.5 microg/m(3)); (2) 2-h O3 (0.6 ppm) and 3-h FA; (3) 5-h FA; and, (4) 2-h O3 and 3-h CB

Repeated inhalation exposure to octamethylcyclotetrasiloxane produces hepatomegaly, transient hepatic hyperplasia, and sustained hypertrophy in female Fischer 344 rats in a manner similar to phenobarbital.

Octamethylcyclotetrasiloxane (D4) has been described as a phenobarbital-like inducer of hepatic enzymes. Phenobarbital (PB) and phenobarbital-like chemicals induce transient hepatic and thyroid hyperplasia and sustained hypertrophy in rats and mice. The extent to which these processes are involved with D4-induced hepatomegaly is not known. The present study has evaluated the effects of repeated inhalation exposure to D4 vapors on hepatic and thyroid cell proliferation and hypertrophy with respect to time and exposure concentration. Female Fischer 344 rats were exposed via whole body inhalation

Alternation of Organ-Specific Exposure in LPS-Induced Pneumonia Mice after the Inhalation of Tetrandrine Is Governed by Metabolizing Enzyme Suppression and Lysosomal Trapping.

The objective of the present study was to define whether inhaled tetrandrine (TET) could be a promising way to achieve the local effect on its therapeutic efficacy based on biodistribution features using the LPS-treated acute lung injury (ALI) model. The tissue distribution profiles of inhaled TET in normal and ALI mouse models showed that pulmonary inflammation led to an altered distribution in a tissue-specific way. More TET accumulated in almost all tissues including in the blood. Among them, the increased exposure in the lungs was significantly higher than in the other tissues. However, th

Size-Dependent Pulmonary Toxicity and Whole-Body Distribution of Inhaled Micro/Nanoplastic Particles in Male Mice from Chronic Exposure.

The ubiquitous presence of micro/nanoplastics (MP/NP) in the atmosphere has raised significant concerns about their potential health risks through inhalation, yet the effects of natural respiratory exposure remain underexplored. This study addresses this critical knowledge void by utilizing a whole-body inhalation exposure system to investigate the distribution, accumulation, and pulmonary toxicity of polystyrene MP/NP (1.5 × 105 particles/m3) in male ICR mice (n = 16/group). Fluorescently labeled MP/NP revealed the highest particle accumulation in the lungs, followed by the bloodstream and sp

Tissue distribution of inhaled micro- and nano-sized cerium oxide particles in rats: results from a 28-day exposure study.

In order to obtain more insight into the tissue distribution, accumulation, and elimination of cerium oxide nanoparticles after inhalation exposure, blood and tissue kinetics were investigated during and after a 28-day inhalation study in rats with micro- and nanocerium oxide particles (nominal primary particle size: < 5000, 40, and 5-10 nm). Powder aerosolization resulted in comparable mass median aerodynamic diameter (1.40, 1.17, and 1.02 μm). After single exposure, approximately 10% of the inhaled dose was measured in lung tissue, as was also estimated by a multiple path particle dosimetry

Exploring the therapeutic potential of crocin, a bioactive component of saffron, in neurocognitive deficit and pulmonary impairment induced by inhalational exposure to petroleum vapours.

BACKGROUND: Long-term recurrent exposure to petroleum fumes poses a substantial occupational risk, specifically among individuals in gasoline stations. They are routinely exposed to the inherent toxic constituents of petrol and diesel, such as hydrocarbons, VOCs, and trace elements, during their routine activities, including fuel dispensing, handling, storage, transport, and refining processes. Due to inadequate safety measures allow these toxicants into the lungs, cross the pulmonary barrier, enter the circulation and subsequently reach the brain through disrupting the lung-brain axis (LBA).
