Human semen quality and sperm DNA damage in relation to urinary metabolites of pyrethroid insecticides.

BACKGROUND: Exposure to synthetic pyrethroid insecticides is widespread, and is expected to increase among the general population due to the need to replace other common insecticides following regulatory use restrictions. On the basis of limited studies, there is animal and human evidence for altered reproductive or endocrine function following pyrethroid exposure. METHODS: The present study measured urinary pyrethroid metabolites [3-phenoxybenzoic acid (3PBA) and cis- and trans-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropane carboxylic acid (CDCCA and TDCCA)], semen quality, sperm motion para

Phosphotriesterase: a complementary tool for the selective detection of two organophosphate insecticides: chlorpyrifos and chlorfenvinfos.

This work shows the possibility of combining the high sensitivity of genetically-modified Drosophila melanogaster acetylcholinesterase (B394) with the ability of phosphotriesterase (PTE) to hydrolyse organophosphate compounds, in the aim of developing a biosensor selective to two insecticides of interest: chlorpyrifos and chlorfenvinfos. The studies clearly demonstrate that chlorfenvinfos is a substrate that acts as competitive inhibitor of PTE, therefore preventing the efficient hydrolysis of other pesticides, including chlorpyrifos. A bi-enzymatic sensor was designed by immobilizing both B39

[Genotoxic effects of insecticides in current use on mucosal epithelial cells from human tonsil tissue].

BACKGROUND: Vector control is of critical medical importance in disease prevention, as reflected in sections 17 and 18 of the German Protection Against Infection Act. In the past, a large number of biocides were found to be hazardous to human health and were banned from the market, subsequently being replaced by new active ingredients and galenic forms. Many of these new insecticides are available in spray or nebuliser form. Whether these preparations have genotoxic effects on mucosal epithelial cells of the upper aerodigestive tract has thus far not been investigated. MATERIALS AND METHODS:

Long-term quantitative ecological assessment of insecticides treatments in four African rivers: a methodological approach.

In West Africa different insecticides had been applied in selected river areas for the reduction of the blackfly populations vectors of Onchocerca volvulus, a parasite causing blindness. To evaluate the possible long term effects of the larvicides on the non target fauna an aquatic monitoring programme has been up from the initial phase of the project. Addressing the attention to the invertebrates data collected in four countries during a maximum period ranging from 1977 to 1996, this paper shows and discusses the data analysis strategy for the measure and interpretation of the biological vari

Arthropod toxins as leads for novel insecticides: an assessment of polyamine amides as glutamate antagonists.

In the search for new toxins, preferably with new sites of action, the polyamine amides represent a new class of compounds with potential as insecticides and as pharmaceutical agents due to their antagonism of ligand-gated cation channels. In particular, they are potent antagonists of the L-glutamate receptors of insect skeletal muscle. In this paper, we report on synthetic studies to produce hybrid analogues based upon the argiotoxin spider toxins and philanthotoxin-433 which is obtained from a solitary, parasitic wasp. We speculate upon possible modes and sites of action for these antagonist

ADDITIONAL STUDIES WITH NEW INSECTICIDES AS RESIDUAL SPRAYS IN BUILDINGS NATURALLY INFESTED WITH ANOPHELES QUADRIMACULTATUS.

Because some species of Anopheles mosquitos have been developing resistance to DDT and dieldrin, field studies have been conducted to evaluate the effectiveness of recently developed insecticides as residual sprays in buildings naturally infested with adults of Anopheles quadrimaculatus. As a deposit at 2 g/m(2), o-isopropoxyphenyl methylcarbamate remained highly effective for more than one year. This compound and 6-chloro-3,4-xylyl methylcarbamate were still producing excellent control 11-12 weeks after application at 1 g/m(2). Other highly effective materials were m-isopropylphenyl methylcar

Binding characteristics of chemosensory protein 11 from Grapholita molesta Busck (Lepidoptera: Tortricidae) to insecticides.

Chemosensory proteins (CSPs) are small soluble proteins that recognize and bind to various small chemical molecules. Although numerous studies have shown that CSPs play an important role in olfactory communication among insects, recent research indicated that CSPs also contribute to insecticide resistance. The antennae-enriched GmolCSP11 from Grapholita molesta Busck exhibits high binding affinities for seven insecticide ligands, as verified by fluorescence competitive binding assays of recombinant protein. GmolCSP11 exhibited the strongest binding affinity for chlorpyrifos (Ki = 1.01 ± 0.00 μ

Design, Synthesis, and Bioactivities of Novel 4-(Trifluoromethyl)nicotinamide Derivatives for Use as Insecticides.

Hemiptera pests damage crops and transmit viruses through piercing-sucking mouthparts, causing enormous economic losses and inducing resistance to insecticides. Novel insecticides are critical to combat rising resistance. Nicotinamidase, a newly validated target present in the insect chordotonal organ, has attracted attention. A series of novel sulfonyl, carbonyl, and amide group-based 4-(trifluoromethyl)nicotinamide derivatives was designed using the prodrug strategy and an intermediate derivatization method. Bioactivity assays indicated that most compounds possessed enhanced insecticidal act

Acute toxicity and sublethal effects of neonicotinoid insecticides on the predatory ladybird Propylea japonica.

BACKGROUND: Neonicotinoid insecticides are widely used for pest control, but their impacts on non-target natural enemies remain insufficiently characterized, particularly at sublethal concentrations. The predatory ladybird Propylea japonica is an important biological control agent in agroecosystems. Here, we evaluated the acute toxicity, ecological risk, sublethal and transgenerational effects, and physiological and transcriptomic responses of P. japonica to multiple neonicotinoid insecticides. RESULTS: Seven neonicotinoid insecticides showed substantial variation in acute toxicity to second-

Residual activity of insecticides against adult Systena frontalis (Coleoptera: Chrysomelidae) under semi-field conditions.

Systena frontalis (Fabricius) (Coleoptera: Chrysomelidae) is a serious insect pest in nursery production. Insecticides are an important control tactic for S. frontalis adults to reduce economic losses associated with its injury. Because densities and activity of S. frontalis adults are influenced by many biotic (e.g., adult emergence timing) and abiotic (e.g., fluctuating temperatures) factors in the nurseries, it is challenging to reliably determine the efficacy and residual activity of insecticides under field conditions. Thus, the objective was to determine the residual activity of common a

Differential effects of pharmaceuticals and insecticides on swimming behaviour and survival in Gammarus pulex.

Many freshwater systems are continuously exposed to waste streams like municipal wastewater and agricultural runoff, leading to exposure to chemicals that can cause mortality and behavioural changes in aquatic organisms. While research has advanced our understanding of pesticide effects on behaviour of aquatic organisms, the impacts of pharmaceuticals are less understood. Psychopharmaceuticals are particularly interesting because they can act on nervous systems, potentially affecting the behaviour of aquatic organisms. Sublethal behavioural effects can be crucial in ecotoxicological research f

Screening of Toxic Effects of Neonicotinoid Insecticides with a Focus on Acetamiprid: A Review.

Recently, neonicotinoids have become the fastest-growing class of insecticides in conventional crop protection, with extensive usage against a wide range of sucking and chewing pests. Neonicotinoids are widely used due to their high toxicity to invertebrates, simplicity, flexibility with which they may be applied, and lengthy persistence, and their systemic nature ensures that they spread to all sections of the target crop. However, these properties raise the risk of environmental contaminations and potential toxicity to non-target organisms. Acetamiprid is a new generation insecticide, which

Zebrafish Larvae Rapidly Recover from Locomotor Effects and Neuromuscular Alterations Induced by Cholinergic Insecticides.

Owing to the importance of acetylcholine as a neurotransmitter, many insecticides target the cholinergic system. Across phyla, cholinergic signaling is essential for many neuro-developmental processes including axonal pathfinding and synaptogenesis. Consequently, early-life exposure to such insecticides can disturb these processes, resulting in an impaired nervous system. One test frequently used to assess developmental neurotoxicity is the zebrafish light-dark transition test, which measures larval locomotion as a response to light changes. However, it is only poorly understood which structur

Nicotinic acetylcholine receptor modulator insecticides act on diverse receptor subtypes with distinct subunit compositions.

Insect nicotinic acetylcholine receptors (nAChRs) are pentameric ligand-gated ion channels mainly expressed in the central nervous system of insects. They are the directed targets of many insecticides, including neonicotinoids, which are the most widely used insecticides in the world. However, the development of resistance in pests and the negative impacts on bee pollinators affect the application of insecticides and have created a demand for alternatives. Thus, it is very important to understand the mode of action of these insecticides, which is not fully understood at the molecular level. In
