Ruthenium red inhibits calcium-induced calcium release from the ryanodine-sensitive intracellular calcium stores; this study sought to evaluate the effects of ruthenium red on agonist-stimulated phasic myometrial contractions. Ruthenium red was found to significantly inhibit in vitro isometric contractions stimulated in response to oxytocin, prostaglandin F2 alpha, aluminum fluoride, potassium chloride, ionomycin, and Bay K 8644. These observations provide support for the hypothesis that calcium-induced calcium release from ryanodine-sensitive calcium stores is an important event during agonis

It remains unknown whether a regimen of a combination of high- and low-intensity resistance training increases muscle size and maximal voluntary isometric contraction (MVC) simultaneously. This study aimed to clarify the effect of the combination of high- and low-intensity resistance training on muscle size and neuromuscular function in the elbow flexors. Sixteen male adults participated in a 9-week isometric training regimen in elbow joint flexion of each arm. We randomly assigned two different training regimens to left and right arms: one aiming to strengthen maximal strength (ST) and the ot

Heart failure (HF) as a result of myocardial infarction (MI) is a major cause of fatality worldwide. However, the cause of cardiac dysfunction succeeding MI has not been elucidated at a sarcomeric level. Thus, studying the alterations within the sarcomere is necessary to gain insights on the fundamental mechansims leading to HF and potentially uncover appropriate therapeutic targets. Since existing research portrays regulatory light chains (RLC) to be mediators of cardiac muscle contraction in both human and animal models, its role was further explored In this study, a detailed characterisatio

BACKGROUND: Greenstick fractures are common forearm injuries in children, with 75-84 % occurring in the distal third of the radius. Conservative treatments such as detachable braces or plaster backslabs permit early physiotherapy intervention, including muscle activation exercises and electrotherapy. This study investigated the biomechanical and electrophysiological effects on fracture healing to improve bone strength and reduce refracture risk, using finite element analysis (FEA) and experimental validation.
OBJECTIVE: To evaluate the effect of controlled isometric muscle contractions and lo

Cross-education has long been an area of interest. The term is credited to Scripture et al. (1894), who initiated the first series of studies in this area. These studies were mainly in the field of psychology and it was known that if a subject practised writing with one hand, skill was gained in writing with the other hand. DeLorme (1945), experimenting with adaption of weight-lifting procedures to the treatment of various disabilities of the skeletomuscular system, observed that unilateral exercise of the progressive-resistance type was associated with a concurrent development of increased st

In vitro experiments define passive force enhancement as the increase in steady-state passive force following the deactivation of an actively stretched muscle, in contrast to a purely passive stretch. This phenomenon, linked to residual force enhancement, is also observed in voluntarily contracted muscles as passive residual torque enhancement (RTEpass). While mechanisms remain unclear, titin stiffness likey plays a key role. Supersonic shear wave elastography (SSI) estimates tissue stiffness via the shear modulus (μ). The study aimed to assess whether RTEpass of the knee extensor muscles is a

OBJECTIVE: To examine electromyography (EMG) activity, passive torque, and stretch perception during static stretch and contract-relax stretch.
DESIGN: Two separate randomized crossover protocols: (1) a constant angle protocol on the right side, and (2) a variable angle protocol on the left side.
SUBJECTS: 10 male volunteers.
INTERVENTION: Stretch-induced mechanical response in the hamstring muscles during passive knee extension was measured as knee flexion torque (Nm) while hamstring surface EMG was measured. Final position was determined by extending the knee to an angle that provoked a s

OBJECTIVE: In contrast to needle electromyography, the diagnostic performance of muscle ultrasound (MUS) for differentiating neurogenic from myopathic disorders remains unclear. We examined the clinical utility of "fascicular twitch," a visible movement of muscle bundles during isometric contraction, and evaluated its ability to differentiate neurogenic from myopathic changes.
METHODS: We quantified fascicular twitches on MUS during isometric contraction as normalized twitch pixels (nTP) using an original image analysis method based on background subtraction. We analyzed echogenicity and nTP

High-definition transcranial direct current stimulation (HD-tDCS) is a potential neuromodulation apparatus for stroke rehabilitation. However, its modulatory effects in stroke subjects is still not well understood. In this paper, the offline modulatory effects of HD-tDCS on the ipsilesional primary motor cortex were investigated by performing wrist isometric contraction tasks before and after HD-tDCS in eleven unilateral chronic stroke subjects using a synchronized HD-tDCS and electroencephalogram/electromyography measurement system. This paper is a randomized, single blinded, and sham-control

A single bout of unaccustomed exercise confers protective effect against muscle damage from a subsequent bout of similar activity, that is, repeated bout effect (RBE). It remains unknown whether varying muscle-specific exercise between sessions alters the magnitude of the RBE. This study examined the effects of muscle-specific exercise variation between consecutive sessions on the RBE. Twenty untrained males (21 ± 2 years) were assigned to one of 2 groups (n = 10 per group): (a) 2 sessions of incline curls, Fixed Exercise or (b) 1 session of incline curls followed by 1 session of preacher curl

KEY POINTS: The neural strategies behind the control of force during muscle pain are not well understood as previous research has been limited in assessing pain responses only during low-force contractions. Here we compared, for the first time, the behaviour of motor units recruited at low and high forces in response to pain. The results showed that motor units activated at low forces were inhibited while those recruited at higher forces increased their activity in response to pain. When analysing lower- and higher-threshold motor unit behaviour at high forces we observed differential changes

The effect of TRH in vitro was observed on electromyograms and isometric tension changes in the uterine horn isolated from the rat. TRH induced transient prolongation of the duration of spike bursts in the electromyogram and an increased tension in contraction of diestrous uterine horns. No distinct response to TRH was elicited in preparations from rats during other estrous stages. TRH produced a contraction associated with a burst of spike potentials in the quiescent horn from the estrogen-primed ovariectomized rat. Priming with progesterone was not a prerequisite for responsiveness to TRH. I

The physiological demands of sailing are highly specific, varying with wind conditions, type of craft, and crew position. In a light wind, the only physiological variable yet shown to influence performance is the resting blood sugar. Under high wind conditions, the skipper should be light (less than 60 kg), but crew members should be heavy (greater than 80 kg). Height does not seem a great advantage to crew, possibly because they then lack the muscular strength to exploit the added leverage. Muscle strength, endurance and a tolerance of anaerobic metabolism are all desirable attributes of crew

Traditional contractility assays using an organ bath setup consist of several chambers (or baths) perfused with temperature-controlled, oxygenated physiological saline. Strips or rings of tissue (usually smooth or cardiac muscle) are mounted within the organ bath between a fixed hook and an isometric force transducer. The contraction force is recorded by the transducer and different parameters of contraction are analyzed. Different experimental protocols can be performed to investigate the effect of drugs and reagents on tissue contractility to investigate tissue physiology or determine the in

The relationship between the concentration of ambenonium (AMB), a selective and reversible acetylcholinesterase (AChE) inhibitor, in plasma and the potentiation of contractile muscle tension was investigated using a sciatic nerve-muscle preparation of rat. The developed isometric contraction was enhanced dose-dependently after i.v. administration of low doses (5-20 nmol/kg) of AMB, but the contraction was weakened when AMB was administered at high doses (100-1000 nmol/kg), and the concentration-effect relationship was bell-shaped. The muscle contraction profile after 50 nmol/kg administration

UNLABELLED: Little is known about the mechanism of thiopental-induced contraction in vascular smooth muscle. This study aimed to clarify this question by conducting isometric tension experiments and (45)Ca(2+) flux measurements in endothelium-denuded rat aortic rings. Thiopental induced a concentration-dependent contraction under basal tension. This contraction was enhanced when rings were precontracted with phenylephrine in the presence of verapamil. In Ca(2+)-free solution, thiopental-induced contraction was reduced but not abolished with high concentrations. Ca(2+) store depletion with a ma

PURPOSE: Low-intensity eccentric contractions with a load corresponding to 10% of maximal voluntary isometric contraction strength (10% EC) attenuate muscle damage in a subsequent bout of higher-intensity eccentric contractions performed within 2 weeks for the elbow flexors, knee flexors and knee extensors. However, it is not known whether this strategy could be applied to other muscles. This study investigated whether 10% EC would confer damage protective effect on high-intensity eccentric contractions (80% EC) for nine different muscle groups.
METHODS: Untrained young men were placed to an

Gentamicin sulfate, an aminoglycoside antibiotic known to cause depression of neuromuscular function, is a drug of choice in intrauterine antibiotic treatment of bovine chronical or subclinical uterine infections but its effects on the contractility of the cow uterus have not been studied. The aim of this study was to characterize, in vitro, the effect of gentamicin sulfate on spontaneous as well as prostaglandin F2alpha (PGF2alpha) and oxytocin-induced contractility of the non-pregnant cow uterus. Myometrial strips were isolated from non-pregnant cows in follicular phase and suspended in a ja

As aged individuals are frequently exposed to short-term disuse caused by disease or musculoskeletal injury, it is important to understand how short-term disuse and subsequent retraining affect lower limb mechanical muscle function. The purpose of the present study was, therefore, to investigate the effect of 4 days of lower limb disuse followed by 7 days of active recovery on mechanical muscle function of the knee extensors in young (24.3±0.9 years, n=11) and old (67.2±1.0 years, n=11) recreationally active healthy males. Slow and moderate dynamic muscle strength were assessed using isokineti
