The discovery of the dipeptide gamma-glutamyltaurine (gamma-GT; glutaurine, Litoralon) in the parathyroid in 1980 and later in the brain of mammals gave rise to studies on intrinsic and synthetic taurine peptides of this type. It was suggested that gamma-glutamyltransferase (GGT; gamma-glutamyltranspeptidase) in the brain is responsible for the in vivo formation of this unusual dipeptide. gamma-GT has been prepared by both synthetic and enzymatic methods. The chemical syntheses included the use of protecting groups and coupling methods. A wide spectrum of analytical and spectroscopic methods w

The structure of the roof of the fourth cerebral ventricle has been studied by scanning and transmission electron microscopy in Rana pipiens from the late embryonic stages, through the larval stages to metamorphosis. Stage 20 and 21 post-hatching embryos have a rhombencephalic roof consisting of undifferentiated embryonic cells. From embryonic stage 22-25 there is progressive differentiation into choroid plexus and posterior tela ependymal cells. Between 1 and 3 interependymal pores occur in the posterior tela and there is a small incipient subarachnoid space dorsal to the posterior tela. Duri

Chemical safety evaluation is in the midst of a transition from traditional whole-animal toxicity testing to molecular pathway-based in vitro assays and in silico modeling. However, to facilitate the shift in reliance on apical effects for risk assessment to predictive surrogate metrics having characterized linkages to chemical mechanisms of action, targeted in vivo testing is necessary to establish these predictive relationships. In this study, we demonstrate a means to predict thyroid-related metamorphic success in the model amphibian Xenopus laevis using relevant biochemical measurements du

BACKGROUND: The transcriptional activity of the thyroid hormone receptors is modulated by the ligand, T3, but they have also activity as aporeceptors, in the unliganded state. Aporeceptor activity is thought to contribute to the severity of profound hypothyroidism. During development thyroid hormone receptors are expressed before onset of thyroid gland function and are present therefore in many tissues mainly as aporeceptors. The question we address is whether thyroid hormone aporeceptors are involved in physiological and/or developmental processes.

Metamorphosis is a classic example of developmental transition, which involves important morphological and physiological changes that prepare the organism for the adult life. It has been very well established that amphibian metamorphosis is mainly controlled by Thyroid Hormone (TH). Here, we show that the heterochronic gene Lin28 is downregulated during Xenopus laevis metamorphosis. Lin28 overexpression before activation of TH signaling delays metamorphosis and inhibits the expression of TH target genes. The delay in metamorphosis is rescued by incubation with exogenous TH, indicating that Lin

Cell death in animals is normally classified as type I (apoptotic), type II (autophagic) or necrotic. Of the biologically controlled types of death, in most embryos apoptosis is the most common, although in metamorphosis and in cells with massive cytoplasm type II is often seen, and intermediate forms are seen. For vertebrate embryos other than mammals, apoptosis is not seen prior to gastrulation but thereafter is used to sculpt the organs of the embryo, while overproduction of cells with subsequent death of excess cells is a common means of generating high specificity with low information cos

The Colorado potato beetle is a notorious agricultural pest with remarkable adaptability and insecticide resistance. This study conducted comprehensive transcriptome analysis across multiple developmental stages and tissues, focusing on the cytochrome P450 monooxygenase gene family. Using RNA-seq data from 65 datasets, we identified significant transcriptional reprogramming during key developmental transitions, particularly from egg to first instar larva and from third instar larva to adult, reflecting critical biological processes, including feeding initiation and metamorphosis. We identified

In order to minimize risks to human and environmental health, chemical safety assessment programs are being reinforced with toxicity tests more specifically designed for detecting endocrine disrupters. This includes the necessity to detect thyroid-disrupting chemicals, which may operate through a variety of modes of action, and have potential to impair neurological development in humans, with resulting deficits of individual and social potential. Mindful of these concerns, the consensus favors in vivo models for both hazard characterization (testing) and hazard identification (screening) steps

Many benthic marine animal populations are established and maintained by free-swimming larvae that recognize cues from surface-bound bacteria to settle and metamorphose. Larvae of the tubeworm Hydroides elegans, an important biofouling agent, require contact with surface-bound bacteria to undergo metamorphosis; however, the mechanisms that underpin this microbially mediated developmental transition have been enigmatic. Here, we show that a marine bacterium, Pseudoalteromonas luteoviolacea, produces arrays of phage tail-like structures that trigger metamorphosis of H. elegans. These arrays comp

Butenolide [5-octylfuran-2(5H)-one] is a very promising antifouling compound. Here, the effects of butenolide on larval behavior and histology are compared in two major fouling organisms, viz. cypris larvae of Balanus amphitrite and swimming larvae of Bugula neritina. Butenolide diminished the positive phototactic behavior of B. amphitrite (EC50=0.82 μg ml(-1)) and B. neritina (EC50=3 μg ml(-1)). Its effect on the attachment of cyprids of B. amphitrite was influenced by temperature, and butenolide increased attachment of larvae of B. neritina to the bottom of the experimental wells. The signal transduction pathway through which excess potassium ion stimulates the larvae of many marine invertebrates to metamorphose is incompletely understood. Recent evidence suggests that dopamine plays important roles in the metamorphic pathway of Crepidula fornicata. Therefore, we asked whether blocking dopamine receptors might prevent excess potassium ion from stimulating metamorphosis in this species. Surprisingly, the effects of the three putative dopamine antagonists tested (all at 10 microM) varied with exposure duration and the age of competent larvae. Chlorpromazine, a nonspecific

The serine/threonine-protein kinase polo plays a crucial role in regulating the cell cycle and mitosis in insects. While the cigarette beetle, Lasioderma serricorne, is a globally distributed pest threatening stored products, the functions of its polo gene remain largely unexplored. This study investigates the polo gene to elucidate its role in the development and reproduction of L. serricorne. The Lspolo gene, which encodes a protein containing a catalytic serine/threonine kinase domain and polo box domains, was identified and cloned. Spatio-temporal expression analyses indicated that Lspolo

BACKGROUND: Flatfish metamorphosis involves dramatic tissue remodeling, including the migration of one eye to the opposite side of the body, enabling the transition from pelagic to benthic life. While this process requires precise transcriptional regulation, the role of epigenetic mechanisms remains poorly understood. Here, we investigate DNA methylation dynamics during turbot metamorphosis using reduced-representation bisulfite sequencing (RRBS) across three key stages: pre-metamorphosis, climax, and post-metamorphosis.
RESULTS: We identified stage-specific methylation patterns, with more th

Amphibians rely exclusively on behavioral thermoregulation to maintain body temperature within species- and developmental stage-specific critical limits. Several members of the bombesin family of peptides and histamine are included in a class of neurochemicals that have potent thermoregulatory effects in ectothermic and endothermic vertebrate species and may be involved in behavioral thermoregulation in amphibians. Because amphibians respond to environmental temperature cues differently in larval versus adult animals, we used immunocytochemistry to study developmental changes in bombesin-like

During metamorphosis, the larval thoracic legs of the moth Manduca sexta are replaced by new adult legs. The leg motoneurons do not die after the loss of the larval muscles, but persist to innervate the new adult leg muscles (Kent and Levine, 1988). The adult muscles form from myoblasts that originate in specific production sites within the legs and migrate to the sites of muscle formation, where they accumulate, proliferate, and fuse to form myofibers (Consoulas et al., 1996b). Throughout adult leg muscle development, there is a close association between nerves and the developing muscles, sug

The clawed toad, Xenopus laevis, responds to surface waves with a very accurate orienting reaction towards the source of the wave. The importance of visual experience during larval development for this orienting reaction guided by the lateral line was investigated by testing the responses in postmetamorphic animals deprived of vision from an early larval stage by enucleation or rearing in darkness. Control animals were enucleated after metamorphosis. The results show that the orienting response in animals enucleated as larvae is less accurate; the slope of the regression of the turning angle o
