Evaluation of frog gastric mucous secretion by 7T magnetic resonance imaging.

This work describes a new method to evaluate the action of drugs on gastric mucus secretion by 7 T MRI. A polyethylene probe was inserted into the stomach of frogs, a water washing cycle was performed, air was inflated into the stomach, and the probe was removed. Drugs were administered topically by mixing them with the water used for stomach washing, and a series of images was collected. In untreated animals, a mucous layer emitting a high intensity signal covered the gastric mucosa. Atropine administration strongly reduced the mucous layer thickness. After pilocarpine administration, MRI ima

[Bronchial hyperresponsiveness and relaxability in patients with mitral stenosis].

To study the differences in bronchial responsiveness to inhaled methacholine and salbutamol between patients with mitral valve disease those with bronchial asthma a bronchial provocation test (Astograph) and pulmonary function tests were done in 25 patients with mitral stenosis (MS) and 18 patients with bronchial asthma (BA). We derived an index of airway reloxability from the effect of salbutamol after precontraction with inhaled methacholine. This index was the change in respiratory conductance (Grs) over time during inhalation of salbutamol (SGrs-d). It was computed as SGrd = delta Grs/delt

Aspecific bronchial hyperreactivity in pulmonary sarcoidosis.

Aspecific bronchial hyperreactivity (A.B.H.) is a condition found mostly in cases of bronchial asthma, but it may also present in other pathologies affecting the respiratory tract. This study examines 30 patients presenting hilo-pulmonary sarcoidosis, 27 asthmatic patients and 25 healthy subjects as controls. The three groups were sex and age matched. No significant difference in smoking habits between controls and sarcoidosis patients was found. Patients were tested for aspecific bronchial hyperreactivity by administering increasing doses of methacholine. Methacholine inhalation was performed

The release of platelet-activating factor into plasma during allergen-induced bronchoconstriction.

Plasma histamine and platelet-activating factor (PAF) were measured in six subjects with mild seasonal asthma before and after allergen-induced bronchoconstriction, and in six other patients with asthma before and after methacholine-induced bronchoconstriction. A significant increase in plasma histamine and PAF levels was found in patients with mild seasonal asthma after allergen-induced bronchoconstriction but not in patients with asthma after bronchoconstriction induced by methacholine. There was a significant correlation between the baseline plasma PAF levels and the degree of bronchial hyp

Elicitation of allergic airway inflammation modulates the cough response via increased prostaglandin I₂ in ovalbumin-sensitized guinea pigs.

BACKGROUND: Asthmatic cough is a common cause of chronic cough, and cough-variant asthma (CVA) and typical bronchial asthma (BA) display distinct pathophysiological characteristics. Eosinophilic airway inflammation is believed to contribute to the persistence and treatment resistance of chronic cough, although the underlying mechanisms remain unclear. This study aimed to elucidate the changes in lipid mediators and cough responses induced by eosinophilic airway inflammation. METHODS: We employed an ovalbumin (OVA)-sensitized guinea pig model to investigate the role of eosinophilic airway infl

Desmopressin nasal spray inhibiting parasympathetic function on isolated tracheal smooth muscle.

Objectives: Nocturia with or without asthma is one of the aging diseases. Desmopressin has been used as a nasal spray for patients who are suffering from nocturia. This study determined the effects of desmopressin on isolated tracheal smooth muscle in vitro. Methods: We evaluated desmopressin's efficiency on isolated rat tracheal smooth muscle. Desmopressin was evaluated for the following effects on tracheal smooth muscle: (1) effect on resting tension; (2) effect on contraction brought on by parasympathetic mimetic 10-6 M methacholine; and (3) effect on electrically produced tracheal smooth m

Effects of airway smooth muscle contraction and inflammation on lung tissue compliance.

There are renewed interests in using the parameter K of Salazar-Knowles' equation to assess lung tissue compliance. K either decreases or increases when the lung's parenchyma stiffens or loosens, respectively. However, whether K is affected by other common features of respiratory diseases, such as inflammation and airway smooth muscle (ASM) contraction, is unknown. Herein, male C57BL/6 mice were treated intranasally with either saline or lipopolysaccharide (LPS) at 1 mg/kg to induce pulmonary inflammation. They were then subjected to either a multiple or a single-dose challenge with methacholi

Sex Modifies Acute Ozone-Mediated Airway Physiologic Responses.

Sex differences clearly exist in incidence, susceptibility, and severity of airway disease and in pulmonary responses to air pollutants such as ozone (O3). Prior rodent O3 exposure studies demonstrate sex-related differences in the expression of lung inflammatory mediators and signaling. However, whether or not sex modifies O3-induced airway physiologic responses remains less explored. To address this, we exposed 8- to 10-week-old male and female C57BL/6 mice to either 1 or 2 ppm O3 or filtered air (FA) for 3 h. At 12, 24, 48, and 72 h following exposure, we assessed airway hyperresponsiveness

Airway Hyperresponsiveness in Asthma: Measurement and Clinical Relevance.

Airway hyperresponsiveness is a characteristic feature of asthma, and its measurement is an important tool in its diagnosis. With a few caveats, methacholine bronchial provocation by a 2-minute tidal breathing method is highly sensitive; a negative test result (PC20 > 16 mg/mL, PD20 > 400 μg) rules out current asthma with reasonable certainty. A PC20 value of less than 1 mg/mL/PD20 value of less than 25 μg is highly specific (ie, diagnostic) but quite insensitive for asthma. For accurate interpretation of the test results, it is important to control and standardize technical factors that have

Airway hyperreactivity is frequent in non-asthmatic children with sickle cell disease.

BACKGROUND: Asthma is associated with poorer outcomes in sickle cell disease (SCD). Whether AHR can exist in SCD as a distinct entity, separate and independent of asthma, is unknown. AIMS: Our goal was to elucidate the prevalence of AHR, as measured by a methacholine challenge test (MCT), in children with SCD who did not have concomitant asthma or any recent history of acute chest syndrome (ACS). To determine if AHR was associated with asthma-like symptoms, we compared the results of the MCT to a validated asthma questionnaire. We also examined if a correlation between AHR and inflammatory ma

History dependence of vital capacity in constricted lungs.

Measurements of dynamic force-length behavior of maximally activated strips of smooth muscle during oscillatory length changes show that force decreases well below the isometric force during the shortening phase of the oscillation. The magnitude of the decrease depends on the rate of shortening; for slower shortening, the decrease is smaller and force is larger. Modeling of expiratory flow, based on these data, predicts that vital capacity in constricted lungs depends on the rate of expiration. In maximally constricted lungs, forced vital capacity (FVC) is predicted to be 16% smaller than cont

Vascular and metabolic effects of methacholine in relation to insulin action in muscle.

AIMS/HYPOTHESIS: Methacholine (MC) is a nitric oxide vasodilator, but unlike other vasodilators, it potentiates insulin-mediated glucose uptake by muscle. The present study aimed to resolve whether this action was the result of a vascular effect of MC leading to increased muscle perfusion or a direct effect of MC on the myocytes. We hypothesise that vascular-mediated insulin-stimulated glucose uptake responses to MC occur at lower doses than direct myocyte MC-mediated increases in glucose uptake. METHODS: The vascular and metabolic effects of this vasodilator were examined in rats in vivo usi

Association between reduced bronchodilatory effect of deep inspiration and loss of alveolar attachments.

BACKGROUND: We have previously shown that the bronchodilatory effect of deep inspiration is attenuated in individuals with COPD. This study was designed to investigate whether the impairment in this effect is associated with loss of alveolar attachments. METHODS: We measured deep inspiration (DI)-induced bronchodilation in 15 individuals with and without COPD (67 +/- 2.2 yrs of age, mean +/- SEM) undergoing lobar resection for peripheral pulmonary nodule. Prior to surgery, we measured TLCO and determined the bronchodilatory effect of deep inspiration after constricting the airways with methac

Vocal cord dysfunction induced by methacholine challenge testing.

STUDY OBJECTIVES: To determine whether methacholine challenge testing (MCT) provokes vocal cord dysfunction (VCD), as evidenced by inspiratory vocal cord closure on direct laryngoscopy, and whether spirometry and flow-volume loops (FVLs) demonstrate any changes that are suggestive of VCD. DESIGN: Prospective, controlled study. SETTING: Army medical center. PATIENTS: Thirty-four subjects all with normal baseline spirometry. Ten subjects had documented evidence of VCD, 12 subjects had exercise-induced asthma (EIA) and reactive MCT, and 12 subjects served as healthy asymptomatic control subjec

[Effect of alpha and beta-adrenergic agonists on methacholine-induced nasal secretion in man].

To examine the roles of alpha-adrenergic, beta-adrenergic and cholinergic agonists on the nasal secretion mechanism, the author repeated nasal methacholine (Mch, 1 time 20 mg) challenges ten times at 10-min intervals in 13 healthy male subjects. As it was difficult to obtain a sufficient amount of nasal secretion for analysis using simple nasal provocation tests with alpha-and beta-adrenergic agonists before repeated Mch challenges, alpha-adrenergic (phenylephrine, Phe, 2 mg) and beta-adrenergic (isoproterenol, Iso, 0.2 mg) agonists were administered into both nostrils. The effects of these ag

Modulation of the effect of atrial natriuretic peptide in human and bovine bronchi by phosphoramidon.

1. We have previously shown that atrial natriuretic peptide causes bronchodilatation and reduces bronchial reactivity when administered intravenously or by inhalation to asthmatic patients. We wished to determine the direct effect of exogenously applied atrial natriuretic peptide on isolated airway and the role of proteases important in atrial natriuretic peptide degradation in other organ systems. 2. The ability of atrial natriuretic peptide (alpha-human atrial natriuretic peptide 28-amino acid) to relax precontracted tissues and to protect against methacholine-induced contraction was studied

Delivery of salbutamol by metered dose inhaler and valved spacer to wheezy infants: effect on bronchial responsiveness.

The efficacy of a new valved spacer device, the Babyhaler inhaler (Glaxo) for administering metered dose inhaler treatment via a facemask to infants was assessed. In a double blind, single dose study, salbutamol (800 micrograms) or placebo were given on separate days to 12 sedated, sleeping, wheezy infants during a symptom free interval. Lung function was measured before and after administration and the bronchial response to aerosol challenge with methacholine was then assessed using the squeeze technique. A small increase in heart rate and a drop in arterial oxygen tension followed salbutamol

Duration of antigen-induced hyperresponsiveness in horses with allergic respiratory disease and possible links with early airway obstruction.

Antigen-induced airway hyperresponsiveness in allergic horses has previously been demonstrated when clinical signs of acute airway obstruction were apparent, as a consequence of exposure of animals to hay and straw for variable periods of time, and repeat measurements of hyperresponsiveness have been made no earlier than 1 week after challenge. In the present study airway responsiveness to methacholine has been measured in normal horses and allergic horses in clinical remission before and 24, 48 and 72 h after a hay and straw challenge of fixed, short, duration (7 h). Correlations between earl
