The gut microbiota mediates the antidepressant effects of imipramine: Evidence from fecal microbiota transplantation and prebiotic inulin intervention.

BACKGROUND: Many antidepressants exert their effects by modulating the gut microbiota, but it is unknown if this mechanism extends to the tricyclic antidepressant imipramine. OBJECTIVE: We probed the extent to which specific gut bacterial taxa mediate the antidepressant effects of imipramine. MATERIAL AND METHODS: Using a chronic unpredictable mild stress (CUMS) mouse model, we administered imipramine, the prebiotic inulin, or their combination, and systematically evaluated depressive-like phenotypes. Gut microbial dynamics were profiled by 16S rRNA sequencing, and fecal microbiota transplan

Gut Microbiota and Metabolites: Orchestrating Depression Pathogenesis Through the Microbiota-Gut-Brain Axis's Neural, Immune, and Metabolic Routes.

Depression (major depressive disorder, MDD) is a globally prevalent, highly disabling, and complex mental disorder whose pathogenesis has not been fully elucidated. In recent years, the role of the gut microbiota in depression via the "microbiota-gut-brain axis" (MGB axis) has attracted increasing attention. A large body of evidence indicates that the gut microbiota and its metabolites can engage in bidirectional communication with the central nervous system through three core pathways-neural, immune, and metabolic-thereby profoundly influencing the onset and progression of depression. This ar

Fecal Microbiota Transplantation and Microbiota-Based Therapeutics in Allogeneic Hematopoietic Stem Cell Transplantation: Current Evidence and Future Directions.

The intestinal microbiome is a key regulator of immune homeostasis, metabolism, and epithelial barrier integrity. In patients with malignant hematological diseases, particularly those undergoing hematopoietic stem cell transplantation, microbiome perturbations by reduced diversity, pathobiont expansion, and loss of beneficial metabolites are common as a consequence of exposure to cytotoxic therapy and broad-spectrum antimicrobials. Accordingly, enteral microbiome manipulation has emerged as a promising strategy. We performed a narrative review of the literature in PubMed/MEDLINE, Embase, and t

Intestinal Microbiota Structure of Xichuan Black-Bone Chickens and Preliminary Evaluation of a Probiotic-Based Fecal Microbiota Substitute.

In this study, we aimed to explore the intestinal microbiota structure of Xichuan black-bone chickens (XBCs) and evaluate the effect of fecal microbiota substitute transplantation (FMST) by comparing it with traditional fecal microbiota transplantation (FMT). Metagenomic sequencing was used to analyze the microbiota composition and diversity of different intestinal segments (duodenum, jejunum, ileum, cecum, and rectum) of adult XBCs. Probiotic strains were subsequently isolated and screened from the cecal contents under anaerobic conditions to prepare FMST preparations. In total, 90 1-day-old

Parkinson's Disease and the Microbiota-Gut-Brain Axis: Metabolites, Mechanisms, and Innovative Therapeutic Strategies Targeting the Gut Microbiota.

The human gut microbiota is diverse and abundant and plays important roles in regulating health by participating in metabolism and controlling physiological activities. The gut microbiota and its metabolites have been shown to affect the functioning of the gut and central nervous system through the microbiota-gut-brain axis. It is well established that microbiota play significant roles in the pathogenesis and progression of Parkinson's disease (PD). Disorders of the intestinal microbiota and altered metabolite levels are closely associated with PD. Here, the changes in intestinal microbiota an

The emerging role of microbiota in neuro-oncology.

Primary and metastatic brain tumours are associated with poor clinical outcomes, largely owing to limited therapeutic advances and an incomplete understanding of brain tumour biology. Increasing evidence suggests that microbial communities play an important part in shaping systemic immunity and tumour biology. Microbes and microbial by-products from the gut and oral cavity can influence the central nervous system through the gut-oral-brain axis, providing potential mechanisms by which distant microbes shape the brain tumour microenvironment. A growing body of literature has identified distinct

Gut microbiota and osteoarthritis: mechanisms and translation.

Osteoarthritis (OA) is increasingly recognised as a whole-joint disease driven by biomechanical stress, metabolic dysfunction, low-grade inflammation and immune dysregulation, yet effective disease-modifying treatments remain unavailable. Growing evidence suggests that gut microbiota dysbiosis may contribute to OA pathogenesis, giving rise to the concept of a functional and potentially targetable gut-joint axis. In this narrative review, we synthesise current evidence linking gut microbial alterations to OA and highlight the immunological mechanisms through which intestinal dysbiosis may influ

Oral microbiota in cardiovascular health and disease.

Hundreds of microbial species inhabit the oral cavity and are essential to local and systemic health. Cardiovascular disease (CVD) causes a significant burden on global health, with its etiology and pathogenesis still not fully understood. Growing evidence indicates an association between the oral microbiome and CVD. However, the variability of populations, follow-up durations, and disease severity between various human investigations led to divergent conclusions. Animal studies have focused on the relationship between individual microbial species and CVD, yet gaps remain in the mechanisms bet

[Microbiota and microbiome of the lacrimal drainage system].

This review analyzes current concepts of the role of the microbiota and microbiome in the physiology and pathology of the human lacrimal drainage system (LDS). The terms are clearly differentiated: microbiota is the collection of living microorganisms, whereas microbiome also includes their genetic material and habitat. The article describes anatomical features of the LDS and involutional changes in adults (atrophy of the lacrimal puncta, canalicular fibrosis, and nasolacrimal duct stenosis), which predispose to tear stagnation and inflammation. The review includes a comparative analysis of th

Gut microbiota and sarcoidosis: a concise review.

Microbial involvement in sarcoidosis pathogenesis is suggested by the observation that histological findings in sarcoid granulomas are like those of leprosy, tuberculosis and parasitic infection. Some studies have shown that the lung microbiome in patients with sarcoidosis is different from healthy individuals. Results are conflicting, reporting an abundance or decrease in bacterial and fungal species. The altered composition of the microbiome in sarcoidosis can contribute to the formation of granulomas, typical lesions of the disease, through interactions with the host immune system. However,

Effects of gut microbiota in breast cancer.

Breast cancer (BC) stands as the predominant form of cancer affecting women globally, with its etiology rooted in a complex interplay of factors. Emerging research underscores the significant impact of microbiota on the development of BC. Evidence points to a correlation between BC and microbial imbalance, not only within the intestinal milieu but also in breast tissue itself. Alterations in the diversity and functionality of bacterial populations in these areas are implicated in the disease's pathogenesis. The intestinal microbiota exerts a pivotal influence on the metabolism of steroid hormon

The intratumor microbiota and thyroid cancer: a review.

Thyroid cancer is a prevalent malignancy with a rising global incidence, driven by factors such as genetic mutations, environmental influences, and gender differences. Despite advancements in diagnostic techniques and treatments, effective therapies for advanced or iodine-refractory thyroid cancer remain limited. Recent discoveries have challenged the long-held belief that tumors are sterile, revealing the presence of intratumor microbiota in various cancers. Intratumor microbiota significantly impact cancer development, immune reactions, and the effectiveness of treatments. This review examin

The microbiota in radiotherapy-induced cancer immunosurveillance.

Radiotherapy has an established role in the clinical treatment of patients with a variety of cancers owing to the ability to preferentially kill malignant cells mostly while sparing their non-malignant counterparts. Results from phase I-II trials also suggest that radiotherapy can have therapeutically relevant immunostimulatory effects, especially when combined with immune-checkpoint inhibitors. Over the past two decades, evidence has emerged showing that intestinal microbial communities have a major influence on the immunological tonus of patients with cancer and can influence sensitivity to

The influence of endocrine disruptors on the gut microbiota.

Endocrine disruptors (EDs) are closely associated with the second brain, the microbiota-derived enteric nervous system, commonly referred to as the gut microbiota. The microbiota plays a crucial role in human health and the development of diseases. In today's industrialized world, the presence of EDs in air, water, and soil leads to primary human exposure through dermal contact and ingestion. The impact of these EDs on the microbiota remains unclear. EDs that disrupt the balance of the gut microbiota may contribute to a range of disorders, including metabolic (obesity, diabetes mellitus), card

Repercussions of gastrointestinal microbiota in postmenopausal osteoporosis.

Osteoporosis, characterized by diminished bone mass and microarchitectural degeneration, increases fracture risk, particularly in postmenopausal women (postmenopausal osteoporosis), leading to reduced quality of life and increased mortality. Recent research has highlighted the gut-bone axis, showing how the gut microbiota influences bone health through immune modulation, endocrine regulation, and calcium absorption. Dysbiosis, an imbalance in gut microbiota (e.g., decreased Bacteroidetes and increased Firmicutes), has been linked to osteoporosis by enhancing osteoclast activity and bone loss.
