Staged deep brain stimulation for refractory craniofacial dystonia with blepharospasm: case report and physiology.

OBJECTIVE AND IMPORTANCE: We report the intraoperative results, subsequent course, and 1-year follow-up evaluation of a patient with medication-refractory craniofacial dystonia for whom we planned bilateral globus pallidus internus (GPi) deep brain stimulation (DBS) implantation but delayed the left GPi DBS implantation because of robust intraoperative effects of right GPi DBS. CLINICAL PRESENTATION: A 47-year-old patient had a 5-year history of progressively severe, bilateral craniofacial dystonia with blepharospasm (Meige's syndrome) that was refractory to medications and to botulinum toxin

Proliferation and Cluster Analysis of Neurons and Glial Cell Organization on Nanocolumnar TiN Sub-Strates.

Biomaterials employed for neural stimulation, as well as brain/machine interfaces, offer great perspectives to combat neurodegenerative diseases, while application of lab-on-a-chip devices such as multielectrode arrays is a promising alternative to assess neural function in vitro. For bioelectronic monitoring, nanostructured microelectrodes are required, which exhibit an increased surface area where the detection sensitivity is not reduced by the self-impedance of the electrode. In our study, we investigated the interaction of neurons (SH-SY5Y) and glial cells (U-87 MG) with nanocolumnar titan

Electrophysiology of the human colon: evidence of segmental heterogeneity.

The electrical properties of epithelial cell membranes in human descending and ascending colon were studied using microelectrodes and the Na channel blocker amiloride. Under control (pre-amiloride) conditions, the transepithelial electrical measurements in the two colonic segments were similar. The mucosal addition of 0.1 mM amiloride to descending colon totally abolished the transepithelial voltage (Vt) and short-circuit current (Isc), and significantly increased the total tissue resistance (Rt) by 19% (p less than 0.005). Intracellular recordings in descending colon obtained with microelectr

Chloride-dependent pH regulation in connective glial cells of the leech nervous system.

We used double-barreled pH-sensitive microelectrodes to study the mechanisms by which the intracellular pH is regulated in the connective glial cells of the medicinal leech. The experiments indicate that a Cl(-)- and HCO3(-)-dependent mechanism mediates some recovery from intracellular alkalosis even in the absence of external Na+. This suggests the presence of Na(+)-independent Cl-/HCO3- exchange in the connective glial membrane. At alkaline pHi, this exchange most likely operates in the direction of net acid loading (i.e. HCO3- efflux).

Advances in In Vitro Models of Neuromuscular Junction: Focusing on Organ-on-a-Chip, Organoids, and Biohybrid Robotics.

The neuromuscular junction (NMJ) is a peripheral synaptic connection between presynaptic motor neurons and postsynaptic skeletal muscle fibers that enables muscle contraction and voluntary motor movement. Many traumatic, neurodegenerative, and neuroimmunological diseases are classically believed to mainly affect either the neuronal or the muscle side of the NMJ, and treatment options are lacking. Recent advances in novel techniques have helped develop in vitro physiological and pathophysiological models of the NMJ as well as enable precise control and evaluation of its functions. This paper re

Exocytosis of SH-SY5Y single cell with different shapes cultured on ITO micro-pore electrode.

Communication between cells by release of specific chemical messengers via exocytosis plays crucial roles in biological process. Catecholamines, like dopamine, epinephrine, and norepinephrine, which are types of neurotransmitters released from cells, can be oxidized and detected by the microelectrodes, and amperometric detection of exocytosis is an effective method for studying the communication between cells. The experimental results depend on many factors, among which the property of the microelectrode, cell states, and their positions to each other are particularly important. A type of indi

Pathophysiology of the basal ganglia and movement disorders: from animal models to human clinical applications.

Electrophysiological studies in control and MPTP treated primates have played a major role in our understanding of the physiology of the basal ganglia and the pathophysiology of Parkinson's disease (PD). Early models emphasized discharge rate and viewed the basal ganglia as a network of boxes (nuclei) connected by excitatory or inhibitory connections. More recent studies view the basal ganglia as neural networks with weak and non-linear interactions in and between the different nuclei. Microelectrode electrophysiological recording enables the high resolution-both in the temporal domain (spike)

Spontaneous and evoked ectopic discharges recorded from single human axons.

A quantitative assessment was made of the firing characteristics of repetitive axonal discharges encountered during microneurographic recordings from human peripheral nerves. Spontaneous activity was recorded from 16 single axons using tungsten microelectrodes inserted percutaneously into fascicles of the median or peroneal nerves in normal subjects. These discharges typically consisted of brief bursts of 2-5 spikes occurring at a frequency of 7-10 Hz. Peak instantaneous frequencies usually exceeded 300 Hz. Based on their similarity with spontaneous high-frequency discharges recorded from sing

pH transients evoked by excitatory synaptic transmission are increased by inhibition of extracellular carbonic anhydrase.

Excitatory synaptic transmission has been associated with a rapid alkalinization of the brain extracellular space. These pH shifts are markedly increased by acetazolamide, an inhibitor of carbonic anhydrase. Although this effect can be readily explained by inhibition of extracellular carbonic anhydrase, this enzyme has been considered strictly intracellular in the central nervous system. To determine whether these alkaline shifts are regulated by extracellular carbonic anhydrase, we studied the effects of a membrane impermeant, dextran-bound inhibitor of this enzyme. Extracellular alkaline tra

Transparent indium-tin oxide electrode patterns for extracellular, multisite recording in neuronal cultures.

Glass plates coated with transparent thin film conductors of indium-tin oxide (ITO), 100 nm thick and 10 microns wide, have been successfully used to record spike potentials from neuronal monolayer cultures. The material is non-toxic to mammalian spinal neurons and is stable under warm culture medium. Laser-deinsulated recording craters that expose 100 mu m2 of ITO yield impedances of 8-10 M omega at 1 kHz with noise levels of 40 muV. Conventional gold plating of the craters reduces these impedances to below 3 M omega. The material is easily etchable and sputtered glass plates of high quality

Link between the ciliary process and lens equatorial current.

Circadian and seasonal rhythmic changes in the intraocular pressure are modulated by the nervous system and expressed in the eye by the release of norepinephrine at adrenergic nerve endings. This study links seasonal changes and the effects of the topical application of norepinephrine with changes in the equatorial current of the lens in frogs. A vibrating probe and microelectrodes were used to study the segment of the K+ electrical loop extending between the internal microelectrode, used for measuring potential difference (PD), and the external vibrating electrode, measuring the equatorial cu

Properties of cAMP-induced transmembrane current in mollusc neurons.

The effects of different substances affecting some or other chains of cAMP metabolism on the properties of transmembrane current induced by cAMP injection have been studied in Helix pomatia neurons. It was found that preinjection of papaverine or adenosine triphosphate from a microelectrode into the neuron increases the cAMP-current amplitude. Injection of dibutyryl-cAMP does not induce transmembrane current by itself but results in a noticeable reversible inhibition of cAMP-current. Extracellular administration of trifluoperazine produces either an increase or a decrease of the cAMP-current a

Experience and safety of intraoperative Neuropixels: a case series of 56 patients.

OBJECTIVE: The Neuropixels probe, a high-density silicon microelectrode array, has been a transformative tool for extracellular recording of large numbers of single neurons across animal models. Traditional tungsten microelectrodes available for clinical neurophysiology typically only record 1-2 neurons at a given time. Human intraoperative Neuropixels recording increases access to single-neuron spiking by an order of magnitude and is poised for rapid adoption. The authors' objective was to determine the safety and yield of intraoperative Neuropixels for large-scale simultaneous neuronal recor

Evaluation of changes in intrarenal oxygenation in rats using multiple gradient-recalled echo (mGRE) sequence.

Changes in intrarenal oxygenation in rats during pharmacological stimuli were evaluated with a multiple gradient-recalled echo (mGRE) sequence. With administration of the loop diuretic furosemide, oxygenation in the medulla improved; acetazolamide, a proximal tubular diuretic, produced no significant change. These results are consistent with our previous studies in humans and resemble earlier studies of medullary oxygenation using oxygen microelectrodes in anesthetized rats. The technique may be useful in the evaluation of therapeutic strategies in animal models of pathophysiological states su

The intracellular chloride activity of glomus cells in the isolated rabbit carotid body.

The resting membrane potential (EM) and intracellular chloride activity (ai(CI] of glomus cells were measured in isolated rabbit carotid bodies using conventional and chloride-selective microelectrodes. EM and ai(CI) were -25.9 +/- 1.4 mV (mean +/- S.E.M., n = 13) and 20.9 +/- 1.9 mV (mean +/- S.E.M., n = 13), respectively. When the chloride equilibrium potential (ECI) was shifted in a positive direction, EM depolarized. It is concluded that chloride ions are not passively distributed across the glomus cell membrane and that they play an important role in determining EM.

Intraoperative Stereotactic Magnetic Resonance Imaging for Deep Brain Stimulation Electrode Planning in Patients with Movement Disorders.

OBJECTIVE: Implantation of deep brain stimulation (DBS) electrodes requires stereotactic imaging. Stereotactic magnetic resonance imaging (MRI) for DBS surgery has become more popular and intraoperative MRI scanners have become more available. We report on our cohort of movement disorder patients who underwent intraoperative stereotactic MRI-only DBS electrode implantation. METHODS: A review of our DBS database for eligible patients over a study period of 8 years was performed. Stereotactic accuracy was calculated as a directional error and the Euclidean distance between planned and controlle

[Effect of stimulation of splanchnic mechanoreceptors on the activity of purkinje cells (author's transl)].

Climbing and mossy fibre activity in Purkinje cells of cat cerebellum (vermis part of lobules V and VI) were recorded by means of extracellular microelectrodes. Responses were obtained by stimulation of different types of mechanoreceptors in the gastro-intestinal tract: muscular receptors connected with C fibres, peritoneal movement receptors connected with A gamma delta or B fibres, Pacinian corpuscules connected with A beta fivres. The characteristics of the splanchnic fibre projections on this cerebellar area were defined. Splanchno-somatic, splanchno-visceral, and splanchno-cortical conver

Interictal Spike and Loss of Hippocampal Theta Rhythm Recorded by Deep Brain Electrodes during Epileptogenesis.

Epileptogenesis is the gradual dynamic process that progressively led to epilepsy, going through the latent stage to the chronic stage. During epileptogenesis, how the abnormal discharges make theta rhythm loss in the deep brain remains not clear. In this paper, a loss of theta rhythm was estimated based on time-frequency power using the longitudinal electroencephalography (EEG), recorded by deep brain electrodes (e.g., the intracortical microelectrodes such as stereo-EEG electrodes) with monitored epileptic spikes in a rat from the first region in the hippocampal circuit. Deep-brain EEG was c

Enhanced characteristics and mechanism of Cu(II) removal from aqueous solutions in electrocatalytic internal micro-electrolysis fluidized-bed.

For the purification of heavy metal wastewater, internal micro-electrolysis (IME) was considered as an effective method but some disadvantage greatly restricts its application. Electrocatalytic internal micro-electrolysis (ECIME) fluidized bed using iron-carbon particles was proposed to avoid disadvantaging of IME. The principal aim of this study was to investigate the enhanced removal characteristics, mechanism, and kinetic behavior of Cu(II) that none clear before. ECIME reactor shows a better copper removal performance and depends much on the polarization of the external electric field (EEF
