Characterization and sequence analysis of extended-spectrum-{beta}-lactamase-encoding genes from Escherichia coli, Klebsiella pneumoniae, and Proteus mirabilis isolates collected during tigecycline phase 3 clinical trials.

In concert with the development of novel beta-lactams and broad-spectrum cephalosporins, bacterially encoded beta-lactamases have evolved to accommodate the new agents. This study was designed to identify, at the sequence level, the genes responsible for the extended-spectrum-beta-lactamase (ESBL) phenotypes of Escherichia coli, Klebsiella pneumoniae, and Proteus mirabilis isolates collected during the global tigecycline phase 3 clinical trials. PCR assays were developed to identify and clone the bla(TEM), bla(SHV), bla(OXA), and bla(CTX) genes from clinical strains. Isolates were also screene

Evaluation of Different Activity of Lactobacillus spp. against Two Proteus mirabilis Isolated Clinical Strains in Different Anatomical Sites In Vitro: An Explorative Study to Improve the Therapeutic Approach.

Urinary tract infections (UTIs) and catheter-associated UTIs (CAUTIs) are the principal hospital-acquired infections. Between these, bacterial prostatitis is believed to be the leading cause of recurrent UTIs in men under 50 years of age and is often unresponsive to antibiotic treatment. Proteus mirabilis is more commonly associated with UTIs in these abnormalities, especially in patients undergoing catheterization. Lactobacillus spp. are an important component of the human microbiota and occur in large quantities in foods. Probiotics are proposed as an alternative to antibiotic therapy in the

The new variant of Salmonella genomic island 1 (SGI1-V) from a Proteus mirabilis French clinical isolate harbours blaVEB-6 and qnrA1 in the multiple antibiotic resistance region.

OBJECTIVES: The clinical strain of Proteus mirabilis VB1248 isolated from a blood culture in August 2009 was multiresistant (i.e. resistant to β-lactams, fluoroquinolones, aminoglycosides and sulphonamides). We searched for the presence of a Salmonella genomic island 1 (SGI1). METHODS: The whole genetic structure surrounding the genes involved in antibiotic resistance was characterized by PCR or gene walking followed by DNA sequencing. RESULTS: The new variant SGI1-V (42.9 kb) was located downstream of the thdF chromosomal gene. Genes sharing homology with phage-related genes were detected o

Bacteriocin isolated from Ralstonia mannitolilytica and bacteriocin-capped silver nanoparticles: Comparative effects on biofilm formation and LuxS Gene's expressions by Proteus mirabilis as an approach to counter MDR catheter infection.

Among undesirables in treating certain infections and diseases is the contamination of catheters, especially of the microbes' resistance to drugs. The situation has necessitated the search for alternative antimicrobial agents. Bacteriocin category, antibiotic-originate, peptide-natured, Ralstonia mannitolilytica microbes-produced, bacteriocin material, and the semi-pure bacteriocin capped silver metal nanoparticles (AgNPs) were used for combating the MDR (multi drug resistance) organism, Proteus mirabilis, which is the third-most common cause of UTI (urinary tract infection), and that is linke

Expression profiling of Wnt/β-catenin, antimicrobial peptides, endoplasmic reticulum stress, and apoptosis signaling pathways and antioxidant/oxidant status in Oreochromis niloticus following Proteus mirabilis challenge: the palliative role of naringenin flavanone.

The most prevalent diseases in fish farming are bacterial infections, which drive considerable financial losses to the aquaculture sector. Figuring out an innovative, eco-friendly antimicrobial is becoming more crucial, especially in light of the escalating resistance of pathogens to the existing pharmaceuticals. Accordingly, this work is the inaugural attempt to look into the antibacterial attributes of a flavanone [naringenin (NG) dissolved in dimethyl sulfoxide (DMSO)] in assisting Oreochromis niloticus (Nile tilapia) to combat a bacterial challenge (Proteus mirabilis). This study was carri

Species interactions in mixed-community crystalline biofilms on urinary catheters.

Previous experimental investigations of the crystalline biofilms that colonize and block urinary catheters have focussed on their formation by pure cultures of Proteus mirabilis. In the urine of patients undergoing long-term catheterization, P. mirabilis is commonly found in mixed communities with other urinary tract pathogens. Little is known about the effect that the other species have on the rate at which P. mirabilis encrusts catheters. In the present study, a set of data on the nature of the bacterial communities on 106 catheter biofilms has been analysed and it was found that while speci

Coriander (Coriandrum sativum L. leaves) improves brain and gut pathology in mouse models of brain-first and gut-first Parkinson's disease.

Recently, an increasing number of studies have focused on the microbiota-gut-brain axis in Parkinson's disease (PD), and a new perspective has emerged regarding the regulation of gut health and the microbiome as potential treatments for PD. We focused on coriander, which is consumed worldwide and exhibits significant anti-inflammatory and antimicrobial activities. In this study, we investigated whether coriander ameliorated PD phenotypes in both brain-first and gut-first PD models. C57BL/6J mice were treated with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) or Proteus mirabilis (P. mira

A shift from acute to chronic spontaneous pyelonephritis in male MM mice associated with a change in the causal micro-organisms.

Proteus mirabilis was the predominant cause of acute diabetes-associated pyelonephritis occurring spontaneously in male MM mice until they were segregated in a new environment. Thereafter Pasteurella pneumotropica and Streptococcus faecalis emerged collectively as the most common causal organisms, the pyelonephritis became more chronic and Proteus mirabilis isolates from faeces and urine produced atypical non-swarming colonies on blood agar plates. This did not account for the reduced pathogenicity of Proteus mirabilis; when MM males were returned to the original environment the pyelonephritis

Lactobacillus taiwanensis BCRC17755 alleviates motor dysfunction and dopaminergic neuronal loss in mouse models of Parkinson's disease.

Parkinson's disease (PD) is a complex progressive neurodegenerative disease characterized by both motor and nonmotor symptoms such as constipation and dyspepsia. Recently, growing evidence has suggested that a specific gut microbiome is associated with the pathophysiology of PD through the microbiota-gut-brain axis. We previously discovered that Proteus mirabilis induced motor impairment and brain dopaminergic neurodegeneration in normal mice. In this context, exploring beneficial microbe would be reasonable strategy to treat PD fundamentally. With that the current study aimed to evaluate whet

Three enzymatic steps required for the galactosamine incorporation into core lipopolysaccharide.

The core lipopolysaccharides (LPS) of Proteus mirabilis as well as those of Klebsiella pneumoniae and Serratia marcescens are characterized by the presence of a hexosamine-galacturonic acid disaccharide (αHexN-(1,4)-αGalA) attached by an α1,3 linkage to L-glycero-D-manno-heptopyranose II (L-glycero-α-D-manno-heptosepyranose II). In K. pneumoniae, S. marcescens, and some P. mirabilis strains, HexN is D-glucosamine, whereas in other P. mirabilis strains, it corresponds to D-galactosamine. Previously, we have shown that two enzymes are required for the incorporation of D-glucosamine into the core

Some observations on the diffusion of antimicrobial agents through the retention balloons of foley catheters.

PURPOSE: We examined the ability of antimicrobial agents to diffuse through the retention balloons of urinary catheters and inhibit their encrustation by Proteus mirabilis. MATERIALS AND METHODS: An agar diffusion screening test was developed to detect agents capable of diffusing through catheter balloons and inhibiting the growth of P. mirabilis. The effect of inflating the balloons with antibacterials on the ability of P. mirabilis to encrust catheters was tested in laboratory models of the catheterized bladder. RESULTS: Of 18 antimicrobial agents active on P. mirabilis only mandelic acid,

Detection of SGI1/PGI1 Elements and Resistance to Extended-Spectrum Cephalosporins in Proteae of Animal Origin in France.

Proteae, and especially Proteus mirabilis, are often the cause of urinary tract infections (UTIs) in humans. They were reported as carriers of extended-spectrum β-lactamase (ESBL) genes, and recently of carbapenemases, mostly carried by the Salmonella genomic island 1 (SGI1) and Proteus genomic island 1 (PGI1). Proteae have also lately become an increasing cause of UTIs in companion animals, but antimicrobial susceptibility data in animals are still scarce. Here, we report the characterization of 468 clinical epidemiologically unrelated Proteae strains from animals collected between 2013 and 2
