Genetic markers and animal models of neurocristopathy.

Neurocristopathy is the disorder in which the series of cell and tissue derived from the neural crest are affected. A variety of neural crest tumors, and systemic neurocristopathies such as neurofibromatosis type 1 (NF1) and type 2 (NF2), multiple endocrine neoplasia type 2 (MEN2) and dysplastic nevus syndrome are included in this category. Genetic abnormalities of specific proto-oncogenes and tumor suppressor genes have been discovered in the neurocristopathies. The NF1 gene has GTP-ase activating protein activity, regulates ras pathway and acts as a potential tumor suppressor. The NF2 gene,

Animal models of spontaneous diabetic kidney disease.

Kidney disease, characterized by proteinuria and glomerular lesions, is a common complication of spontaneous diabetes mellitus in many animal species. It occurs in animals with hypoinsulinemia, hyperinsulinemia, or impaired glucose tolerance. The renal functional and structural abnormalities in spontaneously diabetic animals resemble human diabetic nephropathy in many respects. Mesangial expansion and glomerular basement membrane thickening, two structural hallmarks of diabetic glomerulopathy in humans, are the most frequently encountered lesions in animals. In addition, a nodular form of mesa

Animal models in the study of vomiting.

The emetic responses to various pharmacological agents, cytotoxins, and radiation are compared among animal species. The species included for comparison are the human, nonhuman primate, dog, cat, and ferret. The categories of pharmacologic compounds include both those compounds that act on identified membrane receptors (e.g., cholinergic agonists, catecholamines, and neuroactive peptides) and those that act on unidentified receptors (e.g., cardiac glycosides and Veratrum alkaloids, among others). Emphasis is placed on emetic dose-response relations and threshold ED50 and ED100 values calculate

Animal models of intravenous phencyclinoid self-administration.

Phencyclidine (1-(1-phenylcyclohexyl)piperidine, PCP) self-administration has been demonstrated in rhesus monkeys, baboons, dogs and rats. Generally, an orderly inverted U-shaped dose-response curve for rates of self-injection has been observed. Total drug intake appears to increase slightly with increases in unit dose; however, this increase is much less than might be expected with other CNS depressants such as barbiturates or morphine. Additionally, several arylcyclohexylamine analogues of PCP and some members of the benzomorphan and dioxolane classes referred to collectively as "phencyclino

Laboratory animal models for human scrub typhus.

A wide variety of animals have been utilized in an attempt to provide the information necessary to bring scrub typhus to the point where it is no longer a threat to man. The laboratory mouse is usually the animal of choice for the study of this disease. The discovery that certain strains of inbred mice are genetically resistant to Rickettsia tsutsugamushi, the agent of scrub typhus, has opened new avenues in the study of the immune response to the disease. The cynomolgus monkey, Macaca fascicularis, appears to be the best animal model for the study of scrub typhus as it occurs in humans and sh

Analysis of animal models of macular edema.

Various models of macular edema have been studied; however, frank development of a prototypical cystoid macular edema has not been observed. In humans, cystoid edema is frequently observed in association with other disturbances of the retina. Thus, a basic drawback of the animal models may be that an otherwise healthy retina is capable of resolving the experimentally produced edema, thereby preventing chronic cystoid maculopathy. A review of macular edema models and of experimental retinal and brain edema investigations suggests that blood-retinal (blood-brain) barrier permeability abnormaliti

Animal models of anxiety and benzodiazepine actions.

If rats trained to press a lever for food, then receive a shock to the feet following every response, their behavioural output is severely depressed. This procedure is termed immediately punishment and it was used by Geller and Seifter in the task devised to demonstrate the anxiolytic effect of benzodiazepines. These drugs and a number of others with anxiolytic activity (e. g. barbiturates, ethanol) reverse the suppression induced by the presentation of a highly aversive stimulus, like electric shock. The Geller-Seifter procedure has figured prominently in behavioural studies of benzodiazepine

Animal models of gastrointestinal and liver diseases. Animal models of cystic fibrosis: gastrointestinal, pancreatic, and hepatobiliary disease and pathophysiology.

Multiple organ systems, including the gastrointestinal tract, pancreas, and hepatobiliary systems, are affected by cystic fibrosis (CF). Many of these changes begin early in life and are difficult to study in young CF patients. Recent development of novel CF animal models has expanded opportunities in the field to better understand CF pathogenesis and evaluate traditional and innovative therapeutics. In this review, we discuss manifestations of CF disease in gastrointestinal, pancreatic, and hepatobiliary systems of humans and animal models. We also compare the similarities and limitations of

Tay-Sachs disease models: From cellular and animal models to treatment perspectives.

Tay-Sachs disease (TSD) is a lysosomal storage disorder caused by pathogenic variants in the HEXA gene, resulting in deficient activity of β-hexosaminidase A (Hex-A). Loss of Hex-A function leads to the intralysosomal accumulation of GM2 ganglioside, disrupting lysosomal homeostasis and triggering neurodegenerative processes, including neuronal death, demyelination, neuroinflammation, gliosis, and microglial activation. Current understanding of TSD pathophysiology, as well as the development and preclinical evaluation of therapeutic strategies, has relied heavily on a wide range of cellular an

Transgenic mouse models of tauopathies: prospects for animal models of Pick's disease.

Because filamentous neuronal tau inclusions are neuropathologic hallmarks of Pick's disease and a number of other neurodegenerative disorders known as tauopathies, the authors generated lines of transgenic (Tg) mice that overexpress the shortest human tau isoform in the CNS. These Tg mice accumulated argyrophilic and filamentous tau immunoreactive neuronal inclusions with age in cortex and brainstem as well as in spinal cord, where these inclusions were most abundant and associated with gliosis. The ventral roots of affected spinal cord segments showed axonal degeneration, whereas residual axo

Animal models to study the pathogenesis of human and animal Clostridium perfringens infections.

The most common animal models used to study Clostridium perfringens infections in humans and animals are reviewed here. The classical C. perfringens-mediated histotoxic disease of humans is clostridial myonecrosis or gas gangrene and the use of a mouse myonecrosis model coupled with genetic studies has contributed greatly to our understanding of disease pathogenesis. Similarly, the use of a chicken model has enhanced our understanding of type A-mediated necrotic enteritis in poultry and has led to the identification of NetB as the primary toxin involved in disease. C. perfringens type A food p

Animal models of disease: feline hyperthyroidism: an animal model for toxic nodular goiter.

Since first discovered just 35 years ago, the incidence of spontaneous feline hyperthyroidism has increased dramatically to the extent that it is now one of the most common disorders seen in middle-aged to senior domestic cats. Hyperthyroid cat goiters contain single or multiple autonomously (i.e. TSH-independent) functioning and growing thyroid nodules. Thus, hyperthyroidism in cats is clinically and histologically similar to toxic nodular goiter in humans. The disease in cats is mechanistically different from Graves' disease, because neither the hyperfunction nor growth of these nodules depe

Research progress on animal models of autoimmune gastritis.

Autoimmune gastritis (AIG) is an autoimmune disease characterized primarily by the destruction of gastric parietal cell structure and atrophy of the gastric fundic and body mucosa. The global prevalence of AIG is approximately 3.85%. Its main complications include pernicious anemia, gastric neuroendocrine tumors, and gastric cancer, which pose significant health risks. Currently, targeted treatment options for AIG are lacking worldwide. Animal models of AIG are crucial for investigating its pathogenesis and for developing drug therapies. However, reproducible methods for establishing AIG anima

Animal models for calcium oxalate kidney stone research.

Calcium oxalate (CaOx) stones are the most prevalent form of nephrolithiasis and are characterized by a substantial risk of recurrence. Their pathogenesis arises through distinct etiological pathways, including idiopathic metabolic susceptibility, monogenic disorders such as primary hyperoxaluria, enteric hyperoxaluria, and toxicant-associated injury, and is shaped by two primary papillary mechanisms: Randall's plaque formation and ductal plugging. A central translational limitation is that most experimental systems, particularly chemically induced rodent models, rely on acute hyperoxaluria. T

Advances in animal models of pigmentary skin disorders.

Pigmentary cutaneous disorders are characterized by abnormal decreases or increases in melanin production, leading to altered skin coloration. These conditions include vitiligo, melasma, and post-inflammatory hyperpigmentation. The etiology of these dermatological manifestations is multifactorial, encompassing environmental triggers, hormonal fluctuations, psychological stress, and physical trauma. Although generally non-life-threatening, these disorders significantly impact patients' quality of life due to visible disfigurement and consequent psychosocial distress. This paper reviews the meth

A review of animal models of vascular dementia.

Vascular dementia (VD) is characterized by the presence of cerebrovascular lesions, specifically hemorrhagic or ischemic lesions within the brain parenchyma, with ischemic lesions being more prevalent. The resultant progressive decline in cognitive abilities and memory poses a significant challenge to public health and quality of life. Consequently, a growing number of researchers have focused on exploring the pathogenesis, prevention, and treatment strategies for VD. The establishment of suitable animal models for investigating the underlying mechanisms of VD treatment and assessing therapeut

Animal stroke models and outcome evaluation: A review.

Stroke is among the leading causes of death and disability worldwide, and despite extensive research, prevalence is increasing. This has been partly attributed to systemic flaws in experimental design principles in studies involving stroke, and much work has been done to improve the standards of study design. In this article, different animal models mimicking ischemic and hemorrhagic stroke will be reviewed. Historical models will be discussed, as well as the more commonly employed modern techniques, such as thromboembolic occlusion and intraluminal filament occlusion models. This will be foll

Animal models of Huntington's disease. Pros and cons.

Modelling of human neurological diseases uses a plethora of ever-more sophisticated methods and approaches. For Huntington's disease (HD), which affects specific neuronal types and circuits in the brain, this has meant the use of both neurotoxic compounds and various animal models of different complexity, ranging from rodents to non-primate ones. Genetic models are classified based on the use of specific constructs including gene including gene fragments, full-length, knock-out and knock-in models. In this review, we will discuss the available animal models for HD, highlighting their pros and
