Absence of mutagenic activity of benorylate, paracetamol and aspirin in the Salmonella/mammalian microsome test.

Benorylate and its two major hydrolysis products, paracetamol and aspirin were examined for mutagenicity in the Salmonella/mammalian microsome screening test. The compounds were tested in 6 strains of Salmonella typhimurium (TA1535, TA1537, TA1538, TA100, TA97 and TA98) in the presence and absence of a rat-liver microsome activation system. Benorylate did not show evidence of mutagenic activity in the 6 strains tested with or without metabolic activation at concentrations ranging from 0.006 to 3 mg per plate. Paracetamol and aspirin likewise did not show any evidence of mutagenic activity at c

Mutagenic and cell-transforming activities of triol-epoxides as compared to other chrysene metabolites.

The syn- and anti-isomers of the bay-region diol-epoxides of chrysene and of 3-hydroxychrysene and their metabolic precursors have been investigated for mutagenicity in Salmonella typhimurium (reversion to histidine prototrophy) and V79 Chinese hamster cells (acquirement of resistance to 6-thioguanine) and for transforming activity in M2 mouse prostate cells. Other known and potential chrysene metabolites have been included in mutagenicity experiments. Direct mutagenic activity in S. typhimurium TA 100 exhibited, in order of potency, anti-triol-epoxide greater than syn-triol-epoxide greater th

Cytotoxic and mutagenic effects of 1-nitropyrene and 1-nitrosopyrene in diploid human fibroblasts.

The cytotoxic and mutagenic effects of 1-nitropyrene (1-NP) and its reduced metabolite 1-nitrosopyrene (1-NOP) were determined in diploid human fibroblasts. Conditions for the metabolic activation of the parent compound (1-NP) by human cells in culture were developed. The cytotoxic effect of 1-NP in normal cells was compared with that for repair-deficient xeroderma pigmentosum (XP) cells, and cells from a patient with hereditary cutaneous malignant melanoma (HCMM), which we have shown earlier are abnormally sensitive to 4-nitroquinoline-1-oxide. The slope of the survival curve for XP cells was

Sister-chromatid exchanges in lymphocytes and mutagenicity in urine of nurses handling cytostatic drugs.

Sister-chromatid exchanges (SCE) in peripheral blood lymphocytes and mutagenicity of urine (Ames test) were measured in a group of 21 nurses professionally handling antineoplastic drugs and in a group of 21 unexposed controls. No differences in SCE frequencies and in urinary mutagenic activity between exposed and unexposed groups were detected. A clear positive increase in urinary mutagenicity in the TA98 Salmonella strain was observed with increasing number of cigarettes smoked, whereas no evident influence of smoking on SCE was seen. Age, coffee and alcohol consumption did not show any detec

Structure/activity studies of the nephrotoxic and mutagenic action of cysteine conjugates of chloro- and fluoroalkenes.

The cysteine conjugates of the nephrotoxins hexachlorobutadiene (HCBD), tetrafluoroethylene (TFE) and hexafluoropropene (HFP), together with those of trichloroethylene and perchloroethylene, have been chemically synthesized and a relationship determined between their structures and their nephrotoxicity and mutagenicity in vitro. All of the conjugates had a marked effect on the uptake of both the organic anion p-aminohippuric acid (PAH) and the cation tetraethylammonium bromide (TEA) into rat kidney slices, suggesting activation of the conjugates in the slices to a toxic species which interfere

Mutagenic effects of DNA-containing oncogenic viruses and malignant transformation of mammalian cells.

It was discovered in the 1970s that oncogenic viruses could induce gene mutations in mammalian cells. The phenomenon seems to be widespread: it was observed with all groups of DNA-containing viruses and some retroviruses. The mutagenic effects of the tested viruses at gene level are not locus specific. The viruses induce point mutations, including base substitutions, as well as deletions and insertions. The mutagenic effect of SV40 is controlled by the activity of the early A gene, which encodes the T antigen. Presumably, the process of integration creates the possibility for occurrence of mut

Mutagenic effects of aqueous extracts of Symphytum officinale L. and of its alkaloidal fractions.

Symphytum officinale L. (Boraginaceae) is a medicinal plant widely used in therapy. It roots, described in the Polish Pharmacopoeia as Radix symphyti, are recommended as expectorants, especially for children. Aqueous solutions of three alkaloid fractions obtained from infusions of Symphytum officinale L. root were tested for their antimitotic and mutagenic activity in meristematic cells of the lateral roots of Vicia faba L., var minor. Lasiocarpine, a proven carcinogen, served as a positive control. Mutagenic effects were induced by lasiocarpine, by the alkaloidal fraction I and by diluted inf

Mutagenicity of dichloroacetylene and its degradation products trichloroacetyl chloride, trichloroacryloyl chloride and hexachlorobutadiene.

Dichloroacetylene (DCA) is a highly reactive compound that decomposes rapidly in contact with air into a series of chlorinated aliphatic hydrocarbons (e.g., phosgene, trichloroacetyl chloride, trichloroacryloyl chloride and hexachlorobutadiene). Experiments were performed to compare the mutagenic properties of DCA and its degradation products on the histidine-dependent tester strains TA98 and TA100 of Salmonella typhimurium. In these experiments, DCA vapour was streamed under analytical control through the bacterial suspensions. DCA is soluble in aqueous solution and was stable under the exper

Mutagenicity of pesticides evaluated by means of gene-conversion in Saccharomyces cerevisiae and in Aspergillus nidulans.

Nine pesticides, Afugan, Atrazine, Benomyl, Captan, Daconil, Melthaumittel, Plantvax, Saprol, and Wepsin were tested for the induction of mitotic gene-conversion in two different eucaryotic microorganisms, Saccharomyces cerevisae and Aspergillus nidulans. In S cerevisiae the pesticides were also tested after mouse liver microsomal activation; in A nidulans all tests were performed using both resting and germinating conidia. Among the tested pesticides, only Captan revealed a consistent genetic activity, three times greater than that observed with the standard mutagen methylmethansulphonate. Th

The cytotoxic, mutagenic and clastogenic effects of chromium-containing compounds on mammalian cells in culture.

Examples of chromic and chromate salts have been examined for their effects on a cultured Chinese hamster cell line. The responses studied were cytotoxicity, mutagenesis and clastogenesis. Chromate (hexavalent chromium) salts of both high and medium water solubility were active in producing all three classes of response, whereas an insoluble chromate salt and a soluble chromic (trivalent chromium) salt were inactive. In addition to illustrating the value of using mammalian cells in culture for screening chemicals for biological activity, the results of this study reinforce current views regard

Mutagenic effects of vinyl chloride on Drosophila melanogaster with and without pretreatment with sodium phenobarbiturate.

Treatment of Drosophila males with 1--20% vinyl chloride in air gave an increased frequency of complete and mosaic recessive lethals. Pretreatment with phenobarbiturate for 24 h caused an increase in the mutagenic effect of vinyl chloride. This indicates that the mixed-function oxygenase system can be induced in Drosophila in the same way as in mammals.

Environmental carcinogens often exacerbate endogenous mutagenic processes to enhance tumor promotion.

Environmental exposures significantly influence cancer risk, but their mutational impact remains unclear. We perform whole-exome sequencing of hepatocellular carcinomas (HCCs) from B6C3F1/N mice that arise spontaneously with age (2 years old) or following chronic exposure to one of ten potential human carcinogens that operate through genotoxic or non-genotoxic mechanisms. HCCs from mice exposed to drinking water disinfection byproducts, such as bromochloroacetic acid (BCA) and bromodichloroacetic acid (BDCA), show dose-dependent increases in mutational burden, distinct mutational signatures (B

Mutagenicity of nitrosamines in enhanced Ames tests: A review of strain, metabolism, and solvent effects.

We review the results of the 'Enhanced' Ames Test (EAT) for assessment of the mutagenicity of N-nitrosamines, integrating the data from six key studies. High (>98 %) purities of the nitrosamines were confirmed by LC-MS and glutathione co-incubation to avoid false-positive results. The most sensitive strains were Salmonella typhimurium TA1535 and Escherichia coli WP2 uvrA (pKM101), which detect primarily GC→AT transitions, typically induced by O6-alkylguanine DNA adducts. Strain TA100 gave variable responses and strains TA98 and TA1537 gave negative results. Bioactivation by hamster liver S9 (p

Development of an inhibitor of the mutagenic SOS response that suppresses the evolution of quinolone antibiotic resistance.

Antimicrobial resistance (AMR) is a growing threat to health globally, with the potential to render numerous medical procedures so dangerous as to be impractical. There is therefore an urgent need for new molecules that function through novel mechanisms of action to combat AMR. The bacterial DNA-repair and SOS-response pathways promote survival of pathogens in infection settings and also activate hypermutation and resistance mechanisms, making these pathways attractive targets for new therapeutics. Small molecules, such as IMP-1700, potentiate DNA damage and inhibit the SOS response in methici

Mutagenic effects of ultraviolet radiation and trimethyl psoralen in mycoplasma toward a minimal genome.

Mycoplasmas, autonomously culturable bacteria with the smallest genome, are an important organism to understand the minimal form of life. Mutagenesis using mutagens is a useful methodology for understanding the essential regions of genomic information. Ultraviolet light (UV) and trimethyl psoralen (TMP) are mutagens known to induce various mutations; the latter is reported to specifically induce deletions in nematodes. However, their mutagenic effects on mycoplasma are not known. Here, we exposed Metamycoplasma salivarium to UV-C light or TMP and UV-A as mutagens, and analyzed the mutational p

Microsatellite break-induced replication generates highly mutagenized extrachromosomal circular DNAs.

Extrachromosomal circular DNAs (eccDNAs) are produced from all regions of the eucaryotic genome. We used inverse PCR of non-B microsatellites capable of forming hairpin, triplex, quadruplex and AT-rich structures integrated at a common ectopic chromosomal site to show that these non-B DNAs generate highly mutagenized eccDNAs by replication-dependent mechanisms. Mutagenesis occurs within the non-B DNAs and extends several kilobases bidirectionally into flanking and nonallelic DNA. Each non-B DNA exhibits a different pattern of mutagenesis, while sister clones containing the same non-B DNA also

Ecotoxicity and Mutagenicity Assessment of Novel Antifungal Agents VT-1161 and T-2307.

Antifungal substances are essential for managing fungal infections in humans, animals, and plants, and their usage has significantly increased due to the global rise in fungal infections. However, the extensive application of antifungal agents in pharmaceuticals, personal care products, and agriculture has led to their widespread environmental dissemination through various pathways, such as excretion, improper disposal, and agricultural runoff. Despite advances in wastewater treatment, many antifungal compounds persist in the environment, affecting non-target organisms and contributing to resi

The Mutagenic Plasticity of the Cholera Toxin B-Subunit Surface Residues: Stability and Affinity.

Mastering selective molecule trafficking across human cell membranes poses a formidable challenge in healthcare biotechnology while offering the prospect of breakthroughs in drug delivery, gene therapy, and diagnostic imaging. The cholera toxin B-subunit (CTB) has the potential to be a useful cargo transporter for these applications. CTB is a robust protein that is amenable to reengineering for diverse applications; however, protein redesign has mostly focused on modifications of the N- and C-termini of the protein. Exploiting the full power of rational redesign requires a detailed understandi

Unveiling Urinary Mutagenicity by the Ames Test for Occupational Risk Assessment: A Systematic Review.

Occupational exposure may involve a variety of toxic compounds. A mutagenicity analysis using the Ames test can provide valuable information regarding the toxicity of absorbed xenobiotics. Through a search of relevant databases, this systematic review gathers and critically discusses the published papers (excluding other types of publications) from 2001-2021 that have assessed urinary mutagenicity (Ames test with Salmonella typhimurium) in an occupational exposure context. Due to the heterogeneity of the study methods, a meta-analysis could not be conducted. The characterized occupations were
