ML superfamily proteins, including MD-1, MD-2, Niemann-Pick type C2 (Npc2) protein, GM2 activator protein, phosphatidylinositol/phosphatidylglycerol transfer protein (PG/PI-TP) and mite allergen Der p 2, bind to specific lipids and play important roles in lipid-recognition and metabolism. Among these ML (MD-2-related lipid-recognition) proteins, MD-2 is essential for lipopolysaccharide (LPS) signaling and the following secretion of proinflammatory factors. In this report, we identified the cDNA and gene of an ML protein from an important white shrimp Litopenaeus vannamei and named it LvML. The

The anti-inflammatory properties of a heparin-like compound from the shrimp Litopenaeus vannamei are related. Besides reducing significantly (p<0.001) the influx of inflammatory cells to injury site in a model of acute inflammation, shrimp heparin-like compound was able to reduce the matrix metalloproteinase (MMPs) activity in the peritoneal lavage of inflamed animals. Moreover, this compound also reduced almost 90% the activity of MMP-9 secreted by human activated leukocytes. Negligible anti-coagulant activities in aPPT assay and a poor bleeding potential make this compound a better alternati

Ozone nanobubbles (NB-O3) have recently emerged as a promising technology in aquaculture for improving water quality and disease control. However, their impacts on shrimp remain poorly understood, particularly regarding their effects on biochemical processes. This study evaluated the effects of NB-O3 on growth performance, immune responses, oxidative stress, and HSP70 gene expression in whiteleg shrimp (Penaeus vannamei). Shrimp were cultured for 21 days in control and NB-O3-treated tanks. Growth indices, water quality variables, total hemocyte counts (THC), antioxidant enzyme activities (cata

Vibrio cholerae, a globally significant pathogen, causes both endemic and epidemic cholera and has also been associated with sporadic gastroenteritis and foodborne infections. Shrimp exports are a key source of revenue and employment in Bangladesh. However, Vibrio outbreaks pose serious public health and socio-economic risks through seafood consumption and the spread of antibiotic-resistant genes from aquaculture. The current study investigates the genomic and pathogenic features of the V. cholerae strain associated with seafood, with particular focus on the aquaculture environment of Banglade

This study investigates the enhancement of chitosan-based films by incorporating polyethylene glycol (PEG), deep eutectic solvents (DES), and a DES formulation containing astaxanthin extracted from shrimp shells (DES-Ast). The films were evaluated for visual, structural, mechanical, and functional properties. Neat chitosan films (Ch) exhibited brittleness, high crystallinity (crystallinity index - CrI ∼77.78 %), and rough surfaces (root mean square roughness - Rq = 27.82 nm). The incorporation of PEG and DES reduced surface roughness and crystallinity, with DES demonstrating a stronger plastic

Contamination of seafood products with multi-drug-resistant (MDR) bacteria is considered to be a potential source for the spread of MDR bacteria in communities. However, little is known about the extent of the contamination of seafood, in particular shrimp, with MDR bacteria in Saudi Arabia. In this study, imported frozen shrimp in retail markets were examined for the antimicrobial susceptibility patterns of Escherichia coli. Escherichia coli was isolated from 40 frozen shrimp samples; a total of 25 and 15 shrimp samples were imported from China and Vietnam, respectively. Of the 40 examined fr

Hippo signaling pathway is a serine threonine kinase cascade that is evolutionary conserved with well-established roles in organ size control, development, tumorigenesis and immunity. As its core molecule, Yorkie also plays an important role against pathogen. In this study, we cloned and characterized a Yorkie homolog from Litopenaeus vannamei, designed as LvYKI, which has a 1650 bp open reading frame. It has the characterized domains of Yokie family, and displayed to be close to the insects and crustacean. Quantitative Real-time PCR showed that LvYKI had different regulatory mechanisms in dif

Microbial communities greatly affect rearing water quality and the larvae health during shrimp hatchery periods. In this study, we investigated the microbial communities of rearing water and larvae of Litopenaeus vannamei after treating hatchery water with different kinds of chemical disinfectants: no disinfectants (Con), chlorine dioxide (ClO2), formaldehyde solution (HCHO), bleach powder (CaClO), and iodine (I2). The water and larval samples were collected from nauplius 6 (N6), zoea 1 (Z1), mysis 1 (M1), and postlarvae 1 (P1) shrimp growth periods. 16S rDNA high-throughput sequencing reveale

The PISCF-allatostatins (Manduca sexta- or C-type allatostatins) are a family of pentadecapeptides characterized by a pyroglutamine blocked N-terminus, an unamidated-PISCF C-terminus, and a disulfide bridge between two internal Cys residues. Several isoforms of PISCF-AST are known, all from holometabolous insects. Using a combination of transcriptomics and mass spectrometry, we have identified the first PISCF-type peptides from a non-insect species. In silico analysis of crustacean ESTs identified several Litopenaeus vannamei (infraorder Penaeidea) transcripts encoding putative PISCF-AST precu

Outbreaks of infectious diseases have resulted in high mortality and huge economic losses in penaeid shrimp culture. Interest in understanding shrimp immunity has increased because of its importance in disease control. Here we report cDNA cloning of a Toll receptor from the white shrimp Litopenaeus vannamei. L. vannamei Toll (lToll) is 926 residues, with a putative signal peptide of 19 residues. The protein contains distinct structural/functional motifs of the Toll like receptor (TLR) family, including an extracellular domain containing 16 leucine-rich repeats (LRRs) flanked by cysteine-rich m

This study used a non-covalent complex of whey protein (WP) and caffeic acid (CA) as an interface-stabilizing agent. The interaction between WP and CA was analyzed through spectroscopy and molecular dynamics simulations. A Pickering emulsion (WC-PE) with oregano essential oil (OEO) was created using the WP-CA complex and characterized. This complex was then combined with sodium alginate (SA) to produce calcium alginate gel beads (WC-PE-CAB), which were tested for their ability to preserve shrimp freshness. FT-IR spectroscopy and molecular dynamics showed that CA mainly binds to β-lactoglobulin

In this study, a dual-driven smart and active packaging film was developed by incorporating bovine hide/tannic acid/L-cysteine carbon dots (BTC-CDs) and oxidized Roselle extracts (ORE) into a collagen matrix, aiming to integrate active preservation with dual-mode freshness sensing. BTC-CDs were synthesized via a hydrothermal method, while ORE was prepared through laccase-catalyzed oxidation. Structural characterization revealed that BTC-CDs and ORE primarily interacted with the collagen through Schiff base covalent bonding and hydrogen bonding, resulting in a compact structure of film, which s

Ca2+ homeostasis regulates endoplasmic reticulum (ER) stress and lipid metabolism and is involved in the stress response in mammals. However, how these regulatory pathways respond in aquatic animals exposed to stressful environmental conditions remains unclear. In this study, we used siRNA to inhibit the expression levels of Bip, SERCA, IP3R, and MCU, which are involved in regulating Ca2+ homeostasis in the hepatopancreas of Penaeus vannamei. An ammonia stress experiment was subsequently conducted (stress concentration of 11.3 mg/L) to measure parameters associated with ER stress, Ca2+ homeost

Immunometabolic reprogramming is a hallmark of host defense. Recent studies have shown that hemocyanin regulates the tricarboxylic acid (TCA) cycle in shrimp to enhance immune competence, yet its role in glycolytic regulation remains unclear. Here, we show that RNA interference-mediated silencing of hemocyanin in Penaeus vannamei markedly reduces the expression of glyceraldehyde-3-phosphate dehydrogenase (GAPDH), a key glycolytic enzyme. Hemocyanin knockdown also decreases GAPDH protein abundance and enzymatic activity, accompanied by reduced levels of 1,3-bisphosphoglycerate and NADH. Althoug

BACKGROUND: Due to its rapid lethal effect in the early development stage of shrimp, acute hepatopancreatic necrosis disease (AHPND) has been causing great economic losses, since its first outbreak in southeast China in 2009. Vibrio parahaemolyticus, carrying the pirA and pirB toxin genes is known to cause AHPND in shrimp. The overall objective of this study was to sequence the whole genome of AHPND positive V. parahaemolyticus strains isolated from shrimp (Peneaus monodon) of the south-west region of Bangladesh in 2016 and 2017 and characterize the genomic features and emergence pattern of th
