Sepsis rapidly contributed to multiorgan failure affecting most commonly of the cardiovascular and respiratory systems and yet there were no effective therapies. The current study aimed at providing evidence on the cardioprotection of suppression of 5-Lipoxygenase (5-Lox) and identifying the possible mechanism in the mouse model of sepsis. The cecal ligation-perforation (CLP) model was applied to C57BL/6 wild-type (WT) and 5-Lox deficient (5-Lox-/-) mice to induce sepsis. 5-Lox expression was up-regulated in mouse myocardium and leukotriene B4 (LTB4) level was increased in serum after sepsis.

PURPOSE: The insulin-like growth factor (IGF) pathway is activated in hepatocarcinogenesis. Cixutumumab is a monoclonal antibody against human insulin-like growth factor-1 receptor (IGF-1R). Given the cross-talk between the IGF and VEGF pathways, we performed a phase I study of the combination of cixutumumab and sorafenib in hepatocellular cancer (HCC).
METHODS: Eligible patients with no prior systemic therapy for advanced HCC and Child-Pugh A to B7 were treated with sorafenib 400 mg BID and escalating doses of cixutumumab (2, 4, or 6 mg/kg IV weekly) in a 3 + 3 design. Dose limiting toxicity

The increased risk and prevalence of lacunar stroke and Parkinson's disease (PD) makes the search for better experimental models an important requirement for translational research. In this study we assess ischemic damage of the nigrostriatal pathway in a model of lacunar stroke evoked by damaging the perforating arteries in the territory of the substantia nigra (SN) of the rat after stereotaxic administration of endothelin-1 (ET-1), a potent vasoconstrictor peptide. We hypothesized that transplantation of neural stem cells (NSCs) with the capacity of differentiating into diverse cell types su

2-Methoxyestradiol (2ME2), a naturally occurring metabolite of estradiol, is known to have antiproliferative, antiangiogenic, and antiproapoptotic activities. Mechanistically, 2ME2 has been shown to downregulate hypoxia-inducible factor 1alpha (HIF-1alpha). We hypothesized that hypoxia in the major cerebral arteries might activate a unique signaling pathway, hypoxia-inducible factor-1alpha (HIF-1alpha), to produce or enhance cerebral vasospasm after subarachnoid hemorrhage (SAH). Sprague-Dawley male rats (n = 70) were randomly divided into 5 groups: Sham operated, SAH without treatment, SAH tr

Intermittent explosive disorder (IED), as described in DSM-5, is the categorical expression of pathological impulsive aggression. Previous work has identified neurobiological correlates of the disorder in patterns of frontal-limbic brain activity and dysregulation of serotonergic neurotransmission. Given the importance of short- and-long range white matter connections of the brain in social and emotional behavior, studies of white matter connectivity in impulsive aggression are warranted. Diffusion tensor imaging (DTI) studies in the related conditions of antisocial and borderline personality

BACKGROUND: Initial antibiotics with planned interval appendectomy (interval AP) have been used to treat patients with complicated perforated appendicitis; however, little experience exists with this approach in children with suspected acute perforated appendicitis (SAPA). We sought to determine the outcome of initial antibiotics and interval AP in children with SAPA.
METHODS: Over an 18-month period, 751 consecutive patients underwent appendectomy including 105 patients with SAPA who were treated with initial intravenous antibiotics and planned interval AP ≥ 8 weeks after presentation. All S

Adenosine 3',5'-cyclic monophosphate (cAMP) is known to stimulate exogenous IsK channel current in the Xenopus oocyte expression system. The present study was performed to determine whether elevation of cytosolic cAMP in a native mammalian epithelium known to secrete K+ through endogenously expressed IsK channels would stimulate K+ secretion through these channels. The equivalent short circuit current (Isc) across vestibular dark cell epithelium in gerbil was measured in a micro-Ussing chamber and the apical membrane current (IsK) and conductance (gIsK) of IsK channels was recorded with both t

This study aimed to investigate the effects of lipocalin-2 (LCN2) gene knockout (LCN2KO) on cecal ligation and perforation (CLP)-induced lung injury and ferroptosis in septic mice, and further to clarify the potential role of the Toll-like receptor 4 (TLR4)/nuclear factor κB (NF-κB) pathway. A CLP model was established using LCN2KO mice. The levels of serum interleukin-6 (IL-6) and IL-1β were measured by ELISA. The contents of malondialdehyde (MDA) and glutathione (GSH) in lung tissue were determined using the corresponding assay kits. Reactive oxygen species (ROS) levels in lung tissue were d

Stress adversely affects various aspects of the female reproductive process, including follicle development, maturation, ovulation, luteal activity, and embryo implantation. Restraint stress reduces the number of implantated embryos. The precise mechanisms by which stress affects embryo implantation are not fully understood. In this study, a mouse model of restraint stress was used to investigate the effects of stress on uterine receptivity and the underlying mechanisms. Mice were randomly divided into three groups. The groups were the Control group, Stress group, and Ab-IL-6 treatment group.

Subarachnoid hemorrhage (SAH) is a severe form of hemorrhagic stroke associated with high morbidity and mortality. There are limited pharmacological treatment options available to prevent or treat secondary neurologic injury after SAH. We have previously shown that the immunomodulatory agent fingolimod (FTY720) reduces early adhesion of leukocytes to pial venules, preserves pial arteriolar dilating function, and improves neurologic outcome in a rodent model of SAH. However, the direct effect on cells residing within the brain is less well understood. We employed the endovascular perforation mo

BACKGROUND: About 5%-10% of the world's population is affected by gastric ulcers, which can result in gastrointestinal perforation and bleeding. Consequently, we aimed to investigate whether blocking TNF-α type 1 receptor (TNFR1) with CAY10500 could diminish experimentally induced gastric ulcer (GU) in rats by modulating vascularization.
METHODS: Rats were administered with a single oral dose of 80 mg/kg of indomethacin to produce gastric ulcers. Subsequently, some rats were given 1 mg/kg of CAY10500 orally. Gastric samples were used to assess the genetic expression and protein levels of TNFR

Nanofiltration (NF) is an effective process for micro-/nano-plastics (MNPs) interception, but the impact of accumulated MNPs on the microbial community structure and metabolic pathways of biofilms on NF membranes remains unclear. This provides uncertainty with respect to membrane biofouling behavior and the risks to efficient NF operations. In this study, the size-dependent (20 nm-25 μm) and concentration-dependent (0.1-50 mg·L-1) effects of MNPs on the biofouling of a NF membrane treating secondary wastewater effluent were studied. Three MNPs-tolerant, hypermetabolic and polystyrene-degradabl

BACKGROUND: Subarachnoid hemorrhage (SAH), a severe subtype of stroke, is characterized by notably high mortality and morbidity, largely due to the lack of effective therapeutic options. Although the neuroprotective potential of PPARg and Nrf2 has been recognized, investigative efforts into oroxin A (OA), remain limited in preclinical studies.
METHODS: SAH was modeled in vivo through filament perforation in male C57BL/6 mice and in vitro by exposing HT22 cells to hemin to induce neuronal damage. Following the administration of OA, a series of methods were employed to assess neurological behav

Obstructive sleep apnea can worsen the prognosis of subarachnoid hemorrhage. However, the underlying mechanism remains unclear. In this study, we established a mouse model of subarachnoid hemorrhage using the endovascular perforation method and exposed the mice to intermittent hypoxia for 8 hours daily for 2 consecutive days to simulate sleep apnea. We found that sleep apnea aggravated brain edema, increased hippocampal neuron apoptosis, and worsened neurological function in this mouse model of subarachnoid hemorrhage. Then, we established an in vitro HT-22 cell model of hemin-induced subarach

INTRODUCTION: Existing multimodal pathways for patients undergoing acute high-risk abdominal surgery for intestinal obstruction (IO) and perforated viscus (PV) have focused on rescue in the immediate perioperative period. However, there is little focus on the peri-operative pathophysiology of recovery in this patient group, as done to develop enhanced recovery pathways in elective care. Acute inflammation is the main driver of the perioperative pathophysiology leading to adverse outcomes. Pre-operative high-dose of glucocorticoids provides a reduction in the inflammatory response after surgery

Rheumatoid arthritis (RA) is a chronic autoimmune disorder and the treatment involves the use of traditional and biological disease modifying anti-rheumatic drugs (DMARDs). Recent studies have shown JAK/STAT signaling pathway as potential target for the treatment of RA. Novel JAK/STAT inhibitors viz tofacitinib and baricitinib have been recently approved by FDA for RA treatment and have attained substantial importance. However, the discernible risks of thromboembolism, gastrointestinal (GIT) perforations, hepatotoxicity and serious infections including tuberculosis, herpes zoster associated wi

Dry eye is a common sight-impairing, painful disorder characterized by disruption of the preocular tear film, whose integrity is required for ~70% of the eye's refractive power. A universal feature of clinical dry eye is hyperosmolarity of the tears resulting from their accelerated evaporation due to dysfunction of tear- and oil-producing ocular glands. A key adaptive response to dryness/hyperosmolarity is release of tear-stabilizing mucin by conjunctival goblet cells. Yet the mechanisms mediating this response to hyperosmolarity remain poorly understood. In this study of freshly excised rat c

OBJECTIVES: Neuroinflammation is closely associated with the poor prognosis of patients with subarachnoid hemorrhage (SAH). Lycopene (Lyc), a naturally occurring carotenoid, possesses multiple pharmacological effects, especially its neuroprotective function. However, the role of Lyc in SAH has never been evaluated.
METHODS: In this study, the C57BL/6 mouse SAH model was induced by the endovascular perforation method. Lyc was administered to the mice intragastrically, and tunicamycin, an endoplasmic reticulum stress (ERS) agonist, was injected intravenously. The effects of Lyc on early brain i

The escalating global energy demand underscores the critical need for advanced solutions for energy-efficient buildings. Passive thermal energy storage systems using microencapsulated phase change materials (PCMs) offer promise but face integration challenges in cementitious materials due to weakening mechanical strength, which arises from poor shell strength and weak interfacial bonding with cementitious phases. This study introduces a novel approach for synthesizing functionalized microencapsulated PCMs from fly ash-based cenospheres addressing interfacial compatibility. Cenospheres are perf
