Pharmacological inhibition of mitochondrial carbonic anhydrases protects mouse cerebral pericytes from high glucose-induced oxidative stress and apoptosis.

Diabetes-associated complications in the microvasculature of the brain are caused by oxidative stress, generated by overproduction of reactive oxygen species from hyperglycemia-induced accelerated oxidative metabolism of glucose. Pericytes, essential for the viability of the microvasculature, are especially susceptible to oxidative stress. Mitochondrial carbonic anhydrases, regulators of the oxidative metabolism of glucose, determine the rate of reactive oxygen species production and inhibition of mitochondrial carbonic anhydrases rescues glucose-induced pericyte loss in the diabetic mouse bra

Pericyte loss and endothelial activation define distinct vascular states in cerebral small vessel disease.

Cerebral small vessel disease (cSVD) is a leading cause of stroke and vascular cognitive impairment, yet the cellular mechanisms underlying microvascular dysfunction in human disease remain incompletely understood. In particular, the relationship between pericyte alterations and endothelial activation, two key features of blood-brain barrier dysfunction, remains unresolved. Here, we performed a quantitative single-vessel analysis of the human cortical microvasculature across increasing cSVD severity and ageing. Using multiplex immunohistochemistry combined with spectral unmixing and automated

Profibrotic macrophage-derived CXCL4 promotes pericyte-to-myofibroblast transition after spinal cord injury.

INTRODUCTION: Spinal cord injury (SCI) induces fibrotic scarring that impairs axonal regeneration. Pericytes contribute to scar formation via pericyte-to-myofibroblast transition (PMT), yet the mechanisms underlying PMT in SCI remain unclear. Although CXCL4, a pleiotropic chemokine, is implicated in various fibrotic disorders, its role in driving PMT post-SCI remains unexplored. OBJECTIVES: To investigate whether CXCL4 drives PMT after SCI, elucidate its mechanisms, and assess its therapeutic potential. METHODS: scRNA-seq characterized cell-type dynamics and profibrotic signals in injured mo

Loss of Pericytic Integrin-β1 Exacerbates Blood-Brain Barrier Damage and Hemorrhagic Brain Injury.

BACKGROUND: Pericytes play essential roles in blood-brain barrier regulation and stroke pathogenesis. Given that pericytes are embedded in the ECM (extracellular matrix), it is speculated that ECM-receptor interactions are involved in these functions. Integrin-β1, the most common integrin subunit that can engage multiple ECM proteins, is highly expressed in pericytes. The function of pericytic integrin β1, however, remains unknown. METHODS: To address this question, we generated brain pericyte-specific integrin-β1 knockout mice by crossing the Atp13a5 (ATPase type 13A5)-CreER with the Itgb1 f

MicroRNA210 regulated brain pericyte dysfunction exacerbates hypoxic-ischemic brain injury in neonatal mice.

BACKGROUND: While the role of pericytes in blood-brain barrier (BBB) disruption and neuroinflammation is well-established in adult neurological disorders, their contribution to neonatal brain injury is largely unexplored. Here, we investigated the role of brain pericytes in hypoxic-ischemic (HI) brain injury in the developing brain, with a particular focus on the regulatory role of pericyte-derived microRNA210 (miR210) in pericyte dysfunction. METHODS: HI brain injury was induced on postnatal day 9 transgenic mice, including Atp13a5-tdTomato brain pericyte reporter mice, pericyte-specific diph

Pericyte-derived cancer-associated fibroblasts correlate with poor survival and are enriched after chemoradiotherapy in glioblastoma.

The role of cancer-associated fibroblasts (CAFs) in glioblastoma remains unclear, as their existence in the brain tumor microenvironment is still debated, given that the normal brain parenchyma is devoid of fibroblasts. It is unclear whether cells described as CAFs represent a distinct stromal population or a transcriptional state of perivascular cells such as pericytes. The aim of this study was to determine the identity, origin, and functional relevance of CAFs in glioblastoma. We analyzed 54 single-cell RNA sequencing datasets together with 88 bulk RNA sequencing samples. We identified a co

Where and how do leukocytes stall in retinal microcirculation: A spatiotemporal analysis with a focus on junctions and pericytes.

Irregularities in cellular flow, such as stalls in capillaries, can be potential sources of capillary dysfunction, which significantly impacts oxygen delivery to the central nervous system (CNS). Leukocytes, particularly neutrophils, can act as the stalling elements due to changes in their adhesive or mechanical features. However, the dynamic features of leukocyte stalls in CNS, specifically in the retina, have not been studied in vivo in detail to disclose their association with capillary pericytes and junctions, and their direct influence on red blood cell (RBC) flow. In this study, via in v

Heterogeneity in pericyte inflammatory responses across age and species highlight the importance of human cell models.

Pericytes in the central nervous system are essential for maintaining blood-brain barrier function, regulating blood flow, modulating immune responses, and interacting closely with surrounding cells of the neurovascular unit to support brain homeostasis. Increasing evidence has highlighted their involvement in age-related neuroinflammation, where their dysfunction may contribute to sustained inflammatory states associated with neurodegenerative disorders. Here, we compared inflammatory responses to lipopolysaccharide (LPS) in primary cerebral pericytes from neonatal and adult mice and adult hu

Functionally distinct pericyte subsets differently regulate amyloid-β deposition in patients with Alzheimer's disease.

Although the concept that the blood-brain barrier (BBB) plays an important role in the etiology and pathogenesis of Alzheimer's disease (AD) has become increasingly accepted, little is known yet about how it actually contributes. We and others have recently identified a novel functionally distinct subset of BBB pericytes (PCs). In the present study, we sought to determine whether these PC subsets differentially contribute to AD-associated pathologies by immunohistochemistry and amyloid beta (Aβ) peptidomics. We demonstrated that a disease-associated PC subset (PC2) expanded in AD patients comp

Single cell-resolved study of advanced murine MASH reveals a homeostatic pericyte signaling module.

BACKGROUND & AIMS: Metabolic dysfunction-associated steatohepatitis (MASH) is linked to insulin resistance and type 2 diabetes and marked by hepatic inflammation, microvascular dysfunction, and fibrosis, impairing liver function and aggravating metabolic derangements. The liver homeostatic interactions disrupted in MASH are still poorly understood. We aimed to elucidate the plasticity and changing interactions of non-parenchymal cells associated with advanced MASH. METHODS: We characterized a diet-induced mouse model of advanced MASH at single-cell resolution and validated findings by assayin

Transplantation of hPSC-derived pericyte-like cells promotes functional recovery in ischemic stroke mice.

Pericytes play essential roles in blood-brain barrier (BBB) integrity and dysfunction or degeneration of pericytes is implicated in a set of neurological disorders although the underlying mechanism remains largely unknown. However, the scarcity of material sources hinders the application of BBB models in vitro for pathophysiological studies. Additionally, whether pericytes can be used to treat neurological disorders remains to be elucidated. Here, we generate pericyte-like cells (PCs) from human pluripotent stem cells (hPSCs) through the intermediate stage of the cranial neural crest (CNC) and

Vascular-targeted low dose photodynamic therapy stabilizes tumor vessels by modulating pericyte contractility.

Vascular-targeted low-dose photodynamic therapy (L-PDT) was shown to improve chemotherapy distribution in malignant pleural tumors such as malignant pleural mesothelioma (MPM). However, the mechanisms triggered by L-PDT on the tumor vasculature are still debated. In pericyte and endothelial cell co-cultures, we show that pericytes exhibit enhanced sensitivity towards L-PDT compared to endothelial cells, displaying actin stress fibers and cellular contraction via Rho/ROCK kinase signaling myosin light chain and focal adhesion kinase phosphorylation (MLC-P, FAK-P). We then confirm, in two separa

Vascular Pericyte Impairment and Connexin43 Gap Junction Deficit Contribute to Vasomotor Decline in Diabetic Retinopathy.

Adequate blood flow is essential to brain function, and its disruption is an early indicator in diseases, such as stroke and diabetes. However, the mechanisms contributing to this impairment remain unclear. To address this gap, in the diabetic and nondiabetic male mouse retina, we combined an unbiased longitudinal assessment of vasomotor activity along a genetically defined vascular network with pharmacological and immunohistochemical analyses of pericytes, the capillary vasomotor elements. In nondiabetic retina, focal stimulation of a pericyte produced a robust vasomotor response, which propa

Excessive glycation of pericapillary pericytic matrix components is an essential mechanism of arterial hypertension development in hyperglycemia.

In patients with essential hypertension, inhibition of glucose utilization in tissues was detected similar to that in diabetics. High levels of glycated compounds were found in cerebral, cardiac, and renal tissues of patients dead from coronary heart disease complicated and not complicated from diabetes mellitus and essential hypertension compared to victims dead from traumas. Experimental studies on rabbits and rats showed that the content of glycoconjugates in tissues did not increase in short-term (exogenous or stress-associated) hyperglycemia, while accumulation of glycoconjugates in peric

Changes in pericytes and smooth muscle cells in the kitten model of retinopathy of prematurity: implications for plus disease.

PURPOSE: Dilated and tortuous vessels (plus disease) in ROP is a grim prognostic indicator of visual outcome. The purpose of this study was to determine whether alterations in pericytes and smooth muscle cells (SMCs), are associated with the pathogenesis of ROP, including plus disease. METHODS: Kittens were exposed to either 4 (standard obliterative model) or 2 (modified model) days of hyperoxia, resulting in vaso-obliteration or localized vessel regression, respectively, and returned to room air. The modified model more closely resembles human ROP. Desmin and alpha-smooth muscle actin (SMA)

Endothelial survival factors and spatial completion, but not pericyte coverage of retinal capillaries determine vessel plasticity.

Pericyte loss and capillary regression are characteristic for incipient diabetic retinopathy. Pericyte recruitment is involved in vessel maturation, and ligand-receptor systems contributing to pericyte recruitment are survival factors for endothelial cells in pericyte-free in vitro systems. We studied pericyte recruitment in relation to the susceptibility toward hyperoxia-induced vascular remodeling using the pericyte reporter X-LacZ mouse and the mouse model of retinopathy of prematurity (ROP). Pericytes were found in close proximity to vessels, both during formation of the superficial and th

Transplantation of hiPSC-derived pericytes rescues Alzheimer's disease phenotypes in APOE4/4 mice through IGF2-rich apoptotic vesicles.

BACKGROUND: Effective therapies for Alzheimer's disease (AD) remain to be developed. APOE4 is the strongest genetic risk factor for late-onset AD. Pericyte degeneration and blood-brain barrier (BBB) disruption are thought to be early biomarkers of AD and contribute to cognitive decline in APOE4 carriers, representing potential therapeutic targets. Our previous studies have shown that pericyte transplantation is one of the most effective strategies for BBB restoration, exhibiting great therapeutic potential for APOE4-related BBB damage and AD phenotypes. METHODS: APOE4/4 mice were treated with

Expression of alpha smooth muscle actin decreases with ageing and increases upon lumen obstruction in mouse brain pericytes.

Cerebral pericytes are mural cells covering brain microvessels, organized as ensheathing, mesh and thin-strand pericytes. These latter two, together called capillary pericytes, have low levels of alpha smooth muscle actin (α-SMA), regulating basal vascular tone and applying a slow influence on cerebral blood flow. Pericytes are subject to alterations in ageing which may be even more pronounced in age-related pathologies, including microinfarcts, which usually affect a large number of vessels in the ageing brain. We modelled this condition by injecting 10 µm-size microspheres into the circulati

The α1- and β1-Subunits of Nitric Oxide-Sensitive Guanylyl Cyclase in Pericytes of Healthy Human Dental Pulp.

Nitric oxide-sensitive guanylyl cyclase (NO-GC) is a heterodimeric enzyme with an α- and a β-subunit. In its active form as an α1β1-heterodimer, NO-GC produces cyclic guanosine-3',5'-monophophate (cGMP) to regulate vasodilation and proliferation of vascular smooth muscle cells (VSMCs). In contrast to VSMCs, only a few studies reported on the expression of the NO-GC α1β1-heterodimer in human pericytes. Since NO-GC is a marker for platelet-derived growth factor-β (PDGFRβ)-positive pericytes, we investigated whether NO-GC is expressed in its active α1β1-heterodimer in pericytes of healthy human d

Acorus tatarinowii alleviates D-galactose-induced Alzheimer's-like disease cognitive impairment and Aβ-induced pericytes dysfunction in mice.

Pericytes regulate cerebral blood flow (CBF) and excess amyloid in the brain. Pericyte dysfunction may contribute to the pathology of Alzheimer's disease (AD). Acorus tatarinowii (AT), a Chinese medicine commonly used to treat AD, protects the central nervous system. However, whether AT can regulate pericyte function and ameliorate cognitive dysfunction remains unclear. We employed a novel target recognition assay, quantitative measurement of CBF, hematoxylin and eosin staining, immunofluorescence staining, and Western blot to investigate the role of AT in improving cognitive function in patie
