Alcoholic beer fermentation: impact of glutathione and peroxidase fortification on deoxynivalenol reduction.

The use of contaminated cereals in fermentation processes can elicit a marked response from yeasts in the presence of mycotoxins, as indicated by the production of defense compounds such as peroxidase (PO) and glutathione (GSH) to reduce mycotoxin damage. The objective of this study was to investigate the effect of fortifying GSH and PO levels in alcoholic fermentation as a strategy for reduce concentration deoxynivalenol (DON) during the brewing process. The QuEChERS method was validated for quantifying the mycotoxin and estimating reduction. Fermentation was conducted in a reactor containing

Molecular characterization and functional analysis of thioredoxin peroxidase-2 from Babesia duncani.

Human babesiosis caused by Babesia duncani is an emerging zoonotic disease that poses a growing threat to public health. Since the first reported case in 1991, knowledge of this pathogen has remained limited. Recent completion of the B. duncani genome sequence has accelerated research aimed at elucidating its molecular biology. During its intraerythrocytic life cycle, B. duncani is exposed to oxidative stress generated by redox reactions. In this study, we identified and characterized an antioxidant enzyme from B. duncani-thioredoxin peroxidase-2 (BdTPx-2), a member of the peroxiredoxin family

Evaluating the tissue expression of thyroid peroxidase in chronic spontaneous urticaria patients.

Chronic spontaneous urticaria (CSU) characterized by recurrent wheals and/or angioedema, remains a challenging condition. Autoallergic and autoimmune mechanisms (Type I and IIb) have been recognized for CSU pathogenesis. Studies have demonstrated the association between CSU and serum anti-thyroid peroxidase (TPO) antibodies (Abs). However, the potential for increased TPO expression within affected skin tissue of CSU patients is unclear. Patients diagnosed with CSU were recruited from the UCARE center of Mashhad University of Medical Sciences, the case group. Matched healthy individuals who und

Peroxynitrite detoxification by Trypanosoma cruzi heme peroxidase supports parasite survival in macrophages.

Trypanosoma cruzi, the causative agent of Chagas disease, must overcome a host's nitro-oxidative burst to establish the infection. Among the reactive species generated by immune cells, peroxynitrite stands out as a highly cytotoxic molecule for the parasite. Yet, the parasite's defense mechanisms against peroxynitrite remain incompletely understood. In this work, we demonstrate that the T. cruzi heme-containing ascorbate peroxidase cytochrome c peroxidase (APx-CcP), an antioxidant enzyme previously shown to increase parasite infectivity, acts as a key peroxynitrite-detoxifying enzyme. Using di

Magnetic Platinum-Cobalt Nanoparticles as Peroxidase Mimics for Detection of Cancer Biomarkers.

Inorganic nanomaterial-based peroxidase mimics have recently emerged as promising alternatives to natural peroxidases for enhancing the detection sensitivity of bioassays, such as enzyme-linked immunosorbent assay (ELISA). Among them, magnetically active peroxidase mimics are particularly advantageous due to their ability to facilitate efficient separation and enrichment of target analytes. However, most reported magnetic peroxidase mimics suffer from limited catalytic efficiency and stability. In this study, we introduce platinum-cobalt nanoparticles (Pt-Co NPs) as an innovative class of magn

Colorimetric Detection of Acid Phosphatase Using Nanoparticle CuNCs as Peroxidase Mimicking Activity.

Acid phosphatase (ACP) serves as a critical biomarker for disease diagnosis, yet conventional detection methods face limitations in cost and complexity. Copper nanoclusters (CuNCs), leveraging Earth-abundance, low expense, and noble metal-like catalytic properties, offer an ideal nanozyme platform. This study harnesses the intrinsic peroxidase mimicking activity of CuNCs to construct a visual colorimetric sensor for ACP detection. CuNCs oxidize the substrate 3,3',5,5'-tetramethylbenzidine (TMB) to generate a blue chromogen (λmax = 654 nm). ACP introduction triggers reduction of this chromogen,

Deciphering the Catalytic Mechanism of Peroxidase-like Activity of Iron Sulfide Nanozymes.

Iron sulfide nanomaterials represented by FeS2 and Fe3S4 nanozymes have attracted increasing attention due to their biocompatibility and peroxidase-like (POD-like) catalytic activity in disease diagnosis and treatments. However, the mechanism responsible for their POD-like activities remains unclear. Herein, taking the oxidation of 3,3,5,5-tetramethylbenzidine (TMB) by H2O2 on FeS2(100) and Fe3S4(001) surfaces, the catalytic mechanism was investigated in detail using density functional theory (DFT) calculations and experimental characterizations. Our experimental results showed that the cataly

3D Bioprinting of Sugar Beet Pectin through Horseradish Peroxidase-Catalyzed Cross-Linking.

Horseradish peroxidase (HRP)-mediated hydrogelation, caused by the cross-linking of phenolic groups in polymers in the presence of hydrogen peroxide (H2O2), is an effective route for bioink solidification in 3D bioprinting. Sugar beet pectin (SBP) naturally has cross-linkable phenols through the enzymatic reaction. Therefore, chemical modifications are not required, unlike the various polymers that have been used in the enzymatic cross-linking system. In this study, we report the application of SBP in extrusion-based bioprinting including HRP-mediated bioink solidification. In this system, H2O

Immobilization of horseradish peroxidase with zwitterionic polymer material for industrial phenolic removal.

Horseradish peroxidase (HRP) is a hemoglobin composed of a single peptide chain that catalyzes the oxidation of various substrates such as phenol and aniline in the presence of hydrogen peroxide via its iron-porphyrin catalytic center. This enzyme is widely used in industrial phenol removal, food additives, biomedicine, and clinical test reagents due to its rapid reaction rate and obvious reaction outcomes. However, the large-scale use of HRP in industrial applications still faces numerous challenges, including activity, stability, and sustainability. This study demonstrates that when peroxida

Amorphous copper(II)-cyanoimidazole frameworks as peroxidase mimics for hydrogen sulfide assay.

Metal-organic frameworks with hierarchical porosities and exposed active sites are promising for ideal enzyme mimics. In this work, we developed a simple and feasible air oxidation strategy to prepare amorphous Cu(II)-cyanoimidazole frameworks (aCu(II)-CIFs) using CuI as the metal source in dimethylsulfoxide. Benefiting from coordination unsaturation and hierarchical porosities, aCu(II)-CIFs exhibit inherent peroxidase-mimic activity for rapid colorimetric reaction of 3,3',5,5'-tetramethylbenzidine (TMB). aCu(II)-CIFs were utilized to develop a colorimetric platform for specific H2S assay in t

Catalase-peroxidase StKatG is a bacterial manganese oxidase from endophytic Salinicola tamaricis.

Manganese (Mn) oxides in iron/manganese plaques are widely distributed in the rhizosphere of wetland plants and contribute significantly to elemental cycling and pollutant removal. Mn oxides are primarily produced by bacterial processes using Mn oxidases. However, the molecular mechanism underlying the formation of rhizosphere Mn oxides is still largely unknown. This study identified a manganese-oxidizing enzyme, the catalase-peroxidase StKatG, from an endophytic bacterium Salinicola tamaricis from the wetland plant. The gene encoding StKatG was cloned and overexpressed in Escherichia coli. Th

Efficient Degradation of Tetracycline Antibiotics by Engineered Myoglobin with High Peroxidase Activity.

Tetracyclines are one class of widely used antibiotics. Meanwhile, due to abuse and improper disposal, they are often detected in wastewater, which causes a series of environmental problems and poses a threat to human health and safety. As an efficient and environmentally friendly method, enzymatic catalysis has attracted much attention. In previous studies, we have designed an efficient peroxidase (F43Y/P88W/F138W Mb, termed YWW Mb) based on the protein scaffold of myoglobin (Mb), an O2 carrier, by modifying the heme active center and introducing two Trp residues. In this study, we further ap

Thioredoxin-1 Rescues MPP+/MPTP-Induced Ferroptosis by Increasing Glutathione Peroxidase 4.

Parkinson's disease (PD), a common neurodegenerative disease, is typically associated with the loss of dopaminergic neuron in the substantia nigra pars compacta (SNpc). Ferroptosis is a newly identified cell death, which associated with iron accumulation, glutathione (GSH) depletion, lipid peroxidation formation, reactive oxygen species (ROS) accumulation, and glutathione peroxidase 4 (GPX4) reduction. It has been reported that ferroptosis is linked with PD.Thioredoxin-1 (Trx-1) is a redox regulating protein and plays various roles in regulating the activity of transcription factors and inhibi

Bioinspired Spiky Peroxidase-Mimics for Localized Bacterial Capture and Synergistic Catalytic Sterilization.

Besides the pandemic caused by the coronavirus outbreak, many other pathogenic microbes also pose a devastating threat to human health, for instance, pathogenic bacteria. Due to the lack of broad-spectrum antibiotics, it is urgent to develop nonantibiotic strategies to fight bacteria. Herein, inspired by the localized "capture and killing" action of bacteriophages, a virus-like peroxidase-mimic (V-POD-M) is synthesized for efficient bacterial capture (mesoporous spiky structures) and synergistic catalytic sterilization (metal-organic-framework-derived catalytic core). [Peroxidase activity of octaheme nitrite reductases from bacteria of the Thioalkalivibrio genus].

Closely related penta- and octaheme nitrite reductases catalyze the reduction of nitrite, nitric oxide, and hydroxylamine to ammonium and of sulfite to sulfide. NrfA pentaheme nitrite reductase plays the key role in anaerobic nitrate respiration and the protection of bacterial cells from stresses caused by nitrogen oxides and hydrogen peroxide. Octaheme nitrite reductases from bacteria of the Thioalkalivibrio genus are less studied, and their function in the cell is unknown. In order to estimate the possible role of octaheme nitrite reductases in the cell resistance to oxidative stress, the pe

Peroxidase chemically attached on polymeric micelle and its reaction with phenolic compounds.

Horseradish peroxidase was chemically modified with comb-shaped polymaleic anhydride-alt-1-tetradecene (PMA-TD) in microemulsion systems to produce surface-active peroxidase that has capability to form micellar structures in aqueous solutions and can be concentrated at liquid/liquid interfaces without unfolding of the enzyme. For chemical modification oil-in-water (O/W) and water-in-oil (W/O) microemulsion systems composed of n-butyl acetate and a buffer solution were prepared because n-butyl acetate turned out to be less detrimental to the activity of peroxidase at high degree of modification
