Combined induction by Cu(II) and veratrole enhances the degradation of high molecular weight polyaromatic hydrocarbons by Fusarium dlaminii ZH-H2.

The effect of combined induction by Cu(II) and veratrole on the degradation of high molecular weight polyaromatic hydrocarbons (HMW-PAHs) by Fusarium sp. ZH-H2 was investigated. This strain was characterized as F. dlaminii. The combination treatment of Cu(II) with veratrole (CL) improved the degradation efficiency of a mixture of 10 HMW-PAHs by 16 % compared to the control without inducer (CK), by 5 % compared to single Cu(II) induction (C), and by 12 % compared to single veratrole induction (L). In particular, degradation of benzo(g,hi)perylene (BghiP) was improved by 36 % compared to CK and

Crystal Engineering in Oligorylenes: The Quest for Optimized Crystal Packing and Enhanced Charge Transport.

The crystal structures of organic semiconductors are critical when they are integrated into optoelectronic devices, such as organic field-effect transistors (OFETs). In this study, we introduce a crystal engineering approach that leverages weak, nondirectional dispersion forces and steric effects, working together to govern the molecular packing. We investigated how the substitution at the peri-position affects the crystal structure in a series of oligorylene molecules. Upon elucidation of the crystal structures, we found a distinct difference between symmetrical and unsymmetrical derivatives.

Impacts of crab bioturbation on the fate of polycyclic aromatic hydrocarbons in sediment from the Beitang estuary of Tianjin, China.

Bioturbation by the burrowing crab Helice tiensinensis was investigated to determine its impact on polycyclic aromatic hydrocarbon (PAH)-contaminated estuarine sediments. The concentrations of 16 U.S. Environmental Protection Agency (U.S. EPA) priority PAHs in sediment and pore water from a crab bed (including surface and burrow samples) and a control area, as well as in crabs, were measured. The total concentration of the 16 U.S. EPA priority PAHs in surface sediment of the crab bed (average 2,772 ng/g dry weight) was significantly higher than in the control area (1,173 ng/g dry weight). In t

Modeling the Effects of Fermi Resonances in the CH Stretching Region of the Spectra for Polycyclic Aromatic Hydrocarbon Anions.

A strategy is developed for evaluating spectra of anions of polycyclic aromatic hydrocarbon (PAH) molecules. The high symmetry of these systems and the similarity of the frequencies of the CH wagging and ring distortion vibrations result in normal modes that are highly mixed in character. This results in extensive mixing among the nearly degenerate states with energies in the 2600 to 3200 cm-1 region of the spectrum. When we generate vibrational modes that resemble localized CH stretching, CH wagging, or ring distortion vibrations from linear combinations of the normal modes, we find that the

Method operable design region obtained with a partial least squares model inversion in the determination of ten polycyclic aromatic hydrocarbons by liquid chromatography with fluorescence detection.

A chromatographic method with the Analytical Quality by Design (AQbD) methodology is developed for the simultaneous determination by HPLC-FLD of ten PAHs (naphthalene, phenanthrene, anthracene, fluoranthene, pyrene, chrysene, benzo[a]anthracene, perylene, benzo[b]fluoranthene, and benzo[a]pyrene), widely spread in the environment. The construction of the Method Operable Design Region (MODR) is conducted, for the first time, via the inversion of a multiresponse Partial Least Squares (PLS2) model, which is needed to maintain the correlations among the Critical Method Parameters (CMP), among the

ROS-responsive dexamethasone micelles normalize the tumor microenvironment enhancing hypericin in cancer photodynamic therapy.

The efficacy of photodynamic therapy (PDT) for cancer is limited owing to the abnormality of the tumor microenvironment (TME), such as the dysfunctional tumor vascular system leading to restricted drug distribution in tumor lesions, and hypoxia resulting in hampering the application of the photosensitizer because of the shortage of oxygen. Therefore, normalizing the TME is a novel strategy for enhancing the therapeutic efficacy of PDT. Herein, we designed and fabricated reactive oxygen species (ROS)-responsive micelles with a self-circulating release manner to co-deliver a glucocorticoid stero

Spectroscopic and photostability study of water-soluble hypericin encapsulated with polyvinylpyrrolidone.

Hypericin has gained great attention as a powerful photosensitizing and fluorescent agent for photodynamic therapy (PDT) and fluorescence diagnosis (FD) of cancer. However, native hypericin is hydrophobic and nearly insoluble in aqueous media which hinders its photobiological activity. Herein, we demonstrate the encapsulation of hypericin and polyvinylpyrrolidone (hypericin@PVP) as an attractive class of water-soluble formula of hypericin with improved absorption and emission characteristics in water. The absorption and fluorescence properties of the water-soluble hypericin@PVP were studied at

Effect of light on hypericins contents in fresh flowering top parts and in an extract of St. John's Wort (Hypericum perforatum).

St. John's Wort is a medicinal plant increasingly used for its antidepressive activity. Hypericins are considered as one of the compounds contributing to the activity of the extract. These naphthodianthrones exist in various forms in Hyperici herba. Protopseudohypericin and protohypericin (protopigments) are converted into pseudohypericin and hypericin (pigments) under the action of light. The aim of this work is to study the influence of light on the phototransformation of protopigments into pigments. Two experiments were carried out. The studies were performed on one hand, on plant material

[Reversible increase in photosensitivity to UV-B caused by St. John's wort extract].

A 61-year-old woman with depression developed recurring elevated itching erythematous lesions in light-exposed areas after taking St. John's Wort-extract for three years. Routine patchtesting did not reveal any relevant reactions and photopatch testing was negative. Using a systemic oral photoprovocation test with St. John's Wort, we were able to demonstrate a decrease of the MED-UVB which was reversible after withdrawal of the medication.

Photoactivity of Pluronic® F-127 nanoencapsulated hypericin against Leishmania: A promising approach for cutaneous leishmaniasis.

Hypericin (HYP) is a naturally occurring naphthodianthrone that has been extensively studied, particularly in combination with photodynamic therapy (PDT), for its therapeutic properties. Encapsulation of HYP with Pluronic® F-127 nanoparticles enhances its bioavailability. Leishmaniasis is a neglected disease and a global health problem, with varying clinical forms caused by Leishmania parasites. Current treatments are limited by high toxicity and relapse, highlighting the need for alternative therapeutics. We investigated the activity of free HYP and that encapsulated with Pluronic® F-127 (HYP

Harvest time affects antioxidant capacity, total polyphenol and flavonoid content of Polish St John's wort's (Hypericum perforatum L.) flowers.

The polyphenol content and antioxidant capacity of hyperforin and hypericin-standardized H. perforatum L. extracts may vary due to the harvest time. In this work, ethanol and ethanol-water extracts of air-dried and lyophilized flowers of H. perforatum L., collected throughout a vegetation season in central Poland, were studied. Air-dried flowers extracts had higher polyphenol (371 mg GAE/g) and flavonoid (160 mg CAE/g) content, DPPH radical scavenging (1672 mg DPPH/g), ORAC (5214 µmol TE/g) and FRAP (2.54 mmol Fe2+/g) than lyophilized flowers extracts (238 mg GAE/g, 107 mg CAE/g, 1287 mg DPPH/

Evaluation of hypocrellin A-loaded lipase sensitive polymer micelles for intervening methicillin-resistant Staphylococcus Aureus antibiotic-resistant bacterial infection.

There is an urgent need for new antibacterial strategies to overcome the emergence of antibiotic resistance. Antibacterial photodynamic therapy (APDT) may be an effective method to deliver photosensitizers for the treatment of methicillin-resistant Staphylococcus aureus (MRSA) infections. Here, we report that the photosensitizer hypocrellin A (HA) loaded into lipase-sensitive methoxy poly (ethylene glycol)-block-poly(ε-caprolactone) (mPEG-PCL) micelles showed high anti-MRSA activity in vitro and in vivo by PDT. Once the micelles come into contact with bacteria that secrete lipase, the PCL is d

Topically applied hypericin exhibits skin penetrability on nude mice.

Hypericin, a powerful natural photosensitizer in photodynamic therapy (PDT), is suitable for treating skin diseases involving excess capillary proliferation. In the present study, we aimed to evaluate the skin penetrability of topically applied hypericin, expecting a reduced risk of prolonged skin photosensitivity, which often occurs after systemic administration. Firstly, the Franz diffusion cell assays were performed to evaluate the penetration effects of different enhancers, including menthol, propylene glycol, camphanone, azone, and carbamide. In view of above evaluation results, we select

Light-induced photoactivation of hypericin affects the energy metabolism of human glioma cells by inhibiting hexokinase bound to mitochondria.

Glucose-dependent energy required for glioma metabolism depends on hexokinase, which is mainly bound to mitochondria. A decrease in intracellular pH leads to a release of hexokinase-binding, which in turn decreases glucose phosphorylation, ATP content, and cell proliferation. Thus, intracellular pH might be a target for therapy of gliomas, and a search for agents able to modulate intracellular pH was initiated. Hypericin, a natural photosensitizer, displays numerous biological activities when exposed to light. Its mechanism and site of action at the cellular level remain unclear, but it probab

Improving oleoyl hyaluronan self-assembly and functional properties using zinc ions.

The self-assembly of polymer-based drug carriers plays a critical role in their performance in drug delivery and targeting. In this study, we investigated the effect of various salts on the aggregation behavior of oleoyl-modified hyaluronic acid (OL-HA) and its functional properties. Structural organization was assessed by dynamic light scattering, atomic force microscopy, scanning electron cryomicroscopy, surface tension, rheology and steady-state and time-resolved fluorescence (probes pyrene, Nile red and perylene), revealing that zinc acetate promotes self-assembly compared to water and pho
