Increased blood interleukin-10 mRNA levels in diseased free-ranging harbor porpoises (Phocoena phocoena).

Harbor porpoises from the North and Baltic Seas exhibit a higher incidence of bacterial infections compared to whales from less polluted arctic waters. Toxicological analysis revealed an association between elevated body burdens of environmental contaminants, such as polychlorinated biphenyls (PCB) and polybrominated diphenyl ether (PBDE) and lymphoid depletion in thymus and spleen of these whales. However, it remains undetermined if changes in the immune system are primarily contaminant-induced or a sequel of infectious diseases and emaciation. The aim of the present study was to investigate

Effects of contaminant exposure on reproductive success of ospreys (Pandion haliaetus) nesting in Delaware River and Bay, USA.

Despite serious water-quality problems and pollutant loading and retention, Delaware River and Bay (USA) provide important wildlife habitat. In 2002, we conducted a comprehensive evaluation of contaminant exposure and reproduction of ospreys (Pandion haliaetus) breeding in Delaware River and Bay. Sample eggs were collected from 39 nests and analyzed for organochlorine pesticides, polychlorinated biphenyls (PCBs), and mercury; a subset of 15 eggs was analyzed for perfluorinated compounds and polybrominated diphenyl ethers (PBDEs). The fate of each nest was monitored weekly. Concentrations of 10

Multi-omics investigations reveal the critical role of the liver-microbiota-gut axis in tetrabromobisphenol S-induced liver fibrosis.

Tetrabromobisphenol S (TBBPS), a widely used brominated flame retardant (BFR), is increasingly detected in environmental and biological samples, raising urgent concerns. Although several in vitro studies have investigated the hepatotoxicity of TBBPS, the in vivo effects and mechanisms of chronic exposure, particularly by the liver-microbiota-gut axis, remain largely unexplored. In this study, we administered TBBPS via oral gavage to C57BL/6 J male mice and employed an integrated multi-omics strategy consisting of transcriptomics, serum non-targeted metabolomics, and 16S rDNA gene sequencing. Differential developmental effects and its potential mechanism of long-term exposure to TBBPA in two generations of marine medaka (Oryzias melastigma) during early life stages.

Tetrabromobisphenol A (TBBPA), a most widely used brominated flame retardant, has been detected in worldwide aquatic environments. However, the effects and mechanisms of TBBPA at environmentally realistic levels have not been well characterized in aquatic organisms. This study aims to investigate the impact of TBBPA on developmental toxicity and endocrine system in two generations of marine medaka (Oryzias melastigma) during early life stages. The results revealed that the embryos under exposure to environmentally relevant concentrations of TBBPA (0, 5, 50, and 500 μg/L) resulted in accelerate

Assessment of the disruption effects of tetrabromobisphenol A and its analogues on lipid metabolism using multiple in vitro models.

Tetrabromobisphenol A (TBBPA), a widely-used brominated flame retardant, has been revealed to exert endocrine disrupting effects and induce adipogenesis. Given the high structural similarities of TBBPA analogues and their increasing exposure risks, their effects on lipid metabolism are necessary to be explored. Herein, 9 representative TBBPA analogues were screened for their interference on 3T3-L1 preadipocyte adipogenesis, differentiation of C3H10T1/2 mesenchymal stem cells (MSCs) to brown adipocytes, and lipid accumulation of HepG2 cells. A pilot study of several environmental endocrine disrupting chemicals in children with autism spectrum disorder in south China.

Autism spectrum disorders (ASD) is a group of heterogeneous neurodevelopmental disorders. Evidence has implied that environmental pollutants are important factors related to ASD. In this study, several environmental endocrine-disrupting chemicals, including parabens, benzophenone-type ultraviolet filters, hydroxyl polycyclic aromatic hydrocarbons, triclosan and tetrabromobisphenol A were analyzed in blood plasma in ASD children (n = 34) and the control children (n = 28). The results showed that parabens were the most concentrated chemicals (2.18 ng/mL, median value), followed by hydroxyl polyc

Tissue disposition, excretion and metabolism of 2,2',4,4',6-pentabromodiphenyl ether (BDE-100) in male Sprague-Dawley rats.

The absorption, disposition, metabolism and excretion study of orally administered 2,2',4,4',6-pentabromodiphenyl ether (BDE-100) was studied in conventional and bile-duct cannulated male rats. In conventional rats, >70% of the radiolabelled oral dose was retained at 72 h, and lipophilic tissues were the preferred sites for disposition, i.e. adipose tissue, gastrointestinal tract, skin, liver and lungs. Urinary excretion of BDE-100 was very low (0.1% of the dose). Biliary excretion of BDE-100 was slightly greater than that observed in urine, i.e. 1.7% at 72 h, and glucuronidation of phenolic m

In vivo and in vitro anti-androgenic effects of DE-71, a commercial polybrominated diphenyl ether (PBDE) mixture.

PBDEs have been synthesized in large quantities as flame retardants for commercial products, such as electronic equipment and textiles. The rising in levels of PBDEs in tissues in wildlife species and in human milk and plasma samples over the past several years have raised concerns about possible health effects. Recently, we showed that the PBDE mixture, DE-71, delayed puberty and suppressed the growth of androgen-dependent tissues in male Wistar rat following a peri-pubertal exposure. These effects suggested that DE-71 may be either inducing steroid hormone metabolism or acting as an androgen

TBBPA induced hepatocyte ferroptosis by PCBP1-mediated ferritinophagy.

Tetrabromobisphenol A (TBBPA) is the most widely used brominated flame retardant and has been identified as emerging widespread pollutants. Ferroptosis, a recently characterized form of iron-dependent cell death, is related to a wide range of liver diseases. Ferritinophagy as a novel selective form of autophagy functions in iron processing is essential to induce ferroptosis. Poly(rC)-binding protein 1 (PCBP1) is an iron chaperone involved in iron loading to ferritin. Nevertheless, the potential health risk caused by TBBPA in mammals is unknown. Thus, this study is conducted to explore the mole

TBBPA-BDBPE induces ferroptosis in trophoblasts through chaperone-mediated autophagy of GPX4.

Tetrabromobisphenol A-bis (2,3-dibromopropyl ether) (TBBPA-BDBPE), a novel brominated flame retardant, is increasingly utilized across various industries due to its effective flame-retardant properties. However, its potential gestational toxicity has not been thoroughly investigated, raising concerns about its impact on maternal and fetal health. In this study, we explored the effects of TBBPA-BDBPE on fetal growth restriction (FGR) and placental trophoblast ferroptosis through a comprehensive approach that included both in vivo and in vitro experiments. We employed techniques such as transmis

Association of Prenatal Exposure to Endocrine-Disrupting Chemicals With Liver Injury in Children.

IMPORTANCE: Prenatal exposures to endocrine-disrupting chemicals (EDCs) may increase the risk for liver injury in children; however, human evidence is scarce, and previous studies have not considered potential EDC-mixture effects. Furthermore, the association between prenatal EDC exposure and hepatocellular apoptosis in children has not been studied previously. OBJECTIVE: To investigate associations of prenatal exposure to EDC mixtures with liver injury risk and hepatocellular apoptosis in childhood. DESIGN, SETTING, AND PARTICIPANTS: This prospective cohort study used data collected from Ap

Toxicology of octabromobiphenyl and decabromodiphenyl oxide.

Decabromodiphenyl oxide (DBDPO) and octabromobiphenyl (OBBP) perform well as fire-retardant additives for thermoplastics. Both compounds have low acute oral toxicity and low skin absorption toxicity. They are neither primary skin irritants or skin sensitizers and are only mildly irritating to the eyes. A 30-day dietary feeding study in rats established 8 mg DBDPO/kg-day as an unequivocal no-effect level and 80 mg/kg-day as a marginal effect level. A no-effect level was not established for OBBP in a comparative study. A 2-yr rat study providing 0.1 mg DBDPO/kg-day in the diet revealed the bromi

Exposure of an endangered seabird species to persistent organic pollutants: Assessing levels in blood and link with reproductive parameters.

Ocean contamination, particularly from persistent organic pollutants (POPs), remains a significant threat to marine predators that occupy high trophic positions. Long-lived procellariform seabirds are apex predators in marine ecosystems and tend to accumulate contaminants. Prolonged exposure to pollutants negatively affects their fitness including reproductive success. Low breeding success may represent a hurdle for the restoration of small and endangered seabird populations, including several highly threatened gadfly petrels. Here we investigated the annual variation (2019 and 2022) in organo

Maternal exposure to a brominated flame retardant and genitourinary conditions in male offspring.

BACKGROUND: The upward trend in industrial nations in the incidence of male genitourinary (GU) conditions may be attributed to increased exposure to endocrine disruptors. Polybrominated biphenyl (PBB), a brominated flame retardant, is one such suspected endocrine disruptor. OBJECTIVE: We investigated the relationship between maternal serum levels of PBBs and GU conditions among male offspring exposed in utero. METHODS: In this cohort study of sons born to women accidentally exposed to PBBs during 1973-1974, we examined self-reported data on GU conditions among male offspring in relation to m

Toxicological impact of tetrabromobisphenol-A on pregnant rats: Morphological, biochemical, and histopathological investigations.

Tetrabromobisphenol-A (TBBPA) is found in high concentrations in textiles, plastic polymers, flame retardants, and various industrial products. In this maternofetal study, we aimed to evaluate the potential effects of TBBPA on the mother, fetus, and placenta in pregnant rats. In the experimental design, three different gestational days (GD10, GD15, and GD20) were selected for evaluation in pregnant rats. For each gestational period, one control group (corn oil) and three TBBPA dose groups (20, 200, and 400 mg/kg) were established, resulting in a total of 12 experimental groups. Each group incl

Toxicity of tetrabromobisphenol A to human thyroid follicular epithelial cells through oxidative stress: A combined analysis at the cellular and molecular levels.

Tetrabromobisphenol A (TBBPA), a prominent brominated flame retardant (BFR), is a toxic pollutant. In view of the structural similarity of TBBPA to thyroid hormones, we investigated the toxic effects of TBBPA on human thyroid follicular epithelial cells (Nthy-ori3-1) at concentrations ranging from 0.02 to 100 μM. The findings of this study demonstrated that a 24-hour exposure to 10 μM TBBPA resulted a reduction of cell viability to 50.9 %. Concurrently, intracellular reactive oxygen species (ROS) levels were significantly elevated, and oxidative stress-related proteins, particularly those invo

Mixed exposure to brominated flame retardants adversely affects vaccine-induced antibody titers: Risk prioritization and benchmark dose modeling.

The association between brominated flame retardant (BFRs) exposure and measles, mumps, and rubella vaccine (MMR) antibody responses was investigated using the National Health and Nutrition Examination Survey (2003-2004). Using multivariate linear regression and restricted cubic spline (RCS) model, we found significant inverse association between single BFR exposure (BB-153 and BDE-100) and mumps antibody levels (P < 0.05). Mixed BFR exposure synergistically reduced antibody levels across mumps and rubella vaccines, with BB-153 identified as the dominant contributor for mumps (PIP = 0.460). Ben

Electron paramagnetic resonance evidence of hydroxyl radical generation and oxidative damage induced by tetrabromobisphenol A in Carassius auratus.

Tetrabromobisphenol A (TBBPA) is one of the most widely used brominated flame retardants (BFRs). To confirm its putative oxidative stress-inducing activity, freshwater fish Carassius auratus were injected intraperitoneally with TBBPA. One experiment lasted 3h to 28 days after a single injection of 100mg/kg TBBPA, and the other lasted 24h after a single injection of 0-300 mg/kg TBBPA. Reactive oxygen species (ROS) were trapped by phenyl-tert-butyl nitrone (PBN) and detected by electron paramagnetic resonance (EPR). Protein carbonyl (PCO) and lipid peroxidation product (LPO) content were also de
