Antiserum has been prepared in rabbits against the H and M antigens of H. capsulatum with immunoelectrophoretic precipitin arcs used as vaccines. The antiserum is specific for H. capsulatum in the immunodiffusion test and can be used as reference serum for identifying antibodies to these antigens in sera from suspected cases histoplasmosis. We found that (1) hand m antigens are not located on the surface of yeast-phase cells and (ii) complement fixation releases the antigens reactive in the complement fixation test from yeast-phase cells.

Soluble antigen from bovine blood with a Babesia bigemina parasitemia of 5-6% formed 4 precipitin lines in gel diffusion tests with antiserum from infected cattle. Antigen was obtained from plasma and hemolysate by elution from DEAE cellulose columns and (NH4)2SO4 precipitation as well as from washed parasite-erythrocyte stroma by sonication or freeze-thawing. About 5 ml of antigen suitable for diagnostic tests could be extracted from 200 ml of infected blood.

Serotype-specific polysaccharide antigens of Actinobacillus actinomycetemcomitans ATCC 29523 (serotype a), Y4 (serotype b) and NCTC 9710 (serotype c) were extracted from whole cells by autoclaving. Cross-reactivity in Histoplasma serologic tests was evaluated by using sera from patients with histoplasmosis and other infections. Serum samples from 127 of 134 (95%) patients with histoplasmosis were judged positive by complement fixation tests, and 121 (90%) showed H bands, M bands, or both by immunodiffusion. Of these 134 patients, cross-reactions were seen to Blastomyces dermatitidis in 53 patients (40%), to Coccidioides immitis in 20 patients (16%), and to Aspergillus fumigatus in 3 patients (2%) by complement fixation. Mice were immunized against hepatitis B e antigen (HBeAg) isolated from sera of asymptomatic carriers of hepatitis B virus. The first probable human case of pulmonary anisakiasis is reported. A 37-year-old Japanese person in California developed pleural effusion after consumption of raw salmon at a seafood restaurant in San Francisco. A marked eosinophilia was noted in the blood and pleural exudate. The patient was tested for helminthic infections by intradermal, complement fixation, Ouchterlony, and immunoelectrophoresis tests using antigens of various worm species. A specific precipitin band was recognized only with Anisakis (Type I larvae) antigen.

A technique is described for the detection of immunoglobulins secreted by individual lymphoid cells. Cells were cultured in agarose gel containing dilute radiolabelled anti-immunoglobulin antibodies. The secreted immunoglobulins produced a precipitin lattice which was made visible by washing the gel, followed by autoradiography. Immunoglobulin secreting cells were surrounded by clusters, or `galaxies' of silver grains. Galaxies did not appear in the presence of certain metabolic inhibitors, indicating that the secretion of immunoglobulin was an active process dependent upon cell metabolism.

Specific antibodies were investigated in serums of chicks vaccinated with live vaccine and revaccinated with inactivated vaccine against the infectious bursal disease virus, using three methods. Two families are described in which seven members of a total of 19 were found to have hypersensitivity pneumonitis due to exposure to avian antigens. The reactions of synthetic polypeptides with their antibodies may be followed by passive cutaneous anaphylaxis, but not by the reverse technique. The reactions were optimal when 18 hours elapsed between the injection of the serum and that of the antigen. Materials which were non-immunogenic as followed by the precipitin test, were shown to be non-immunogenic also by PCA technique. The antisera to various synthetic polypeptide antigens cross-reacted with many chemically related polypeptides. As little as 0.12 μg. of antibody could be detected in 1 ml. of diluted antisera by the PCA technique.

Sixty-seven asthmatic individuals treated with either beclomethasone diproprionate or flunisolide were sequentially evaluated for up to 32 months to determine the incidence of oropharyngeal candidiasis as well as laboratory parameters which might be predictive of this complication. Throat cultures and measurements of Candida antibody by immunodiffusion and radioimmunoassay were performed and compared over time and treatment groups. Unlike other studies, pre-treatment Candida precipitins did not predict increased risk for clinical thrush nor did quantitative determinations of Candida antibody.

Serum samples were collected from 24 confirmed cases of typhoid fever and 23 normal healthy controls. The influence of blood and blood-product therapy was studied in two groups of children: 1) 90 children who had exchange transfusion after birth because of serologic incompatibility (aged 5 months to 5 years).
