Apaf1 (CED-4 homolog) regulates programmed cell death in mammalian development.

The cytosolic protein APAF1, human homolog of C. elegans CED-4, participates in the CASPASE 9 (CASP9)-dependent activation of CASP3 in the general apoptotic pathway. We have generated by gene trap a null allele of the murine Apaf1. Homozygous mutants die at embryonic day 16.5. Their phenotype includes severe craniofacial malformations, brain overgrowth, persistence of the interdigital webs, and dramatic alterations of the lens and retina. Homozygous embryonic fibroblasts exhibit reduced response to various apoptotic stimuli. In situ immunodetection shows that the absence of Apaf1 protein preve

Developing a national HIV/AIDS Prevention Program through state health departments.

The Centers for Disease Control (CDC) shaped the basic development and direction of the HIV/AIDS Prevention Program through technical support and financial assistance for State and local health departments and other organizations. Through this provision of support, CDC has responded to the course of the human immunodeficiency virus (HIV)-acquired immunodeficiency syndrome (AIDS) epidemic by creating programs to preserve the safety of the blood supply, by developing counseling and testing centers, by promoting "safer sex," by promoting health education and risk reduction, by evaluating existing

Development and operation of a program to obtain volunteer bone marrow donors unrelated to the patient.

Because of recent increases in bone marrow transplantation, the lack of available matched sibling donors has become a problem. This study investigated the feasibility of obtaining volunteer bone marrow donors unrelated to the patient. A program was developed to inform potential donors and obtain their consent for listing in a bone marrow donor registry. Sixty-six percent of whole blood or apheresis donors agreed to participate in the program. There were no demographic differences between those who accepted or declined to participate. Religion, experiences with medical practices, and the attitu

Development of an in vitro assay for screening programmed death receptor-1/programmed cell death ligand 1 monoclonal antibody therapy in dogs.

Immunomodulatory antibody drugs that modulate the function of immune checkpoint molecules, such as programmed death receptor-1 (PD-1) and programmed cell death ligand 1 (PD-L1), have been established as new cancer treatments in human medicine. In recent years, there have also been reports on antibodies that inhibit immune checkpoint molecules in dogs, and clinical trials using such antibodies for canine cancer have been gradually increasing in number. Because inhibitory antibodies restore T-cell function by inhibiting the binding of PD-1 on T cells and its ligand PD-L1, the quality of antibody

Development of an evidence-based hepatitis C education program to enhance public health literacy in the Australian prison sector: The Hepatitis in Prisons Education program (HepPEd).

BACKGROUND: Australia's prisons have a high chronic hepatitis C (HCV) prevalence (8 %). Antiviral therapies and prison-based hepatitis services are available, but only a minority of those eligible are being treated. Improving the HCV public health literacy of the prison sector via targeted education may overcome key barriers to scale-up treatment. This paper describes the: i) HCV public health literacy of the prison setting; ii) barriers and solutions for HCV education and service engagement; iii) HCV education program co-design and development processes; and iv) HepPEd resources. METHODS: A

Patient-centered development of a cognitive behavioral pain intervention for cystic fibrosis: The PACE CF program.

BACKGROUND: Despite treatment advances, the majority of adults with cystic fibrosis (CF) experience chronic pain, negatively impacting quality of life. CF Foundation guidelines call for a multi-modal approach to pain management. While nonpharmacologic behavioral interventions are an effective and safe component of such an approach, there are no existing behavioral interventions that have been designed specifically for adults with CF who experience chronic pain. OBJECTIVE: We created a novel cognitive-behavioral and mind-body intervention tailored to the needs, preferences, and experiences of

Phosphorylated TYK2 orchestrates the pathogenic program of CD4 + T cells in the development of CNS autoimmunity.

BACKGROUND: The imbalance between pathogenic Th1/Th17 cells and regulatory T cells (Tregs) is a central mechanism in central nervous system (CNS) autoimmune diseases, including autoimmune uveitis (AU) and multiple sclerosis. Tyrosine kinase 2 (TYK2) mediates signaling downstream of multiple cytokines implicated in CD4 + T cell differentiation, yet its subset-specific activation and therapeutic potential in CNS autoimmunity remain unclear. Here, we investigated the subset-specific activation of TYK2 and evaluated the therapeutic potential of selective TYK2 inhibition in CNS autoimmune diseases.

Piezo1 incorporates mechanical force signals into the genetic program that governs lymphatic valve development and maintenance.

The lymphatic system plays crucial roles in tissue homeostasis, lipid absorption, and immune cell trafficking. Although lymphatic valves ensure unidirectional lymph flows, the flow itself controls lymphatic valve formation. Here, we demonstrate that a mechanically activated ion channel Piezo1 senses oscillating shear stress (OSS) and incorporates the signal into the genetic program controlling lymphatic valve development and maintenance. Time-controlled deletion of Piezo1 using a pan-endothelial Cre driver (Cdh5[PAC]-CreERT2) or lymphatic-specific Cre driver (Prox1-CreERT2) equally inhibited l
