Background: Hypoxic brain injury is one of the major causes of neurodevelopmental impairment and cardiovascular disability. LncRNA SNHG1 works as a critical factor in hypoxic induced injury, however, the potential mechanism is still not known well. Methods: The expression level of small nucleolar RNA host gene 1 (SNHG1) and miR-140-5p was detected by qRT-PCR. The western blot assay was performed to measure the level of Bcl-XL and apoptosis-related proteins. The target relationship between lncRNA SNHG1 and miR-140-5p, as well as miR-140-5p and Bcl-XL was detected by dual luciferase reporter gene

Erythroid differentiation-associated gene (EDAG) is a haematopoietic tissue-specific transcription regulator that plays a key role in maintaining the homeostasis of haematopoietic lineage commitment. In acute myeloid leukaemia (AML) patients, the high expression level of EDAG is associated with poor prognosis. NPM1 (nucleophosmin/B23), a ubiquitous nucleolar phosphoprotein, comprises a multifunctional protein that is involved in several cellular processes, including ribosome biogenesis, centrosome duplication, cell cycle progression, cell growth and transformation. Various studies have implica

RNA modifications play a fundamental role in cellular function. Pseudouridylation, the most abundant RNA modification, is catalyzed by the H/ACA small ribonucleoprotein (snoRNP) complex that shares four core proteins, dyskerin (DKC1), NOP10, NHP2, and GAR1. Mutations in DKC1, NOP10, or NHP2 cause dyskeratosis congenita (DC), a disorder characterized by telomere attrition. Here, we report a phenotype comprising nephrotic syndrome, cataracts, sensorineural deafness, enterocolitis, and early lethality in two pedigrees: males with DKC1 p.Glu206Lys and two children with homozygous NOP10 p.Thr16Met.

Traditionally recognized for guiding rRNA modifications, small nucleolar RNAs (snoRNAs) are increasingly appreciated as key regulators of drug response. However, snoRNA and drug association data remain limited, and computational approaches capable of capturing their complex, high-order relationships are scarce. Here, we present HMHLVI, a hybrid multi-view hypergraph learning with variational inference framework that integrates sequence, structural, and association information to predict snoRNA-drug response associations. HMHLVI constructs three higher-order networks called attribute, topology,

Analyzing the pathogenic mechanism of dry eye disease provides a new research point for the treatment of patients. A total of 86 patients with dry eye disease and the same number of healthy individuals were recruited as the patient group and normal group. The levels of small nucleolar RNA host gene 7 (SNHG7) in tear samples and human conjunctival epithelial cells (HCECs) were determined by a quantitative real-time PCR (RT-qPCR). Meanwhile, TNF-α and IL-6 mRNA levels in HCECs were also detected by RT-qPCR. The molecular mechanism of SNHG7 was analyzed by biological prediction and luciferase act

Progressive encephalopathy with oedema, hypsarrhythmia, and optic atrophy (PEHO) syndrome is an early childhood onset, severe autosomal recessive encephalopathy characterized by extreme cerebellar atrophy due to almost total granule neuron loss. By combining homozygosity mapping in Finnish families with Sanger sequencing of positional candidate genes and with exome sequencing a homozygous missense substitution of leucine for serine at codon 31 in ZNHIT3 was identified as the primary cause of PEHO syndrome. ZNHIT3 encodes a nuclear zinc finger protein previously implicated in transcriptional re

In addition to their role in protein translation, tRNAs can be cleaved into shorter, biologically active fragments called tRNA fragments (tRFs). Specific tRFs from spermatocytes can propagate metabolic disorders in second generations of mice. Thus, tRFs in germline cells are a mechanism of epigenetic inheritance. It has also been shown that stress and toxins can cause alterations in tRF patterns. We were therefore interested in whether injecting illicit drugs, a major stressor, impacts tRFs in germline cells. We sequenced RNA from spermatocytes and from semen-derived exosomes from people who i

MiR-181b-5p plays a critical role in the pyroptosis and injury of kidney tubular cells in diabetic kidney disease (DKD). The long noncoding RNA (lncRNA) small nucleolar RNA hostgene 7 (SNHG7) has been shown to bind to and inhibit the function of miR-181b-5p. However, the precise role of SNHG7 in DKD remains unclear. To address this, the current study measured the expression levels of SNHG7, miR-181b-5p, and Toll-like receptor 4 (TLR4) using RT-qPCR analysis of renal biopsies from both normal individuals and patients with DKD. An in vitro DKD model was subsequently established by exposing HK-2

Angelman syndrome (AS) is a debilitating neurodevelopmental disorder caused by loss of maternally-inherited UBE3A. In neurons, paternally-inherited UBE3A is silenced in cis by a long non-coding RNA called Ube3a-ATS. Here, we found that Neisseria meningitidis Cas9 with two mutations (D15A and H587A) in the nuclease domains (dNmCas9) can unsilence the dormant paternal Ube3a allele in mouse and human neurons when targeted to Snord115 snoRNA genes located in introns of Ube3a-ATS. Importantly, dNmCas9 disrupted Ube3a-ATS with a non-template bias and in the absence of a chromatin modifying domain, s

In a search for genes specifically expressed in Reed-Sternberg (RS) cells of Hodgkin lymphoma (HL), we applied the serial analysis of gene expression (SAGE) technique on the HL-derived cell line DEV. Genes highly expressed in DEV were subjected to an RT-PCR analysis to confirm the SAGE results. For one of the genes, a high expression was observed in DEV and other HL-derived cell lines but not in non-Hodgkin lymphoma (NHL)-derived cell lines and normal controls, suggesting an HL-specific expression. This gene corresponds to the human BIC gene, a member of the noncoding mRNA-like molecules. RNA

MicroRNA (miR) 15a‑5p can promote ischemia/reperfusion (I/R)‑induced apoptosis of cerebral vascular endothelial cells, which is inhibited by long non‑coding RNAs (lncRNAs). The present study investigated the potential of lncRNAs targeting miR‑15a‑5p to regulate oxygen‑glucose deprivation and reoxygenation (OGD‑R)‑induced apoptosis of human brain microvascular endothelial cells (hBMECs). hBMECs were transfected with or without miR‑15a‑5p or its mutant, together with p‑small nucleolar RNA host gene 16 (SNHG16) or its mutant. Following OGD‑R, proliferation, apoptosis and miR‑15a‑5p, SNHG16 and Bc

PURPOSE: Diffuse intrinsic pontine gliomas (DIPGs) are the most fatal primary brainstem tumors in pediatric patients. The identification of new molecular features, mediating their formation and progression, as non-coding RNAs (ncRNAs), would be of great importance for the development of effective treatments.
METHODS: We analyzed the DIPGs transcriptome with the HTA2.0 array and it was compared with pediatric non-brainstem astrocytoma expression profiles (GSE72269).
RESULTS: More than 50% of the differentially expressed transcripts were ncRNAs and based on this, we proposed a DIPGs ncRNA sign

Prader-Willi syndrome (PWS), a disorder of genomic imprinting, is characterized by neonatal hypotonia, hypogonadism, small hands and feet, hyperphagia and obesity in adulthood. PWS results from the loss of paternal copies of the cluster of SNORD116 C/D box snoRNAs and their host transcript, 116HG, on human chromosome 15q11-q13. We have investigated the mechanism of repression of the maternal SNORD116 cluster and 116HG. Here, we report that the zinc-finger protein ZNF274, in association with the histone H3 lysine 9 (H3K9) methyltransferase SETDB1, is part of a complex that binds to the silent m

Prader-Willi syndrome (PWS) is a rare genetic neurodevelopmental and neurobehavioral disorder characterized by hyperphagia and behavioral, endocrine, arousal, and cognitive dysregulation. Although caused by loss of paternally expressed, maternally imprinted genes in 15q11-q13, the mechanisms linking this lesion to the broad clinical phenotype remain incompletely understood. This review synthesizes evidence that central monoaminergic dysfunction, encompassing serotonin, dopamine, and norepinephrine, provides an integrative, though not exclusive, mechanistic framework for core features of PWS. Psoriasis is a chronic autoimmune and autoinflammatory disorder defined by abnormal skin cell turnover and inflammation, resulting in the formation of plaques on the skin. Although biologic therapies targeting interleukin (IL)-17 and IL-23 have significantly improved the treatment landscape for moderate-to-severe psoriasis, they are not effective for all patients. This highlights the need for additional therapeutic strategies. In recent years, exploring novel treatment avenues such as targeting IL-21, small nucleolar RNA (snoRNA) Snora73, the gut microbiome, and natural remedies have shown inc

BACKGROUND: Non-small cell lung cancer (NSCLC) remains a leading cause of cancer-related mortality with limited treatment efficacy. Small nucleolar RNAs (snoRNAs) have emerged as potential key players in tumorigenesis, but their roles in NSCLC are largely unexplored. Our previous study identified SCARNA20 as significantly downregulated in NSCLC tissues, suggesting a potential tumor-suppressive function. Thus, in order to further explore the role of SCARNA20 in NSCLC, we conducted this study to verify its function.
METHODS: The biological functions of SCARNA20 were investigated in vitro using
