Mechanism-aware and explainable artificial intelligence for drug release prediction in polymeric drug delivery systems.

Predicting drug release from polymer-based delivery systems remains challenging because release behavior depends on nonlinear interactions among formulation composition, polymer properties, drug characteristics, processing conditions, and time. This study develops an interpretable artificial intelligence framework for predicting cumulative drug release from PLGA nanoparticle formulations prepared by nanoprecipitation. A curated dataset containing 4,909 formulation-release observations was used to integrate formulation-informed feature engineering, temporal representation learning, symbolic reg

Deep latent variable modelling for clinically significant subgroup discovery among transfusion recipients.

BACKGROUND: Transfusion recipients are a heterogeneous group of patients, yet identifying these groups has traditionally relied on human-driven univariate analyses and domain knowledge instead of analyzing multivariate characteristics of individuals. Electronic health records (EHR) combined with unsupervised machine learning enables robust, data-driven way for phenotyping populations, providing finer-grained view on subgroup characteristics. METHODS: We introduce an extension to the Variational Autoencoder (VAE) framework and apply the model to EHR data of 19,629 adult transfusion recipients.

Self-Concentration Detection Based on Doubled Amplitude/Phase Processing in Node PDE Modular Models.

Reliable classification of brain sequence cases is a challenging problem due to signal ambiguity and noise. Personal concentration is primarily determined by the base frequency of electroencephalogram (EEG) waves, i.e., the task rests on appropriate modelling and recognition of patterns in the corresponding human (in)activity (e.g., reading, relaxation, solving maths problems, etc.). Five underlying types of frequency (alpha, beta, gamma, delta, and theta) were considered as secondary input wave parameters in complex-valued node extensions to the prime amplitude in processing signals. Self-opt

Language-model-based patient embedding using electronic health records facilitates phenotyping, disease forecasting, and progression analysis.

Current studies regarding the secondary use of electronic health records (EHR) predominantly rely on domain expertise and existing medical knowledge. Though significant efforts have been devoted to investigating the application of machine learning algorithms in the EHR, efficient and powerful representation of patients is needed to unleash the potential of discovering new medical patterns underlying the EHR. Here, we present an unsupervised method for embedding high-dimensional EHR data at the patient level, aimed at characterizing patient heterogeneity in complex diseases and identifying new

Canvas Beyond Words: Teaching Machines to Sketch Empowers Their Spatial Intelligence.

How to encode and evaluate spatial intelligence in foundation models remains an open challenge. Existing approaches often rely on textual proxies and VQA-style evaluation to assess visual-spatial intelligence (VSI), which can obscure geometric structure, encourage linguistic shortcuts, and hinder attribution to genuinely spatial reasoning abilities. To address this limitation, we introduce spatial intelligence grid (SIG), a structured grid-based schema that explicitly represents object layouts, inter-object relations, and physically grounded priors. As a complementary modality to text, SIG off

A foundation model for brain tumor MRI analysis: WHO grading and subtype classification.

OBJECTIVES: This study aims to develop a self-supervised foundational model based on routine MRI and to evaluate its performance in tasks of brain tumor grading and pathological subtype classification. METHODS: We developed the Unified Multimodal Brain Imaging Foundation (UMBIF) model by performing self-supervised learning on 51,029 MRI images from multiple institutions. The model was first trained using a contrastive masked image modeling task to extract robust feature representations. Next, UMBIF was fine-tuned for downstream tasks-glioma grading and histological classification-using multi-

WITHDRAWN: Molecular basis of BACE1 modulation revealed by machine learning, molecular simulations, and experimental validation.

Источник пометил публикацию как отозванную

Alzheimer's disease (AD) remains one of the most prevalent and debilitating neurodegenerative disorders worldwide, with no currently available disease-modifying treatments. β-site amyloid precursor protein cleaving enzyme 1 (BACE1) catalyzes the rate-limiting step in amyloid-β (Aβ) production and represents a validated therapeutic target for AD intervention. In this study, we developed an integrated computational framework combining machine learning-based virtual screening, molecular docking, and molecular dynamics simulations with experimental validation using CCK-8 assays and Western blot an

Comprehensive transcriptomic analysis and machine learning reveal unique gene expression profiles in patients with immune-mediated necrotizing myopathy.

BACKGROUND: Immune-mediated necrotizing myopathy (IMNM) is an autoimmune myopathy characterized by severe proximal weakness and muscle fiber necrosis, yet its pathogenesis remains unclear. So far, there are few bioinformatics studies on underlying pathogenic genes and infiltrating immune cell profiles of IMNM. Therefore, we aimed to characterize differentially expressed genes (DEGs) and infiltrating cells in IMNM muscle biopsy specimens, which may be useful for elucidating the pathogenesis of IMNM. METHODS: Three datasets (GSE39454, GSE48280 and GSE128470) of gene expression profiling related

Deep learning of joint myelin and T1w MRI features in normal-appearing brain tissue to distinguish between multiple sclerosis patients and healthy controls.

Myelin imaging is a form of quantitative magnetic resonance imaging (MRI) that measures myelin content and can potentially allow demyelinating diseases such as multiple sclerosis (MS) to be detected earlier. Although focal lesions are the most visible signs of MS pathology on conventional MRI, it has been shown that even tissues that appear normal may exhibit decreased myelin content as revealed by myelin-specific images (i.e., myelin maps). Current methods for analyzing myelin maps typically use global or regional mean myelin measurements to detect abnormalities, but ignore finer spatial patt

Contrastive pretraining improves deep learning classification of endocardial electrograms in a preclinical model.

BACKGROUND: Rotors and focal ectopies, or "drivers," are hypothesized mechanisms of persistent atrial fibrillation (AF). Machine learning algorithms have been used to identify these drivers, but the limited size of current driver data sets constrains their performance. OBJECTIVE: We proposed that pretraining using unsupervised learning on a substantial data set of unlabeled electrograms could enhance classifier accuracy when applied to a smaller driver data set. METHODS: We used a SimCLR-based framework to pretrain a residual neural network on 113,000 unlabeled 64-electrode measurements from

Emerging frontiers for AI in single-cell genomics.

We are just over a decade into the single-cell revolution in biology, which has been coupled to the growing use of artificial intelligence (AI) in single-cell genomics. Most early single-cell RNA embedding models and subsequent "foundation models"-representational models pretrained on large single-cell datasets-did not gain wide traction among experimental biologists. However, as massive single-cell RNA-sequencing (scRNA-seq) atlas efforts ramp up, there is a clear rationale for exploiting AI models to interpret new datasets in the context of vast existing data resources. Meanwhile, the advent

English-Persian cross-language plagiarism detection: Multilingual deep learning approach.

Cross-lingual plagiarism detection (CLPD) remains a critical challenge in Natural Language Processing, particularly for low-resource language pairs where translation and paraphrasing obscure source materials. This study introduces a robust framework for Persian-English CLPD by integrating state-of-the-art sentence representations with optimized machine learning classifiers. We utilized the BAAI/bge-m3 model to generate high-dimensional embeddings, employing cosine similarity to measure contrastive linguistic distances. To evaluate classification performance, we compared various architectures,

Shared-task Self-supervised Learning for Estimating Free Movement Unified Parkinson's Disease Rating Scale III.

The Unified Parkinson's Disease Rating Scale (UP-DRS) is used to recognize patients with Parkinson's disease (PD) and rate its severity in clinical settings. Machine learning and wearables can reduce the need for clinical examinations and provide a reliable estimation of the severity of PD at home. This work introduces a multi-channel convolutional neural network to estimate UPDRS part III from motion data recorded by two wearable sensors, considering the gyroscope signals and their spectrogram representations. A novel shared-task self-supervised learning is then employed to leverage the knowl

Explainable Multi-Modal Deep Learning for Recording-Level Classification of Respiratory Audio Signals Under Internal and Domain-Shift Evaluation.

Respiratory diseases are a major global health challenge. However, identification of respiratory diseases is often limited by subjectivity, environmental noise and inter-clinician variability. This study presents an explainable multimodal deep learning framework for recording-level multiclass classification of respiratory audio signals. The proposed system integrates two complementary representations-a spectro-temporal encoder based on a CNN-BiLSTM-attention architecture and a handcrafted acoustic-feature encoder capturing acoustic descriptors commonly used in respiratory-audio analysis, inclu

An Experimental Analysis on Multicepstral Projection Representation Strategies for Dysphonia Detection.

Biometrics-based authentication has become the most well-established form of user recognition in systems that demand a certain level of security. For example, the most commonplace social activities stand out, such as access to the work environment or to one's own bank account. Among all biometrics, voice receives special attention due to factors such as ease of collection, the low cost of reading devices, and the high quantity of literature and software packages available for use. However, these biometrics may have the ability to represent the individual impaired by the phenomenon known as dys

Representation Learning Approaches to Detect False Arrhythmia Alarms from ECG Dynamics.

The high rate of intensive care unit false arrhythmia alarms can lead to disruption of care and slow response time due to desensitization of clinical staff. We study the use of machine learning models to detect false ventricular tachycardia (v-tach) alarms using ECG waveform recordings. We propose using a Supervised Denoising Autoencoder (SDAE) to detect false alarms using a low-dimensional representation of ECG dynamics learned by minimizing a combined reconstruction and classification loss. We evaluate our algorithms on the PhysioNet Challenge 2015 dataset, containing over 500 records (over

Comparison of feature representations in MRI-based MCI-to-AD conversion prediction.

Alzheimer's disease (AD) is a progressive neurological disorder in which the death of brain cells causes memory loss and cognitive decline. The identification of at-risk subjects yet showing no dementia symptoms but who will later convert to AD can be crucial for the effective treatment of AD. For this, Magnetic Resonance Imaging (MRI) is expected to play a crucial role. During recent years, several Machine Learning (ML) approaches to AD-conversion prediction have been proposed using different types of MRI features. However, few studies comparing these different feature representations exist,
