Inhibition of the mitochondrial complex-1 protects against carbon tetrachloride-induced acute liver injury.

Mitochondrial dysfunction has been documented to play a crucial role in the pathogenesis of liver injury. In the present study, we investigated the role of rotenone, a mitochondrial complex-1 inhibitor, in carbon tetrachloride (CCl4) -induced acute liver injury, as well as the underlying mechanisms. Before CCl4 administration, the mice were pretreated with rotenone at a dose of 250 ppm in food for three days. Then CCl4 was administered to the mice for 16 h by intraperitoneal injection. The liver injury, mitochondrial status, oxidative stress, and inflammation were examined. Strikingly, CCl4 tr

The expression of MCOLN1 in preeclampsia maintains the balance of autophagy and pyroptosis induced by reactive oxygen species.

Reactive oxygen species (ROS) can stimulate autophagy and pyroptosis, which have been slightly reported in preeclampsia (PE). Mucolipin TRP Cation Channel 1 (MCOLN1) is a sensor for autophagy to receive ROS. This study aimed to investigate the role of MCOLN1 on ROS-mediated autophagy and pyroptosis in an in vitro model of PE. HTR-8/SVneo cells were treated with different concentrations of rotenone and pre-transfected with plasmids overexpressing or silencing MCOLN1. Thiazolyl blue tetrazolium bromide was used to examine the effects of rotenone and MCOLN1 on trophoblasts. ROS levels were detect

Methyl jasmonate abrogates rotenone-induced parkinsonian-like symptoms through inhibition of oxidative stress, release of pro-inflammatory cytokines, and down-regulation of immnopositive cells of NF-κB and α-synuclein expressions in mice.

Oxidative stress and neuroinflammation play key roles in the initiation and progression of Parkinson's disease (PD), a neurodegenerative disorder, associated with the loss of nigrostriatal dopaminergic pathway. Thus, compounds that can mitigate oxidative stress and neuroinflammation are being investigated as promising agents for the treatment of PD. This study was designed to evaluate the effects of methyl jasmonate (MJ), a potent antioxidant and anti-inflammatory compound on parkinsonian-like symptoms and the underlying biochemical changes induced by rotenone (Rot) in mice. To this end, the e

Hemorphin LVV-H3 attenuates calcineurin activity and regulates cytokine levels in experimental Parkinson's disease.

Dysregulation of intracellular Ca2+ levels and activation of the Ca2+/calmodulin-dependent phosphatase calcineurin (CaN) contribute to neurodegeneration and inflammatory responses associated with the Parkinson's disease (PD) pathogenesis. Hence, regulation of CaN activity may be beneficial in PD. Hemorphins, endogenous hemoglobin-derived peptides, play modulatory role in the brain and immune system in experimental pathologies. The effect of hemorphin LVV-H3 on CaN activity and proinflammatory cytokines (IL-2, TNFα) in the CNS (brain, spinal cord), peripheral tissues (thymus, spleen) and plasma

Involvement of Raphe Nuclei in Depressive-like Behaviors and Short-Term Memory in an Animal Model of Parkinson's Disease.

Parkinson's disease (PD) is a neurodegenerative disease characterized by motor symptoms and nonmotor disturbances such as fatigue, cognitive impairment, constipation, anxiety, and depression. The dopaminergic neurodegeneration is a hallmark of PD; however, the serotonergic system also presents alterations. Dorsal raphe nucleus (DRN) and median raphe nucleus (MNR) neurons are responsible for the central biosynthesis of serotonin (5-HT). We aimed to investigate whether the rotenone model would affect serotonergic neurons of the DRN and MRN and reduce 5-HT levels in target structures related to d

Differential Alterations in the Expression of AMPA Receptor and Its Trafficking Proteins in the Hippocampus Are Associated with Recognition Memory Impairment in the Rotenone-Parkinson's Disease Mouse Model: Neuroprotective Role of Bacopa monnieri Extract CDRI 08.

Parkinson's disease (PD), an age-associated neurodegenerative motor disorder, is associated with dementia and cognitive decline. However, the precise molecular insight into PD-induced cognitive decline is not fully understood. Here, we have investigated the possible alterations in the expression of glutamate receptor and its trafficking/scaffolding/regulatory proteins underlying the memory formation and neuroprotective effects of a specialized Bacopa monnieri extract, CDRI-08 (BME) in the hippocampus of the rotenone-induced PD mouse model. Our Western blotting and qRT-PCR data reveal that the

Carbon monoxide abrogates ischemic insult to neuronal cells via the soluble guanylate cyclase-cGMP pathway.

PURPOSE: Carbon monoxide (CO) is an accepted cytoprotective molecule. The extent and mechanisms of protection in neuronal systems have not been well studied. We hypothesized that delivery of CO via a novel releasing molecule (CORM) would impart neuroprotection in vivo against ischemia-reperfusion injury (IRI)-induced apoptosis of retinal ganglion cells (RGC) and in vitro of neuronal SH-SY5Y-cells via activation of soluble guanylate-cyclase (sGC). METHODS: To mimic ischemic respiratory arrest, SH-SY5Y-cells were incubated with rotenone (100 nmol/L, 4 h) ± CORM ALF186 (10-100 µmol/L) or inactiv

Crosstalk between SIRT1/Nrf2 signaling and NLRP3 inflammasome/pyroptosis as a mechanistic approach for the neuroprotective effect of linagliptin in Parkinson's disease.

In recent years, special attention has been paid to highlighting the antiparkinsonian effect of linagliptin. However, the mechanism of its action has not yet been well investigated. The present study aimed to verify the neuroprotective effect of linagliptin in the rotenone model of Parkinson's disease (PD) and further explore its potential molecular mechanisms. Rats were intoxicated with rotenone (2 mg/kg/day; sc) and treated with linagliptin (10  mg/kg/day; po) for 14 consecutive days. The present finding showed that linagliptin ameliorated the histopathological changes of rotenone on substan

Therapeutic potential of neural stem cell-derived exosomes and PRP in Parkinson's disease: A novel regenerative approach.

INTRODUCTION: Parkinson's disease (PD) is a neurological ailment characterized by oxidative stress, neuroinflammation, and the progressive loss of dopaminergic neurons in the substantia nigra. The current treatments lack disease-modifying capabilities. The current study investigated the neuroprotective and regenerative efficacy of a combination of neural stem cell-derived exosomes (NMSC-EXO) and PRP in a rotenone-induced rat model of PD. MATERIAL AND METHODS: Thirty male Wistar rats were randomly assigned to three equal groups (n = 10/group). Group I (negative Control): received subcutaneous

Inhibition of complex I regulates the mitochondrial permeability transition through a phosphate-sensitive inhibitory site masked by cyclophilin D.

Inhibition of the mitochondrial permeability transition pore (PTP) has proved to be an effective strategy for preventing oxidative stress-induced cell death, and the pore represents a viable cellular target for drugs. Here, we report that inhibition of complex I by rotenone is more effective at PTP inhibition than cyclosporin A in tissues that express low levels of the cyclosporin A mitochondrial target, cyclophilin D; and, conversely, that tissues in which rotenone does not affect the PTP are characterized by high levels of expression of cyclophilin D and sensitivity to cyclosporin A. Methylene blue provides behavioral and metabolic neuroprotection against optic neuropathy.

Methylene blue (MB) is a diaminophenothiazine with potent antioxidant and unique redox properties that prevent morphologic degenerative changes in the mouse retina induced by rotenone, a specific mitochondrial complex I inhibitor. This study evaluated pigmented rats to determine whether MB's neuroprotective effects against rotenone-mediated retinal neurotoxicity have functional relevance and whether these effects are mediated by an improvement in neuronal energy metabolism in vivo. Visual function was behaviorally assessed by determining differences in the illuminance sensitivity threshold pre

Mitochondrial complex inhibitors preferentially damage substantia nigra dopamine neurons in rat brain slices.

Using a rat brain slice preparation, we investigated the role of energy impairment on the selective loss of dopamine neurons in the substantia nigra (SN). Brain slices (400 microm) were incubated at 35 degrees C for 2 h in the presence or absence of mitochondrial complex inhibitors, rotenone, MPP+, 3-nitropropionic acid, and antimycin A. Slices were also incubated in rotenone with excitatory amino acid (EAA) receptor antagonists, MK-801 and CNQX, to determine whether rotenone-induced damage was mediated by EAAs. The slices were then fixed, recut into 30-microm sections, and immunolabeled for t

Inhibition of mitochondrial complex-1 restores the downregulation of aquaporins in obstructive nephropathy.

Obstructive kidney disease is a common complication in the clinic. Downregulation of aquaporins (AQPs) in obstructed kidneys has been thought as a key factor leading to the polyuria and impairment of urine-concentrating capability after the release of kidney obstruction. The present study was to investigate the role of mitochondrial complex-1 in modulating AQPs in obstructive nephropathy. Following 7-day unilateral ureteral obstruction (UUO), AQP1, AQP2, AQP3, and vasopressin 2 (V2) receptor were remarkably reduced as determined by qRT-PCR and/or Western blotting. Notably, inhibition of mitoch

p-Coumaric acid potential in restoring neuromotor function and oxidative balance through the Parkin pathway in a Parkinson disease-like model in Drosophila melanogaster.

p-Coumaric acid is a significant phenolic compound known for its potent antioxidant activity. Thus, this study investigated the effects of p-coumaric acid on the behavioral and neurochemical changes induced in Drosophila melanogaster by exposure to rotenone in a Parkinson disease (PD)-like model. The flies were divided into four groups and maintained for seven days on different diets: a standard diet (control), a diet containing rotenone (500 μM), a control diet to which p-coumaric acid was added on the fourth day (0.3 μM), and a diet initially containing rotenone (500 μM) with p-coumaric acid

Arterial baroreflex dysfunction fails to mimic Parkinson's disease in rats.

Patients with Parkinson's disease (PD) often have attenuated baroreflex function, which may occur before the onset of PD-associated movement disorders. The aim of the present study was to test whether impaired arterial baroreflex (ABR) function could contribute to the pathogenesis of PD. 6-Hydroxydopamine (8 mug in 4 mul) was microinjected into the left substantia nigra of rats to establish unilateral PD models, and bilateral PD models were established in rats by administration of rotenone by osmotic minipump for four weeks, at a dose of 2.5 mg . kg(-1) . day(-1). An ABR dysfunction model was

ATP13A2-mediated endo-lysosomal polyamine export counters mitochondrial oxidative stress.

Recessive loss-of-function mutations in ATP13A2 (PARK9) are associated with a spectrum of neurodegenerative disorders, including Parkinson's disease (PD). We recently revealed that the late endo-lysosomal transporter ATP13A2 pumps polyamines like spermine into the cytosol, whereas ATP13A2 dysfunction causes lysosomal polyamine accumulation and rupture. Here, we investigate how ATP13A2 provides protection against mitochondrial toxins such as rotenone, an environmental PD risk factor. Rotenone promoted mitochondrial-generated superoxide (MitoROS), which was exacerbated by ATP13A2 deficiency in S

The effect of mitochondrial inhibitors on membrane currents in isolated neonatal rat carotid body type I cells.

Inhibitors of mitochondrial energy metabolism have long been known to be potent stimulants of the carotid body, yet their mechanism of action remains obscure. We have therefore investigated the effects of rotenone, myxothiazol, antimycin A, cyanide (CN(-)) and oligomycin on isolated carotid body type I cells. All five compounds caused a rapid rise in intracellular Ca(2+), which was inhibited on removal of extracellular Ca(2+). Under current clamp conditions rotenone and CN(-) caused a rapid membrane depolarization and elevation of [Ca(2+)](i). Voltage clamping cells to -70 mV substantially att

Partial mitochondrial complex I inhibition induces oxidative damage and perturbs glutamate transport in primary retinal cultures. Relevance to Leber Hereditary Optic Neuropathy (LHON).

Leber Hereditary Optic Neuropathy (LHON) is a maternally inherited form of visual loss, due to selective degeneration of retinal ganglion cells. Despite the established aetiological association between LHON and mitochondrial DNA mutations affecting complex I of the electron transport chain, the pathophysiology of this disorder remains obscure. Primary rat retinal cultures were exposed to increasing concentrations of rotenone to titrate complex I inhibition. Neural cells were more sensitive than Müller glial cells to rotenone toxicity. Rotenone induced an increase in mitochondrial-derived free

Correlational assessment of the effects of JM-20 in a rat model of parkinsonism.

We previously demonstrated that JM-20, a molecule with neuroactive functions, protects rats against rotenone and 6-hydroxydopamine (6-OHDA) neurotoxicity. In addition, we demonstrated that JM-20 inhibits the aggregation and cytotoxicity of alpha-synuclein in vitro. In this study, we performed correlation studies between morphological and molecular variables, as well as the motor behavior of parkinsonian rats (6-OHDA and rotenone lesion) treated with JM-20 at different doses (oral with gavage). Our results showed that higher asymmetry evaluated in the cylinder test correlated with greater redox
