Navigating cross-reactivity and host species effects in a serological assay: A case study of the microscopic agglutination test for Leptospira serology.

BACKGROUND: Serology (the detection of antibodies formed by the host against an infecting pathogen) is frequently used to assess current infections and past exposure to specific pathogens. However, the presence of cross-reactivity among host antibodies in serological data makes it challenging to interpret the patterns and draw reliable conclusions about the infecting pathogen or strain. METHODOLOGY/PRINCIPAL FINDINGS: In our study, we use microscopic agglutination test (MAT) serological data from three host species [California sea lion (Zalophus californianus), island fox (Urocyon littoralis)

Detection of coccidioidal antibodies by 33-kDa spherule antigen, Coccidioides EIA, and standard serologic tests in sera from patients evaluated for coccidioidomycosis.

During a 9-month study of patients being evaluated for coccidioidomycosis, 1 or more serum samples were obtained from 138 patients with an illness suggestive of recent infection. In this group, standard immunodiffusion tests of unconcentrated sera were positive for 25; 49 additional patients had at least 1 reactive test result by newer enzyme-linked serologic tests. At least 11 of these 49 patients had coccidioidomycosis as determined by culture or subsequent standard serologic tests. Patients with coccidioidomycosis identified only by newer tests had fewer or milder clinical abnormalities tha

Applications of SARS-CoV-2 serological testing: impact of test performance, sample matrices, and patient characteristics.

Laboratory testing has been a key tool in managing the SARS-CoV-2 global pandemic. While rapid antigen and PCR testing has proven useful for diagnosing acute SARS-CoV-2 infections, additional testing methods are required to understand the long-term impact of SARS-CoV-2 infections on immune response. Serological testing, a well-documented laboratory practice, measures the presence of antibodies in a sample to uncover information about host immunity. Although proposed applications of serological testing for clinical use have previously been limited, current research into SARS-CoV-2 has shown gro

Assessment of milk ring test and some serological tests in the detection of Brucella melitensis in Syrian female sheep.

Brucella melitensis infection prevalence among Syrian female sheep, to evaluate a number of serological tests and to discuss some epidemiological aspects of brucellosis, was studied. A total of 2,580 unvaccinated Syrian female sheep sera samples were tested for B. melitensis antibodies detection using four serological methods: the Rose Bengal test (RBT), the serum agglutination test (SAT), the complement fixation test (CFT) and an indirect enzyme-linked immunosorbent assay (iELISA). In addition, 2,375 milk samples were collected, then milk ring test (MRT) and bacterial isolation test were empl

Serological diagnosis of Erysipelothrix rhusiopathiae: a comparative study between the growth inhibition test and the complement fixation test.

A Complement Fixation Test (CFT) was developed for the diagnosis of Erysipelas infections. The results of that work were compared with those of the Growth Inhibition Test (GIT), and they showed that the CFT test was very useful to determine whether an animal had been vaccinated or was still suffering from the disease. The CFT is as reliable as the GIT in detecting recent as well as chronic infections, but its value is limited where animals are examined for their response to a vaccination. The test is quick, easy, and safe to conduct.

Utility of Serological Tests in the Era of Molecular Testing for Diagnosis of Human Brucellosis in Endemic Area with Limited Resources.

BACKGROUND: The culture has always been the gold standard test for diagnosis of human brucellosis but the conventional Brucella diagnostic tests viz. serology and culture are often beset with poor specificity & sensitivity respectively. The culture positivity rates for Brucella vary from 92% for bone marrow to 10% for non-blood samples and also dependent on the type of sample. The primary immune-determinant for Brucella species is the cell wall surface lipopolysaccharide, which is antigenically similar to other gram-negative rods. Hence, Brucella serological tests cross react with Escherichia

Comparison of passive haemagglutination test with Widal agglutination test for serological diagnosis of typhoid fever in an endemic area.

A passive haemagglutination test, using sheep red blood cells sensitised with Salmonella typhi lipopolysaccharide, was compared with the Widal test for the serological diagnosis of typhoid fever in an endemic area. The results obtained on sera from 152 patients with bacteriologically confirmed typhoid and 183 patients who did not have typhoid were analysed in terms of sensitivity, specificity, simplicity, and rapidity of the respective tests. The passive haemagglutination test was found to be more sensitive (80%) than the S typhi O antigen (71%) but marginally less sensitive than the H antigen

Agar-gel precipitin-inhibition test for coccidioidomycosis. II. Serological test antigen studies in agar-gel.

In this study of saprophytic and parasitic growth-phase extracts of Coccidioides immitis, an antigen from the spherule culture supernatant fluid detected a specific antibody in heretofore serologically negative suspect coccidioidomycosis human sera when diffused in agar-gel. This antigen-antibody reaction occurred also in some of the serologically positive human coccidioidomycosis sera. This study indicates that this antigen-antibody reaction should be utilized as a possible routine serological test for complete serodiagnosis.

Serological and Clinical Evaluation of Incompatible Crossmatches Encountered in Pre-transfusion Testing.

Background Pre-transfusion testing is essential for preventing incompatible red cell transfusions and associated adverse reactions. Despite modern serological advancements, unexpected alloantibodies, autoantibodies, and monoclonal antibody interference continue to contribute to incompatible crossmatches. This study evaluates the serological patterns, clinical associations, and diagnostic considerations of incompatible crossmatches encountered in routine transfusion practice. Methods This cross-sectional study was conducted at a tertiary-care transfusion service of Swami Rama Himalayan Universi

Red cell cryopreservation for serologic testing: bulk freezing with polyvinylpyrrolidone (PVP).

BACKGROUND/AIM: Red cell cryopreservation is essential for immunohematology reference laboratories. Various cryopreservation methods maintain the structural integrity of red cells, such as bulk freezing with glycerol and droplet freezing with polyvinylpyrrolidone (PVP). In this paper, we provide a technical note with real-world data and report our experience with a modified PVP bulk-freezing method in a transfusion medicine setting. MATERIALS AND METHODS: Red cells were separated from whole blood samples and incubated for 1 h at room temperature with PVP with slow agitation or no agitation be

Candidate antigens for serological testing (ELISA) and diagnosis of canine visceral leishmaniasis.

The diagnosis of canine visceral leishmaniasis (CVL) relies on parasitological, molecular methods and serological. However, there are difficulties in using these methods to monitor the disease, and there is a need for more efficient diagnostic alternatives for dogs with suspected visceral leishmaniasis, also using clinical aspects of the animals, which that can be useful for surveillance and control. This study aimed to evaluate the serological diagnostic potential of recombinant antigens in order to improve CVL diagnosis. Disease diagnosis, clinical evaluation, and collection of biological sa

Navigating the Diagnostic Challenges of Mycoplasma and Legionella Coinfection Through Serological Testing.

Diagnosing community-acquired pneumonia (CAP) is increasingly challenging, especially with the emergence of atypical pathogens such as Mycoplasma pneumoniae and Legionella pneumophila. This report presents the case of a 60-year-old male exhibiting lethargy and decreased oral intake, with a medical history marked by chronic kidney disease and benign prostate hyperplasia. Despite a positive Legionella urine antigen, the clinical and radiological profile did not align with Legionella pneumonia. Elevated M. pneumoniae IgM antibody titers further complicated the diagnostic scenario. We explore the
