Natural product anticipation through synthesis.

Natural product synthesis remains one of the most vibrant and intellectually rewarding areas of chemistry, although the justifications for pursuing it have evolved over time. In the early years, the emphasis lay on structure elucidation and confirmation through synthesis, as exemplified by celebrated studies on cocaine, morphine, strychnine and chlorophyll. This was followed by a phase where the sheer demonstration that highly complex molecules could be recreated in the laboratory in a rational manner was enough to justify the economic expense and intellectual agonies of a synthesis. Since the

Evaluation of some neuropharmacological effects of Caladium bicolor aiton (araceae) leaf extracts in mice.

Caladium bicolor Aiton (Araceae) is used in ethnomedicine for the treatment of boils, wound ulcers and convulsion. This study investigated the effects of the leaf extracts on some neuropharmacological parameters. The leaves were collected, dried, powdered and then extracted by maceration in methanol to yield the whole extract (WE). Extraction was also done using n-hexane, ethyl acetate and methanol in a Soxhlet apparatus to obtain n-hexane (HE), ethyl acetate (EA) and methanol (ME) extracts. Preliminary phytochemical screening was done using the whole extract. Some neuropharmacological evaluat

Structure and Pharmacologic Modulation of Inhibitory Glycine Receptors.

Glycine receptors (GlyR) are inhibitory Cys-loop ion channels that contribute to the control of excitability along the central nervous system (CNS). GlyR are found in the spinal cord and brain stem, and more recently they were reported in higher regions of the CNS such as the hippocampus and nucleus accumbens. GlyR are involved in motor coordination, respiratory rhythms, pain transmission, and sensory processing, and they are targets for relevant physiologic and pharmacologic modulators. Several studies with protein crystallography and cryoelectron microscopy have shed light on the residues an

Evaluation of Behavioral and Pharmacological Effects of Hydroalcoholic Extract of Valeriana prionophylla Standl. from Guatemala.

There are few studies on the pharmacological properties of Valeriana prionophylla Standl. (VP), known as "Valeriana del monte", and used in Mesoamerican folk medicine to treat sleep disorders. This study examines the pharmacological effects of the hydroalcoholic extract of the dry rhizome using the open field, rota rod, elevated plus-maze (EPM), forced swimming (FST), strychnine- and pentobarbital-induced sleeping time, PTZ-induced seizures, and the inhibitory avoidance tests. VP did not show any protective effect against PTZ-induced convulsions. In the EPM, exhibited an anxiolytic-like effect

Analgesic action of nicotine on tibial nerve transection (TNT)-induced mechanical allodynia through enhancement of the glycinergic inhibitory system in spinal cord.

The activation of cholinergic pathways by nicotine elicits various physiological and pharmacological effects in mammals. For example, the stimulation of nicotinic acetylcholine receptors (nAChRs) leads to an antinociceptive effect. However, it remains to be elucidated which subtypes of nAChR are involved in the antinociceptive effect of nicotine on nerve injury-induced allodynia and the underlying cascades of the nAChR-mediated antiallodynic effect. In this study, we attempted to characterize the actions of nicotine at the spinal level against mechanical allodynia in an animal model of neuropa

Inhibitory effects of corymine, an alkaloidal component from the leaves of Hunteria zeylanica, on glycine receptors expressed in Xenopus oocytes.

We previously reported that corymine, an alkaloidal compound extracted from the leaves of Hunteria zeylanica native to Thailand, potentiated convulsions induced by either picrotoxin or strychnine. Therefore, to clarify the mechanism of action of corymine, the effects of corymine on gamma-aminobutyric acid (GABA) and glycine receptors were examined. We used Xenopus oocytes expressing these receptors and the two-electrode voltage-clamp method. The receptors expressed in oocytes injected with rat brain and spinal cord RNA showed the pharmacological properties of GABAA and glycine receptors, respe

Anatomical, physiological, and theoretical basis for the antiepileptic effect of vagus nerve stimulation.

The vagus is a mixed nerve carrying somatic and visceral afferents and efferents. The majority of vagal nerve fibers are visceral afferents and have a wide distribution throughout the central nervous system (CNS) either monosynaptically or via the nucleus of the solitary tract. Besides activation of well-defined reflexes, vagal stimulation produces evoked potentials recorded from the cerebral cortex, the hippocampus, the thalamus, and the cerebellum. Activation of vagal afferents can depress monosynaptic reflexes, decrease the activity of spinothalamic neurons, and increase pain threshold. Dep

A common structural model for central nervous system drugs and their receptors.

On the basis of the hypothesis that there is a common structural basis for central nervous system (CNS) drug action consisting primarily of an aromatic group and a nitrogen atom, a four-point model for a common pharmacophore is defined with use of five semirigid CNS-active drug molecules: morphine, strychnine, LSD, apomorphine, and mianserin. Two of the points of the model represent possible hydrophobic interactions between the aromatic group and the receptor, while the other two represent hydrogen bonding between the nitrogen atom and the receptor. The model is then extended by the inclusion

Pharmacological disinhibition enhances paced breathing following complete spinal cord injury in rats.

Respiratory dysfunction is one of the most devastating and life-threatening deficits that occurs following cervical spinal cord injury (SCI). Assisted breathing with mechanical ventilators is a necessary part of care for many cervical injured individuals, but it is also associated with increased risk of secondary complications such as infection, muscle atrophy and maladaptive plasticity. Pre-clinical studies with epidural stimulation (EDS) have identified it as an alternative/additional method to support adequate lung ventilation without mechanical assistance. The full potential of EDS, howeve

Adulterants in illicit drugs: a review of empirical evidence.

Widespread public perception is that illicit drugs contain substances that are a serious risk to health, even though adulterants are often not considered in clinical or forensic toxicology. This review attempts to present an evidence-based overview of adulterants in illicit drugs, and their associated toxicity. Adulterants are deliberately added to increase bulk, enhance or mimic a pharmacological effect, or to facilitate drug delivery. Those present unintentionally are as a result of poor manufacturing techniques. From the reports gathered, adulterants are predominantly substances which are r

Chloride-dependent acute excitotoxicity in adult rat retinal ganglion cells.

Mechanisms of excitotoxic degeneration of retinal ganglion cells (RGCs) remain controversial, due to the lack of suitable in vitro experimental systems for evaluation of RGC death. In this study, we investigated acute excitotoxicity in RGCs using eyecup preparations obtained from adult rats, with special reference to ionic dependence of N-methyl-D-aspartate (NMDA) and kainate toxicity. Retrograde labeling of RGCs with a fluorescent tracer diamidino yellow, combined with labeling of dead cells by propidium iodide, enabled us to discriminate dead RGCs from other cells in the ganglion cell layer.

Identification of active thoracic spinal segments responsible for tonic and bursting sympathetic discharge in neonatal rats.

The isolated thoracic cord of a neonatal rat in vitro generates tonic sympathetic activities in the splanchnic nerves. This tonic sympathetic nerve discharge (SND) has a prominent quasi-periodic oscillation at approximately 1-2 Hz. Bath application of bicuculline and strychnine, which removes endogenous GABA(A) and glycine receptor activities, transforms the quasi-periodic tonic SND into synchronized bursts (bSND). Picrotoxin, another GABA(A) receptor antagonist, also induces bSND. Serial transections of the thoracic cord (T1-12) were performed to identify the cord segments responsible for the

CNS inhibitory effects of barakol, a constituent of Cassia siamia Lamk.

The present study determined the pharmacological profile of barakol, a major constituent of Cassia siamea Lamk., in rodent behavioral and neurochemical tests. Barakol reduced spontaneous locomotor activity, increased the number of sleeping animals and prolonged the thiopental-induced sleeping time, indicating a sedative effect. As for interactions between barakol and convulsants (pentylenetetrazole (PTZ), picrotoxin, bicuculline and strychnine), only a high dose (100 mg/kg, i.p.) of barakol slightly prolonged the latency of clonic convulsion induced by picrotoxin. This suggests that the sedati

Contralateral sound suppresses distortion product otoacoustic emissions through cholinergic mechanisms.

Presentation of an acoustic signal to one ear can suppress sound-evoked activity recorded at the opposite ear. The suppression appears to be mediated by medial olivocochlear (MOC) efferent neurons synapsing with outer hair cells (OHCs) and acting through the MOC neurotransmitter, acetylcholine (ACh). The purpose of the present investigation was to study the suppression of distortion product otoacoustic emissions (DPOAEs) by contralateral sound and to examine whether the suppression could be blocked by known antagonists of olivocochlear (OC) efferent activity. Urethane-anesthetized guinea pigs

Studies on the mechanism of hypoglycemia induced by intrathecal morphine: dissociation from behavioral effects, effects of tolerance and depletion of liver glycogen.

To characterize further the hypoglycemic effect of intrathecally (i.t.) administered morphine, species and drug specificity, effects of morphine-induced tolerance and pentobarbital-induced anesthesia and effects on liver glycogen were studied in nonfasted animals. In rats, morphine (125 micrograms i.t.) produced the same behavioral toxicity (scratching, biting, seizures) and hypoglycemia as previously reported in mice. In mice, the glycine antagonist strychnine (5 micrograms i.t.) and the morphine metabolite morphine-3-glucuronide (2 micrograms i.t.) mimicked the behavioral, but not the hypogl

Avermectin B1a modulation of gamma-aminobutyric acid/benzodiazepine receptor binding in mammalian brain.

The anthelminthic natural product avermectin B1a (AVM) modulates the binding of gamma-aminobutyric acid (GABA) and benzodiazepine (BZ) receptor ligands to membrane homogenates of mammalian brain. The potent (EC50 = 40 nM) enhancement by AVM of [3H]diazepam binding to rat or bovine brain membranes resembled that of barbiturates and pyrazolopyridines in being inhibited (partially) by the convulsants picrotoxin, bicuculline, and strychnine, and by the anticonvulsants phenobarbital and chlormethiazole. The maximal effect of AVM was not increased by pentobarbital or etazolate. However, AVM affected

Medicinal plants--old and new.

The historic role of plants in healing declined early in the twentieth century with the ascendency of synthetic drugs, even though a number of basic medical tools, such as opium, strychnine, and cocaine, are of botanical origin. In recent years, interest in natural products has been restored dramatically by the discovery of penicillin, plant-derived tranquilizers, and plant precursors of cortisone. Contrary to previous beliefs, botanical drugs are proving more economical than synthetics and hold forth encouraging prospects of inhibiting or destroying tumors without undue damage to healthy tiss

EXPERIMENTAL STUDIES ON THE RELATION OF THE PITUITARY BODY TO RENAL FUNCTION.

1. The inconstant results of past observations on the relation of pituitary extracts to renal activity have been due chiefly to unsuitable methods. 2. A standard curve of artificially induced polyuria may be plotted for rabbits, giving 200 cc. of water by mouth. 3. Extracts of the pars intermedia and posterior lobe of the hypophysis, given by mouth, subcutaneously, or intravenously, are able definitely to check polyuria thus induced. Extracts of the anterior lobe show a similar effect, but only to a slight degree. 4. This antidiuretic effect is constant, and is independent of (a) changes in bl
