Swedish Alzheimer's disease variant perturbs activity of retrograde molecular motors and causes widespread derangement of axonal transport pathways.

Experimental studies in flies, mice, and humans suggest a significant role of impaired axonal transport in the pathogenesis of Alzheimer's disease (AD). The mechanisms underlying these impairments in axonal transport, however, remain poorly understood. Here we report that the Swedish familial AD mutation causes a standstill of the amyloid precursor protein (APP) in the axons at the expense of its reduced anterograde transport. The standstill reflects the perturbed directionality of the axonal transport of APP, which spends significantly more time traveling in the retrograde direction. This ine

WDR72 regulates vesicle trafficking in ameloblasts.

As the hardest tissue in the human body, tooth enamel formation is a highly regulated process involving several stages of differentiation and key regulatory genes. One such gene, tryptophan-aspartate repeat domain 72 (WDR72), has been found to cause a tooth enamel defect when deleted or mutated, resulting in a condition called amelogenesis imperfecta. Unlike the canonical genes regulating tooth development, WDR72 remains intracellularly and is not secreted to the enamel matrix space to regulate mineralization, and is found in other major organs of the body, namely the kidney, brain, liver, and

The roles of ABCA12 in keratinocyte differentiation and lipid barrier formation in the epidermis.

ABCA12 is a member of the large superfamily of ATP-binding cassette (ABC) transporters, which bind and hydrolyze ATP to transport various molecules across limiting membranes or into vesicles. The ABCA subfamily members are thought to be lipid transporters. ABCA12 is a keratinocyte transmembrane lipid transporter protein associated with the transport of lipids in lamellar granules to the apical surface of granular layer keratinocytes. Extracellular lipids, including ceramide, are thought to be essential for skin barrier function. ABCA12 mutations are known to underlie the three main types of au

Melatonin localization in human salivary glands.

BACKGROUND: Circulating melatonin is believed to reach body fluids by crossing passively the cell membranes, but alternative ways for melatonin transport also are hypothesized. This investigation was carried out to furnish ultrastructural evidences for melatonin transport by salivary gland cells in order to indicate plausible routes by which circulating melatonin can reach saliva. METHODS: Bioptic samples of parotid, submandibular and labial glands were processed for the electron microscopy and treated to demonstrate melatonin reactivity by the immunogold staining method. RESULTS AND CONCLUS

Exploitation of the endoplasmic reticulum by bacterial pathogens.

The endoplasmic reticulum (ER) has unique properties that are exploited by microbial pathogens. Exotoxins secreted by bacteria take advantage of the host transport pathways that deliver proteins from the Golgi to the ER. Transport to the ER is necessary for the unfolding and translocation of these toxins into the cytosol where their host targets reside. Intracellular pathogens subvert host vesicle transport to create ER-like vacuoles that support their intracellular replication. Investigations on how bacterial pathogens can use the ER during host infection are providing important details on tr

[Therapeutic mechanism of Astragalus polysaccharides in bladder dysfunction in DBA mice based on regulatory role of myosin Va in neurotransmission].

This study aimed to investigate the therapeutic mechanism of Astragalus polysaccharides(APS) in improving bladder function in DBA mice by focusing on the repair of neurotransmission processes via the myosin Va pathway, specifically addressing its role in transport and docking of neurotransmitter vesicles. DBA mice were used to assess bladder dysfunction based on metabolic cage monitoring and urodynamics. The experiment included a control group, a DBA group, APS low-, medium-, and high-dose(0.256, 0.513, and 1.026 g·kg~(-1)) groups, and a positive control mecobalamin(0.195 g·kg~(-1)) group. Aft

ULK1 phosphorylates Sec23A and mediates autophagy-induced inhibition of ER-to-Golgi traffic.

BACKGROUND: Autophagy is an inducible autodigestive process that allows cells to recycle proteins and other materials for survival during stress and nutrient deprived conditions. The kinase ULK1 is required to activate this process. ULK1 phosphorylates a number of target proteins and regulates many cellular processes including the early secretory pathway. Recently, ULK1 has been demonstrated to phosphorylate Sec16 and affects the transport of serotonin transporter at the ER exit sites (ERES), but whether ULK1 may affect the transport of other cargo proteins and general secretion has not been f

Engineering of Glioblastoma-Derived Biomimetic Vesicles and Their Structural and Molecular Features.

Biomimetic nanosystems and vesicles have arisen as a novel approach to design vesicular transport systems with diverse therapeutic potential. The 'biomimetic' strategy involves the integration of cell membrane components into lipid bilayers, conferring them with biological properties originating from the cell of origin. Until now, most studies have primarily focused on the evaluation of the biological activity and function of different biomimetic nanosystems with limited exploration of the engineering parameters selected and little characterization of their features at the molecular level. Double-labeled rabies virus: live tracking of enveloped virus transport.

Here we describe a strategy to fluorescently label the envelope of rabies virus (RV), of the Rhabdoviridae family, in order to track the transport of single enveloped viruses in living cells. Red fluorescent proteins (tm-RFP) were engineered to comprise the N-terminal signal sequence and C-terminal transmembrane spanning and cytoplasmic domain sequences of the RV glycoprotein (G). Two variants of tm-RFP were transported to and anchored in the cell surface membrane, independent of glycosylation. As shown by confocal microscopy, tm-RFP colocalized at the cell surface with the RV matrix and G pro

Organoid-Derived Extracellular Vesicles: From Biogenesis and Cargo Mechanisms Toward Therapeutic Applications.

Organoids are three-dimensional in vitro models that mimic the anatomical and functional complexity of biological tissues, evolving into a powerful platform for studying human development and disease. Organoid-derived extracellular vesicles have been revealed as important intercellular messengers within organoid systems. Participating in the transport of functional substances such as proteins, lipids, and nucleic acids, these lipid-coated nanometer-scale carriers play a role in regulating biological responses. Unlike extracellular vesicles derived from 2D culture, organoid-derived extracellula

Influence of colchicine and vinblastine on the intracellular migration of secretory and membrane glycoproteins: II. Inhibition of secretion of thyroglobulin in rat thyroid follicular cells as visualized by radioautography after 3H-fucose injection.

Young (40 gm) rats were given a single intravenous injection of colchicine (4.0 mg) or vinblastine (2.0 mg). At 10 min after colchicine and 30 min after vinblastine administration, the rats were injected with 3H-fucose. Control rats received 3H-fucose only. All rats were sacrificed 90 min after 3H-fucose injection and their tissues processed for radioautography. In thyroid follicular cells of control animals, at this time interval, 57% of the total label was associated with colloid and secretory vesicles in the apical cytoplasm while 27% was localized in the Golgi apparatus and neighboring ves

Intestinal goblet cells sample and deliver lumenal antigens by regulated endocytic uptake and transcytosis.

Intestinal goblet cells maintain the protective epithelial barrier through mucus secretion and yet sample lumenal substances for immune processing through formation of goblet cell associated antigen passages (GAPs). The cellular biology of GAPs and how these divergent processes are balanced and regulated by goblet cells remains unknown. Using high-resolution light and electron microscopy, we found that in mice, GAPs were formed by an acetylcholine (ACh)-dependent endocytic event remarkable for delivery of fluid-phase cargo retrograde into the trans-golgi network and across the cell by transcyt

Role of microvesicles in acute kidney injury.

The main function of microvesicles (MVs) is signaling through specific interactions with target cells and transferring gene products. Therefore, they may participate in physiological and pathological processes. Gaining further insights into the molecular specificity of MVs has allowed identifying the cellular source and may provide new diagnostic tools in the future. Indeed, an increasing body of evidence indicates that MVs may offer prognostic information in various diseases such as chronic inflammation, cardiovascular and renal diseases, pathological pregnancy, tumors, and sepsis. Localization and activities of lysosomal enzymes in jejunal and ileal epithelial cells of the young pig.

The localization and the specific activities of lysosomal enzymes in jejunal and ileal epithelium of suckling pigs were determined. Acid phosphatase was found in pinocytotic vesicles and vacoles at both intestinal sites in pigs examined during (less than or equal to 1 day old) or shortly after (2 days old) closure. These pinocytolically active vesicles and vacuoles were determined to be phagolysosomes. In the ileum, phagolysosomes persisted well after closure, at an age (10 days) when ileal epithelium retains the capacity for pinocytotic uptake of immunoglobulin, but does not transport it to b

Endocytosis of cationized ferritin to vesicles in the Golgi region of the glomus cells dissociated from the adult rat carotid body.

Glomus cells were dissociated from the carotid bodies of adult rats by enzymatic digestion with collagenase. The cells were then incubated at 37 degrees C for 30 minutes to 3 hours in the continuous presence of cationized ferritin (CF) as a membrane marker and extracellular tracer to study the intracellular route of endocytosis in this cell type. After 30 minutes of incubation with CF, occasional solitary CF-containing vesicles were observed at the cell periphery and also in the Golgi region. After 2-3 hours of incubation with CF, cell viability was still preserved and CF-labeled vesicles were

KIF1A-associated neurological disorders: therapeutic opportunities and challenges.

KIF1A-Associated Neurological Disorder (KAND) is a rare, progressive neurodegenerative condition caused by variants in the KIF1A gene, which encodes a kinesin-3 motor protein essential for anterograde axonal transport of synaptic vesicles, dense core vesicles, and organelles in neurons. KAND comprises a broad spectrum of overlapping neurological phenotypes, including hereditary spastic paraplegia, intellectual disability, peripheral neuropathy, optic nerve atrophy, epilepsy, and progressive motor decline. Pathogenic variants in KIF1A disrupt the balance of intracellular transport and neuronal

Mechanisms of Neurological Dysfunction in GOSR2 Progressive Myoclonus Epilepsy, a Golgi SNAREopathy.

Successive fusion events between transport vesicles and their target membranes mediate trafficking of secreted, membrane- and organelle-localised proteins. During the initial steps of this process, termed the secretory pathway, COPII vesicles bud from the endoplasmic reticulum (ER) and fuse with the cis-Golgi membrane, thus depositing their cargo. This fusion step is driven by a quartet of SNARE proteins that includes the cis-Golgi t-SNARE Membrin, encoded by the GOSR2 gene. Mis-sense mutations in GOSR2 result in Progressive Myoclonus Epilepsy (PME), a severe neurological disorder characterise

Trafficking the NGF signal: implications for normal and degenerating neurons.

Nerve growth factor (NGF) activates TrkA to trigger signaling events that promote the survival, differentiation and maintenance of neurons. The mechanism(s) that controls the retrograde transport of the NGF signal from axon terminals to neuron cell bodies is not known. The 'signaling endosome' hypothesis stipulates that NGF, TrkA and signaling proteins are retrogradely transported on endocytic vesicles. Here, we provide evidence for the existence of signaling endosomes. Following NGF treatment, clathrin-coated vesicles (CCVs) contain NGF bound to TrkA together with activated signaling proteins

Venture from the Interior-Herpesvirus pUL31 Escorts Capsids from Nucleoplasmic Replication Compartments to Sites of Primary Envelopment at the Inner Nuclear Membrane.

Herpesviral capsid assembly is initiated in the nucleoplasm of the infected cell. Size constraints require that newly formed viral nucleocapsids leave the nucleus by an evolutionarily conserved vescular transport mechanism called nuclear egress. Mature capsids released from the nucleoplasm are engaged in a membrane-mediated budding process, composed of primary envelopment at the inner nuclear membrane and de-envelopment at the outer nuclear membrane. Once in the cytoplasm, the capsids receive their secondary envelope for maturation into infectious virions. Two viral proteins conserved througho

A calcium pump in plasma membrane vesicles from Leishmania braziliensis.

A subcellular fraction highly enriched in plasma membrane vesicles was prepared from Leishmania promastigotes. This fraction showed (Ca2+ + Mg2+)-ATPase activity. This, however, represented a small fraction (about 25%) of the overall ATPase activity. The Ca2(+)-ATPase showed general characteristics common to plasma membrane ATPases involved in Ca2+ transport. Thus, the Ca2(+)-ATPase was activated by Ca2+ with a high affinity (Km about 0.7 microM), saturating at about 5 microM Ca2+. Furthermore, it was stimulated by calmodulin (about 70-80% with 5 micrograms/ml) and almost fully inhibited by tr
