The role of dirt floors and of firewood in rural dwellings in the epidemiology of Chagas' disease in Costa Rica.

In an endemic area of Chagas' disease in Costa Rica, 50 houses infested with Triatoma dimidiata were re-examined after a period of 14-17 years. Criteria used were two socioeconomic parameters which are closely associated with the presence of the bugs: colonies of triatomines inside houses are favored by the presence of a dirt floor, and stored firewood is an excellent refuge for insects outdoors. Indoor infestation was completely eliminated from nine of 13 houses in which the floors had been changed from dirt to concrete during this period, and nearly eliminated from the other four, supporting

Nuclear Morphofunctional Organization and Epigenetic Characteristics in Somatic Cells of T. infestans (Klug, 1834).

Triatoma infestans (Klug) is an insect recognized as not only an important vector of South American trypanosomiasis (Chagas disease) but also a model of specific cellular morphofunctional organization and epigenetic characteristics. The purpose of the present review is to highlight certain cellular processes that are particularly unveiled in T. infestans, such as the following: (1) somatic polyploidy involving nuclear and cell fusions that generate giant nuclei; (2) diversification of nuclear phenotypes in the Malpighian tubules during insect development; (3) heterochromatin compartmentalizati

Effect of ectoparasitic Pimeliaphilus plumifer mites (Acari: Pterygosomatidae) on Meccus pallidipennis (Hemiptera: Reduviidae) and several other Chagas' disease vectors under laboratory conditions.

Several biological parameters were evaluated to determine the capacity of Pimeliaphilus plumifer as biological control agent of Triatominae bugs. When P. plumifer and bugs of a variety of triatomine species were forced together in cages in the laboratory, the incidence of mite infestation was the following: Meccus pallidipennis > M. bassolsae > Triatoma rubida > M. longipennis > M. picturatus, and practically no mites were found on T. infestans and Rhodnius prolixus. Adults and hexapod larvae of P. plumifer were the only stages found to parasitize on M. pallidipennis. Fourth and fifth instar n

Patterns of change in vector-borne diseases.

The statuses of vector-borne diseases have changed over recent years. How a few such diseases have changed and the primary causes of change (urbanization, increased conflict, changes in water-resource management, ecological and environmental change, and reduced health service resourcing) are the subjects of the present review. The key impacts which these primary causes have on selected vectors and the infections they transmit are tabulated. The success of vector-control programmes against onchocerciasis and Chagas disease is discussed, and the methods used to evaluate the epidemiological impac

Nationwide molecular epidemiology of Trypanosoma cruzi TcI in El Salvador using multi-locus sequence typing.

Chagas disease, caused by the protozoan parasite Trypanosoma cruzi, remains a major public health problem in Latin America. In endemic countries, transmission to humans is primarily mediated by infected triatomine bugs. In this study, we investigated the molecular epidemiology of T. cruzi in Triatoma dimidiata, the principal vector in El Salvador. A total of 145 T. cruzi DNA samples were obtained from T. dimidiata collected from anthropic (domestic and peri-domestic) environments nationwide. Discrete typing unit analysis followed by multi-locus sequence typing (MLST) using thirteen housekeepin

Triatomicidal effect of new spot-on formulations applied to poultry in semi-field conditions.

Chagas disease is an endemic disease affecting ten million people in the American continent. Produced by a parasite transmitted by triatomine insects, the main actions for reducing the incidence of this disease are focused on the control of insect vectors. This type of control has produced highly effective results within rural homes, but not in peridomestic areas (kitchens, warehouses, hen houses and other buildings not attached to the houses). The object of the present study was to assess the triatomicidal effect of new spot-on formulations developed by our laboratory in a semi-rural environm

Susceptibility and resistance to insecticides of Chagas disease vectors.

Chemical control of Chagas disease vectors appears to be the best practical way to reduce the incidence of the disease. DDT was initially tested in the 1950s for the campaigns of control of Chagas disease vectors. Its low level of effectiveness against triatomine caused the failure of these control actions. HCH was then introduced in the southern cone and Dieldrin in the north of Latinoamerica. Starting in the late 1960s anticholinesterasic organophosphorus and carbamate compounds were introduced in the control of Chagas vectors. The use of pyrethroid compounds began in 1980. This family of in

Chagas infection transmission control: situation of transfusional transmission in Brazil and other countries of Latin America.

The transmission of the transfusion-associated Chagas disease is an important mechanism of its dissemination in several Latin American countries. The transmission risk depends on five factors: prevalence of infection in blood donors, degree of serological coverage, sensibility of used tests, safety of obtained results and infection risk. The Southern Cone Iniciative set off by the Pan-American Health Organization, in 1991, is contributing to the implementation of blood law in each endemic country, and to reduce the risk of transfusional transmission of this horrible disease. Despite the clear

Molecular epidemiology of Trypanosoma cruzi I infection in northern Argentina.

Trypanosoma cruzi I (TcI) infects a broad range of mammalian and triatomine species across the Americas and displays broad genetic diversity. The transmission cycles of TcI remain poorly understood. We investigated whether TcI parasites from Didelphis albiventris opossums were linked to the TcI parasites found in domestic dogs, cats and peridomestic Triatoma sordida from the Argentinean Chaco or were linked to other T. cruzi genotypes in opossums. We analyzed the intergenic region of the spliced leader (SL-IR) of the mini-exon gene sequence from direct triatomine samples and culture isolates f

Venomous bites to the external genitalia: an unusual cause of acute scrotum.

The acute scrotum generates a long list of differential diagnoses. An unusual etiology includes insect envenomation, which typically is an acute process with rapid onset of symptomatologies. Two patients with genital envenomation are reported. We review the reported cases at our institution with all types of bites and stings. Symptoms of pain and pruritus, and signs of ecchymosis and edema preceding exfoliating dermatitis were evident in both cases. Mild analgesics and antihistamines promoted resolution in each instance.

One Health Interactions of Chagas Disease Vectors, Canid Hosts, and Human Residents along the Texas-Mexico Border.

BACKGROUND: Chagas disease (Trypanosoma cruzi infection) is the leading cause of non-ischemic dilated cardiomyopathy in Latin America. Texas, particularly the southern region, has compounding factors that could contribute to T. cruzi transmission; however, epidemiologic studies are lacking. The aim of this study was to ascertain the prevalence of T. cruzi in three different mammalian species (coyotes, stray domestic dogs, and humans) and vectors (Triatoma species) to understand the burden of Chagas disease among sylvatic, peridomestic, and domestic cycles. METHODOLOGY/PRINCIPAL FINDINGS: To d

Control of Chagas disease.

The Southern Cone Initiative (Iniciativa de Salud del Cono Sur, INCOSUR) to control domestic transmission of Trypanosoma cruzi is a substantial achievement based on the enthusiasm of the scientific community, effective strategies, leadership, and cost-effectiveness. INCOSUR triggered the launch of other regional initiatives in Central America and in the Andean and Amazon regions, which have all made progress. The Central American Initiative targeted the elimination of an imported triatomine bug (Rhodnius prolixus) and the control of a widespread native species (Triatoma dimidiata), and faced c

Targeted deletion of the gp72 gene decreases the infectivity of Trypanosoma cruzi for mice and insect vectors.

The infective behavior of a mutant Trypanosoma cruzi clone, carrying a targeted deletion of the gp72 gene, was studied in the insect vector Triatoma infestans and in mice. After feeding T. infestans with complement-resistant forms (CRF) of Ynull and wild-type clones, it was observed that the number of parasites released in the bug's feces was reduced to less than 1% in the mutant clone. Both gp72-null and wild-type clones had a low infectivity for mice in comparison with other T. cruzi isolates, probably as a consequence of prolonged in vitro culture. Therefore, the behavior of both clones was

Triatomines: Trypanosomatids, Bacteria, and Viruses Potential Vectors?

Triatominae bugs are the vectors of Chagas disease, a major concern to public health especially in Latin America, where vector-borne Chagas disease has undergone resurgence due mainly to diminished triatomine control in many endemic municipalities. Although the majority of Triatominae species occurs in the Americas, species belonging to the genus Linshcosteus occur in India, and species belonging to the Triatoma rubrofasciata complex have been also identified in Africa, the Middle East, South-East Asia, and in the Western Pacific. Not all of Triatominae species have been found to be infected w

A Case of Cardboard Boxes Likely Facilitating the Biting of a Patient by Trypanosoma cruzi-Infected Triatomine Bugs.

Chagas disease is a vector-borne and potentially fatal parasitic disease that is transmitted by the triatomine bug, a nocturnal feeding, flying arthropod, often referred to by its colloquial name, the "kissing bug." Vector-borne transmission is considered the most important means of spreading Chagas disease in endemic and nonendemic areas. Corrugated cardboard boxes may accelerate the spread of these insect vectors to nonendemic areas through their ability to harbor and transport small terrestrial arthropods such as silverfish, termites, and cockroaches. We report the case of a patient living

American trypanosomiasis.

American trypanosomiasis is a parasitic disease caused by the flagellate protozoan Trypanosoma cruzi. Chagas disease is endemic in Latin America, where an estimated 10-14 million people are infected, and an emerging disease in Europe and the USA. Trypanosoma cruzi is transmitted by blood-sucking bugs of the family Reduviidae. Rhodnius prolixus, Panstrongylus megistus, Triatoma infestans, and T. dimidiata are the main vectors in the sylvatic cycle. Non vector-borne transmission includes blood transfusion, congenital and oral transmission, transplantation, and accidental infections. Most cases o

On the possibility of autochthonous Chagas disease in Roraima, Amazon region, Brazil, 2000-2001.

Chagas disease has been almost entirely eradicated from the arid zones in Central and Northeastern Brazil where rare or no autochthonous cases have been reported. However, in the last 10 years the disease has increasingly been registered in the Amazon Region. Aiming to investigate the possibility of the occurrence of autochthonous cycle of Chagas disease in Roraima, triatomine collections, vectorial susceptibility studies (this one to be shown elsewhere), parasitological and serological analyses were conducted in three agricultural settlement areas (Rorainópolis, Passarão Project and Ilha Comm

Epidemiology of Chagas disease in Ecuador. A brief review.

Chagas disease is a complex public health problem that has been underestimated in Ecuador. Here we review the relevant published information, and present unpublished and new data that help to understand the current Chagas disease epidemiological situation and its evolution in the country. Three main characteristics have been identified: (i) persistence of Trypanosoma cruzi transmission in already known foci; (ii) a marked endemicity in some urban areas of Guayaquil; and (iii) the transformation of new Amazon foci into truly endemic areas. The situation in other suspect areas remains uncertain.

Viral metagenome characterization reveals species-specific virome profiles in Triatominae populations from the southern United States.

Kissing bugs (Triatominae) are hematophagous insects and the principal vectors of Trypanosoma cruzi, the causative agent of Chagas disease. While their bacterial microbiomes have received considerable attention, the diversity of viruses associated with these insects remains poorly understood. To address this gap, we investigated the metavirome of five Triatominae species from the southern United States (Triatoma rubida, T. sanguisuga, T. gerstaeckeri, T. indictiva, and Hospesneotomae protracta), sampled in Texas, New Mexico, and Arizona. We sequenced 23 samples, including abdomen, gut and repr
