Mycotoxin contamination remains a major challenge to food and feed safety, agricultural productivity, and postharvest resource utilization. Although numerous mycotoxin-transforming enzymes have been discovered and increasingly improved through protein engineering, only a limited number have progressed from laboratory characterization to practical food or feed applications. This translational gap arises because catalytic efficiency alone does not address matrix interference, processing stress, scalable production, formulation, degradation-product safety, and application-specific regulatory requ

T-2 toxin is one of the most potent and commonly occurring secondary metabolites of type A trichothecene mycotoxins. Although it has been associated with cardiomyocyte necrosis, the underlying mechanisms of T-2 toxin-induced myocardial injury, particularly combined with low selenium conditions, remain unclear. This study investigates the role of MLKL, the terminal effector of the Receptor interacting protein kinase-1 and -3/mixed lineage kinase domain-like protein (RIP1-RIP3-MLKL) necroptotic pathway, in cardiac injury induced by T-2 toxin alone or in combination with low selenium. Histopathol

T-2 toxin, a trichothecene mycotoxin, is a common contaminant in food and animal feed, and is also present in processed cereal products. The most common route of T-2 toxin exposure in humans is through dietary ingestion. The cytotoxic effects of T-2 toxin include modifications to feeding behavior, nervous disorders, cardiovascular alterations, immunosuppression, and hemostatic derangements. However, to date, effects on the central nervous system (CNS) have rarely been reported. In the present study, female Wistar rat were given a single dose of T-2 toxin at 2 mg/kg b.w. and were sacrificed at

Satratoxin G (SG) is a macrocyclic trichothecene mycotoxin produced by Stachybotrys chartarum, the "black mold" suggested to contribute etiologically to illnesses associated with water-damaged buildings. Using an intranasal instillation model in mice, we found that acute SG exposure specifically induced apoptosis of olfactory sensory neurons (OSNs) in the olfactory epithelium. Dose-response analysis revealed that the no-effect and lowest-effect levels at 24 hr postinstillation (PI) were 5 and 25 microg/kg body weight (bw) SG, respectively, with severity increasing with dose. Apoptosis of OSNs

Deoxynivalenol (DON), a mycotoxin produced by fungi of the genus Fusarium, is ubiquitously distributed in feed and poses a substantial threat to human and animal health. Pigs exhibit particular sensitivity to DON, which can induce reproductive impairment. While the ovarian toxicity of DON has been extensively investigated, its effects on the uterus remain largely understudied. We established a DON-exposed piglet model and observed severe oxidative damage in the uterus. Reactive oxygen species (ROS) regulate protein function by catalyzing cysteine modifications in vivo. Deoxynivalenol (DON) is one of the most common mycotoxins distributed in food and feed, which causes severe liver injury in humans and animals. Cold atmospheric plasma (CAP) has received much attention in mycotoxin degradation due to the advantages of easy operation, high efficiency, and low temperature. So far, the majority of studies have focused on the degradation efficiency and mechanism of CAP on DON, while there is still little information available on the hepatotoxicity of DON after CAP treatment. Herein, this study aimed to investigate the effect of CAP on DON-induced hepatotoxicity bo

The objective of this study was to evaluate the efficacy of mycotoxin binders in reducing the adverse effects of co-occurring dietary aflatoxin B1 (AFB1), deoxynivalenol (DON) and ochratoxin A (OTA) on laying hens. Three hundred and sixty 26-week-old Roman laying hens were randomly allocated into four experimental groups with 10 replicates of nine birds each. The four groups received either a basal diet (BD; Control), a BD supplemented with 0.15 mg/kg AFB1 + 1.5 mg/kg DON + 0.12 mg/kg OTA (Toxins), a BD + Toxins with Toxo-HP binder (Toxins + HP), or a BD + Toxins with TOXO XL binder (Toxins +

Microtubule is a well-known structural protein participating in cell division, motility and vesicle traffic. In this study, we found that β2 -tubulin, one of the microtubule components, plays an important role in regulating secondary metabolite deoxynivalenol (DON) biosynthesis in Fusarium graminearum by interacting with isocitrate dehydrogenase subunit 3 (IDH3). We found IDH3 negatively regulate DON biosynthesis by reducing acetyl-CoA accumulation in F. graminearum and DON biosynthesis was stimulated by exogenous acetyl-CoA. In addition, the expression of IDH3 significantly decreased in the c

Dietary deoxynivalenol (DON) impairs the intestinal immune system and digestive functions of broiler chickens. However, little is known whether increasing doses of DON similarly affect the intestinal functions in different segments of the small intestine in chickens and whether a second oral challenge may potentiate those effects. The present objective was to investigate the effect of increasing dietary DON concentrations on the relative expression of genes for tight junction proteins, mucins, toll-like receptors (TLR), and cytokines in duodenum and jejunum, jejunal mucosal permeability, as we

A feeding trial was conducted to evaluate the effects of moderate dietary concentrations of deoxynivalenol (DON) during a 21-day feeding experiment on the performance of broilers. Fifteen 1-day-old broiler chicks were randomly divided into two groups. The control group was fed non-contaminated diet. Another group of broilers was fed a diet naturally contaminated with 5 mg DON/kg diet. Deoxynivalenol had no effect (p > 0.05) on feed consumption, feed conversion, body-weight gain, live body weight or mortality. The absolute and relative weight of the organs (gizzard, pancreas, heart, spleen, col

In an experiment to determine the effect of corn vs. grain sorghum on performance of laying hens, egg production decreased significantly in the grain sorghum-fed hens in each of two trials starting 24 weeks after the trials began. Necropsy of chickens fed both diets revealed buccal ulceration at the ventral aspect of the oral cavity and squamous metaplasia of the esophageal glands and submaxillary salivary glands. Lesions were much more severe in the sorghum-fed birds than in the corn-fed birds. Analysis of the grain sorghum and corn revealed the presence of mycotoxins. Zearalenone and deoxyni

T-2 toxin, a highly toxic trichothecene mycotoxin widely found in food and feed, poses a significant threat to human health as well as livestock and poultry industry. Liver, being a crucial metabolic organ, is particularly susceptible to T-2 toxin induced damage characterized by inflammation and oxidative stress. Despite the role of Sirtuin 5 (SIRT5) in mitigating liver injury has been confirmed, its specific impact on T-2 toxin induced liver injury remains to be elucidated. The objective of this study was to investigate the protective role of SIRT5 against T-2 toxin induced liver injury in mi

Deoxynivalenol (DON) is a globally prevalent mycotoxin that threatens food and feed safety via severe multi-organ toxicity. Previous studies indicate that DON induces cellular energy metabolism dysregulation by triggering oxidative stress and impairing mitochondrial function. During this process, nicotinamide adenine dinucleotide (NAD+), a central coenzyme in cellular energy metabolism, frequently exhibits significantly decreased intracellular levels or even complete depletion. However, the molecular mechanisms underlying the disruption of NAD+ homeostasis by DON exposure, as well as the devel

BACKGROUND: Deoxynivalenol (DON), a mycotoxin produced by Fusarium species, posed significant threats to food safety and human health due to its widespread prevalence and detrimental effects. Upon exposure, the liver, which played a crucial role in detoxifying DON, experienced depleted antioxidant levels and heightened inflammatory responses. Bone marrow mesenchymal stem cell (BMSC)-derived exosomes (BMSC-exos) exhibited therapeutic potential by promoting cellular repair and delivering bioactive substances, such as cerium dioxide nanoparticles (CeO₂ NPs), which are recognized for their ability

The foodborne mycotoxin deoxynivalenol (DON) produced by Fusarium species threats animal and human health through disruption of the intestinal barrier. Targeting the gut microbiota and its products appears as a promising strategy to mitigate DON intestinal toxicity. In this study, we investigated whether the bacterial metabolite butyrate could alleviate epithelial barrier disruption induced by DON. We used a model of cell monolayers derived from porcine jejunum organoids allowing to reproduce the cellular complexity of the intestinal epithelium. Our results show that DON dose-dependently disru

Using alternative ingredients or low-quality grain grades to reduce feeding costs for pig diets can introduce mycotoxins such as deoxynivalenol (DON) into feed, which is known to induce anorexia, inflammation, and oxidative stress. Adding vitamin 25(OH)D3 or vitamins E and C to the feed could increase piglets' immune system to alleviate the effects of DON. This study used 54 pigs (7.8 ± 0.14 kg) in 27 pens (2 pigs/pen) with a vitamin 25(OH)D3 or vitamin E-C supplementation, or their combination, in DON-contaminated (5.1 mg/kg) feed ingredients over 21 days followed by a lipopolysaccharide (LPS

In recent years, the deleterious effects attributed to mycotoxins, in particular on the intestine, faced increased attention and it was shown that deoxynivalenol (DON) causes adverse effects on gut health. In this context, it has been repeatedly reported that DON can alter the intestinal morphology, disrupt the intestinal barrier and reduce nutrient absorption. The underlying mechanism of a compromised intestinal barrier caused by DON in chickens has yet to be illustrated. Although, DON is rapidly absorbed from the upper parts of the small intestine, the effects on the large intestine cannot b

Fusarium head blight (FHB) is a worldwide devastating disease of wheat, barley and other small grain cereals and caused primarily by Fusarium graminearum. Carbendazim (MBC) is one of the fungicides widely used to control FHB in China. In the present study, we have identified the F. graminearum locus FGSG_04220 as the sequence homolog for Saccharomyces cerevisiae ScSWI6, named FgSWI6 hereafter. Deletion of FgSWI6 causes mycelium of F. graminearum become sensitive to MBC in liquid medium. In addition, deletion of FgSWI6 reduces mycelial growth as well as production and development of conidia. F.

Mycotoxins are secondary metabolites produced by microfungi that are capable of causing disease and death in humans and other animals. This work was aimed at investigation of influence of mouldy wheat contaminated by pathogenic fungi producing mycotoxins on metallothionein levels in hepatic tissue of rats. The rats were administrating feed mixtures with different contents of vitamins or naturally mouldy wheat for 28 days. It was found that the wheat contained deoxynivalenol (80 +/- 5 microg per kg of mouldy wheat), zearalenone (56 +/- 3 microg/kg), T2-toxin (20 +/- 2 microg/kg) and aflatoxins
