Optimized Flow Cytometry Incorporating t-SNE Enables Minimal Residual Disease Assessment in a Philadelphia Chromosome-positive Acute Lymphoblastic Leukemia at or Near the Lower Detection Limit.

Unsupervised machine learning shows significant promise for evaluating multicolor flow cytometry, particularly in advancing minimal residual disease (MRD) analysis. We implemented t-distributed stochastic neighbor embedding (t-SNE)-assisted MRD analysis in a challenging case of Philadelphia chromosome-positive acute lymphoblastic leukemia. This approach enabled us to identify MRD on an unbiased basis with a sensitivity comparable to that of genetic analysis while simultaneously reducing the workload. t-SNE is a valuable tool for the detection and verification of MRD, with its effectiveness sig

An unsupervised approach for projection binning to reduce motion artifacts in free-breathing animal models.

BACKGROUND: Dynamic imaging holds great potential in the diagnosis and comprehensive evaluation of different diseases by capturing mechanical and dynamic characteristics of moving organs. Nonetheless, motion artifacts notably impair image quality, hindering accurate and localized analysis-particularly in free-breathing scenarios. In preclinical studies, traditional methods often necessitate artificial breathing control or use invasive techniques that do not permit functional lung assessment under normal physiological conditions, potentially biasing results and restricting longitudinal studies.

Machine Learning-Based Cytokine Endotyping of Thyroid Eye Disease.

PURPOSE: Thyroid eye disease (TED) exhibits profound phenotypic heterogeneity that conventional binary thyrotropin receptor antibody fails to capture. We aimed to deconstruct this heterogeneity by establishing a cytokine-derived molecular taxonomy. METHODS: We profiled 102 plasma cytokines in 101 TED patients using a Luminex assay. Integrative machine learning identified key cytokine signatures. Unsupervised K-means clustering defined four discrete endotypes. A diagnostic nomogram (named the cytokine-derived cluster [CKC] score) was developed and evaluated in an independent external cohort.

Traceability and discrimination of opium poppy shell analogues using HS-GC-IMS combined with machine learning algorithms.

Illegal adulteration has been a critical issue in food safety, emerging as a focal point in forensic science. This situation has led to an increased demand for effective detection and monitoring technologies. Opium poppy shells are a critical source of drugs, and the accurate tracing and identification of their analogues are essential in drug-related cases. The features of volatile compounds in six opium poppy shell analogues (OPSA) were characterized using headspace-gas chromatography-ion mobility spectrometry (HS-GC-IMS) in this study, and an accurate model for origin tracing was established

Machine learning identifies a distinct microbiota signature in immune checkpoint inhibitor colitis compared with inflammatory bowel disease.

BACKGROUND: Immune checkpoint inhibitors (ICIs) have revolutionized oncology by enhancing antitumor immune responses. However, their use is frequently associated with immune-mediated adverse events, including colitis (ICIs-colitis). This condition shares clinical and histological features with inflammatory bowel diseases (IBD), such as Crohn's disease (CD) and ulcerative colitis (UC). Emerging evidence highlights the gut microbiota's role in both ICIs-colitis and IBD pathogenesis. This study aimed to determine whether a distinct microbiota profile, assessed via machine learning, could differen

Multimodal data-driven clustering analysis of heart failure patients identifies two distinct patterns of heart failure with preserved ejection fraction: Results from the PACIFIC-PRESERVED study.

Heart failure with preserved ejection fraction (HFpEF) accounts for over half of heart failure cases but remains poorly defined due to its clinical heterogeneity. In the prospective PACIFIC-Preserved cohort (NCT04189029, n = 155), we applied multimodal deep phenotyping, including high-throughput proteomics, and unsupervised machine learning to identify biologically distinct HFpEF subgroups. Multiview clustering revealed two major HFpEF phenogroups: PEF1, characterized by inflammatory signaling, TNF receptor activation, cardio-skeletal injury, and greater renal dysfunction; and PEF2, marked by

Unsupervised single-domain generalization for tissue classification via progressive domain transformation.

Tissue classification is one of the fundamental tasks in computational pathology, but domain shifts in digital pathology images limit the generalization of classification models. Domain generalization has emerged as a leading solution to address this gap, with related research often using multiple public datasets to demonstrate model generalization ability across different sources. To further explore this, we introduce the GDPH-CRC-HE-MS dataset, consisting of 101 H&E-stained colorectal cancer slides from Guangdong Provincial People's Hospital, scanned by 1 to 6 different scanners. Revisiting Abnormalities of Ventricular Depolarization: Redefining Phenotypes and Associated Outcomes Using Tree-Based Dimensionality Reduction.

BACKGROUND: Abnormal ventricular depolarization, evident as a broad QRS complex on an ECG, is traditionally categorized into left bundle-branch block (LBBB) and right bundle-branch block or nonspecific intraventricular conduction delay. This categorization, although physiologically accurate, may fail to capture the nuances of diseases subtypes. METHODS: We used unsupervised machine learning to identify and characterize novel broad QRS phenogroups. First, we trained a variational autoencoder on 1.1 million ECGs and discovered 51 latent features that showed high disentanglement and ECG reconstr

Unsupervised identification of disease states from high-dimensional physiological and histopathological profiles.

The liver and kidney in mammals play central roles in protecting the organism from xenobiotics and are at high risk of xenobiotic-induced injury. Xenobiotic-induced tissue injury has been extensively studied from both classical histopathological and biochemical perspectives. Here, we introduce a machine-learning approach to analyze toxicological response. Unsupervised characterization of physiological and histological changes in a large toxicogenomic dataset revealed nine discrete toxin-induced disease states, some of which correspond to known pathology, but others were novel. Analysis of dyna

Embryo selection at the cleavage stage using Raman spectroscopy of day 3 culture medium and machine learning: a preliminary study.

BACKGROUND: Blastocyst transfer has been associated with shorter leukocyte telomere length in ART-conceived children, suggesting that extended embryo culture may accelerate aging in offspring. Selecting Day 3 embryos with high developmental potential for transfer could address this issue. The aim of this study is to investigate whether machine learning combined with Raman spectroscopy of spent Day 3 culture medium can serve as a potential method for predicting extended embryo culture outcomes, thereby enabling embryo selection at the cleavage stage. Methods: This prospective study analyzed 172

Machine Diagnostics and Machine Phenotyping of Migraine: A HUNT Study.

BACKGROUND AND OBJECTIVES: In the absence of biomarkers, the true biological footprint of migraine remains incompletely understood. It could perhaps be best characterized using machine learning models of multimodal data. The aim of this study was to (1) develop diagnostic models of migraine using multimodal data and (2) identify data-driven migraine phenotypes. METHODS: This was a cross-sectional machine learning analysis of demographics, self-reported clinical and headache data, and genome-wide genotype data from the Trøndelag Health Study (data collected 1995-1997 and 2006-2008). A two-stage learning framework for piecewise affine system identification.

This paper proposes a fully data-driven and end-to-end adaptive framework for the complete black-box identification of piecewise affine ARX (PWARX) systems, in which model structure discovery, submodel parameter estimation, recursive parameter fine-tuning, and explicit polyhedral partition identification are integrated into a unified two-stage learning architecture. In Stage 1, an unsupervised process determines the number and order of submodels, computes consistent initial submodel parameter estimates, and constructs a compact labeled training set. In Stage 2, a self-tuning semi-supervised su
